최근 몇 년 동안 3차원(3D) 스페로이드 배양 시스템은 암 연구, 약물 발견 및 독성학의 다양한 영역을 연구하는 중요한 도구가 되었습니다. 이러한 배양은 2차원(2D) 세포 배양 단층과 복잡한 기관 사이의 간극을 메우기 때문에 큰 관심을 불러일으킨다1.

동물 모델은 인간의 생물학과 질병을 연구하기 위해 오랫동안 사용되어 왔습니다3. 이와 관련하여 인간과 동물 사이에는 본질적인 차이가 있기 때문에 이러한 모델은 외계 연구에 완전히 적합하지 않습니다. 3D 배양 스페로이드 및 오가노이드는 생체 내에서 발생하는 다양한 세포 유형 간의 조직과 유사한 구조, 상호 작용 및 누화를 연구하는 유망한 도구이며 동물 모델을 줄이거나 대체하는 데 기여할 수 있습니다. 그들은 간 질환의 발병 기전과 약물 스크리닝 플랫폼을 연구하는 데 특히 관심이 있습니다4.
3D 스페로이드 배양은 2D 배양을 위한 종양 세포 단층을 준비하는 데 필요한 트립신 처리 또는 콜라게나제 처리의 필요성을 줄여 세포와 환경 간의 불연속성을 제거할 수 있기 때문에 암 연구에 특히 중요합니다. 종양 스페로이드는 정상 세포와 악성 세포가 주변 환경으로부터 어떻게 신호를 받고 반응하는지에 대한 연구를 가능하게 하며5 종양 생물학 연구의 중요한 부분이다.
단층에 비해 다양한 세포 유형으로 구성된 3D 배양은 구조적 및 기능적 특성에서 종양 조직과 유사하므로 종양 세포의 전이 및 침윤을 연구하는 데 적합합니다. 이것이 바로 이러한 스페로이드 모델이 암 연구를 가속화하는 데 기여하는 이유입니다6.
스페로이드는 또한 조직 및 장기 생물학, 특히 인간에서 연구하기가 매우 어렵기 때문에 인간 오가노이드를 만드는 기술을 개발하는 데 도움이 됩니다. 줄기세포 배양이 진행됨에 따라 줄기세포와 조직 전구체로 구성된 오가노이드와 같은 3D 배양을 개발할 수 있을 뿐만 아니라 실제 장기와 같은 기능적 특성을 가진 장기의 다양한 유형의 성숙(조직) 세포를 개발할 수 있어 장기 발달, 질병을 모델링하는 데 사용할 수 있을 뿐만 아니라 재생 의학7에서도 유용하다고 간주될 수 있다.
1차 인간 간세포는 일반적으로 인간 간세포, 간 기능 및 약물 유발 독성의 시험관 내 생물학을 연구하는 데 사용됩니다. 인간 간세포의 배양은 두 가지 주요 단점을 가지고 있는데, 첫째, 인간 간세포와 같은 일차 조직의 가용성이 제한적이고, 둘째, 간세포가 2D 배양에서 빠르게 역분화되어 특정 간세포 기능을 상실하는 경향이 있다8. 3D 간 배양은 이와 관련하여 우수하며 최근에는 분화된 인간 배아 줄기 세포(hESC) 또는 유도 만능 줄기 세포(hiPSC)로 만들어졌습니다9. 생체공학적 간 3D 스페로이드는 간 질환 치료를 위한 약물 발견뿐만 아니라 간의 발달, 독성, 유전 및 감염성 질환을 연구하는 데 특히 중요합니다10. 마지막으로, 급성 간 질환의 치사율이 거의 80%에 달한다는 사실을 알기 때문에 임상적으로 사용될 수 있는 가능성도 있으며, 생체 인공 간 및/또는 간 스페로이드는 적절한 기증자를 찾을 때까지 부분적인 간 기능을 제공함으로써 이러한 환자들을 잠재적으로 구제할 수 있다11.
우리는 혈액 유래 만능 줄기 세포(BD-PSC)를 사용하여 4000 내지 1 x 106 세포를 포함하는 다양한 크기의 스페로이드를 제조하고 광학 현미경 및 면역형광을 통해 분석하는 인간 간 스페로이드 생성을 위한 프로토콜을 확립했습니다. 또한 해독 과정을 통해 세포 및 약물 대사에 중요한 역할을 하는 시토크롬 P450 계열에 속하는 시토크롬 P450 3A4(CYP3A4) 및 2E1(CYP2E1) 효소의 발현을 평가하여 간세포 특이적 기능의 능력을 테스트했습니다12.
