The central focus of the present experiment is the isolation, expansion, and adipogenic induction of APC from PVAT depots. Here we present a protocol for the isolation of APC based on the identification of cells expressing the surface markers CD34 and PDGFR&#945;. Perivascular Adipose Tissue (PVAT), due to its close proximity to blood vessels, is a major paracrine signaling component in vasculature function1. Expansion of this adipose tissue is dependent on the phenotype of the Adipocyte Progenitor Cells (APC) present2,3. Isolation of cells from adipose tissues is difficult as primary adipocytes are fragile, buoyant, and range in size. Certain isolation techniques can also alter cell phenotype and morphology by increasing inflammatory protein synthesis and reducing adipogenic gene expression4, emphasizing the importance of a protocol that maintains the integrity of the cells.
Culture of primary cells and specific preadipocyte subpopulations gives a reductionist approach to in vivo growth and maintains equivalent cellular genetic makeup5, although working time with these cells is limited due to deterioration with aging, or senescence6. Preadipocytes from various adipose depots, including subcutaneous and omental depots, also demonstrate differences in proliferation7, which emphasizes the importance of collecting cells from specific anatomical sites. Precursor cells from non-PVAT white adipose depots have been characterized in previous studies7,8,9, but less is known about PVAT APC phenotypes.
The techniques described here allow for the analysis of specific and defined APC populations with minimal impact on their morphology, viability, and potential to proliferate and differentiate. Magnetic-activated Cell Sorting (MCS) is amenable to downstream applications, such as culture, as the beads dissolve without altering the cell. MCS is also economical, and once the antibody concentrations have been standardized, the need for flow cytometry assays is minimal. In vitro studies with PVAT precursors can also give a glimpse of the potential that these primary cells may have.

Expansion of Perivascular Adipose Tissue (PVAT), a major regulator of vascular function through paracrine signaling, is directly related to the development of hypertension during obesity. The extent of hypertrophy and hyperplasia depends on depot location, sex, and the type of Adipocyte Progenitor Cell (APC) phenotypes present. Techniques used for APC and preadipocytes isolation in the last 10 years have drastically improved the accuracy at which individual cells can be identified based on specific cell surface markers. However, isolation of APC and adipocytes can be a challenge due to the fragility of the cell, especially if the intact cell must be retained for cell culture applications.
Magnetic-activated Cell Sorting (MCS) provides a method of isolating greater number of viable APC per weight unit of adipose tissue. APC harvested by MCS can be used for in vitro protocols to expand preadipocytes and differentiate them into adipocytes through use of growth factor cocktails allowing for analysis of the prolific and adipogenic potential retained by the cells. This experiment focused on the aortic and mesenteric PVAT depots, which play key roles in the development of cardiovascular disease during expansion. These protocols describe methods to isolate, expand, and differentiate a defined population of APC. This MCS protocol allows isolation to be used in any experiment where cell sorting is needed with minimal equipment or training. These techniques can aid further experiments to determine the functionality of specific cell populations based on the presence of cell surface markers.

The overall goals of these isolation and differentiation protocols are to obtain adipocyte progenitor cells from perivascular adipose tissues, and to induce their differentiation into mature adipocytes. This method allows the analysis of specific and defined adipocyte progenitor cell populations from perivascular adipose tissue, with minimal impact on their morphology, viability, and potential to proliferate and differentiate. The main advantage of this technique is that it allows an increased isolation of antigen-presenting cells per gram of adipose tissue, compared to fluorescence-activated cell sorting.After confirming a lack of response to toe pinch, make a vertical midline incision along the sternum to the perineal area of the rat, to access the abdominal cavity. Expose the superior mesenteric artery, the small mesenteric resistance vessels, and the thoracic aorta, and sever all of the connections to the mesentery and aorta. Collect the gonadal adipose tissue and transfer the isolated fat pads into a new container of Krebs-Ringer bicarbonate buffer, or KRBB solution, supplemented with 10 millimolar heaps.Transfer the vessels to a petri dish containing KRBB solution, and place the dish under a dissecting microscope. Remove perivascular adipose tissue from the small mesenteric resistance vessels and aorta. In a biosafety hood, transfer about 50 milligrams of tissue into a 1.7 milliliter tube containing 1 milliliter of collagenase type I solution, and mince the tissue into 1 to 3 millimeter pieces.It is important to adequately mince the adipose tissue to allow the collagenase to thoroughly infiltrate the connective tissue. Incubate the fragments at 37 degrees Celsius with shaking for 1 hour. Then sequentially filter the digested material through individual 100-and 40-micron cell strainers into a 50-milliliter tube.Collect the resulting filtrate by centrifugation, and resuspend the pellet in 1 milliliter of erythrocyte lysis buffer. Transfer the cell suspension into a new 1.7 milliliter Microfuge tube for a 5-minute incubation at room temperature, protected from light. Then collect the stromal vascular fraction cells with another centrifugation and resuspend the pellet in stromal vascular fraction basal medium for counting.To isolate the adipocyte progenitor cells by magnetic-activated cell sorting, collect the cells by centrifugation, and resuspend the pellet in magnetic-activated cell sorting blocking buffer at a 1 times 10 to the 6th cells per milliliter concentration for a 20-minute incubation at 4 degrees Celsius. It is essential that all steps of the isolation are done at 4 degrees Celsius, and not over ice. Next, incubate the cells with 5 microliters of FITC-conjugated mouse anti-CD34 for 30 minutes.Collect the cells by centrifugation. Then incubate the cells with 4 microliters of anti-FITC MicroBeads in 96 microliters of magnetic-activated cell sorting buffer for 5 minutes in the dark. While the cells are incubating, place a multi-sort column into the magnetic separator with a 5-milliliter waste tube under the column.Rinse the column with 500 microliters of magnetic-activated cell sorting buffer, and discard the effluent and waste tube. Then place a new tube under the column, and load the cells onto the top of the column, collecting the unlabeled CD34 negative cells in the new collection tube. When all of the cells have passed through the top of the column, wash the column 3 times with 500 microliters of degassed magnetic-activated cell sorting buffer, continuing to pool the eluate in the collection tube.After the last wash, transfer the column into a new collection tube, and use the column plunger to flush 1 milliliter of magnetic-activated cell sorting buffer through the column into the tube, to collect the CD34 positive cells. Collect the adipocyte progenitor cells by centrifugation. Then incubate the CD34 positive fraction in 10 microliters of rabbit anti-platelet-derived growth factor receptor alpha for 30 minutes.At the end of the incubation, centrifuge the cells again, and incubate the pellet in 4 microliters of anti-rabbit IgG MicroBeads in 96 microliters of magnetic-activated cell sorting buffer for magnetic bead isolation of the platelet-derived growth factor receptor alpha positive cells, as just demonstrated. To culture the CD34 positive platelet-derived growth factor receptor alpha positive adipocyte progenitor cells, seed the cells in individual 6-well tissue culture plates in basal medium, and place the plates in a 37 degree Celsius and 5%carbon dioxide cell culture incubator. After 3 serial passages, seed the cells into black 96-well tissue culture plates at 1 times 10 to the 2 cells per well, and evaluate their proliferation at 8, 24, 48, and 96 hours at 5 times 10 to the 4th cells per well, in 24-well assay plates, according to standard proliferation assay protocols.To induce adipogenesis, when the adipocyte progenitor cell culture reaches confluency, feed the cells with adipocyte progenitor basal medium for 48 hours, then treat the cultures with bone morphogenic protein 4 for another 48 hours. On the third day, replace the medium with adipocyte progenitor cell induction medium, without IBMX and examethasone for 14 days. The degree of adipogenesis in the cultures can then be assessed at 1 times 10 to the 4th cells per well in 48-well tissue culture plates, according to standard adipogenesis assay protocols.Although both methods demonstrate a similar cell population distribution and viability, magnetic-activated cell sorting isolation produces a greater number of cells for culture, compared to fluorescence-activated cell sorting. In this representative experiment, the in vitro proliferation of magnetic-activated cell sorted stromal vascular fraction and adipocyte progenitor cells, isolated from the thoracic aorta, small mesenteric resistance vessels, and gonadal adipose tissue of male rats, was evaluated at 8, 24, 48, and 96 hours after plating, using a quantitative DNA assay. No site differences in stromal vascular fraction expansion rates were observed at any time point, except for the adipocyte progenitor cells from the thoracic aorta, which demonstrated less proliferation by 96 hours, compared to stromal vascular cells from the same site.Stimulation of confluent adipocyte progenitor cells with bone morphogenic protein 4 for 48 hours induces adipocyte differentiation, evidenced by a greater lipid accumulation in droplets, as evaluated by both fluorescent lipid uptake assay, and Oil Red O staining. Once mastered, this technique can be completed in 8 hours, if it is performed correctly. While attempting this procedure, it is important to remember to maintain 4 degree Celsius temperatures during the isolation steps.After watching this video you should have a good understanding of how to isolate and differentiate adipocyte progenitor cells.

Isolation and Expansion of Mesenchymal Stem/Stromal Cells Derived from Human Placenta Tissue

Mesenchymal stem/stromal cells (MSC) are promising candidates for use in cell-based therapies. In most cases, therapeutic response appears to be cell-dose dependent. Human term placenta is rich in MSC and is a physically large tissue that is generally discarded following birth. Placenta is an ideal starting material for the large-scale manufacture of multiple cell doses of allogeneic MSC. The placenta is a fetomaternal organ from which either fetal or maternal tissue can be isolated. This article describes the placental anatomy and procedure to dissect apart the decidua (maternal), chorionic villi (fetal), and chorionic plate (fetal) tissue. The protocol then outlines how to isolate MSC from each dissected tissue region, and provides representative analysis of expanded MSC derived from the respective tissue types. These methods are intended for pre-clinical MSC isolation, but have also been adapted for clinical manufacture of placental MSC for human therapeutic use.

Isolation of Perivascular Multipotent Precursor Cell Populations from Human Cardiac Tissue

Multipotent mesenchymal stem/stromal cells (MSC) were conventionally isolated, through their plastic adherence, from primary tissue digests whilst their anatomical tissue location remained unclear. The recent discovery of defined perivascular and MSC cell marker expression by perivascular cells in multiple tissues by our group and other researchers has provided an opportunity to prospectively isolate and purify specific homogenous subpopulations of multipotent perivascular precursor cells. We have previously demonstrated the use of fluorescent activated cell sorting (FACS) to purify microvascular CD146+CD34- pericytes and vascular CD34+CD146- adventitial cells from human skeletal muscle. Herein we describe a method to simultaneously isolate these two perivascular cell subsets from human myocardium by FACS, based on the expression of a defined set of cell surface markers for positive and negative selections. This method thus makes available two specific subpopulations of multipotent cardiac MSC-like precursor cells for use in basic research and/or therapeutic investigations.

Human cardiac tissue harbours multipotent perivascular precursor cell populations that may be suitable for myocardial regeneration. The technique described here allows for the simultaneous isolation and purification of two multipotent stromal cell populations associated with native blood vessels, i.e. CD146+CD34- pericytes and CD34+CD146- adventitial cells, from the human myocardium.

Induction of Acute Skeletal Muscle Regeneration by Cardiotoxin Injection

Skeletal muscle regeneration is a physiological process that occurs in adult skeletal muscles in response to injury or disease. Acute injury-induced skeletal muscle regeneration is a widely used, powerful model system to study the events involved in muscle regeneration as well as the mechanisms and different players. Indeed, a detailed knowledge of this process is essential for a better understanding of the pathological conditions that lead to skeletal muscle degeneration, and it aids in identifying new targeted therapeutic strategies. The present work describes a detailed and reproducible protocol to induce acute skeletal muscle regeneration in mice through a single intramuscular injection of cardiotoxin (CTX). CTX belongs to the family of snake venom toxins and causes myolysis of myofibers, which eventually triggers the regeneration events. The dynamics of skeletal muscle regeneration is evaluated by histological analysis of muscle sections. The protocol also illustrates the experimental procedures for dissecting, freezing, and cutting the Tibialis Anterior muscle, as well as the routine Hematoxylin &#38; Eosin staining that is widely used for subsequent morphological and morphometric analysis.

Chondrogenic Differentiation Induction of Adipose-derived Stem Cells by Centrifugal Gravity

Impaired cartilage cannot heal naturally. Currently, the most advanced therapy for defects in cartilage is the transplantation of chondrocytes differentiated from stem cells using cytokines. Unfortunately, cytokine-induced chondrogenic differentiation is costly, time-consuming, and associated with a high risk of contamination during in vitro differentiation. However, biomechanical stimuli also serve as crucial regulatory factors for chondrogenesis. For example, mechanical stress can induce chondrogenic differentiation of stem cells, suggesting a potential therapeutic approach for the repair of impaired cartilage. In this study, we demonstrated that centrifugal gravity (CG, 2,400 &#215; g), a mechanical stress easily applied by centrifugation, induced the upregulation of sex determining region Y (SRY)-box 9 (SOX9) in adipose-derived stem cells (ASCs), causing them to express chondrogenic phenotypes. The centrifuged ASCs expressed higher levels of chondrogenic differentiation markers, such as aggrecan (ACAN), collagen type 2 alpha 1 (COL2A1), and collagen type 1 (COL1), but lower levels of collagen type 10 (COL10), a marker of hypertrophic chondrocytes. In addition, chondrogenic aggregate formation, a prerequisite for chondrogenesis, was observed in centrifuged ASCs.

Identification Of Erythromyeloid Progenitors And Their Progeny In The Mouse Embryo By Flow Cytometry

Macrophages are professional phagocytes from the innate arm of the immune system. In steady-state, sessile macrophages are found in adult tissues where they act as front line sentinels of infection and tissue damage. While other immune cells are continuously renewed from hematopoietic stem and progenitor cells (HSPC) located in the bone marrow, a lineage of macrophages, known as resident macrophages, have been shown to be self-maintained in tissues without input from bone marrow HSPCs. This lineage is exemplified by microglia in the brain, Kupffer cells in the liver and Langerhans cells in the epidermis among others. The intestinal and colon lamina propria are the only adult tissues devoid of HSPC-independent resident macrophages. Recent investigations have identified that resident macrophages originate from the extra-embryonic yolk sac hematopoiesis from progenitor(s) distinct from fetal hematopoietic stem cells (HSC). Among yolk sac definitive hematopoiesis, erythromyeloid progenitors (EMP) give rise both to erythroid and myeloid cells, in particular resident macrophages. EMP are only generated within the yolk sac between E8.5 and E10.5 days of development and they migrate to the fetal liver as early as circulation is connected, where they expand and differentiate until at least E16.5. Their progeny includes erythrocytes, macrophages, neutrophils and mast cells but only EMP-derived macrophages persist until adulthood in tissues. The transient nature of EMP emergence and the temporal overlap with HSC generation renders the analysis of these progenitors difficult. We have established a tamoxifen-inducible fate mapping protocol based on expression of the macrophage cytokine receptor Csf1r promoter to characterize EMP and EMP-derived cells in vivo by flow cytometry.

Generation of Scaffold-free, Three-dimensional Insulin Expressing Pancreatoids from Mouse Pancreatic Progenitors In Vitro

The pancreas is a complex organ composed of many different cell types that work together to regulate blood glucose homeostasis and digestion. These cell types include enzyme-secreting acinar cells, an arborized ductal system responsible for the transportation of enzymes to the gut, and hormone-producing endocrine cells. 
Endocrine beta-cells are the sole cell type in the body that produce insulin to lower blood glucose levels. Diabetes, a disease characterized by a loss or the dysfunction of beta-cells, is reaching epidemic proportions. Thus, it is essential to establish protocols to investigate beta-cell development that can be used for screening purposes to derive the drug and cell-based therapeutics. While the experimental investigation of mouse development is essential, in vivo studies are laborious and time-consuming. Cultured cells provide a more convenient platform for screening; however, they are unable to maintain the cellular diversity, architectural organization, and cellular interactions found in vivo. Thus, it is essential to develop new tools to investigate pancreatic organogenesis and physiology.
Pancreatic epithelial cells develop in the close association with mesenchyme from the onset of organogenesis as cells organize and differentiate into the complex, physiologically competent adult organ. The pancreatic mesenchyme provides important signals for the endocrine development, many of which are not well understood yet, thus difficult to recapitulate during the in vitro culture. Here, we describe a protocol to culture three-dimensional, cellular complex mouse organoids that retain mesenchyme, termed pancreatoids. The e10.5 murine pancreatic bud is dissected, dissociated, and cultured in a scaffold-free environment. These floating cells self-assemble with mesenchyme enveloping the developing pancreatoid and a robust number of endocrine beta-cells developing along with the acinar and the duct cells. This system can be used to study the cell fate determination, structural organization, and morphogenesis, cell-cell interactions during organogenesis, or for the drug, small molecule, or genetic screening.

Efficient Generation of Pancreas/Duodenum Homeobox Protein 1+ Posterior Foregut/Pancreatic Progenitors from hPSCs in Adhesion Cultures

Human pluripotent stem cell (hPSC)-derived pancreatic cells are a promising cell source for regenerative medicine and a platform to study human developmental processes. Stepwise directed differentiation that recapitulates developmental processes is one of the major ways to generate pancreatic cells including pancreas/duodenum homeobox protein 1+ (PDX1+) pancreatic progenitor cells. Conventional protocols initiate the differentiation with small colonies shortly after the passage. However, in the state of colonies or aggregates, cells are prone to heterogeneities, which might hamper the differentiation to PDX1+ cells. Here, we present a detailed protocol to differentiate hPSCs into PDX1+ cells. The protocol consists of four steps and initiates the differentiation by seeding dissociated single cells. The induction of SOX17+ definitive endoderm cells was followed by the expression of two primitive gut tube markers, HNF1&#946; and HNF4&#945;, and eventual differentiation into PDX1+ cells. The present protocol provides easy handling and may improve and stabilize the differentiation efficiency of some hPSC lines that were previously found to differentiate inefficiently into endodermal lineages or PDX1+ cells.

Isolation, Culture, and Adipogenic Induction of Neural Crest Original Adipose-Derived Stem Cells from Periaortic Adipose Tissue

An excessive amount of adipose tissue surrounding the blood vessels (perivascular adipose tissue, also known as PVAT) is associated with a high risk of cardiovascular disease. ADSCs derived from different adipose tissues show distinct features, and those from the PVAT have not been well characterized. In a recent study, we reported that some ADSCs in the periaortic arch adipose tissue (PAAT) descend from the neural crest cells (NCCs), a transient population of migratory cells originating from the ectoderm.
In this paper, we describe a protocol for isolating red fluorescent protein (RFP)-labeled NCCs from the PAAT of Wnt-1 Cre+/-;Rosa26RFP/+ mice and inducing their adipogenic differentiation in vitro. Briefly, the stromal vascular fraction (SVF) is enzymatically dissociated from the PAAT, and the RFP+ neural crest derived ADSCs (NCADSCs) are isolated by fluorescence activated cell sorting (FACS). The NCADSCs differentiate into both brown and white adipocytes, can be cryopreserved, and retain their adipogenic potential for ~3&#8211;5 passages. Our protocol can generate abundant ADSCs from the PVAT for modeling PVAT adipogenesis or lipogenesis in vitro. Thus, these NCADSCs can provide a valuable system for studying the molecular switches involved in PVAT differentiation.

We present a protocol for the isolation, culture, and adipogenic induction of neural crest derived adipose-derived stem cells (NCADSCs) from the periaortic adipose tissue of Wnt-1 Cre+/-;Rosa26RFP/+ mice. Generation and Expansion of Human Cardiomyocytes from Patient Peripheral Blood Mononuclear Cells

Generating patient-specific cardiomyocytes from a single blood draw has attracted tremendous interest in precision medicine on cardiovascular disease. Cardiac differentiation from human induced pluripotent stem cells (iPSCs) is modulated by defined signaling pathways that are essential for embryonic heart development. Numerous cardiac differentiation methods on 2-D and 3-D platforms have been developed with various efficiencies and cardiomyocyte yield. This has puzzled investigators outside the field as the variety of these methods can be difficult to follow. Here we present a comprehensive protocol that elaborates robust generation and expansion of patient-specific cardiomyocytes from peripheral blood mononuclear cells (PBMCs). We first describe a high-efficiency iPSC reprogramming protocol from a patient&#39;s blood sample using non-integration Sendai virus vectors. We then detail a small molecule-mediated monolayer differentiation method that can robustly produce beating cardiomyocytes from most human iPSC lines. In addition, a scalable cardiomyocyte expansion protocol is introduced using a small molecule (CHIR99021) that could rapidly expand patient-derived cardiomyocytes for industrial- and clinical-grade applications. At the end, detailed protocols for molecular identification and electrophysiological characterization of these iPSC-CMs are depicted. We expect this protocol to be pragmatic for beginners with limited knowledge on cardiovascular development and stem cell biology.

Development of Organoids from Mouse Pituitary as In Vitro Model to Explore Pituitary Stem Cell Biology

The pituitary is the master endocrine gland regulating key physiological processes, including body growth, metabolism, sexual maturation, reproduction, and stress response. More than a decade ago, stem cells were identified in the pituitary gland. However, despite the application of transgenic in vivo approaches, their phenotype, biology, and role remain unclear. To tackle this enigma, a new and innovative organoid in vitro model is developed to deeply unravel pituitary stem cell biology. Organoids represent 3D cell structures that, under defined culture conditions, self-develop from a tissue's (epithelial) stem cells and recapitulate multiple hallmarks of those stem cells and their tissue. It is shown here that mouse pituitary-derived organoids develop from the gland's stem cells and faithfully recapitulate their in vivo phenotypic and functional characteristics. Among others, they reproduce the activation state of the stem cells as in vivo occurring in response to transgenically inflicted local damage. The organoids are long-term expandable while robustly retaining their stemness phenotype. The new research model is highly valuable to decipher the stem cells' phenotype and behavior during key conditions of pituitary remodeling, ranging from neonatal maturation to aging-associated fading, and from healthy to diseased glands. Here, a detailed protocol is presented to establish mouse pituitary-derived organoids, which provide a powerful tool to dive into the yet enigmatic world of pituitary stem cells.
