Enfermedad humana cultivos neuronales específicas son esenciales para la generación de modelos in vitro para enfermedades neurológicas humanas. Sin embargo, la falta de acceso a los cultivos primarios neuronales adultas humanas plantea desafíos únicos. La evolución reciente de las células madre pluripotentes inducidas (IPSC) ofrece un enfoque alternativo para derivar culturas neuronales de fibroblastos de la piel a través de paciente específico IPSC, pero este proceso es laborioso, requiere conocimientos especiales y grandes cantidades de recursos, y puede tomar varios meses. Esto impide la amplia aplicación de esta tecnología para el estudio de enfermedades neurológicas. Para superar algunos de estos problemas, hemos desarrollado un método para obtener células madre neurales directamente de la sangre periférica humano adulto, sin pasar por el proceso de derivación de IPSC. Las células progenitoras hematopoyéticas enriquecidas a partir de sangre periférica humana adulta se cultivaron in vitro y se transfectaron con los vectores de virus Sendai que contienen factores de transcripción Sox2, Oct3/4, Klf4 y c-Myc. Los resultados de transfección en cambios morfológicos en las células que se seleccionan aún más mediante el uso de medio progenitora neural humano que contiene factor de crecimiento de fibroblastos básico (bFGF) y factor de crecimiento endotelial vascular (VEGF). Las células resultantes se caracterizan por la expresión de marcadores de células madre neuronales, tales como nestina y SOX2. Estas células madre neurales podrían diferenciarse aún más a las neuronas, oligodendrocitos astroglia y en medio de diferenciación especificado. Usando muestras de sangre periférica humana de fácil acceso, este método podría utilizarse para obtener células madre neuronales para una mayor diferenciación de las células neuronales para el modelado in vitro de los trastornos neurológicos y puede avanzar en estudios relacionados con la patogénesis y el tratamiento de esas enfermedades.
