Un parámetro importante que determina el éxito de un procedimiento de injerto óseo es la vascularización de la zona que rodea el injerto. La hipótesis de que la implantación de un autoinjerto de hueso induciría una mayor regeneración ósea mediante la formación de vasos sanguíneos abundantes. Para investigar el efecto del injerto sobre la neovascularización en el sitio del defecto, hemos desarrollado un enfoque micro-tomografía computarizada (μCT) para caracterizar los vasos sanguíneos de nueva formación, lo que implica la perfusión sistémica del animal con un agente de contraste de polimerización. Este método permite el análisis detallado vascular de un órgano en su totalidad. Además, la perfusión sanguínea se evaluó a través de imágenes de fluorescencia (FLI) de un agente fluorescente transmitida por la sangre. La formación de hueso se cuantificó por FLI utilizando una sonda de hidroxiapatita de orientación y análisis μCT. Reclutamiento de células madre se siguió por imágenes de bioluminiscencia (BLI) de ratones transgénicos que expresan la luciferasa bajo el control del promotor de la osteocalcina.Aquí describimos y demostramos preparación del aloinjerto, la cirugía defecto de la bóveda craneal, los protocolos de exploración μCT para el estudio y análisis de la neovascularización la formación de hueso (incluyendo la perfusión in vivo de agente de contraste), y el protocolo para el análisis de datos. El análisis de alta resolución 3D de la vasculatura demostró significativamente mayor angiogénesis en animales con autoinjertos implantados, especialmente con respecto a la formación arteriola. En consecuencia, la perfusión sanguínea fue significativamente mayor en el grupo de autoinjerto por el 7º día después de la cirugía. Se observó mineralización ósea superior y la formación de hueso mayor medido en animales que recibieron autoinjertos. Implantación autoinjerto residente inducida reclutamiento de células madre a la sutura ósea injerto-huésped, donde las células diferenciadas en células formadoras de hueso entre el 7 y el 10 º día postoperatorio. Este hallazgo significa que la formación de hueso mejorada puede ser atribuida ala alimentación vascular aumentada que caracteriza a la implantación autoinjerto. Los métodos presentados pueden servir como una herramienta óptima para estudiar la regeneración ósea en términos de formación de hueso bien delimitada y neovascularización.

La neovascularización se evaluó mediante análisis volumétrico μCT y FLI utilizando un agente de transmisión sanguínea fluorescente para cuantificar la perfusión sanguínea.
