骨移植手順の成功を決定する主要なパラメータは、移植片の周囲の血管新生です。我々は、骨自家移植片の移植が豊富な血管形成によってより大きな骨再生を誘導するという仮説を立てました。欠損部位での血管新生に対する移植片の効果を調べるために、我々は、新たに重合し、造影剤を用いた動物の全身潅流を伴う血管形成特徴付けるマイクロコンピュータ断層撮影（μCT）アプローチを開発しました。この方法は、その全体の臓器の詳細な血管解析を可能にします。加えて、血液灌流は、血液由来の蛍光剤の蛍光イメージング（FLI）を用いて評価しました。骨形成は、ヒドロキシアパタイト標的化プローブおよびμCT分析を用いてFLIにより定量しました。幹細胞の動員は、オステオカルシンプロモーターの制御下でルシフェラーゼを発現するトランスジェニックマウスの生物発光イメージング（BLI）によりモニターしました。ここでは、説明し、同種移植片の準備、頭蓋冠の欠陥手術、血管新生の研究と骨（造影剤の in vivo灌流を含む）の形成分析、およびデータ分析のためのプロトコルのμCTスキャンプロトコルを示しています。 血管系の3次元高分解能分析は、特に細動脈の形成に関して、移植された自家移植片を有する動物において有意に高い血管形成を示しました。したがって、血液灌流は、手術後7 日目により自家群で有意に高かったです。私たちは優れた骨の石灰化を観察し、自家移植を受けた動物においてより大きな骨形成を測定しました。自家移植は、細胞が7 番目と10 番目の術後日の骨形成細胞に分化し、移植片-宿主骨の縫合糸に常駐幹細胞の動員を誘導しました。この知見は、強化された骨形成は、に起因し得ることを意味自家移植を特徴付ける増強血管給。図示の方法はしっかり有界骨形成及び新血管形成の点で骨再生を研究するための最適なツールとして機能することができます。
