Esta investigación recibió la aprobación del Comité de Ética de Experimentos con Animales de la Universidad Médica de Kunming (kmmu20220945). Todos los experimentos se adhirieron a las pautas establecidas para el cuidado de los animales. Se recolectaron células endoteliales linfáticas leptomeníngeas (LLC) de ratones machos C57Bl/6J de 6-8 semanas de edad y con un peso de entre 20-25 g. Estos ratones fueron adquiridos de la Universidad Médica de Kunming en Kunming, China. Todo el procedimiento experimental se llevó a cabo bajo estrictas condiciones estériles. Todos los pasos de centrifugación se realizan a temperatura ambiente, a menos que se especifique lo contrario.
1. Preparación de reactivos e instrumentos
NOTA: Todos los pasos que involucren soluciones deben llevarse a cabo dentro de un gabinete de riesgo biológico de clase II.
<ol><li>Prepare el tampón de lavado mezclando solución salina tamponada con fosfato (PBS) con calcio y magnesio, suero fetal bovino (FBS) al 10% y penicilina-estreptomicina (P/S) al 1% (ver Tabla de materiales). Mantener este tampón frío a 4 °C. Es fundamental prepararlo fresco el día de su uso y desgasificar el tampón.</li>
	<li>Prepare las enzimas digestivas combinando 10 ml de PBS (sin calcio ni magnesio) con 2 ml de tripsina al 0,25%, 1 ml de 20 mg/ml de papaína y 200 μl de colagenasa I (ver Tabla de materiales). NOTA: Asegure el uso de alícuotas de volúmenes apropiados para evitar ciclos repetidos de congelación y descongelación. Almacenar estas alícuotas a -20 °C.</li>
	<li>Prepare el medio de cultivo utilizando el kit de medio de crecimiento de células endoteliales disponible en el mercado, que incluye VEGF-C (100 ng/mL) y 1% de P/S (consulte la Tabla de materiales).</li>
	<li>Prepare el tampón de detención complementando el medio de Eagle modificado de Dulbecco (DMEM) con un 10% de FBS para detener el proceso de digestión.</li>
	<li>Prepare instrumentos quirúrgicos estériles: estos instrumentos deben incluir tijeras, pinzas de punta dentada y pinzas de punta fina.</li>
</ol>2. Recubrimiento del matraz
<ol><li>Recubra el matraz T25 con una solución de fibronectina de 100 μg/ml (véase la tabla de materiales) e incube durante la noche a 37 °C. Antes de usar, retire la solución de recubrimiento con una pipeta.</li>
	<li>Lave el matraz tres veces con PBS, posteriormente aspire el PBS y deje que el matraz T25 se seque al aire.</li>
</ol>3. Disociación de leptomeninges
NOTA: Utilice siempre tampones y soluciones preenfriados a 4 °C.
<ol><li>Anestesiar a los ratones con exceso de inhalación de isoflurano al 4% y luego sacrificarlos por decapitación rápida utilizando tijeras previamente limpias con etanol al 70%.</li>
	<li>Incida con cuidado la piel a lo largo de la línea media, comenzando desde la abertura en la parte posterior del cráneo y extendiéndose hacia el área frontal.</li>
	<li>Retire delicadamente el cráneo, levantándolo suavemente con las tijeras para evitar dañar las leptomeninges, asegurándose de obtener todo el cerebro, incluidas las leptomeninges.</li>
	<li>Sumerge todo el cerebro en un tampón de lavado y enjuágalo suavemente para eliminar la sangre de la superficie.</li>
	<li>Transfiera el cerebro a una placa de Petri estéril sin picar y extraiga las leptomeninges de la superficie del cerebro usando pinzas de punta fina bajo un microscopio.</li>
	<li>Cortar el tejido de las leptomeninges en fragmentos con unas microtijeras estériles.</li>
</ol>4. Digestión enzimática de Leptomeninges
<ol><li>Añadir 10 ml de la mezcla enzimática (paso 1.2) a los fragmentos e incubar a 37 °C durante 15 min. Agite suavemente para asegurarse de que los fragmentos se desprendan del fondo del tubo. Después, añade 10 ml de tampón de parada (paso 1.4) para detener la digestión.</li>
	<li>Centrifugar a 300 x g durante 5 min a 4 °C y retirar cuidadosamente el sobrenadante con una pipeta.</li>
	<li>Agregue 10 ml de PBS frío y pase la mezcla a través de un colador de 70 μm a un tubo estéril de 50 ml para filtrar los grumos.</li>
</ol>5. Expansión celular
<ol><li>Colocar 1 x 10 5 células porcm2 en un matraz T25 recubierto de fibronectina (paso 2) con5 ml de medio de cultivo.</li>
	<li>Incubar las células a 37 °C con 5% de CO2 durante 24 h. Después, retire el medio para eliminar las células no adheridas.</li>
	<li>Mantener el cultivo reemplazando el 50% del medio cada dos días. Repite este proceso dos o tres veces.</li>
</ol>6. Selección de celdas magnéticas
NOTA: Trabaje rápidamente, mantenga la frialdad de las células y utilice soluciones preenfriadas para evitar el etiquetado de células no específicas.
<ol><li>Preparación de instrumentos y reactivos: conecte un separador magnético a un soporte separador magnético (consulte la tabla de materiales). Conecte una columna de selección al separador magnético y coloque un filtro de células de 70 μm encima de la columna de selección. Coloque un tubo de 50 ml debajo de la columna de selección para recoger el flujo.</li>
	<li>Digestión enzimática: una vez que las células alcancen el 80% de confluencia, aspirar el medio y enjuagar las células con PBS. Añadir tripsina al 0,25% para separar las células adherentes e incubar a 37 °C durante 5 min. Detenga la digestión agregando el tampón de parada. Centrifugar las células a 300 x g durante 5 min a temperatura ambiente y retirar el sobrenadante.</li>
	<li>Incubación de anticuerpos: resuspender 1 x 107 células totales en 100 μL de PBS y añadir 10 μL de anticuerpo LYVE-1 (ver Tabla de Materiales). Mezclar bien e incubar durante 30 minutos en la oscuridad a 4 °C.<ol><li>Girar las células a 300 x g durante 5 min y desechar el sobrenadante. Enjuagar las células introduciendo 1 mL de PBS y centrifugar a 300 x g durante 5 min. Retire completamente el sobrenadante.</li>
	</ol></li>
	<li>Etiquetado de microperlas: resuspender las células en 100 μL de PBS y añadir 20 μL de microperlas (ver Tabla de Materiales). Mezclar bien e incubar durante 30 min en la oscuridad a 4 °C.<ol><li>Girar las celdas a 300 x g durante 5 min y decantar el sobrenadante. Posteriormente, limpiar las células con 1 mL de PBS mediante centrifugación a 300 x g durante 5 min. Finalmente, retire completamente el sobrenadante.</li>
	</ol></li>
	<li>Exclusión magnética negativa: resuspender las células en 4 mL de PBS. Pase las células a través de un filtro de células de 70 μm para eliminar los grumos. Prepare la columna de selección (consulte la Tabla de materiales) enjuagándola con 3 ml de PBS, luego aplique la suspensión de celdas en la columna de selección.<ol><li>Realice los pasos de lavado con 3 ml de PBS y recoja las células negativas de LYVE-1 en un tubo de 50 ml colocado debajo de la columna de selección para permitir que pasen.</li>
	</ol></li>
	<li>Selección magnética positiva: pipetear 6 mL de PBS en la columna de selección. Elimine instantáneamente las celdas marcadas magnéticamente empujando firmemente el émbolo en la columna de selección para obtener LLEC positivos para LYVE-1. NOTA: Espere siempre hasta que el depósito de la columna de selección esté vacío antes de continuar con el siguiente paso.</li>
</ol>7. Cultura de los LLECs
<ol><li>Centrifugar los LLEC positivos LYVE-1 a 300 x g durante 5 minutos y, a continuación, retirar con cuidado el sobrenadante.</li>
	<li>Colocar 1 x 105 células porcm2 en un matraz T25 recubierto de fibronectina con 5 mL de medio de cultivo.</li>
	<li>Mantenga el cultivo reemplazando el 50% del medio cada dos días. Cuando las células alcancen el 80% de confluencia, separe las células con tripsina al 0,25% y realice el paso celular. Utilice las celdas para experimentos posteriores después de 2-3 pasadas.</li>
</ol>8. Análisis de citometría de flujo
NOTA: El análisis citométrico de flujo se realizó siguiendo el procedimiento15 descrito anteriormente.
<ol><li>Añadir tripsina al 0,25% para separar las células adherentes e incubar a 37 °C durante 5 min. Luego, agregue el tampón de parada para detener la digestión.</li>
	<li>Centrifugar las células a 300 x g durante 5 min y aspirar completamente el sobrenadante. Resuspender 1 x 106 células en 100 μL de PBS.</li>
	<li>Añadir 1 μL de anticuerpo LYVE-1. Mezclar bien e incubar durante 10 minutos en la oscuridad a temperatura ambiente.</li>
	<li>Vuelva a suspender las células en una cantidad adecuada de tampón PBS para su análisis mediante citometría de flujo y el software FlowJo (consulte la tabla de materiales).</li>
</ol>9. Tinción inmunofluorescente
NOTA: La tinción inmunofluorescente se realizó siguiendo el procedimiento descrito anteriormente16.
<ol><li>Lave las células con PBS tres veces. Fije las celdas con paraformaldehído (PFA) al 4% durante 10 minutos a temperatura ambiente. Deseche el PFA y lave las células con PBS tres veces.</li>
	<li>Aplique el tampón de bloque durante 1 h a temperatura ambiente. Retire el búfer de bloque de los cubreobjetos.</li>
	<li>Agregue el anticuerpo LYVE-1 en el tampón de tinción a las células e incube en la oscuridad. Lave las celdas tres veces con PBS.</li>
	<li>Agregue un anticuerpo secundario apropiado (ver Tabla de materiales) en el tampón de tinción a las células e incube en la oscuridad. Lave las celdas tres veces con PBS.</li>

Las células endoteliales linfáticas leptomeníngeas (LLECs) recientemente descubiertas forman una malla de células individuales dentro de las leptomeninges, exhibiendo un patrón de distribución distinto en comparación con las células endoteliales linfáticas periféricas 1,2. Las funciones celulares y las implicaciones clínicas asociadas con los LLEC siguen siendo en gran medida un territorio inexplorado. Con el fin de allanar el camino para la investigación funcional de los LLECs, es imperativo establecer un modelo in vitro para su estudio. Por lo tanto, este estudio ha diseñado un protocolo integral para el aislamiento y el cultivo primario de los LLECs.
Los ratones son el modelo animal preferido debido a su idoneidad para la manipulación genética en la investigación de enfermedades. Estudios anteriores han aislado con éxito células endoteliales linfáticas de varios tejidos de ratón, incluidos los ganglios linfáticos3, el tejido mesentérico4, el tejido dérmico5, los linfáticos colectores6 y el tejido pulmonar7. Además, los esfuerzos de investigación han llevado al establecimiento de líneas celulares aracnoideas de rata y líneas celulares capilares linfáticas de rata11,12. A pesar de la existencia de técnicas de cultivo de explantes para leptomeninges13, existe una necesidad urgente de un protocolo estandarizado para el aislamiento y cultivo de LLECs. En consecuencia, este estudio ha cosechado y cultivado con éxito LLECs disociando meticulosamente leptomeninges bajo la guía de un microscopio y promoviendo la expansión de LLECs mediante el uso del factor de crecimiento endotelial vascular-C (VEGF-C). El marcador distintivo de las células endoteliales linfáticas es el receptor hialurónico de vasos linfáticos-1 (LYVE-1)14. Este protocolo de varios pasos aísla selectivamente los LLEC positivos para LYVE-1 mediante MACS y, posteriormente, verifica su pureza mediante análisis citométrico de flujo y tinción inmunofluorescente.

Las células endoteliales linfáticas leptomeníngeas (LLECs) son una población celular intracraneal recientemente descubierta con una distribución única claramente distinta de las células endoteliales linfáticas periféricas. Su función celular y sus implicaciones clínicas siguen siendo en gran medida desconocidas. The newly discovered leptomeningeal lymphatic endothelial cells (LLECs) form a meshwork of individual cells within the leptomeninges, exhibiting a distinct distribution pattern when compared to peripheral lymphatic endothelial cells1,2. The cellular functions and clinical implications associated with LLECs remain largely uncharted territory. In order to pave the way for functional research on LLECs, it is imperative to establish an in vitro model for their study. Therefore, this study has devised a comprehensive protocol for the isolation and primary culture of LLECs.
Mice are the preferred animal model due to their suitability for genetic manipulation in disease research. Previous studies have successfully isolated lymphatic endothelial cells from various mouse tissues, including lymph nodes3, mesenteric tissue4, dermal tissue5, collecting lymphatics6, and lung tissue7. These isolation procedures have primarily relied on techniques such as magnetic-activated cell sorting (MACS) and flow cytometry sorting8,9,10. Additionally, research efforts have led to the establishment of rat arachnoid cell lines and rat lymphatic capillary cell lines11,12. Despite the existence of explant culture techniques for leptomeninges13, there exists an urgent need for a standardized protocol for the isolation and culture of LLECs. Consequently, this study has successfully harvested and cultured LLECs by meticulously dissociating leptomeninges under the guidance of a microscope and promoting LLECs expansion through the use of vascular endothelial growth factor-C (VEGF-C). The distinctive marker for lymphatic endothelial cells is lymphatic vessel hyaluronic receptor-1 (LYVE-1)14. This multi-step protocol selectively isolates LYVE-1-positive LLECs using MACS and subsequently verifies their purity through flow cytometric analysis and immunofluorescent staining.
The primary steps of this multi-step protocol can be summarized as follows: flask coating, dissociation of leptomeninges, enzymatic digestion of leptomeninges, cell expansion, magnetic cell selection, and subsequent culture of LLECs. Finally, the purity of the isolated LLECs is confirmed through flow cytometric analysis and immunofluorescent staining. The overarching aim of this study is to present a reproducible, multi-step protocol for the isolation of LLECs from mouse leptomeninges and their subsequent in vitro culture. This protocol is poised to greatly facilitate investigations into the cellular functions and clinical implications of LLECs.

Leptomeningeal lymphatic endothelial cells (LLECs) are a recently discovered intracranial cellular population with a unique distribution clearly distinct from peripheral lymphatic endothelial cells. Their cellular function and clinical implications remain largely unknown. Consequently, the availability of a supply of LLECs is essential for conducting functional research in vitro. However, there is currently no existing protocol for harvesting and culturing LLECs in vitro.
This study successfully harvested LLECs using a multi-step protocol, which included coating the flask with fibronectin, dissecting the leptomeninges with the assistance of a microscope, enzymatically digesting the leptomeninges to prepare a single-cell suspension, inducing the expansion of LLECs with vascular endothelial growth factor-C (VEGF-C), and selecting lymphatic vessel hyaluronic receptor-1 (LYVE-1) positive cells through magnetic-activated cell sorting (MACS). This process ultimately led to the establishment of a primary culture. The purity of the LLECs was confirmed through immunofluorescence staining and flow cytometric analysis, with a purity level exceeding 95%. This multi-step protocol has demonstrated reproducibility and feasibility, which will greatly facilitate the exploration of the cellular function and clinical implications of LLECs.

Mouse Leptomeninges Dissociation and Enzymatic Digestion for Efficient Culturing of Leptomeningeal Cells

To begin, carefully incise the skin of the sacrificed mouse, starting from the skull's opening and extending towards the frontal area. Using scissors, delicately lift and remove the skull without damaging the leptomeninges, collecting the entire brain. Submerge the whole brain in the washing buffer, and gently flush it to remove surface blood.Transfer the brain into a sterile Petri dish without chopping. Then, under a microscope, use fine-point tweezers to extract the leptomeninges from the brain's surface. Using sterile microscissors, cut the leptomeninges tissue into fragments.Repair the digestive enzyme mix using the given components. Add 10 milliliters of mix to the fragments and incubate at 37 degrees Celsius for 15 minutes. Gently agitate to detach the fragments from the bottom of the tube.Then, add 10 milliliters of stopping buffer. Centrifuge the suspension at 300 g for five minutes at four degrees Celsius. And using a pipette, carefully remove the supernatant.Add 10 milliliters of cold PBS, and filter any clumps through a 70 micron strainer into a sterile 50 milliliter tube. Plate 10 to the fifth cells per square centimeter into a fibronectin-coated T25 flask with five milliliters of culture medium, and incubate at 37 degrees Celsius with 5%carbon dioxide for 24 hours. After incubation, replace the medium with a fresh medium to eliminate non-attached cells.

Establishing Primary In Vitro Culture of Leptomeningeal Lymphatic Endothelial Cells (LLECs)

Begin by attaching a magnetic separator to its stand. Connect a selection column to the magnetic separator and place a 70 micron cell strainer on top of the selection column. Place a 50 milliliter tube beneath the selection column to collect the flow.Once the mouse brain cells reach 80%confluency, aspirate the medium and rinse the cells with PBS. Add 0.25%trypsin to detach the adherence cells and incubate at 37 degrees Celsius for five minutes. Then add a stopping buffer.Centrifuge the cell suspension at 300 G for five minutes, and remove the supernatant. For antibody staining, resuspend 10 to the seven cells in 100 microliters of PBS. Then add 10 microliters of LYVE-1 antibody solution and mix thoroughly.Incubate the mixture for 30 minutes in the dark at four degrees Celsius. After incubation, centrifuge the cells and discard the supernatant. Then rinse the cells by adding one milliliter of PBS and centrifuging at 300 G for five minutes.For microbead labeling, resuspend the pellet in 100 microliters of PBS and 20 microliters of microbeads. Make swell and incubate for 30 minutes in the dark at four degrees Celsius. Then centrifuge the suspension and wash the pellet with one milliliter of PBS.Again, centrifuge and resuspend the pellet in four milliliters of PBS for magnetic negative exclusion. Pass the cell suspension through a 70 micron strainer to eliminate clumps. Prepare the selection column by rinsing it with three milliliters of PBS.Then add cell suspension into the selection column. Wash the column with three milliliters of PBS and collect LYVE-1 negative cells into a 50 milliliter tube. For magnetic positive selection, pipette six milliliters of PBS into the selection column.Then flush the magnetically labeled cells by firmly pushing the plunger into the selection column to obtain LYVE-1 positive LLECs. Next, centrifuge the positive cell suspension and remove the supernatant. Plate 10 to the fifth cells per square centimeter into a T25 flask with five milliliters of culture medium.Maintain the culture by replacing 50%of the medium every alternate day. When the cells reach 80%confluency, detach them as demonstrated previously before performing cell passage. Flow cytometry analysis revealed that the percentage of LYVE-1 positive cells in passage two was not significantly different from passage three after MACS, indicating a purity greater than 95%Immunofluorescent staining confirmed co-staining of LYVE-1 with PDPN, VEGFR-3 and PROX1, establishing the identity of LLECs.LYVE-1 positive cells did not express F48 and platelet derived growth factor beta, effectively distinguishing LLECs from macrophages and fibroblasts. Leptomeningeal cells before MACS displayed heterogeneous morphology ranging from round spheres to fused fiber shapes. However, post MACS, LLECs exhibited typical endothelial-like features such as spindle and cobblestone shapes.
