The objective of this work is to establish an optical imaging technique that would enable monitoring of the integration of mesenchymal stem cells (MSC) in arthritic joints. Our approach is based on first developing a labeling technique of MSC with the fluorescent dye DiD followed by tracking the cell migration kinetics from the spatial distribution of the DiD fluorescence in optical images (OI). The experimental approach involves first the in vitro OI of MSC labeled with DiD accompanied by fluorescence microscopy measurements to establish localization of the signal within the cells. Thereafter, DiD-labeled MSC were injected into polyarthritic, athymic rats and the signal localization within the experimental animals was monitored over several days. The experimental results indicate that DiD integrated into the cell membrane. DiD-labeled MSC localization in the arthritic ankle joints was observed with OI indicating that this method can be applied to monitor MSC in arthritic joints.

An optical imaging method to monitor stem cell migration in a model of immune-mediated arthritis.

L&#x26;#39;objectif de ce travail est d&#x26;#39;&#xE9;tablir une technique d&#x26;#39;imagerie optique qui permettrait le suivi de l&#x26;#39;int&#xE9;gration des cellules souches m&#xE9;senchymateuses (CSM) dans les articulations arthritiques. Notre approche est bas&#xE9;e sur le premier point d&#x26;#39;une technique d&#x26;#39;&#xE9;tiquetage des MSC avec le colorant fluorescent saviez suivie par le suivi des cin&#xE9;tiques de migration de cellules de la distribution spatiale de la fluorescence dans DiD images optiques (OI). L&#x26;#39;approche exp&#xE9;rimentale consiste tout d&#x26;#39;abord in vitro OI de MSC &#xE9;tiquet&#xE9;s avec DID accompagn&#xE9;e par des mesures de microscopie par fluorescence d&#x26;#39;&#xE9;tablir la localisation du signal dans les cellules. Das Ziel dieser Arbeit ist es, ein optisches bildgebendes Verfahren zu etablieren, das &Uuml;berwachung der Integration von mesenchymalen Stammzellen (MSC) in arthritischen Gelenken erm&ouml;glichen w&uuml;rde. Unser Ansatz basiert auf der ersten Entwicklung eine Kennzeichnung Technik des MSC mit der Leuchtstofff&auml;rbung gefolgt von tracking-die Zelle Migration Kinetik von der r&auml;umlichen Verteilung der die Fluoreszenz in optische Bilder (OI) haben. Der experimentelle Ansatz beinhaltet zun&auml;chst die in-vitro-OI des MSC mit beschriftet begleitet von Fluoreszenz-Mikroskopie-Messungen zur Lokalisierung des Signals innerhalb der Zellen zu etablieren. Danach hat-mit der Bezeichnung MSC wurden in Polyarthritic injiziert, athymischen Ratten und die Signal-Lokalisierung in den Versuchstieren wurde &uuml;ber mehrere Tage beobachtet. Die experimentellen Ergebnisse zeigen, die in die Zellmembran integriert. El objetivo de este trabajo es establecer una t&#xE9;cnica de imagen &#xF3;ptica que permita el seguimiento de la integraci&#xF3;n de las c&#xE9;lulas madre mesenquimales (MSC) en las articulaciones artr&#xED;ticas. Nuestro enfoque se basa en la primera el desarrollo de una t&#xE9;cnica de etiquetado del MSC con el colorante fluorescente DiD seguido por el seguimiento de la cin&#xE9;tica de la migraci&#xF3;n de c&#xE9;lulas de la distribuci&#xF3;n espacial de la DiD de fluorescencia en las im&#xE1;genes &#xF3;pticas (OI). El m&#xE9;todo experimental implica primero la in vitro de OI de MSC marcado con hizo acompa&#xF1;ado por mediciones de microscop&#xED;a de fluorescencia para establecer la localizaci&#xF3;n de la se&#xF1;al dentro de las c&#xE9;lulas. MSC A partir de entonces, DID-etiquetados se inyectaron en las ratas, los poliartr&#xED;ticas at&#xED;micos y la localizaci&#xF3;n de la se&#xF1;al dentro de los animales de experimentaci&#xF3;n, se control&#xF3; durante varios d&#xED;as.
