Dans la dernière décennie, les techniques de manipulation et d&#39;imagerie de molécules simples ont vu de grands progrès en termes de résolution spatiale et temporelle. La combinaison des techniques de manipulation et d&#39;imagerie est à la base de puissants outils qui permettent maintenant le contrôle de l&#39;état mécanique d&#39;un polymère biologique unique, tels que ADN, ARN ou du cytosquelette des filaments, et la localisation simultanée de protéines individuelles en interaction avec le même polymè...

Molécule unique (SM) techniques ont considérablement développé au cours des trente dernières années pour répondre à la nécessité de surmonter certaines des limites de mesures de la solution, en vrac traditionnels 1-3. La manipulation de molécules biologiques simples a créé la possibilité de mesurer les propriétés mécaniques des biopolymères 4 et de contrôler les paramètres mécaniques de protéine-protéine 5 et interactions protéine-ADN 6,7. SM détection de fluorescence, d&#39;autre part, représente un outil très polyvalent pour l&#39;étude de l&#39;activité de la protéine in vitro et in vivo, ce qui conduit à la possibilité de localisation et de suivi de molécules individuelles avec une précision nanométrique. Par ajustement de l&#39;appareil d&#39;étalement de point de fonction-SM de l&#39;image, en fait, on peut réaliser avec une précision de localisation en fonction principalement de rapport signal-sur-bruit (SNR) et d&#39;atteindre une limite de l&#39;ordre de 8,9 nanomètre. Récemment, la combinaison de ces deux techniques a produit une nouvelle génération de dispositifs expérimentaux de 10 à 14 permettant la manipulation simultanée d&#39;un substrat biologique (par exemple un filament d&#39;actine ou une molécule d&#39;ADN) et la détection / localisation d&#39;un partenaire d&#39;interaction enzyme (par exemple myosine ou d&#39;une protéine de liaison à l&#39;ADN). Les avantages de ces techniques reposent principalement sur la possibilité d&#39;exercer un contrôle mécanique sur le polymère piégé, ainsi enabling l&#39;étude de la dynamique d&#39;interaction par rapport à des forces ou des couples. En outre, la méthode permet de mesurer les réactions biochimiques loin de la surface, en évitant l&#39;une des principales limites des méthodes classiques SM, à savoir, la nécessité pour l&#39;immobilisation des molécules à l&#39;étude sur une surface (lame de verre ou microsphères). La combinaison des deux techniques de molécules simples, il faut surmonter plusieurs difficultés techniques, principalement en raison des exigences de stabilité mécanique et SNR suffisant (en particulier en exigeant la localisation avec une précision nm) 15. En particulier, lors de l&#39;accouplement détection par fluorescence SM avec des pincettes optiques, la réduction du bruit et photoblanchiment des lasers infrarouges piégeage 16 et le contrôle des tampons biochimiques pour l&#39;assemblage des complexes biologiques et la performance des mesures expérimentales 11 sont d&#39;une importance primordiale. Ici, nous décrivons les méthodes pour effectuer avec succèsmesures dans une configuration de localisation double piégeage / SM fluorescence. La méthodologie est illustrée par l&#39;exemple de la protéine répresseur lactose (Lad) marqué par fluorescence (avec Atto532) et détecté comme il se lie à une molécule d&#39;ADN (coincé entre deux pinces optiques) contenant des séquences spécifiques Laci liaison (c.-à-opérateurs). Nous démontrons l&#39;efficacité de la méthode de détection de la liaison à l&#39;ADN de LacI et la diffusion le long de son contour dans le processus de recherche de cible. La méthode est applicable à n&#39;importe quelle combinaison de séquences d&#39;ADN et de protéine de liaison à l&#39;ADN, ainsi que pour d&#39;autres systèmes (microtubules ou des filaments d&#39;actine et de protéines interagissant avec les moteurs).

combining single-molecule manipulation and imaging for the study of protein-dna interactions

The paper describes the combination of optical tweezers and single molecule fluorescence detection for the study of protein-DNA interaction. The method offers the opportunity of investigating interactions occurring in solution (thus avoiding problems due to closeby surfaces as in other single molecule methods), controlling the DNA extension and tracking interaction dynamics as a function of both mechanical parameters and DNA sequence. The methods for establishing successful optical trapping and nanometer localization of single molecules are illustrated. We illustrate the experimental conditions allowing the study of interaction of lactose repressor (lacI), labeled with Atto532, with a DNA molecule containing specific target sequences (operators) for LacI binding. The method allows the observation of specific interactions at the operators, as well as one-dimensional diffusion of the protein during the process of target search. The method is broadly applicable to the study of protein-DNA interactions but also to molecular motors, where control of the tension applied to the partner track polymer (for example actin or microtubules) is desirable.
