Here we describe a molecular readout of long-term olfactory adaptation in Caenorhabditis elegans. The Protein Kinase G, EGL-4, is necessary for stable adaptation responses in the primary sensory neuron pair called AWC. During prolonged odor exposure EGL-4 translocates from the cytosol to nucleus of the AWC.

Hyo Jin Park 1, Ji Hoon Oh 1, Sang-Jun Ha 1
1Department of Biochemistry, College of Life Science & Biotechnology, Yonsei University

Here, we describe a protocol to analyze the phenotype of regulatory T (Treg) cells isolated from naïve and chronic lymphocytic choriomeningitis virus-infected mice. In addition, we provide a process to evaluate the suppressive activity of the Treg cells.

Uihan Kim 1, Jaewoo Song 2, Suho Ryu 1, Soocheol Kim 1, Chulmin Joo 1
1School of Mechanical Engineering, Yonsei University, 2Department of Laboratory Medicine, Yonsei University

A photo-thermal angular light scattering (PT-AS) sensor enables the rapid and chemical-free hemoglobin assay of nanoliter-scale blood samples. Here, details of the PT-AS setup and a measurement protocol for the hemoglobin concentration in blood are provided. Representative results for anemic blood samples are also presented.

Cultivation of Caenorhabditis elegans in Three Dimensions in the Laboratory
Tong Y. Lee 1, Kyoung-hye Yoon 1, Jin I. Lee 1
1Division of Biological Science and Technology, Yonsei University

We present a simple method to construct 3D nematode cultivation systems called NGT-3D and NGB-3D. These can be used to study nematode fitness and behaviors in habitats that are more similar to natural Caenorhabditis elegans habitats than the standard 2D laboratory C. elegans culture plates.

1College of Pharmacy, Yonsei University, 2College of Medicine, Yonsei University, 3Yonsei Institute of Pharmaceutical Sciences, 4Polyscientech Co. Ltd

We report a method for drug sorption evaluation using a pump and model drugs (diazepam and tacrolimus). After drug analysis using high-performance liquid chromatography, drug concentrations and sorption levels in the tubes of administration sets are calculated.

Recombinant Protein Expression, Crystallization, and Biophysical Studies of a Bacillus-conserved Nucleotide Pyrophosphorylase, BcMazG
Meong Il Kim 1, Choongdeok Lee 1, Minsun Hong 1
1Division of Biological Science and Technology, Yonsei University

Bacillus anthracis is the obligate pathogen of fatal inhalational anthrax, so studies of its genes and proteins are strictly regulated. An alternative approach is to study orthologous genes. We describe biophysicochemical studies of a B. anthracis ortholog in B. cereus, bc1531, requiring minimal experimental equipment and lacking serious safety concerns.

We performed unilateral carotid artery occlusion on postnatal day 7-10 CD-1 mouse pups to create a neonatal hypoxic-ischemic (HI) model and investigated the effects of HI brain injury. We studied neurobehavioral functions in these mice compared to non-operated normal mice.

Preparation of DMMTAV and DMDTAV Using DMAV for Environmental Applications: Synthesis, Purification, and Confirmation
Hosub Lee 1, Youn-Tae Kim 2, Seulki Jeong 1, Hye-On Yoon 1
1Korea Basic Science Institute, Seoul Center, 2Natural Science Research Institute, Yonsei University

This article presents modified experimental protocols for dimethylmonothioarsinic acid (DMMTAV) and dimethyldithioarsinic acid (DMDTAV) synthesis, inducing dimethylarsinic acid (DMAV) thiolation through mixing of DMAV, Na2S, and H2SO4. The modified protocol provides an experimental guideline, thereby overcoming limitations of the synthesis steps that could have caused experimental failures in quantitative analysis.
