Phillip Huyett 1, Mark Gilbert 1, Lijun Liu 2, Robert L. Ferris 2, Seungwon Kim 1
1Department of Otolaryngology, University of Pittsburgh Medical Center, 2University of Pittsburgh Cancer Institute, Hillman Cancer Center

Perineural invasion (PNI) is a common feature of head and neck squamous cell carcinoma (HNSCC), conferring lower survival rates. Its mechanisms are poorly understood. Utilizing neurites generated from murine dorsal root ganglia confined to a semisolid matrix, the pathways involved in the PNI of HNSCC cell lines can be investigated.

Model of Ischemic Heart Disease and Video-Based Comparison of Cardiomyocyte Contraction Using hiPSC-Derived Cardiomyocytes
Yun Liu *1, Yin Liang *1, Mengxue Wang *1, Chen Wang 1, Heng Wei 2, Keiji Naruse 1, Ken Takahashi 1
1Department of Cardiovascular Physiology, Graduate School of Medicine, Dentistry and Pharmaceutical Sciences, Okayama University, 2Institute of Laboratory Animals, Graduate School of Medicine, Kyoto University

We present a model of ischemic heart disease using cardiomyocytes derived from human induced pluripotent stem cells, together with a method for quantitative evaluation of tissue damage caused by ischemia. This model can provide a useful platform for drug screening and further research on ischemic heart disease.

Surgically Induced Cardiac Volume Overload by Aortic Regurgitation in Mouse
Jian Wu *1, Shijun Wang *1, Le Kang *1, Chunjie Yang *1, Jianguo Jia 1, Xingxu Wang 2, Ge Wei 1, Xuan Li 1, Jieyun You 2, Yunzeng Zou 1
1Shanghai Institute of Cardiovascular Diseases, Zhongshan Hospital and Institutes of Biomedical Sciences, Fudan University, 2Department of Cardiovascular Medicine, Shanghai East Hospital, School of Medicine, Tongji University

This protocol presents a practical guide on the surgery for creation of aortic regurgitation (AR) in the mouse. Assessment of the AR mouse by echocardiography and invasive hemodynamic measurement recapitulates its clinically relevant characteristics of volume overload-induced eccentric hypertrophy, suggesting its promising application in the study of cardiac hypertrophy.
