PRB (Retinoblastom-Protein) hat eine zentrale Rolle in der Kontrolle von den G-1-S-Phasen&uuml;bergang des Zellzyklus, die teilweise durch die Regulierung der E2F Transkriptionsfaktoren vermittelt wird. Beim S-Phase Eintritt ist umfassend, pRB phosphoryliert die in Zug Versionen gebunden E2Fs, den Ausdruck der Gene, die f&uuml;r die S-Phase Fortschreiten zu fahren. In der vorliegenden Studie zeigen wir, dass E2F1-unterh&auml;lt die F&auml;higkeit zur Interaktion mit PpRB (Hyperphosphorylated pRB). Diese Interaktion ist abh&auml;ngig von der &#39;spezifischen&#39; E2F1-Bindungsstelle befindet sich in der C-terminalen Dom&auml;ne PRB. Eine einzigartige Region der markierten Feld Dom&auml;ne des E2F1 wendet sich die &#39;spezifische&#39; Website, um die Interaktion mit PpRB zu vermitteln. Die mechanistische Grundlage f&uuml;r die Interaktion zwischen E2F1 und PpRB ist subtil. Eine einzelne Substitution zwischen Valin und Prolin R&uuml;ckst&auml;nde im markierten Feld unterscheidet E2F1s F&auml;higkeit zur Interaktion mit PpRB in der Unf&auml;higkeit des E2F3 an der &#39;spezifischen&#39; Website in PpRB gebunden. Die E2F1-pRB-Interaktion am Standort &#39;spezifischen&#39; unterh&auml;lt auch die F&auml;higkeit, die transkriptionelle Aktivierung E2F1 Zielgene zu regulieren. Diese Daten zeigen einen Mechanismus, durch welchen, den E2F1 Verordnung von pRB beibehalten kann, wenn pRB Hyperphosphorylated und vermutlich inaktiv sein.

The pRB (retinoblastoma protein) has a central role in the control of the G(1)-S phase transition of the cell cycle that is mediated in part through the regulation of E2F transcription factors. Upon S-phase entry pRB is phosphorylated extensively, which in turn releases bound E2Fs to drive the expression of the genes required for S-phase progression. In the present study, we demonstrate that E2F1-maintains the ability to interact with ppRB (hyperphosphorylated pRB). This interaction is dependent upon the 'specific' E2F1-binding site located in the C-terminal domain of pRB. A unique region of the marked box domain of E2F1 contacts the 'specific' site to mediate the interaction with ppRB. The mechanistic basis of the interaction between E2F1 and ppRB is subtle. A single substitution between valine and proline residues in the marked box distinguishes E2F1's ability to interact with ppRB from the inability of E2F3 to bind to the 'specific' site in ppRB. The E2F1-pRB interaction at the 'specific' site also maintains the ability to regulate the transcriptional activation of E2F1 target genes. These data reveal a mechanism by which E2F1 regulation by pRB can persist, when pRB is hyperphosphorylated and presumed to be inactive.

The biochemical basis of CDK phosphorylation-independent regulation of E2F1 by the retinoblastoma protein.

La pRB (prot&#xE9;ine du r&#xE9;tinoblastome) a un r&#xF4;le central dans le contr&#xF4;le de la G (1)-S transition de phase du cycle cellulaire qui est m&#xE9;di&#xE9;e en partie gr&#xE2;ce &#xE0; la r&#xE9;gulation de facteurs de transcription E2F. Ces donn&#xE9;es r&#xE9;v&#xE8;lent un m&#xE9;canisme par lequel E2F1 r&#xE9;gulation par pRB peut persister, quand pRB est hyperphosphoryl&#xE9;e et pr&#xE9;sum&#xE9;es &#xEA;tre inactif.

Il pRB (proteina retinoblastoma) ha un ruolo centrale nel controllo della G (1)-transizione di fase S del ciclo cellulare che &egrave; in parte mediato attraverso la regolazione dei fattori di trascrizione E2F. Al momento dell&#39;ingresso in fase S pRB &egrave; fosforilata estesamente, che in giro comunicati associato E2Fs a guidare l&#39;espressione dei geni necessari per la progressione in fase S. Nel presente studio, dimostreremo che E2F1-mantiene la capacit&agrave; di interagire con ppRB (iperfosforilata pRB). Questa interazione &egrave; dipendente del sito vincolante E2F1 &#39;specifico&#39; si trova nel dominio C-terminale del pRB. Una regione unica del dominio casella di E2F1 contatti il sito &#39;specifico&#39; per mediare l&#39;interazione con ppRB. La base dell&#39;interazione tra E2F1 e ppRB meccanicistica &egrave; sottile. Una singola sostituzione tra valina e prolina residui nella casella contrassegnato distingue la capacit&agrave; di E2F1 di interagire con ppRB dall&#39;incapacit&agrave; di E2F3 da associare al sito &#39;specifico&#39; in ppRB. L&#39;interazione E2F1-pRB presso il sito di &#39;specifico&#39; mantiene anche la capacit&agrave; di regolare l&#39;attivazione trascrizionale dei geni bersaglio E2F1. Questi dati rivelano un meccanismo da cui E2F1 regolamento da pRB pu&ograve; persistere, quando pRB &egrave; iperfosforilata e si presume sia inattivo.

La pRb (prote&#xED;na retinoblastoma) tiene un papel central en el control de la G (1)-S transici&#xF3;n de fase del ciclo celular que est&#xE1; mediada, en parte, a trav&#xE9;s de la regulaci&#xF3;n de los factores de transcripci&#xF3;n E2F. Tras la fase S-entrada pRB es fosforilada extensamente, que a su vez libera E2Fs unidos a conducir la expresi&#xF3;n de los genes necesarios para la progresi&#xF3;n de la fase S. En el presente estudio, hemos demostrado que E2F1-mantiene la capacidad de interactuar con ppRB (hyperphosphorylated PRB). Esta interacci&#xF3;n es dependiente de la &#x26;#39;espec&#xED;fica&#x26;#39; E2F1-sitio de uni&#xF3;n situado en el dominio C-terminal de pRb. Una regi&#xF3;n &#xFA;nica en el dominio de casilla de E2F1 en contacto con el &#x26;quot;espec&#xED;fico&#x26;quot; del sitio para mediar en la interacci&#xF3;n con ppRB. La base mec&#xE1;nica de la interacci&#xF3;n entre E2F1 y ppRB es sutil. Una sola sustituci&#xF3;n entre los residuos de valina y prolina en la casilla que distingue la capacidad de E2F1 para interactuar con ppRB de la incapacidad de E2F3 para enlazar con el &#x26;quot;espec&#xED;fico&#x26;quot; en el sitio de ppRB. La interacci&#xF3;n E2F1-pRB en el &#x26;quot;espec&#xED;fico&#x26;quot; sitio tambi&#xE9;n mantiene la capacidad de regular la activaci&#xF3;n transcripcional de los genes diana E2F1. Estos datos revelan un mecanismo por el cual la regulaci&#xF3;n de E2F1 pRB puede persistir, cuando pRb se hyperphosphorylated y presume que es inactivo.
