Assays for Validating Histone Acetyltransferase Inhibitors
Aaron R. Waddell 1, Daiqing Liao 1
1Department of Anatomy and Cell Biology, and UF Health Cancer Center, University of Florida College of Medicine

Inhibitors of histone acetyltransferases (HATs, also known as lysine acetyltransferases), such as CBP/p300, are potential therapeutics for treating cancer. However, rigorous methods for validating these inhibitors are needed. Three in vitro methods for validation include HAT assays with recombinant acetyltransferases, immunoblotting for histone acetylation in cell culture, and ChIP-qPCR.

This method introduces a simple technique for the detection of endogenous monoamine release using acute brain slices. The setup uses a 48-well plate containing a tissue holder for monoamine release. Released monoamine is analyzed by HPLC coupled with electrochemical detection. Additionally, this technique provides a screening method for drug discovery.

A Guide to Examining Intramuscular Fat Formation and its Cellular Origin in Skeletal Muscle
Connor D. Johnson 1, Lylybell Y. Zhou 1, Daniel Kopinke 1,2
1Department of Pharmacology and Therapeutics, University of Florida College of Medicine, 2Myology Institute, University of Florida College of Medicine

The replacement of healthy muscle tissue with intramuscular fat is a prominent feature of human diseases and conditions. This protocol outlines how to visualize, image, and quantify intramuscular fat, allowing the rigorous study of the mechanisms underlying intramuscular fat formation.
