動物実験は、中南大学動物管理委員会によって承認されました。すべての研究は、実験動物の福祉と倫理の原則に沿って実施されました。
1.動物の給餌と麻酔の手順
<ol><li>280〜320 gのSprague-Dawleyオスラットをグループ化し、12時間/ 12時間の明暗サイクルで維持し、自由に餌 と水にアクセスできます。ラットが6日間順応した後、研究を実施します。</li>
	<li>ラットが足または尾のつまみに反応しなくなるまで、誘導ボックス内の0.6 L / minの気流で3%イソフルランでラットを麻酔します。.流量を30秒間維持します。 注:鎮痛剤は、神経行動学的評価におけるラットの反応を妨げるため、使用されませんでした。</li>
</ol>2. 術前設定
<ol><li>硬度35D(スポンジの重量35kg/m3)で、長さと幅は同じで厚さは12cmのスポンジを、トップカバーのないアクリルボックス(15 cm x 22 cm x 43 cm)に入れます。</li>
	<li>錫箔(厚さ20μm)を切り取り、粘着テープでアクリル箱に貼り付けると、ネズミの体重を支えることができる破壊可能な表面を形成します。さらに、ラットの頭を配置するための指定された場所となる約10cmのカットラインをマークします。</li>
	<li>アイロンスタンドを使用して、PVCチューブを所定の位置にしっかりと固定します。直径18ミリメートルで重さ550グラムの穴あき重りを準備します。ポリ塩化ビニルまたはPVCチューブ内の1メートルの高さで釣り糸に重りを取り付けます。そして、スズ箔の3センチ上のガイドチューブの位置を調整します。</li>
	<li>ヘルメットと枕を用意します。直径10mm、厚さ3mmのステンレス製の円盤を使ってヘルメットを作ります。くさび形のスポンジ枕を用意して、ネズミの頭の下に置き、重力方向に対して垂直になるようにします。 注:衝撃装置の概略図を 図1に示します。ヘルメットは、衝撃の場所を特定し、外力の分散を強化する目的を果たします。枕は、均一で安定した損傷を保証するために利用されています。</li>
</ol>3. mTBI誘導
<ol><li>麻酔をかけたラットを胸の錫箔の上にすばやく置きます。 注:mTBI誘導には2人のオペレーターが必要であり、1人は準備用、もう1人は検証用です。</li>
	<li>準備:枕をネズミの下に置き、ネズミの頭がホイル紙と平行になるようにします。ヘルメットをネズミの耳に合わせ、所定の位置に固定します。</li>
	<li>確認:塩ビパイプがヘルメットの真上に配置されていることを確認します。両方のオペレーターが正しい設定を確認したら、次の手順に進みます。</li>
	<li>頭の回転の誘導:重りを解放し、落下させてラットの頭にぶつからせ、スポンジへの落下と180°の回転を誘発します。</li>
	<li>ネズミを清潔なケージに仰向けに置きます。</li>
</ol>4.偽の誘導
<ol><li>ラットは、以前のmTBI誘導の説明と同じ方法で扱いますが、頭部に衝撃を与えないでください。</li>
</ol>5.検証手順:急性神経行動学的評価
注: 以下の評価は、神経学的重症度スコア9 と Flierl et al.10 によるプロトコルに基づいて修正されました。これらの評価はすべて、ラットが正立反射を回復してから10分後に実施されました。
<ol><li>意識喪失時間:ラットが麻酔されてから正動反射が回復するまでの時間を記録します。 注:直立反射は、ネズミを仰向けにしたときにひっくり返るプロセスです。矯正反射の喪失は人道的なエンドポイントと見なされるべきであり、動物は施設のガイドラインに従って安楽死させる必要があります。</li>
	<li>最初のシーク行動時間:ラットが麻酔されてから、初めてシーク行動を示すまでの時間を記録します。 注:行動を求めることは、環境への関心の表れであり、生理学的反応です。</li>
	<li>脱出する能力<ol><li>出口(長さ12.5 cm、幅9 cm)のある円形の装置(直径0.5 m、高さ0.3 m)の真ん中にラットを置きます。</li>
		<li>ネズミが円から出るのにかかる時間を記録します。 注意: ラットが180秒以内に円から出ない場合は、時間を180秒として記録します。</li>
	</ol></li>
	<li>ビームバランス能力試験<ol><li>それに応じて、3 cm、2 cm、および1.5 cm幅のビームにラットを1分間置きます。</li>
		<li>ネズミがビーム上で安定した姿勢でバランスを保っている場合は、0としてスコアを付けます。</li>
		<li>ネズミがビームの側面をつかんだら、1のスコアを与えます。ネズミが梁を抱きしめ、片方の手足が梁から落ちた場合、2としてスコアを付けます。</li>
		<li>ネズミがビームを抱きしめ、2本の手足がビームから落ちるかスピンした場合(>60秒)、3とスコアを付けます。</li>
		<li>ネズミが梁の上でバランスをとろうとして落ちた場合(40秒>)、4と評価します。</li>
		<li>ネズミがビームの上でバランスをとろうとして落ちた場合(>20秒)、5とスコアを付けます。</li>
		<li>ネズミがバランスをとろうとしたり、梁にぶら下がったりせず、20秒以内に落ちた場合は、6とスコアを付けます。 注:ビームバランステストは、事前試験を必要としません。</li>
	</ol></li>
</ol>6.検証手順:神経行動評価
注:行動実験の前に、ラットはストレスと新規性の混乱を最小限に抑えるために、3日間連続して毎日2分間処理されました。すべての行動実験は、実験開始前に動物を試験環境に60分間置いて行った。
<ol><li>運動協調能力(ビーム課題)<ol><li>実験装置<ol><li>ネズミを平均台の一端(長さ1.5 m、床から75 cm)に置きます。もう一方の端にエスケープボックス(斜めの寝具ホームケージ)を置きます。</li>
			<li>試験中に転倒した場合のネズミの怪我の潜在的なリスクを軽減するために、ビームの下にフォームパッドを配置します。</li>
			<li>ビデオカメラの電源を入れます。</li>
			<li>受傷後または偽治療後の特定の時点(例:1日目、3日目、7日目)に検査日をスケジュールします。</li>
		</ol></li>
		<li>研修期間(2日間)<ol><li>4cm幅の梁を3回連続して横切るようにラットを訓練し、続いて2cm幅の梁を2回試します。</li>
			<li>トレーニング中は、ネズミが干渉することなく簡単に横切れるようになるまで、梁を横切ってネズミを優しく誘導します。</li>
		</ol></li>

rat model of closed-head mild traumatic injury and its validation

動物モデルは、軽度外傷性脳損傷(mTBI)の理解を深め、臨床研究を導くために重要です。有意義な洞察を得るためには、安定した再現性のある動物モデルの開発が不可欠です。本研究では、Sprague-Dawleyラットを用いたクローズドヘッドmTBIモデルと代表的な検証法について詳細に記述し、モデリング効果を検証した。このモデルでは、550gの重りを100cmの高さから破壊可能な表面のラットの頭に直接落下させ、その後180度回転させます。損傷を評価するために、ラットは損傷後10分後に、意識喪失時間、最初のシーク行動時間、脱出能力、およびビームバランス能力テストを含む一連の神経行動学的評価を受けました。受傷後の急性期および亜急性期には、運動協調能力(ビーム課題)、不安(オープンフィールドテスト)、学習能力と記憶能力(モリスウォーターメイズテスト)を評価するための行動テストが実施されました。クローズドヘッドmTBIモデルは、死亡率を最小限に抑え、実際の状況を再現した一貫した損傷反応を示しました。この検証手法は、モデル開発を効果的に検証し、モデルの安定性と一貫性を確保しました。

Mild traumatic brain injury (mTBI), or concussion, is the most prevalent type of injury and can lead to various short-term and chronic symptoms1. These symptoms may include dizziness, headaches, depression, and anhedonia, among others, leading to significant suffering for individuals affected by mTBI2. Since most mTBIs are caused by blunt force trauma3, it becomes imperative to develop animal models that accurately mimic such injuries. These models are essential for gaining a better understanding of the injury and its underlying mechanisms, offering a controlled environment with reduced variability and heterogeneity compared to human studies.
Numerous well-established rodent models have been developed for traumatic brain injury (TBI), including fluid percussion injury (FPI)4, controlled cortical impact (CCI)5, weight-drop injury6, blast traumatic brain injury7, and others. However, these models primarily focus on replicating moderate to severe TBI scenarios. In contrast, the experimental models specifically designed to simulate mTBI have received relatively less attention and remain underexplored8. Therefore, there is a critical need to establish a stable and reproducible animal model that accurately represents mTBI. Such a model would significantly enhance our understanding of the neurobiological and behavioral consequences associated with mTBI.
One cannot distinguish the functional deficits in mTBI rats compared to normal rats via casual observation after the effects of anesthesia have worn off. Therefore, it is necessary to administer specific tests. In humans, a wide range of clinical assessments are used to evaluate patients9,10,11. Similarly, establishing a successful model in the rat model also requires using rapid assessment tools to determine its validity.
In this study, we present a closed-head mTBI rat model, enabling the investigation of mTBI in a manner that closely resembles the human condition. The detailed description of the model and its validation procedure provides a comprehensive understanding of the experimental approach utilized in studying mTBI.

In this study, we introduced a closed-head, mild traumatic brain injury rat model, and provided a comprehensive overview of its validation. Our goal was to offer a straightforward approach for replicating this type of brain injury and a rapid assessment method for confirming the success of the model. This model successfully simulates a closed-head, mild traumatic brain injury without the need for scalp incision or skull opening, provide a more accurate representation of the impact scenario observed in human cases.The avoidance of scalp incision helps to prevent inflammatory responses that may not align with the active situation. Mild traumatic brain injury lacks subjective and accurate diagnostic methods. Animal models are crucial for advancing our understanding of mild traumatic brain injury (mTBI) and guiding clinical research. To achieve meaningful insights, developing a stable and reproducible animal model is essential. In this study, we report a detailed description of a closed-head mTBI model and a representative validation method using Sprague-Dawley rats to verify the modeling effect. The model involves dropping a 550 g mass weight from a height of 100 cm directly onto the head of a rat on a destructible surface, followed by a 180-degree turn. To assess the injury, rats underwent a series of neurobehavioral assessments 10 min post-injury, including time of loss of consciousness, first seeking-behavior time, escape ability, and beam balance ability test. During the acute and subacute stages following the injury, behavioral tests were conducted to assess motor coordination ability (Beam task), anxiety (Open Field test), and learning and memory abilities (Morris Water Maze test). The closed-head mTBI model produced a consistent injury response with minimal mortality and replicated real-life situations. The validation method effectively verified the model development and ensured the stability and consistency of the model.

Here, we present a closed-head mild traumatic brain injury (mTBI) rat model and its validation exhibiting remarkable similarity to human mTBI concerning behavioral manifestations during the acute and subacute stages.

Experimental Set Up to Create a Rat Model of Mild Traumatic Brain Injury (MTBI)  

To begin, place a 12-centimeter thick sponge with a 35D hardness in an acrylic box, lacking a top cover. Then, cut a sheet of tinfoil and adhere it to the acrylic box with adhesive tape to create a fragile surface capable of supporting the weight of a rat. Also, mark a 10-centimeter long line as the designated location for positioning the rat's head.Using an iron stand, firmly secure a polyvinyl chloride or PVC tube in place. Prepare a perforated weight weighing 550 grams with an 18-millimeter diameter. Attach the weight to a fishing line at a height of one meter inside the PVC tube and adjust the position of the guide tube three centimeters above the tin foil.Next, make a helmet from a stainless steel disc. Prepare a wedge-shaped sponge pillow to place under the rat's head. Swiftly position the anesthetized rat on its chest in the tinfoil.Slide the pillow beneath the rat, ensuring its head remains parallel to the foil surface. Align the helmet with the rat's ears, and securely fasten it in place. Confirm that the PVC pipe is directly above the helmet.Then, release the weight and allow it to impact the rat's head resulting in a fall onto the sponge and a 180 degree rotation. Afterwards, position the rat on its back inside a clean cage.

After inducing a mild traumatic brain injury in an anesthetized rat, position it on its back inside a clean cage. Record the duration from when the rat is anesthetized to when it regains the righting reflex. Also record the time it takes for the rat to exhibit its first seeking behavior from the moment of anesthesia.Next place the rat in the center of a circular apparatus with an exit. Measure and record the time it takes for the rat to exit the circle. Then place the rat successively on three beams of varying length each for a duration of one minute.Score the rat on a scale of zero to six, depending on their performance on the beam. mTBI rats exhibited a significantly prolonged recovery from unconsciousness. The seeking behavior increased significantly in the mTBI group during the recovery period.Also, the mTBI rats took longer to exit the circle compared to sham rats. The beam balance test showed a significant injury effect in wide beam tasks indicating balance impairment in mTBI rats 10 minutes post impact. The 2 and 1.5 centimeter wide beams effectively differentiated between sham and mTBI groups in three independent experiments.

After inducing a mild traumatic brain injury in an anesthetized rat, position it on its back inside a clean cage. Place foam padding beneath the balance beam to reduce the risk of injury to the rats in case of falls during the test. Place an escape box at one end of the beam.Activate the video camera to record the test. Once the animal recovers, position the rats at the other end of the balance beam. Position the rats on the two centimeter wide beam for a series of five consecutive trials.Record the initiation and conclusion of each trial when the rat's nose crosses the starting and finishing lines respectively. Prepare the open field arena, ensuring it is clean and devoid of any prior odor cues. Divide the arena into three distinct zones:a central inner zone, a middle zone, and an outer zone.Place a rat in the center of the open field arena and initiate the timer. Allow the rat to freely explore the arena for a duration of five minutes. After this period, carefully and gently return the rat to its home cage.Quantify the total distance covered by the rat during the five-minute exploration period. Additionally, determine the amount of time the rat spends in the three zones. Ensure that the water maze apparatus is in proper working condition.Place the platform at a depth of 2.5 centimeters below the water surface. Dye the water black and position cues in the four cardinal directions. Establish a monitoring system to record and observe the behavior of the rats.Then introduce the rat into the water maze. If the rat is unable to reach the platform within a two-minute timeframe, gently guide it using a wooden stick. Allow the rat to become accustomed to the maze environment while standing on the platform for a duration of 20 seconds, then remove it.On the sixth day, which is the probe test day, remove the platform from the water maze and position the rat in the same quadrant for a duration of two minutes. Employ the monitoring system to continuously observe and record the rat's behavior. Motor coordination was assessed pre and post-anesthesia or injury using the beam task.At day one post-injury, mTBI rats showed significantly more hindlimb slips compared to shams. However, after two days, hindlimb slips returned to sham levels. All six mTBI rats had more post-impact hindlimb slips with no differences in beam traversal times between mTBI and sham rats.Distance traveled showed no significant differences between sham and mTBI groups. Anxiety-like behavior was evident in mTBI rats at three and seven days post-injury, spending less time in the center zone during the open field test. Morris water maze results indicated impaired spatial learning and memory in mTBI rats.They took longer to find the hidden platform and spent less time searching for the removed platform during the probe trial without differences in swimming speed suggesting no impact on spontaneous locomotor function.
