In the Global Polio Eradication Initiative, laboratory diagnosis plays a critical role by isolating and identifying poliovirus (PV) from the stool samples of patients with acute flaccid paralysis (AFP). In this study, we developed a particle agglutination (PA) method with a soluble human PV receptor (hPVR) in the form of an immunoadhesin (PVR-IgG2a) for the simple and rapid identification of PV. Sensitized gelatin particles with PVR-IgG2a showed specific agglutination with the culture fluid of PV-infected cells within 2 h of reaction in a one-step procedure. Detection limits for type 1, 2, and 3 PV(Sabin) strains were 1.5 x 10(6) 50% cell culture infectious doses (CCID(50)), 5.3 x 10(5) CCID(50), and 9.1 x 10(5) CCID(50), respectively. Wild-type PVs and PV isolates from acute flaccid paralysis cases examined were identified correctly with this PA method, except for some samples with a mixture of different serotypes of PVs, where a minor population of PV failed to be detected. These results suggest that this PA method is useful for the simple and rapid identification of PV, although the sensitivity was not high enough to detect a minor population of PV (<1/10 of the major population) among mixed PVs.

Development of a particle agglutination method with soluble virus receptor for identification of poliovirus.

Dans le Global Polio Eradication Initiative, le diagnostic de laboratoire joue un r&#xF4;le crucial en isolement et l&#x26;#39;identification du poliovirus (PV) &#xE0; partir des &#xE9;chantillons de selles de patients atteints de paralysie flasque aigu&#xEB; (PFA). Dans cette &#xE9;tude, nous avons d&#xE9;velopp&#xE9; une agglutination de particules (PA) m&#xE9;thode avec un r&#xE9;cepteur humain soluble PV (HPVR) sous la forme d&#x26;#39;une immunoadh&#xE9;sine (PVR-IgG2a) pour l&#x26;#39;identification simple et rapide des PV. Sensibilis&#xE9;es particules de g&#xE9;latine avec PVR-IgG2a montr&#xE9; agglutination sp&#xE9;cifique avec le liquide de culture de cellules infect&#xE9;es par le PV au bout de 2 h de r&#xE9;action &#xE0; une proc&#xE9;dure en une seule &#xE9;tape. Les limites de d&#xE9;tection de type 1, 2 et 3 PV (Sabin) souches &#xE9;taient de 1,5 x 10 (6) 50% des doses infectieuses de culture de cellules (CCID (50)), 5,3 x 10 (5) CCID (50), et 9,1 x 10 (5) CCID (50), respectivement. De type sauvage et les isolats PV PV &#xE0; partir de cas de paralysie flasque aigu&#xEB; a examin&#xE9; ont &#xE9;t&#xE9; correctement identifi&#xE9;s par cette m&#xE9;thode PA, &#xE0; l&#x26;#39;exception de quelques &#xE9;chantillons avec un m&#xE9;lange de diff&#xE9;rents s&#xE9;rotypes de PVs, o&#xF9; une population mineure de PV n&#x26;#39;ont pas pu &#xEA;tre d&#xE9;tect&#xE9;. In der Global Polio Eradication Initiative spielt die Labordiagnostik eine entscheidende Rolle von Isolierung und Identifizierung von Polioviren (PV) aus Stuhlproben von Patienten mit akute schlaffe L&auml;hmung (AFP). In dieser Studie entwickelten wir eine Partikel-Agglutination (PA)-Methode mit einem l&ouml;slichen menschlichen PV-Rezeptor (hPVR) in Form eines Immunoadhesin (PVR-IgG2a) f&uuml;r die einfache und schnelle Identifizierung der PV. Sensibilisierte Gelatine-Teilchen mit PVR-IgG2a zeigten spezifische Agglutination mit der Kultur-Fl&uuml;ssigkeit der PV-infizierten Zellen innerhalb von 2 h Reaktion in einem einstufigen Verfahren. Nachweisgrenzen f&uuml;r Typ 1, 2 und 3 PV(Sabin) St&auml;mme wurden 1,5 x 10 6 50 % Zelle Kultur infekti&ouml;se Dosis (CCID(50)), 5,3 x 10 Absatz 5 CCID(50) und 10 Absatz 9.1 X 5 CCID(50), beziehungsweise. Wildtyp PVs und PV isoliert von akute schlaffe L&auml;hmung, die untersuchten F&auml;lle wurden korrekt mit dieser PA-Methode, au&szlig;er f&uuml;r einige Proben mit einer Mischung aus verschiedenen Serotypen des PVs, wo eine kleine Population von PV konnte nicht erkannt werden. Nell&#39;iniziativa di eradicazione globale della Polio, diagnosi di laboratorio svolge un ruolo critico di isolare e identificare il poliovirus (PV) dai campioni di feci di pazienti con acute flaccid paralysis (AFP). In questo studio, abbiamo sviluppato un metodo di agglutinazione (PA) delle particelle con un recettore solubile di PV umano (hPVR) sotto forma di un immunoadhesin (PVR-IgG2a) per l&#39;identificazione rapida e semplice di PV. Particelle di gelatina sensibilizzate con PVR-IgG2a ha mostrato specifica agglutinazione con il liquido di coltura di cellule infettate da PV entro 2 h di reazione in un procedimento a fase unica. En la Iniciativa Mundial de Erradicaci&#xF3;n de la Poliomielitis, el diagn&#xF3;stico de laboratorio desempe&#xF1;a un papel fundamental mediante el aislamiento y la identificaci&#xF3;n de poliovirus (PV) de las muestras de heces de pacientes con par&#xE1;lisis fl&#xE1;ccida aguda (PFA). En este estudio, hemos desarrollado una aglutinaci&#xF3;n de part&#xED;culas (PA) con un receptor soluble humana PV (hPVR) en forma de una inmunoadhesina (PVR-IgG2a) para la identificaci&#xF3;n r&#xE1;pida y sencilla de la energ&#xED;a fotovoltaica. Sensibilizadas part&#xED;culas de gelatina con PVR-IgG2a mostraron aglutinaci&#xF3;n espec&#xED;fica con el fluido de cultivo de c&#xE9;lulas infectadas con PV dentro de 2 h de reacci&#xF3;n en un procedimiento de una etapa. Los l&#xED;mites de detecci&#xF3;n para el tipo 1, 2 y 3 cepas PV (Sabin) fueron de 1,5 x 10 (6) 50% dosis de cultivo de c&#xE9;lulas infecciosas (CCID (50)), 5,3 x 10 (5) CCID (50), y 9,1 x 10 (5) CCID (50), respectivamente. De tipo salvaje PV y FV aislados de casos de par&#xE1;lisis fl&#xE1;ccida aguda examinados fueron correctamente identificados con este m&#xE9;todo PA, a excepci&#xF3;n de algunas muestras con una mezcla de diferentes serotipos de PV, en donde una poblaci&#xF3;n menor de la energ&#xED;a fotovoltaica no pudieron ser detectados. Estos resultados sugieren que este m&#xE9;todo PA es &#xFA;til para la identificaci&#xF3;n simple y r&#xE1;pida de la energ&#xED;a fotovoltaica, aunque la sensibilidad no era lo suficientemente alta como para detectar una poblaci&#xF3;n menor de PV (&#x26;lt;1/10 de la poblaci&#xF3;n mayor) entre PVs mixtos.
