The aim of this research was to identify subunit immunogens that can generate enhanced CD8 T cell and TH1 responses against Mycobacterium tuberculosis. A genomic comparison of the M. tuberculosis H(37)R(V) and M. bovis BCG identified 61 proteins that are unique to H(37)R(V). Further screening of these 61 proteins using in silico analyses mimicking proteasomal digestion, transporter-associated antigen processing and H-2 antigen presentation identified 13 proteins with high densities of predicted MHC class I epitopes. Two native proteins, Rv1986c and Rv3875, were selected on the basis of their secreted or transmembrane characteristics and relatively lower frequencies of predicted MHC class II epitopes. To further enhance the CD8 T cell and TH1 responses, a hybrid protein, H32, was constructed by combining the nucleotide sequences encoding the MHC class I antigen-rich segment of Rv1986c and the entire Rv3875 sequence. The two native proteins and the hybrid were used to immunize C57BL/6 and Balb/c mice, which was followed by pulmonary instillation with irradiated M. tuberculosis H(37)R(V). All three proteins elicited elevated IFN-gamma responses, with the hybrid showing significant increases over the native proteins in both mice. This strategy of immunogen selection might be used to improve the current subunit vaccines against M. tuberculosis as well as other intra-cellular pathogens.

Immunization with hybrid recombinant Mycobacterium tuberculosis H37Rv proteins increases the TH1 cytokine response in mice following a pulmonary instillation of irradiated mycobacteria.

Le but de cette recherche &eacute;tait d&#39;identifier des immunog&egrave;nes de sous-unit&eacute; qui peuvent g&eacute;n&eacute;rer des cellules T CD8 am&eacute;lior&eacute;e et r&eacute;ponses TH1 contre Mycobacterium tuberculosis. Deux prot&eacute;ines indig&egrave;nes, Rv1986c et Rv3875, ont &eacute;t&eacute; choisies pour leurs caract&eacute;ristiques s&eacute;cr&eacute;t&eacute;es ou transmembranaires et relativement basses fr&eacute;quences pr&eacute;dites MHC classe II &eacute;pitopes. Ziel dieser Forschung war Untereinheit Immunogenen zu identifizieren, die verbesserte CD8-T-Zell und TH1 Antworten gegen Mycobacterium Tuberculosis generieren k&ouml;nnen. Die Tuberkulose M. H(37)R(V) und M. Bovis BCG genomischer Vergleich identifiziert 61 Proteine, die nur f&uuml;r H(37)R(V) verwendet werden. Zur weiteren Verbesserung der CD8-T-Zelle und TH1 Antworten wurde ein Hybrid-Protein, H32, errichtet durch die Kombination der Nukleotid Sequenzen, die Codierung der MHC Klasse I Antigen-reiche Segment der Rv1986c und die gesamte Rv3875-Sequenz. Die zwei native Proteine und der Hybrid wurden zur C57BL/6 und dem pulmonalen Instillation mit bestrahlten M. Tuberkulose H(37)R(V) folgte Balb/c M&auml;usen zu immunisieren. Alle drei Proteine, die dadurch hervorgelockt erh&ouml;ht IFN-Gamma Antworten, mit dem Hybrid zeigen deutliche Steigerungen &uuml;ber die native Proteine in beide M&auml;use. Diese Strategie der Immunogen Auswahl kann verwendet werden, um die aktuellen Untereinheit Impfstoffe gegen M. Tuberkulose sowie andere intra-cellular Krankheitserreger zu verbessern.

L&#39;obiettivo della ricerca era identificare immunogeni subunit&agrave; che possono generare una maggiore delle cellule T CD8 e risposte TH1 contro il Mycobacterium tuberculosis. Un confronto genomic di M. tuberculosis H(37)R(V) e M. bovis BCG identificato 61 proteine che sono uniche per H(37)R(V). Ulteriormente lo screening di queste 61 proteine utilizzando in silico analisi imitando proteosomale digestione, elaborazione dell&#39;antigene associato al trasportatore e antigene 13 proteine H-2 presentazione identificato con alta densit&agrave; di classe MHC predetto gli epitopi. Due proteine native, Rv1986c e Rv3875, sono state selezionate in base alle loro caratteristiche secrete o transmembrane e frequenze relativamente basse del predetti MHC classe II epitopi. Per migliorare ulteriormente le cellule T CD8 e le risposte TH1, fu costruita una proteina ibrida, H32, combinando il nucleotide sequenze codifica il MHC classe I segmento di antigene-ricco di Rv1986c e l&#39;intera sequenza di Rv3875. Le due proteine native e l&#39;ibrido sono stati utilizzati per immunizzare C57BL/6 e topi Balb/c, che &egrave; stata seguita da instillazione polmonare con irradiati M. tuberculosis H(37)R(V). Tutte le tre proteine ha suscitate elevate risposte IFN-gamma, con l&#39;ibrido che mostrano un aumento significativo sopra le proteine native in entrambi i topi. Questa strategia di selezione immunogeno potrebbe essere utilizzata per migliorare le attuali vaccini di subunit&agrave; contro M. tuberculosis, come pure altri patogeni intracellulare.

Una comparaci&oacute;n gen&oacute;mica de H(37)R(V) M. tuberculosis y M. bovis BCG hab&iacute;a identificado 61 prote&iacute;nas que son exclusivas de H(37)R(V). Mayor proyecci&oacute;n de estas 61 prote&iacute;nas en an&aacute;lisis de silico m&iacute;mica proteasomal digesti&oacute;n, procesamiento de ant&iacute;geno transportador y H-2 ant&iacute;geno presentaci&oacute;n identificado 13 prote&iacute;nas con altas densidades de predicha MHC de clase I epitopos. Dos prote&iacute;nas nativas, Rv1986c y Rv3875, fueron seleccionadas por sus caracter&iacute;sticas secretadas o transmembrana y frecuencias relativamente bajas de predicha MHC clase II ep&iacute;topos. Para mejorar las c&eacute;lulas T CD8 y respuestas TH1, construy&oacute; una prote&iacute;na h&iacute;brida, H32, combinando el nucle&oacute;tido secuencias de codificaci&oacute;n del MHC de clase I rico ant&iacute;geno de Rv1986c y toda la secuencia de Rv3875. A close interconnection between nutrient metabolism and virulence factor expression contributes to the pathophysiology of Pseudomonas aeruginosa as a successful pathogen. P. aeruginosa fatty acid (FA) degradation is complicated with multiple acyl-CoA synthetase homologs (FadDs) expressed in vivo in lung tissue during cystic fibrosis infections. The promoters of two genetically linked P. aeruginosa fadD genes (fadD1 and fadD2) were mapped and northern blot analysis indicated they could exist on two different transcripts. These FadDs contain ATP/AMP signature and FA-binding motifs highly homologous to those of the Escherichia coli FadD. Upon introduction into an E. coli fadD(-)/fadR(-) double mutant, both P. aeruginosa fadDs functionally complemented the E. coli fadD(-)/fadR(-) mutant, allowing degradation of different chain-length FAs. Chromosomal mutagenesis, growth analysis, induction studies, and determination of kinetic parameters suggested that FadD1 has a substrate preference for long-chain FAs while FadD2 prefers shorter-chain FAs. When compared to the wild type strain, the fadD2 mutant exhibited decreased production of lipase, protease, rhamnolipid and phospholipase, and retardation of both swimming and swarming motilities. Interestingly, fadD1 mutant showed only increased swarming motility. Growth analysis of the fadD mutants showed noticeable deficiencies in utilizing FAs and phosphatidylcholine (major components of lung surfactant) as the sole carbon source. This defect translated into decreased in vivo fitness of P. aeruginosa in a BALB/c mouse lung infection model, supporting the role of lipids as a significant nutrient source for this bacterium in vivo.

Ein enger Verbund zwischen N&auml;hrstoff-Stoffwechsel und Virulenz Faktor Ausdruck tr&auml;gt zur Pathophysiologie von Pseudomonas Aeruginosa als ein erfolgreicher Erreger. P. Aeruginosa Fetts&auml;uren (FA) Abbau ist mit mehreren-Acyl-CoA Synthetase UCP1 (FadDs) ausgedr&uuml;ckt in Vivo in Lungengewebe bei Mukoviszidose Infektionen kompliziert. Die Promotoren der zwei genetisch verkn&uuml;pften p. Aeruginosa Triathlonmeisterschaft Genen (fadD1 und fadD2) wurden zugeordnet und Nordfleck Untersuchung ergab, dass sie auf zwei verschiedene Transkripte existieren k&ouml;nnte. Diese FadDs enthalten ATP/AMP-Signatur und FA-Bindung Motive hoch Homolog zu denen der Escherichia coli Triathlonmeisterschaft. Bei der Einf&uuml;hrung in ein E. Coli fadD(-)/fadR(-) Doppel-Mutant erg&auml;nzt beide p. Aeruginosa FadDs funktional die E. Coli fadD(-)/fadR(-) Mutant, wodurch Abbau von unterschiedlichen Kettenl&auml;nge FAs. Ermittlung der kinetischen Parameter chromosomalen Mutagenese, Wachstum-Analyse und Induktion Studien vorgeschlagen, dass FadD1 eine Substrat-Vorliebe f&uuml;r langkettige FAs, hat w&auml;hrend FadD2 k&uuml;rzere langkettige FAs bevorzugt. Im Vergleich zu der Wildtyp-Stamm zur&uuml;ckgegangen der fadD2-Mutant ausgestellt Produktion von Lipase, Protease, Rhamnolipid und Phospholipase und Retardierung sowohl Schwimmen und Schw&auml;rmen von Motilities. Interessanterweise erh&ouml;ht fadD1 Mutant zeigte nur schw&auml;rmen Motilit&auml;t. Wachstum Triathlonmeisterschaft Mutanten zeigte deutliche M&auml;ngel bei der Nutzung von FAs und Phosphatidylcholin (Hauptkomponenten der Lunge Tensid) als einziger Kohlenstoffquelle. Dieser Fehler in verringerter in-vivo-Tauglichkeit von p. Aeruginosa in einem BALB/c Maus Lunge Infektion Modell, unterst&uuml;tzt die Rolle von Lipiden als eine bedeutende N&auml;hrstoffquelle f&uuml;r dieses Bakterium in-vivo &uuml;bersetzt.

Mehrere Triathlonmeisterschaft Coenzym Synthetases tragen zur differenziellen Fetts&auml;ure Abbau- und Virulenz in Pseudomonas Aeruginosa.

Una stretta interconnessione tra il metabolismo dei nutrienti ed espressione di fattore di virulenza contribuisce alla patofisiologia di Pseudomonas aeruginosa come un agente patogeno di successo. Degradazione degli acidi grassi (FA) di p. aeruginosa &egrave; complicato con multipli di acil-CoA sintetasi omologhi (FadDs) espresse in vivo nel tessuto polmonare durante le infezioni di fibrosi cistica. I promotori di due geni fadD geneticamente collegato p. aeruginosa (fadD1 e fadD2) sono stati mappati e northern blot analisi indicato che potrebbe esistere su due diverse trascrizioni. Questi FadDs contengono ATP/AMP firma e motivi FA altamente omologhi a quelli di Escherichia coli FadD. Introduzione in un mutante doppio di e. coli fadD(-)/fadR(-), entrambi fadDs di p. aeruginosa completato funzionalmente il mutante di fadD(-)/fadR(-) e. coli, permettendo la degradazione di diversa lunghezza catena FAs. Mutagenesi cromosomica, analisi di crescita, studi di induzione e determinazione dei parametri cinetici suggerito che FadD1 ha una preferenza di substrato per catena lunga FAs mentre FadD2 preferisce pi&ugrave; corta catena FAs. Rispetto al ceppo wild type, il mutante fadD2 esposto diminuita produzione di lipasi, proteasi, rhamnolipid e fosfolipasi e il ritardo di nuoto e brulicante di motilities. &Egrave; interessante notare che, fadD1 mutante ha mostrato solo aumentata motilit&agrave; brulicante. Analisi della crescita dei mutanti fadD ha mostrato evidenti carenze nell&#39;utilizzazione FAs e fosfatidilcolina (principali componenti del surfattante polmonare) come l&#39;unica fonte di carbonio. Questo difetto &egrave; tradotto in diminuzione fitness in vivo di p. aeruginosa in un BALB/c polmone infezione modello murino, sostenendo il ruolo dei lipidi come fonte di nutrienti importante per questo batterio in vivo.

Degradaci&oacute;n de &aacute;cidos grasos (FA) de p. aeruginosa es complicada con varios hom&oacute;logos de la sintetasa acil-CoA (FadDs) expresadas en vivo en tejido pulmonar en la fibrosis qu&iacute;stica infecciones. Los promotores de dos genes de fadD vinculado gen&eacute;ticamente p. aeruginosa (fadD1 y fadD2) fueron mapeados y an&aacute;lisis de northern blot indic&oacute; que podr&iacute;an existir en dos expedientes distintos. Mutag&eacute;nesis cromos&oacute;mica, an&aacute;lisis de crecimiento, estudios de inducci&oacute;n y determinaci&oacute;n de par&aacute;metros cin&eacute;ticos sugirieron que FadD1 tiene una preferencia del substrato por FAs de cadena larga, mientras que FadD2 prefiere FAs de cadena m&aacute;s corta. En comparaci&oacute;n con la cepa de tipo salvaje, el mutante fadD2 exhibido disminuida producci&oacute;n de proteasa, lipasa y fosfolipasa, rhamnolipid y el retraso de nadar y enjambre de motilities. Curiosamente, fadD1 mutante mostr&oacute; s&oacute;lo aument&oacute; pululaban motilidad.
