This present work describes the scalable synthetic methods of oxidation and reduction reactions for producing graphene oxide and reduced graphene oxide nanomaterials. Cascade design of Mn(VII)-based oxidation reaction is applied to convert graphite into multilayer graphite oxide powder, the precursor of single-layer graphene oxide nanosheets. Since exothermic heats, relevant chemical ratios, and combinative strategy are utilized, the cascade design process saves considerable amounts of heating energy, chemical reagents, and synthetic time. In the next stage, the hydrated multilayer structure of graphite oxide is exfoliated in water with the support of sonication. Graphene oxide nanosheets are electrostatically stabilized and chemically reduced using highly basic ammonia solution at pH &#62;11&#160;and a temperature of 90 &#176;C. Alkaline ammonium hydroxide solution is considered an environmentally friendly and inexpensive chemical agent for synthesizing reduced graphene oxide nanosheets assembled in a hydrogel structure. Hydrated reduced graphene oxide nanosheets in the supramolecular hydrogel can be ultrasonically exfoliated to produce homogeneous aqueous dispersions. The general protocol of oxidation-reduction reactions sequentially synthesizes graphite oxide powder, graphene oxide nanosheet, reduced graphene oxide hydrogel, and reduced graphene oxide nanosheet for various scientific research and multidisciplinary applications. Prospective development of the synthetic approach from laboratory scale to industrial production is envisioned to elaborate on the potential.

Analytical methods can range from detailed regulatory documents to simpler summaries. Regulatory methods may include information on amenable analytes, supported matrices, required reagents, statistical performance, interlaboratory validation, and other specifics. Summaries typically provide a general overview of reagents, instrumentation, and often a short list of analytes. Analytical methods from U.S. government bodies, including the U.S. Environmental Protection Agency (USEPA), U.S. Geological Survey (USGS), U.S. Department of Agriculture (USDA), the Food and Drug Administration (FDA), and others, offer detailed procedural information. Instrument vendors such as Agilent, Shimadzu, Thermo Fisher Scientific, Sciex, and others also provide access to hundreds of application notes, which may be considered summary methods. This study has developed a cheminformatics-enabled database of methods in which chemicals are extracted from method documents, with identifiers (names and/or Chemical Abstracts Service registry numbers (CASRN)) mapped to chemical structures. The resulting database, containing approximately 7,000 methods, is searchable by identifier, chemical structure, and structural similarity, and is supplemented by approximately one million public domain spectra (LC/MS, GC/MS, NMR, and IR). The application supports searching of analytical methods and filtering based on analytes, functional usage, method sources, and other related metadata.

Pot marigold (Calendula officinalis L.) with multiple uses, notably its medicinal applications due to its richness and diversity in secondary metabolites, such as flavonoids, which possess anti-inflammatory and antioxidant properties.This study analyzed plant growth and biomass distribution, as well as total flavonoid concentration in tubular, ligulate flowers, and flower heads throughout floral development.&#160;Floral initiation and differentiation occurred five weeks after transplantation, following a sigmoid pattern of biomass accumulation in its organs as well as in plant height. Aerial organs reached a maximum biomass of 76 g/ g/plant on day 69. The maximum absolute growth rate was 2.38 g/day at 62 days, while the highest relative growth rate was recorded in reproductive organs on day 48 (0.137 g/g/day). Flavonoids, expressed as quercetin equivalents, showed higher concentrations during stages ranging from flower buds to fully opened flower heads (145-177 mg/g of dry matter). These findings emphasize the importance of harvesting at optimal stages to maximize flavonoid content and leverage their potential for therapeutic and commercial applications.

A fast, efficient method has been developed on the GAIA synthesis module for automated gallium-68 radiolabeling of 3BP-3940, a molecular imaging probe targeting the fibroblast activation protein for positron emission tomography imaging of the tumor microenvironment. The reaction conditions involved acetate buffer (final concentration: 0.1 M), methionine as an anti-radiolysis agent (final concentration: 5.4 mg/mL), and 30 &#181;g of 3BP-3940, with heating for 8 min at 98 &#176;C. A final purification step on a C18 cartridge was necessary to obtain a radiolabeled product of high purity. In contrast, the generator-produced 68Ga was used directly without a concentration step on a cation exchange cartridge. The production of three validation batches confirmed the method&#39;s reliability, allowing the synthesis of [68Ga]Ga-3BP-3940 in 22.3 &#177; 0.6 min with high radiochemical purity (RCP), as determined by both radio-HPLC (99.1% &#177; 0.1%) and radio-TLC (99.2% &#177; 0.1%). The average radiochemical yield, based on RCP values measured by radio-HPLC, was 74.4% &#177; 3.3%. The stability of the radiolabeled product was demonstrated for up to 4 h after preparation. This protocol provides a reliable, rapid, and efficient methodology for the preparation of [68Ga]Ga-3BP-3940, which can easily be transposed to a clinical setting.

The increasing incidence of HIV-1 drug resistance presents a challenge to the effectiveness of combination antiretroviral therapy, particularly in Southern Africa. The development of resistance to non-nucleotide reverse transcriptase inhibitors (NNRTIs) threatens the long-term success of antiretroviral therapy. In 2019, antimicrobial resistance directly accounted for an estimated 1.27 million deaths globally. This study employed an in-silico approach to investigate NNTRI drugs and their derivatives. Techniques used included density functional theory calculations, molecular docking, enumeration, quantitative structure-activity relationship (QSAR) analysis, molecular dynamics simulation (MDS), and molecular mechanics with generalized Born and surface area methods. The analysis focused on various pyrimidine derivatives and six NNRTI drugs, examining their interactions with the HIV-1 protein (PDB code 1HQU).
A QSAR model was developed to predict the biological activity of the six NNRTIs under investigation. Using 94 pyrimidine derivatives, the QSAR model achieved an R2 OF 0.822 and a Q2 of 0.815, indicating a high level of predictive accuracy.
MDS were conducted to assess the stability of various ligands and their newly developed alternatives, ensuring they remained bound to the protein&#39;s active site over a 200-nanosecond simulation timeframe. Etravirine exhibited root mean square deviation (RMSD) fluctuations of approximately 4.5 &#197;, while its enumerated derivatives showed RMSD fluctuations of 3.5 &#197;. Through molecular docking, MDS, and free energy computations, enumerated Etravirine demonstrated the best performance, with an activity value of 7.373 and a docking score of -10.517 kcal/mol. Furthermore, the calculated free energy of binding for enumerated Etravirine was -89.684 kcal/mol, outperforming other ligands under investigation. The significant improvement suggests that the modified Etravirine holds promising potential as a novel agent in antiretroviral therapy.
The obtained lower RMSD value, the enhanced amino acid interactions, and the highest binding free energy indicate that enumerated Etravirine could serve as a viable alternative for HIV/AIDS treatment.

This study investigates the preparation of polyacrylonitrile (PAN) nanofibers through electrospinning to create highly porous and strong materials for applications in water purification, electrocatalysis, and biomedicine. The uniformly white PAN nanofiber mats were cut into 2 cm x 2 cm coupons to ensure consistency. After electrospinning, these nanofibers were coated with an electroactive polymer (EAP) using chemical vapor deposition, with iron (III) chloride (FeCl3) serving as an oxidant for polymerizing 3,4-ethylenedioxythiophene (EDOT) into poly(3,4-ethylenedioxythiophene) (PEDOT). The study examined the impact of different FeCl3 concentrations on PEDOT deposition on the PAN coupons. PEDOT deposition led to an increase in coupon weight. Scanning electron microscopy (SEM) revealed increases in the diameter of the nanofibers treated with increasing FeCl3 oxidant concentration, although higher FeCl3&#160;concentrations caused inter-fiber bridging, implying a concomitant decrease in inter-fiber spacing. Energy dispersive X-ray spectroscopy (EDS) was used to confirm the presence of Fe, Cl, and S in the nanofibers, with sulfur content rising with FeCl3&#160;concentration used, suggesting increased PEDOT deposition efficiency with increasing oxidant concentration. Mechanical testing showed that PEDOT-coated PAN fibers had improved tensile strength and toughness in the hydrated state compared to pure PAN nanofibers. These results highlight the crucial role of FeCl3&#160;concentration in influencing the morphology and properties of PAN-PEDOT composites, enhancing their suitability for applications such as water purification, tissue engineering, biosensing, catalysis, and energy storage.

Fluid-cell Raman Spectroscopy (FCRS) enables the real-time and space-resolved (operando) study of reaction mechanisms, kinetics, and their mutual interactions with transport processes during silicate glass corrosion at the micrometer scale and at elevated temperatures. This manuscript provides a detailed protocol for setting up an FCRS experiment, exemplified by a corrosion experiment with a ternary Na borosilicate glass and a 0.5 M NaHCO3 solution at a temperature of 86 &#177; 1 &#176;C. The protocol involves (i) sample preparation, (ii) assembly of the fluid cell, and (iii) setting of Raman measurement parameters for collecting Raman spectra across the sample/solution interface in regular time intervals. The results from the experiment show the formation of a water-rich zone between a silica-based surface alteration layer (SAL) and the pristine glass, which is an intrinsic feature of an interface-coupled dissolution-precipitation model for the formation of a SAL during silicate glass corrosion. The ability to track the reaction and transport processes during the corrosion of silicate glasses and potentially of other transparent materials, spatially resolved and in real-time, represents a unique strength of this technique, overcoming the disadvantages of conventional analysis of multi-step quenching experiments. The corrosion of the top side of the glass sample represents a current issue, reducing spatial resolution at depth due to precipitation within the laser pathway. This is caused by a solution-filled gap between the sapphire window of the fluid cell lid and the top side of the monolith, which is difficult to avoid during the experimental setup. This must be taken into account when choosing the depth at which the measurement should be made. In a few cases, the formation of air bubbles was observed, which disrupted or even led to the termination of the experiment. However, this can be avoided by carefully setting up the experiment, which requires little practice.

This paper presents a robot-based experimental program aimed at developing an efficient and fast colorimetric gas sensor. The program employs an automated Design-Build-Test-learning (DBTL) approach, which optimizes the search process iteratively while optimizing multiple recipes for different concentration intervals of the gas. In each iteration, the algorithm generates a batch of recipe suggestions based on various acquisition functions, and with the increase in the number of iterations, the values of weighted objective function for each concentration interval significantly improve.
The DBTL method begins with parameter initialization, setting up the hardware and software environment. Baseline tests establish performance standards. Subsequently, the DBTL method designs the following round of optimization based on the proportion of recipes in each round and tests performance iteratively. Performance evaluation compares baseline data to assess the effectiveness of the DBTL method. If the performance improvement does not meet expectations, the method will be performed iteratively; if the objectives are achieved, the experiment concludes. The entire process maximizes system performance through the DBTL iterative optimization process.
Compared to the traditional manual developing process, the DBTL method adopted by this experimental process uses multi-objective optimization and various machine learning algorithms. After defining the upper and lower limits of component volume, the DBTL method dynamically optimizes iterative experiments to obtain the optimal ratio with the best performance. This method greatly improves efficiency, reduces costs, and performs more efficiently within the multi-formulation variable space when finding the optimal recipe.

Efficient Synthesis of Carbon-11 Radiotracers Using the Loop Method Methylation

A successful positron emission tomography imaging program involving carbon-11 radiotracers demands fast, efficient, and reliable synthesis methods, requiring an on-site cyclotron and radiochemistry group, as well as clinical staff trained to operate under the unique constraints of the carbon-11 radionuclide. This study examines the merits and advantages of a captive solvent &#39;loop method&#39; of radiolabeling four tracers with the carbon-11 radionuclide, producing the radioligands [11C]ER-176, [11C]MRB, [11C]mHED, and [11C]PiB.
The &#39;loop method&#39; is compared against the traditional reactor-based method of carbon-11 methylation in the course of synthesizing the same radiotracers on the identical automated platform. Further, a complete overview of the clinical research preparation of the [11C]ER-176 radiotracer is presented. As demonstrated by the production of [11C]ER-176, the captive solvent &#39;loop method&#39; of heterogeneous alkylation proved to be more efficient, with excellent radiochemical purity (99.6 &#177; 0.6%, n = 25), higher and more consistent radiochemical yield (end of synthesis (EOS) = 5.4 &#177; 2.2 GBq, n = 25) compared to the reactor method (EOS = 1.6 &#177; 0.5 GBq, n = 6), increased molar activity (loop method = 194 &#177; 66 GBq/&#181;mol, n = 25; reactor method = 132 &#177; 78 GBq/&#181;mol, n = 6), along with an average 5 min shorter reaction sequence.

Fusidic Acid-Loaded Hydrogel Films with Aloe vera for Enhanced Drug Delivery

A new fusidic acid-loaded hydrogel film was prepared via the solvent casting technique using alginate and Aloe vera. The hydrogel films were optimized using different ratios of sodium alginate, Aloe vera, and glycerin. The films containing 10% glycerin (w/w of alginate) exhibited the best appearance. Incorporating Aloe vera influenced the thickness, swelling behavior, water vapor permeability, and drug release profile of the hydrogel films. Higher Aloe vera content resulted in thicker films (up to a certain ratio), increased swelling, reduced water vapor permeability, and a prolonged drug release of up to 93% over 12 h. Fourier-transform infrared spectroscopy (FTIR) analysis confirmed the presence of key functional groups and the interaction between the hydrogel components. The study suggests that the combination of sodium alginate, Aloe vera, and glycerin can improve the mechanical properties and drug release profiles of hydrogel films, making them a promising option for enhanced topical drug delivery and wound healing applications.
