Nous avons développé un modèle ayant une pertinence clinique pleinement opérationnel de la DGPC dans une souris. Avec plus d’une trentaine de souches de souris ayant des maladies cardiovasculaires, notre modèle pourrait être un point de départ pour le développement de nouveaux protocoles éventuels associés au doryphore. En outre, en raison de la pléthore des réactifs spécifiques de souris et souris knockout-out, ce modèle ne peut pas seulement remplacer l’actuel modèle de rat de CPB mais facilitera la ...

Depuis l’introduction d’extra-corporelle (CEC) dans la clinique, il a joué un rôle essentiel dans la chirurgie cardiaque1. En chirurgie cardiaque moderne, prolongée de la DGPC est indispensable pour effectuer des reconstructions aortiques et procédures combinées. Bien que les progrès technologiques ont été énormes, l’utilisation de la circulation extracorporelle est associée avec intra - et postopératoires systémique et endommager les organes locaux2,3.
Grands modèles animaux ont été développés pour étudier le rôle de la DGPC sur les processus physiologiques4,5. Bien que ces modèles ont fourni à l’aperçu de certains de la DGPC associés à des complications, ils sont extrêmement coûteux et outils moléculaires (p. ex. anticorps) sont très limitées. Une alternative plus rentable a été développée chez de petits animaux. Depuis leur mise au point, plusieurs études ont été menées afin d’optimiser un modèle de la DGPC aux rats et lapins5,6,7,8,9. Ces modèles fournissent une bonne base pour des mesures des processus physiopathologique de la maladie ; Cependant, ils sont encore insuffisants pour étudier l’immunologie cellulaire et humorale en raison de l’absence d’anticorps compétents et réactifs. Cela porte atteinte à leur rôle dans ce domaine de recherche.
Nous avons récemment développé un modèle murin de la DGPC. En raison d’une grande variété de réactifs spécifiques de souris et souris génétiquement modifiés, les modèles murins sont en général le modèle de choix pour les recherches physiologiques, moléculaires et immunologiques10,11. Par conséquent, notre modèle facilitera l’étude du CPB en ce qui concerne les diverses maladies concomitantes comme il existe de nombreuses souches de souris disponibles avec des maladies ayant une pertinence clinique12,13. En conséquence, le présent document explique, en détail, comment effectuer CPB chez la souris. Oxygène et paramètres hémodynamiques sont surveillés étroitement après l’arrestation de profondes respiratoire et circulatoire.

cardiopulmonary bypass in a mouse model: a novel approach

Comme extracorporelle prolongée devient plus essentiel pendant les interventions cardiaques, une demande croissante de clinique se pose pour l’optimisation de la procédure et pour minimiser les lésions organiques résultant de la circulation extracorporelle prolongée. L’objectif de cette étude était de démontrer un modèle entièrement fonctionnel et cliniquement pertinent de la circulation extracorporelle chez une souris. Nous rapportons sur la conception de l’appareil, optimisation de circuit de perfusion et techniques microchirurgicales. Ce modèle est un modèle aiguë, ce qui n’est pas compatible avec la survie en raison de la nécessité de multiples dessins de sang. En raison de l’éventail d’outils disponibles pour la souris (par exemple, marqueurs, Ejecteurs, etc.), ce modèle facilitera l’enquête sur les mécanismes moléculaires de lésions organiques et l’effet de la circulation extracorporelle par rapport aux autres comorbidités.

Ce protocole décrit le circuit de perfusion, les interventions chirurgicales et la surveillance des paramètres physiologiques au cours de la DGPC de souris. Lorsque exécutée par une microsurgeon adéquatement qualifié, on obtient les résultats régulièrement et de façon reproductible.
