Treatment options for androgen-independent prostate cancer cells are limited. Therefore, it is critical to identify agents that induce death of both androgen-responsive and androgen-insensitive cells. Here we demonstrate that a product of plant cell walls, pectin, is capable of inducing apoptosis in androgen-responsive (LNCaP) and androgen-independent (LNCaP C4-2) human prostate cancer cells. Commercially available fractionated pectin powder (FPP) induced apoptosis (approximately 40-fold above non-treated cells) in both cell lines as determined by the Apoptosense assay and activation of caspase-3 and its substrate, poly(ADP-ribose) polymerase. Conversely, citrus pectin (CP) and the pH-modified CP, PectaSol, had little or no apoptotic activity. Glycosyl residue composition and linkage analyses revealed no significant differences among the pectins. Mild base treatment to remove ester linkages destroyed FPP's apoptotic activity and yielded homogalacturonan (HG) oligosaccharides. The treatment of FPP with pectinmethylesterase to remove galacturonosyl carboxymethylesters and/or with endopolygalacturonase to cleave nonmethylesterified HG caused no major reduction in apoptotic activity, implicating the requirement for a base-sensitive linkage other than the carboxymethylester. Heat treatment of CP (HTCP) led to the induction of significant levels of apoptosis comparable to FPP, suggesting a means for generating apoptotic pectic structures. These results indicate that specific structural elements within pectin are responsible for the apoptotic activity, and that this structure can be generated, or enriched for, by heat treatment of CP. These findings provide the foundation for mechanistic studies of pectin apoptotic activity and a basis for the development of pectin-based pharmaceuticals, nutraceuticals, or recommended diet changes aimed at combating prostate cancer occurrence and progression.

Por lo tanto, es fundamental para identificar a agentes que inducen la muerte de las c&eacute;lulas sensibles al andr&oacute;geno tanto andr&oacute;geno-insensibles. Aqu&iacute; demostramos que un producto de las paredes celulares, pectina, es capaz de inducir apoptosis en andr&oacute;geno-sensible (LNCaP) y c&eacute;lulas de c&aacute;ncer de pr&oacute;stata humano andr&oacute;geno-independiente (LNCaP C4-2). Polvo de pectina fraccionada comercialmente disponibles (FPP) indujo apoptosis (aproximadamente 40-fold anteriores no tratadas c&eacute;lulas) en ambas l&iacute;neas celulares seg&uacute;n lo determinado por el an&aacute;lisis de Apoptosense y activaci&oacute;n de la caspasa-3 y su sustrato, ribosa polimerasa. Por el contrario, pectina c&iacute;trica (CP) y el CP pH-modificado, PectaSol, ten&iacute;a poca o ninguna actividad apopt&oacute;tica. An&aacute;lisis de composici&oacute;n y acoplamiento de residuo glycosyl revelaron diferencias significativas entre las pectinas. Tratamiento de base suave para quitar v&iacute;nculos &eacute;ster hab&iacute;a destruido actividad apopt&oacute;tica FPP y rindi&oacute; homogalacturonan oligosac&aacute;ridos (HG). El tratamiento de la FPP con pectinmethylesterase para eliminar galacturonosyl carboxymethylesters y/o endopolygalacturonase de allegarse nonmethylesterified HG no caus&oacute; ninguna reducci&oacute;n en actividad apopt&oacute;tica, implicando la exigencia de un v&iacute;nculo sensible a la base que no sea el carboxymethylester. Tratamiento t&eacute;rmico de CP (HTCP) condujo a la inducci&oacute;n de niveles significativos de apoptosis comparable a la FPP, sugiriendo un medio para generar estructuras p&eacute;cticas apopt&oacute;tica. Photosystem II (PSII) is the membrane protein complex that catalyzes the photo-induced oxidation of water at a manganese-calcium active site. Light-dependent damage and repair occur in PSII under conditions of high light stress. The core reaction center complex is composed of the D1, D2, CP43, and CP47 intrinsic polypeptides. In this study, a new chromophore formed from the oxidative post-translational modification of tryptophan is identified in the CP43 subunit. Tandem mass spectrometry peptide sequencing is consistent with the oxidation of the CP43 tryptophan side chain, Trp-365, to produce N-formylkynurenine (NFK). Characterization with ultraviolet visible absorption and ultraviolet resonance Raman spectroscopy supports this assignment. An optical assay suggests that the yield of NFK increases 2-fold (2.2 Â± 0.5) under high light illumination. A concomitant 2.4 Â± 0.5-fold decrease is observed in the steady-state rate of oxygen evolution under the high light conditions. NFK is the product formed from reaction of tryptophan with singlet oxygen, which can be produced under high light stress in PSII. Reactive oxygen species reactions lead to oxidative damage of the reaction center, D1 protein turnover, and inhibition of electron transfer. Our results are consistent with a role for the CP43 NFK modification in photoinhibition.

Photosystem II (PSII) ist die Membran Proteinkomplex, die Foto-induzierte Oxidation von Wasser an einem Mangan-Calcium-aktiv-Standort katalysiert. Licht-abh&auml;ngige-Sch&auml;digung und Reparatur auftreten in PSII unter Bedingungen der hohen leichte Belastungen. Der Kern Reaktion Mitte Komplex besteht aus der D1, D2, CP43 und CP47 systeminterne Polypeptide. In dieser Studie wird eine neue Chromophor bildete der oxidativen POSTTRANSLATIONELLE Ver&auml;nderung von Tryptophan in der CP43-Untereinheit identifiziert. Tandem-Massenspektrometrie-Peptid-Sequenzierung steht im Einklang mit der Oxidation der CP43-Tryptophan-Seitenkette, Trp-365, N-Formylkynurenine (NFK) zu produzieren. Charakterisierung mit Ultraviolett sichtbare Absorption und UV-Resonanz-Raman-Spektroskopie unterst&uuml;tzt diese Zuordnung. Ein optischer Assay legt nahe, dass der Ertrag von NFK 2-fach erh&ouml;ht (2,2 &plusmn; 0,5) unter hohen Licht Beleuchtung. Eine damit einhergehende 2,4 &plusmn; 0.5-fold Abnahme wird in der station&auml;ren bei der Sauerstoff-Evolution unter den hohen Lichtverh&auml;ltnissen beobachtet. NFK ist das Produkt gebildet durch Reaktion von Tryptophan mit Singulett-Sauerstoff, die unter hohen Licht Stress in PSII produziert werden k&ouml;nnen. Reaktive Sauerstoff Spezies Reaktionen f&uuml;hren zu oxidativen Sch&auml;den der Reaktion Mitte, D1-Protein-Umsatz und Hemmung der Elektronentransfer. Unsere Ergebnisse sind konsistent mit eine Rolle f&uuml;r die CP43 NFK-&Auml;nderung in Photoinhibition.

Riparazione e danno luce-dipendente si verificano ISOFORMS in condizioni di elevato stress leggero. Il complesso del centro di reazione core &egrave; composto dei polipeptidi intrinseche D1, D2, CP43 e CP47. In questo studio, un nuovo cromoforo formato dalla modificazione ossidativa alberino-di traduzione di triptofano &egrave; identificato la subunit&agrave; CP43. Sequenziamento di peptide spettrometria di massa tandem &egrave; coerenza con l&#39;ossidazione della catena laterale CP43 triptofano, Trp-365, per produrre N-formylkynurenine (NFK). Caratterizzazione con assorbimento ultravioletto visibile e ultravioletta risonanza spettroscopia Raman supporta questa assegnazione. Un test ottico suggerisce che il rendimento del NFK aumenta 2 volte (2.2 &plusmn; 0,5) sotto illuminazione luce alta. Fotosistema II (PSII) es el complejo de prote&iacute;na de membrana que cataliza la oxidaci&oacute;n inducida por foto de agua en un sitio activo de manganeso-calcio. Reparaci&oacute;n y da&ntilde;os dependientes de la luz ocurren en PSII bajo condiciones de alto estr&eacute;s ligero. El complejo de centro de reacci&oacute;n de n&uacute;cleo se compone de los polip&eacute;ptidos intr&iacute;nsecos D1, D2, CP43 y CP47. En este estudio, se identifica un nuevo crom&oacute;foro formado a partir de la modificaci&oacute;n poste-de translaci&oacute;n oxidativa de tript&oacute;fano en la subunidad CP43. Secuenciaci&oacute;n de p&eacute;ptidos de espectrometr&iacute;a de masas en t&aacute;ndem es consistente con la oxidaci&oacute;n de la cadena lateral del tript&oacute;fano CP43, Trp-365, para producir N-formylkynurenine (NFK). Caracterizaci&oacute;n con absorci&oacute;n ultravioleta visible y ULTRAVIOLETA resonancia espectroscop&iacute;a Raman apoya esta asignaci&oacute;n. Un an&aacute;lisis &oacute;ptico sugiere que el rendimiento de NFK aumenta 2 (2.2 &plusmn; 0.5) bajo iluminaci&oacute;n luz alta. Un &plusmn; 2.4 concomitante disminuci&oacute;n 0.5-fold se observa en la tasa de estado estacionario de la evoluci&oacute;n de ox&iacute;geno en las condiciones de luz altas. Nuestros resultados son consistentes con un papel para la modificaci&oacute;n de CP43 NFK en fotoinhibici&oacute;n.
