ES-Zellen haben das Potenzial, in die Zellen aus aller Keimblätter, die sie zu einem attraktiven Werkzeug für die Entwicklung neuer Therapien ist zu differenzieren. In der Regel folgt die Differenzierung von ES-Zellen das Konzept zum ersten generieren unreifen Vorläuferzellen, die dann vermehrt und differenziert zu reifen zellulären Phänotypen können. Dies gilt auch für ES-Zellen abgeleiteten Neurogenese, in denen die Entwicklung von neuralen Zellen folgt zwei wichtige Schritte: Erstens, die Gewinnung und Erweiterung von unreifen Vorstufen neuroepithelial und zweitens, deren Differenzierung zu reifen Nervenzellen. Eine gängige Methode, um neuronale Vorläuferzellen aus ES-Zellen produzieren auf Embryoidkörper (EB)-Bildung, die die Differenzierung von Zellen aus aller Keimblätter einschließlich Neuroektoderms verrät basiert. Eine alternative und effizientere Methode, um neuro-Zell-Entwicklung zu induzieren verwendet Stroma-Zellen abgeleiteten induzierende Aktivität (SDIA), die durch Co-Kultivierung ES-Zellen mit Totenkopf aus Knochenmark stammende Stromazellen (1) erreicht werden kann. Beide, EB Bildung und SDIA zeigen die Entwicklung der Rosette-ähnliche Strukturen, die vermutlich neuronale ähneln Rohr-und / oder Neuralleiste-like Vorfahren. Die neuralen Vorläuferzellen isoliert werden ausgebaut und weiter differenziert in bestimmten Neuronen und Gliazellen mit definierten Kulturbedingungen. Hier beschreiben wir die Erzeugung und Isolierung solcher Rosetten in Co-Kultur Experimente mit der stromalen Zelllinie MS5 (2-5).
