Die Fähigkeit von menschlichen embryonalen Stammzellen (hESC) in verschiedenen Zelltypen zu differenzieren, eine neue Ära der In-vitro-Toxizitätstests 1, Krankheitsmodelle und der regenerativen Medizin 2 eröffnet. Die Stammzellen werden mit der Fähigkeit, selbst zu replizieren, um ihre pluripotenten Zustand zu halten und sich in spezialisierte Zellen 3,4 unterscheiden dotiert. Die Eigenschaften der hESC (Fähigkeit, zu allen wichtigen Zelltypen differenzieren) werden auch in anderen menschlichen pluripotenten Stammzellen, wie Menschen induzierte pluripotente Stammzellen (hiPSC) oder Zellen, die durch Kerntransfer erzeugt 5 gefunden. Zum Beispiel haben viele verschiedene Linien hESC in Neuronen 6, Nierenzellen 7, Neuralleistenzellen 8, Kardiomyozyten 9-12 oder Leberzellen wie Zellen 13,14 differenziert. Darüber hinaus kann hESC spontan in Zellen aller drei Keimblätter 15-18 in Embryoidkörpern (EBS) 19,20 unterscheiden. Early embryonalen Entwicklung durch unterschiedliche Expression verschiedener Gene zu den verschiedenen Keimblätter, die auf mRNA-Ebene durch Transkriptomik mit Microarray-Technologie 15 aufgenommen wurde, im Zusammenhang geregelt. Diese Bemühungen führten zu der Gründung der Organspezifische toxikologische Modelle auf Basis von hESC / hiPSC und Transkriptom-Analyse (für einen Überblick siehe 21,22). Diese Modelle haben Vorteile gegenüber der traditionellen Verwendung von Versuchstieren für toxikologische Untersuchungen, wie präklinische Studien mit Labortieren sind nicht immer prädiktiv für die Sicherheit der Menschen. Das Medikament induzierte Toxizitäten bei Patienten angetroffen werden oft metabolische oder Signalprozesse, die zwischen Menschen und Versuchstieren unterscheiden verwandt. Die Spezies Unterschied hat die zuverlässige Früherkennung von Entwicklungstoxizität beim Menschen verhindert und zum Beispiel Arzneimittel, wie Thalidomid 23,24 und 25,26 Diethylstilbestrol wurden aus dem Markt aufgrund Teratogenität zurückgezogen. ThaliDomide hat keine Entwicklungstoxizität bei Ratten und Mäusen gezeigt. Umweltchemikalien wie Methylquecksilber 27 resultierte in der pränatalen Entwicklungstoxizität in Bezug auf das Nervensystem in verschiedenen Arten, aber menschlichen Erscheinungsformen sind schwer zu in Tiere. Um das Problem der Spezies-Spezifität Probleme anzugehen, Wissenschaftler, die im Rahmen verschiedener Projekte auf Basis von Stammzellen wie ReProTect, ESNATS, Detektiv etc. sind in die Entwicklung der verschiedenen Modelle für embryonale Toxizität, Neurotoxizität, Kardiotoxizität, Lebertoxizität und Nephrotoxizität mit menschlichen Giftstoffe zu vermuten engagiert Auswirkungen auf den Menschen. Im Rahmen des europäischen Konsortiums Projekt &quot;embryonalen Stammzellen-basierten Roman alternativer Test (ESNATS)&quot; fünf Testsysteme wurden eingerichtet. Ein Testsystem die sogenannte UKK (U niversitäts k linikum K ÖLN) Testsystem zum Teil erfasst frühen embryonalen Entwicklung. In diesem system menschlichen embryonalen H9-Zellen werden in drei Keimblätter (Ektoderm, Entoderm und Mesoderm) 15 und Keim spezifischen Signaturen von Transkriptomik gefangen genommen worden Profile mit Hilfe der Affymetrix-Microarray-Plattform unterschieden. Verschiedenen Entwicklungsgiftstoffen wie Thalidomid 28, Valproinsäure, Methylquecksilber 16,17 oder Cytosinarabinosid 15 haben in diesem System getestet und Giftstoff spezifische Gen-Signaturen erhalten wurden. In einem zweiten Testsystem, das so genannte UKN1 (U NIVERSITÄT K onsta n z) Testsystem 1 werden H9-Zellen zu neuroektodermalen Progenitorzellen (NEP) für 6 Tage differenziert. Dies wird durch eine hohe Expression der neuronalen Marker-Gen wie PAX6 und OTX2 belegt. Während der Differenzierung für 6 Tage wurden NEP-Zellen zu Entwicklungsneurotoxisch, wie VPA, Methylquecksilber ausgesetzt. Giftstoff spezifische deregulierten Transkriptomik Profile wurden obtainiert als auch mit Hilfe der Affymetrix-Microarray-Plattform 16,29. Die neue Vision für die Toxikologie des 21. Jahrhunderts sieht vor, dass Testsysteme nicht nur zu erhalten phänotypische Beschreibungen wie Histopathologie in vivo oder Transkriptom Änderungen am Ende des langfristigen Giftstoff Inkubationen. Vielmehr legt nahe, dass Assays stellen mechanistische Informationen 3, und dass diese Informationen, so genannte abgebildet werden negative Ergebnis Wegen (AOP), die eine wissenschaftliche Begründung für den Ex-Effekte 30 bereitzustellen. Um solche Informationen zu liefern, haben die zur Prüfung verwendeten Systeme werden in hohem Grade Qualität kontrolliert 31, wie beispielsweise durch robuste Standardarbeitsanweisungen dokumentiert. Darüber hinaus müssen die zeitabhängigen Änderungen mit hoher Auflösung abgebildet werden. Dies erfordert Testsysteme mit synchronisierten Änderungen 32. Die hier beschriebenen UKN1 und UKK-Test-Systeme für diese Anforderungen optimiert.

human pluripotent stem cell based developmental toxicity assays for chemical safety screening and systems biology data generation

Effiziente Protokolle für die menschliche pluripotente Stammzellen zu verschiedenen Geweben in Kombination mit differen -omics Technologien eröffnet neue Horizonte für in-vitro-Toxizitätsprüfung von potentiellen Medikamenten. Um eine solide wissenschaftliche Grundlage für solche Tests liefern, wird es wichtig sein, um quantitative Informationen über den zeitlichen Verlauf der Entwicklung und auf die zugrunde liegenden Regulationsmechanismen durch die Systembiologie zu gewinnen. Zwei Tests wurden daher hier für diese Anforderungen abgestimmt. In der UKK-Test-System werden menschliche embryonale Stammzellen (hESC) (oder anderen pluripotenten Zellen) links nach spontan differenzieren für 14 Tage in embryonale Körper, um die Erzeugung von Zellen aller drei Keimblätter zu ermöglichen. Dieses System rekapituliert Schlüsselschritte der frühen embryonalen Entwicklung, und es kann humanspezifischen frühen embryonalen Schädigung / Teratogenität vorherzusagen, wenn Zellen auf Chemikalien während der Differenzierung ausgesetzt. Die UKN1 Testsystem basiert auf hES bezogen auf einen Differen populatIonen neuroektodermalen Vorläufer (NEP) Zellen für 6 Tage. Dieses System rekapituliert frühen neuronalen Entwicklung und prognostiziert frühen Entwicklungsneurotoxizität und epigenetische Veränderungen durch Chemikalien ausgelöst. Beide Systeme in Kombination mit Transkriptom Mikroarray Studien eignen sich zur Identifizierung von Biomarkern Toxizität. Darüber hinaus können sie in Kombination verwendet, um Daten für die Systembiologie Analyse zu erzeugen. Diese Testsysteme haben Vorteile gegenüber den traditionellen toxikologische Untersuchungen, die große Mengen an Tieren. Die Testsysteme können zu einer Verringerung der Kosten für die Wirkstoffentwicklung und chemische Sicherheitsbewertung beitragen. Ihre Kombination beleuchtet vor allem auf Verbindungen, die Entwicklung des Nervensystems gezielt beeinflussen können.

Methylquecksilberbelastung in UKK Testsystem Zytotoxizitätstest wurde mit H9 EBs geführt, um einen IC 10 -Wert (Verringerung der Lebensfähigkeit um 10%) für die Zytotoxizität von Methylquecksilber (Figur 1) erhalten.
