The present study describes assessing autonomic responses to passive motion in rodents using elevator vertical motion and Ferris-wheel rotation. These equipment and procedures can be easily adopted to other rodents and several modifications of the assays exist to confirm vestibular functioning in different circumstances, such as during in pharmacological challenge or surgical interventions. Motion sickness (MS) due to abnormal visuo-vestibular stimulation leads to autonomic reaction, eliciting symptoms such epigastric discomfort, nausea and/or vomiting1. According to current theories, motion sickness may be caused by a sensory conflict or neuronal mismatch from receiving integrated motion information that differs from the anticipated internal model of the environment2,3 or postural instability as would occur on a yawing ship4,5. Despite significant advances in the field of motion sickness and vestibular autonomic functioning6,7,8,9,10,11,12, future research can be aided by standardized evaluation protocols. Assessing the autonomic effects of standard passive motions will greatly benefit investigations into the causes and prevention of motion sickness. The overall goal of this study is to assess the autonomic and behavioral effects of passive motion in rodents. Animal models, such as rodents, allow easy experimental manipulation (e.g., passive motion and pharmaceutical) and behavioral evaluation, which can be used to study the etiology of motion sickness. Here, we present a detailed battery for testing the effects of passive motion and the integrity of vestibular functioning.
The present study details two assays, elevator vertical motion (EVM) and Ferris-wheel rotation (FWR), that induce autonomic reactions to the passive motion. The assays are coupled to three quantitative behavioral measures, the balance beam (on mice13 and rats14,15,16,17), open-field examination, and defecation counting. The EVM (similar to the pitch and roll of a ship encountering a wave) assesses vestibular functioning by stimulating the otolith sensory organs that encode linear accelerations (i.e., the saccule that responds to movements in the vertical plane)18. The FWR (centrifugal rotation or sinusoidal motion) device stimulates the otolith organs by linear acceleration and the semicircular canals by angular acceleration19,20. The Ferris-wheel/centrifugal rotation device is unique in its autonomic assessment. To date, the only similar device in the literature is the off-vertical axis rotation (OVAR) turntable, which is used to examine the vestibulo-ocular reflex (VOR)18,21,22, conditioned avoidance23,24, and the effects of hypergravity25,26,27. The EVM assay and the FWR device assay induce vestibular stimulation leading to autonomic reactions. We couple the EVM and FWR to quantitative measurements such as balance beam, defecation counting, and open-field analysis28,29,30, to ensure robust and reproducible results. Similar to those previously described in mice13 and rats14,15,16,17, the balance beam assay is a 1.0 m long beam suspended 0.75 m from the ground between two wooden stools using a simple black-box modification at the goal end (finish). The balance beam has been used to assess anxiety (obscure black box)14,17, traumatic injury15,16,17, and here, autonomic reactions affecting balance. We have performed defecation counting for assessing the autonomic response in the motion sickness model previously, and it is a reliable quantitative measurement that is easily performed and unequivocally assessed6,8,9,11. The open-field analysis employs a simple black box open-&#64257;eld behavior assessment using Ethovision28, Bonsai30, or a simple video analysis in Matlab29 to quantify behavior such as motion. In the current protocol, we use the total distance traveled, but we note several different paradigms exist (e.g., elongation, zone of movement, velocity, etc.)28,29,30. Collectively, these procedures form a short battery of assessments for the examination and evaluation of autonomic reactions to passive motion, for example in motion sickness6,7,8,9,10,11. The present assays can be adapted to a variety of animal models.

The overall goal of this study is to assess the autonomic and behavioral effects of passive motion in rodents using the elevator vertical motion and Ferris-wheel rotation devices. These assays can help confirm the integrity and normal functioning of the autonomic nervous system. They are coupled to quantitative measures based on defecation counting, open-field examination, and balance beam crossing. The advantages of these assays are their simplicity, reproducibility, and quantitative behavioral measures. The limitations of these assays are that the autonomic reactions could be epiphenomena of non-vestibular disorders and that a functioning vestibular system is required. Examination of disorders such as motion sickness will be greatly aided by the detailed procedures of these assays.

Figure 2 demonstrates representative balance beam results of time taken to transverse. Rats were trained for 3 consecutive days in order to achieve stable performance on the balance beam10. The subsequent day, rats were evaluated for balance beam performance. In the y-axis of the figure, we have the number of seconds taken for rodents to cross the balance beam for Ferris-wheel, elevator vertical motion, and control groups for demonstrative purposes.

These protocols are important for establishing the integrity and normal functioning of the autonomic system to assess autonomic nervous system functionality and to reveal vestibular system disorders. A vestibular functioning or motor coordination deficit can signal damage to the autonomic system. These techniques can reveal the integrity of autonomic functioning.Behavioral techniques can be susceptible to variabilities introduced by the experimenter. We recommend the same researcher conduct the techniques throughout the experiment and to treat every rodent identically. For elevator vertical motion analysis, place up to four two-month-old 200 to 250 gram rats in a 22.5 by 26 by 20 centimeter plexiglass box and securely fashion the box.Place the box onto the elevator pad of an elevator vertical motion device and turn on the device at the lowest setting for acclimatization. Next, set the amplitude to 22 centimeters up and 22 centimeters down from neutral and start the device with an initial cycle period at 2, 500 milliseconds for five minutes, 2, 000 milliseconds for five minutes, and 1, 500 milliseconds for five minutes. Then set the device to 1, 000 milliseconds for two hours before slowing the device in reverse using periods of 1, 500 milliseconds for five minutes, 2, 000 milliseconds for five minutes, and 2, 500 milliseconds for five minutes.For a Ferris-wheel rotation procedure, place the plexiglass container on a wooden bench and place the rats into the container with the long axis of their bodies perpendicular to the horizontal rotation rod of the Ferris-wheel. After closing the box securely, place a second set of rats with a similar mass into a second plexiglass container with the long axis of the body perpendicular to the horizontal rotation rod on the second arm of the Ferris-wheel rotation device. After securely closing the box, start the Ferris-wheel rotation in a clockwise direction at 16 degrees per second squared to reach an angular velocity of 120 degrees per second.When the Ferris-wheel has reached peak velocity, begin to decelerate at 48 degrees per second squared to reach zero degrees per second. After a one-second pause, have the container continue to rotate in the counterclockwise direction at 16 degrees per second squared to reach an angular velocity of 120 degrees per second before decelerating at 48 degrees second squared to reach zero degrees per second. Then continue the clockwise-counterclockwise rotation for two hour per session for approximately 720 rotations.For balance beam training, first place two wooden 0.75 meter high stools into the experimental field approximately 110 centimeters apart. Place a plastic black 15 by 15 by eight centimeter box on the finish tool and place a narrow 2.5 by 130 centimeter wooden beam between the two stools leaving a 100 centimeter distance between the stool edges from the start stool to the finish stool. Place a lamp at the start stool and turn on the lamp.Then turn off the room light so that the room is as dark as possible and introduce the first rat to the beam in the lighted corner. If the rat does not cross of its volition, gently prompt the animal to cross the beam. After three consecutive days of training, place the trained rat onto the start stool near the light and immediately start a stopwatch.The rat should cross the balance beam rapidly and enter the black box on the finish stool. Stop the stopwatch as soon as the animal's nose enters the dark box on the finish stool. For defecation counting after motion treatment, transfer the rats from the plexiglass container into individual 40 by 40 by 45 centimeter open field boxes and count the number of feces pellets in the plexiglass box attributed to each rodent.For an open field behavior test, use an infrared video camera to record the animals for three minutes and use the resulting video footage to determine the total distance traveled for each animal. Here, representative balance beam results of the time taken to traverse the balance beam after three consecutive days of training are shown. Elevator vertical motion, changes in the Hertz vertical motion, and Ferris-wheel rotation significantly increased defecation.In addition, elevator vertical motion, changes in the Hertz vertical motion, and Ferris-wheel rotation significantly decreased the open field distance traveled. The balance beam training is critical as the rats must be motivated and confident when crossing the beam. Otherwise, their balance will not be measured during the evaluation period.Following the behavioral procedures, the effects of different drugs of interest on autonomic functioning and cell signaling can be assessed using these vestibular stimulation assays.

A Procedure to Observe Context-induced Renewal of Pavlovian-conditioned Alcohol-seeking Behavior in Rats

Environmental contexts in which drugs of abuse are consumed can trigger craving, a subjective Pavlovian-conditioned response that can facilitate drug-seeking behavior and prompt relapse in abstinent drug users. We have developed a procedure to study the behavioral and neural processes that mediate the impact of context on alcohol-seeking behavior in rats. Following acclimation to the taste and pharmacological effects of 15% ethanol in the home cage, male Long-Evans rats receive Pavlovian discrimination training (PDT) in conditioning chambers. In each daily (Mon-Fri) PDT session, 16 trials each of two different 10 sec auditory conditioned stimuli occur. During one stimulus, the CS+, 0.2 ml of 15% ethanol is delivered into a fluid port for oral consumption. The second stimulus, the CS-, is not paired with ethanol. Across sessions, entries into the fluid port during the CS+ increase, whereas entries during the CS- stabilize at a lower level, indicating that a predictive association between the CS+ and ethanol is acquired. During PDT each chamber is equipped with a specific configuration of visual, olfactory and tactile contextual stimuli. Following PDT, extinction training is conducted in the same chamber that is now equipped with a different configuration of contextual stimuli. The CS+ and CS- are presented as before, but ethanol is withheld, which causes a gradual decline in port entries during the CS+. At test, rats are placed back into the PDT context and presented with the CS+ and CS- as before, but without ethanol. This manipulation triggers a robust and selective increase in the number of port entries made during the alcohol predictive CS+, with no change in responding during the CS-. This effect, referred to as context-induced renewal, illustrates the powerful capacity of contexts associated with alcohol consumption to stimulate alcohol-seeking behavior in response to Pavlovian alcohol cues.

Operant Procedures for Assessing Behavioral Flexibility in Rats

Executive functions consist of multiple high-level cognitive processes that drive rule generation and behavioral selection. An emergent property of these processes is the ability to adjust behavior in response to changes in one&#8217;s environment (i.e., behavioral flexibility). These processes are essential to normal human behavior, and may be disrupted in diverse neuropsychiatric conditions, including schizophrenia, alcoholism, depression, stroke, and Alzheimer&#8217;s disease. Understanding of the neurobiology of executive functions has been greatly advanced by the availability of animal tasks for assessing discrete components of behavioral flexibility, particularly strategy shifting and reversal learning. While several types of tasks have been developed, most are non-automated, labor intensive, and allow testing of only one animal at a time. The recent development of automated, operant-based tasks for assessing behavioral flexibility streamlines testing, standardizes stimulus presentation and data recording, and dramatically improves throughput. Here, we describe automated strategy shifting and reversal tasks, using operant chambers controlled by custom written software programs. Using these tasks, we have shown that the medial prefrontal cortex governs strategy shifting but not reversal learning in the rat, similar to the dissociation observed in humans. Moreover, animals with a neonatal hippocampal lesion, a neurodevelopmental model of schizophrenia, are selectively impaired on the strategy shifting task but not the reversal task. The strategy shifting task also allows the identification of separate types of performance errors, each of which is attributable to distinct neural substrates. The availability of these automated tasks, and the evidence supporting the dissociable contributions of separate prefrontal areas, makes them particularly well-suited assays for the investigation of basic neurobiological processes as well as drug discovery and screening in disease models.

The ability to assess executive functions such as behavioral flexibility in rats is useful for investigating the neurobiology of cognition in both intact animals and disease models. Here we describe automated tasks for assessing strategy shifting and reversal learning, which are particularly sensitive to disruptions in prefrontal cortical networks.

How to Study Placebo Responses in Motion Sickness with a Rotation Chair Paradigm in Healthy Participants

Placebo responses occur in every medical intervention when patients or participants expect to receive an effective treatment to relieve symptoms. However, underlying mechanisms of placebo responses are not fully understood. It has repeatedly been shown that placebo responses are associated with changes in neural activity but for many conditions it is unclear whether they also affect the target organ, such as the stomach in motion sickness. Therefore, we present a methodology for the multivariate assessment of placebo responses by subjective, behavioral and objective measures in motion sickness with a rotation chair paradigm. The physiological correlate of motion sickness is a shift in gastric myoelectrical activity towards tachygastria that can be recorded with electrogastrography. The presented study applied the so-called balanced placebo design (BPD) to investigate the effects of ginger compared to placebo and the effects of expectations by verbal information. However, the study revealed no significant main or interactional effects of ginger (as a drug) or information on outcome measures but showed interactions when sex of participants and experimenters are taken into considerations. We discuss limitations of the presented study and report modifications that were used in subsequent studies demonstrating placebo responses when rotation speed was lowered. In general, future placebo studies have to identify the appropriate target organ for the studied placebo responses and to apply the specific methods to assess the physiological correlates.

Multifunctional Setup for Studying Human Motor Control Using Transcranial Magnetic Stimulation, Electromyography, Motion Capture, and Virtual Reality

The study of neuromuscular control of movement in humans is accomplished with numerous technologies. Non-invasive methods for investigating neuromuscular function include transcranial magnetic stimulation, electromyography, and three-dimensional motion capture. The advent of readily available and cost-effective virtual reality solutions has expanded the capabilities of researchers in recreating &#8220;real-world&#8221; environments and movements in a laboratory setting. Naturalistic movement analysis will not only garner a greater understanding of motor control in healthy individuals, but also permit the design of experiments and rehabilitation strategies that target specific motor impairments (e.g. stroke). The combined use of these tools will lead to increasingly deeper understanding of neural mechanisms of motor control. A key requirement when combining these data acquisition systems is fine temporal correspondence between the various data streams. This protocol describes a multifunctional system&#8217;s overall connectivity, intersystem signaling, and the temporal synchronization of recorded data. Synchronization of the component systems is primarily accomplished through the use of a customizable circuit, readily made with off the shelf components and minimal electronics assembly skills.

Transcranial magnetic stimulation, electromyography, and 3D motion capture are commonly used non-invasive techniques for investigating neuromuscular function in humans. Substantiating Appropriate Motion Capture Techniques for the Assessment of Nordic Walking Gait and Posture in Older Adults

Nordic walking (NW) has become a safe and simple form of exercise in recent years, and in studying this gait pattern, various data collection techniques have been employed, each with positives and negatives. The aim was to determine the effect of NW on older adult gait and posture and to determine optimal use of different data collection systems in both short and long duration analysis. Gait and posture during NW and normal walking were assessed in 17 healthy older adults (age: 69 &#177; 7.3). Participants performed two trials of 6 Minute Walk Tests (6MWT) (1 with poles (WP) and 1 without poles (NP)) and 6 trials of a 5m walk (3 WP and 3 NP). Motion was recorded using two systems, a 6-sensor accelerometry system and an 8-camera 3-dimensional motion capture system, in order to quantify spatial-temporal, kinematic, and kinetic parameters.
With both systems, participants demonstrated increased stride length and double support and decreased gait speed and cadence WP compared to NP (p &#60;0.05). Also, with motion capture, larger single support time was found WP (p &#60;0.05). With 3-D capture, smaller hip power generation and moments of force were found at heel contact and pre-swing as well as smaller knee power absorption at heel contact, pre-swing, and terminal swing WP compared to NP, when assessed over one cycle (p &#60;0.05). Also, WP yielded smaller moments of force at heel contact and terminal swing along with larger moments at mid-stance of a gait cycle (p &#60;0.05). No changes were found for posture.
NW seems appropriate for promoting a normal gait pattern in older adults. Three-dimensional motion capture should primarily be used during short duration gait analysis (i.e. single gait cycle), while accelerometry systems should be primarily employed in instances requiring longer duration analysis such as during the 6MWT.

Visualizing Motion Patterns in Acupuncture Manipulation

Acupuncture manipulation varies widely among practitioners in clinical settings, and it is difficult to teach novice students how to perform acupuncture manipulation techniques skillfully. The Acupuncture Manipulation Education System (AMES) is an open source software system designed to enhance acupuncture manipulation skills using visual feedback. Using a phantom acupoint and motion sensor, our method for acupuncture manipulation training provides visual feedback regarding the actual movement of the student's acupuncture manipulation in addition to the optimal or intended movement, regardless of whether the manipulation skill is lifting, thrusting, or rotating. Our results show that students could enhance their manipulation skills by training using this method. This video shows the process of manufacturing phantom acupoints and discusses several issues that may require the attention of individuals interested in creating phantom acupoints or operating this system.

The Rodent Psychomotor Vigilance Test (rPVT): A Method for Assessing Neurobehavioral Performance in Rats and Mice

The human Psychomotor Vigilance Test (PVT) is a widely used procedure for measuring changes in fatigue and sustained attention. The present article describes a rodent version of the PVT&#8212;termed the &#34;rPVT&#34;&#8212;that measures similar aspects of attention (i.e., performance accuracy, motor speed, premature responding, and lapses in attention). Data are presented that demonstrate both the short- and long-term usefulness of the rPVT when employed with laboratory rats. Rats easily learn the rPVT, and learning to perform the basic procedure takes less than two weeks of training. Once acquired, rat performances in the rPVT show a high degree of similarity to these same performance measures in the human PVT, including similarities in, lapses in attention, reaction times, vigilance decrements across session time (i.e., the human &#34;time-on-task&#34; effects), and the response-stimulus interval (RSI) effect described for humans. Thus the rPVT can be an extremely valuable tool for assessing the effects of a wide range of variables on sustained attention quite similar to human PVT performances, and thus can be useful for developing novel treatments for neurobehavioral dysfunctions.

Behavioral Disturbances: An Innovative Approach to Monitor the Modulatory Effects of a Nutraceutical Diet

In dogs, diets are often used to modulate behavioral disturbances related to chronic anxiety and stress caused by intense and restless activity. However, the traditional ways to monitor behavioral changes in dogs are complicated and not efficient. In the current clinical evaluation, a new, simple monitoring system was used to assess the effectiveness of a specific diet in positively modulating the intense and restless activity of 24 dogs of different ages and breeds. This protocol describes how to easily and rapidly evaluate improvement in a set of symptoms related to generalized anxiety by using a specific sensor, a mobile phone app, a wireless router, and a computer. The results showed that dogs treated with specific diets showed significant improvement in the times spent active and at rest after 10 days (p &lt; 0.01 and p &lt; 0.05, respectively). These dogs also showed an overall significant improvement in clinical and behavioral symptoms. A specific sensor, along with its related hardware, was demonstrated to successfully monitor behavioral changes relating to movement in dogs.

A Wireless, Bidirectional Interface for In Vivo Recording and Stimulation of Neural Activity in Freely Behaving Rats

In vivo electrophysiology is a powerful technique to investigate the relationship between brain activity and behavior at a millisecond and micrometer scale. However, current methods mostly rely on tethered cable recordings or only use unidirectional systems, allowing either recording or stimulation of neural activity, but not at the same time or same target. Here, a new wireless, bidirectional device for simultaneous multichannel recording and stimulation of neural activity in freely behaving rats is described. The system operates through a single portable head stage that both transmits recorded activity and can be targeted in real-time for brain stimulation using a telemetry-based multichannel software. The head stage is equipped with a preamplifier and a rechargeable battery, allowing stable long-term recordings or stimulation for up to 1 h. Importantly, the head stage is compact, weighs 12 g (including battery) and thus has minimal impact on the animal&#180;s behavioral repertoire, making the method applicable to a broad set of behavioral tasks. Moreover, the method has the major advantage that the effect of brain stimulation on neural activity and behavior can be measured simultaneously, providing a tool to assess the causal relationships between specific brain activation patterns and behavior. This feature makes the method particularly valuable for the field of deep brain stimulation, allowing precise assessment, monitoring, and adjustment of stimulation parameters during long-term behavioral experiments. The applicability of the system has been validated using the inferior colliculus as a model structure.

Measuring Active and Passive Tameness Separately in Mice

Domesticated animals such as dogs and laboratory mice show a high level of tameness, which is important for humans to handle them easily. Tameness has two behavioral components: a reluctance to avoid humans (passive tameness) and a motivation to approach humans (active tameness). To quantify these components in mice, we previously developed behavioral tests for active tameness, passive tameness, and the willingness to stay on a human hand, each designed to be completed within 3 min. The data obtained were used for selective breeding, with a large number of mice analyzed per generation. The active tameness test measures the movement of the mouse toward a human hand and the contact it engages in. The passive tameness test measures the duration of time that a mouse tolerates human touch. In the stay-on-hand test, a mouse is placed on a human hand and touched slowly using the thumb of that hand; the duration of time that the animal remains on the hand is measured. Here, we describe the test set-up and apparatus, explain the procedures, and discuss the appropriate data analysis. Finally, we explain how to interpret the results.
