Phosphacan is a chondroitin sulfate proteoglycan representing the secreted extracellular part of a transmembrane receptor protein tyrosine phosphatase (RPTP-beta). These isoforms have been implicated in cell-extracellular matrix signaling events associated with myelination, axon growth, and cell migration in the developing central nervous system and may play critical roles in the context of brain pathologies. Recently, we have reported the identification of a new isoform of phosphacan, the phosphacan short isoform (PSI), the expression of which peaks in the second postnatal week. PSI interacts with the neuronal receptors L1 and F3/contactin and can promote neurite growth of cortical neurons. In this study, we have assessed, by in situ hybridization, the expression profile of PSI in the rat brain at postnatal day 7. PSI is largely expressed in the gray matter of the developing cerebral cortex in which it colocalizes with phosphacan, whereas the expression of RPTPbeta receptor forms is restricted to the ventricular area in which PSI has not been observed. Neurons from all layers of the cortex express PSI. In the cerebellum, on the other hand, no expression of PSI has been detected, although the other phosphacan/RPTP-beta isoforms show strong PSI expression here. Overall, our study suggests that PSI is expressed during the postnatal period in differentiated neurons of the cortex but is absent from structures in which proliferation and migration occur. The significance of these observations is discussed in the context of previous models of phosphacan/RPTP-beta functions.

Cortical neurons express PSI, a novel isoform of phosphacan/RPTPbeta.

Phosphacan est un prot&#xE9;oglycane chondro&#xEF;tine sulfate, repr&#xE9;sentant la partie extracellulaire s&#xE9;cr&#xE9;t&#xE9;e d&#x26;#39;une phosphatase transmembranaire du r&#xE9;cepteur prot&#xE9;ine tyrosine (RPTP-b&#xEA;ta). Ces isoformes ont &#xE9;t&#xE9; impliqu&#xE9;s dans les &#xE9;v&#xE9;nements cellule-matrice extracellulaire de signalisation associ&#xE9;es &#xE0; la my&#xE9;linisation, la croissance axonale et la migration cellulaire dans le d&#xE9;veloppement du syst&#xE8;me nerveux central et peut jouer un r&#xF4;le critique dans le contexte de pathologies c&#xE9;r&#xE9;brales. R&#xE9;cemment, nous avons signal&#xE9; l&#x26;#39;identification d&#x26;#39;une nouvelle isoforme phosphacan, l&#x26;#39;isoforme courte phosphacan (PSI), l&#x26;#39;expression de qui culmine dans la deuxi&#xE8;me semaine postnatale. PSI interagit avec le L1 neuronale r&#xE9;cepteurs et F3/contactin et peut favoriser la croissance des neurites des neurones corticaux. Dans cette &#xE9;tude, nous avons &#xE9;valu&#xE9;, par hybridation in situ, le profil d&#x26;#39;expression de l&#x26;#39;ISP dans le cerveau de rat au jour postnatal 7. PSI est largement exprim&#xE9; dans la mati&#xE8;re grise du cortex c&#xE9;r&#xE9;bral en d&#xE9;veloppement dans lequel il co-localise avec phosphacan, alors que l&#x26;#39;expression de formes de r&#xE9;cepteurs RPTPbeta est limit&#xE9; &#xE0; la zone ventriculaire dans lequel PSI n&#x26;#39;a pas &#xE9;t&#xE9; observ&#xE9;e. Les neurones de toutes les couches de la PSI cortex expresse. Dans le cervelet, d&#x26;#39;autre part, aucune expression de PSI a &#xE9;t&#xE9; d&#xE9;tect&#xE9;e, bien que l&#x26;#39;autre phosphacan / RPTP-b&#xEA;ta isoformes montrent une expression PSI fort ici. Dans l&#x26;#39;ensemble, notre &#xE9;tude sugg&#xE8;re que PSI est exprim&#xE9; au cours de la p&#xE9;riode postnatale dans les neurones diff&#xE9;renci&#xE9;s du cortex, mais est absent de structures dans lesquelles la prolif&#xE9;ration et la migration se produisent. La signification de ces observations est discut&#xE9;e dans le contexte des mod&#xE8;les pr&#xE9;c&#xE9;dents de phosphacan / RPTP-b&#xEA;ta fonctions.

Corticale neurones expriment PSI, une nouvelle isoforme de Phosphacan / RPTPbeta

Kortikalen Neuronen Express PSI, ein Roman Isoform Phosphacan / RPTPbeta.

Phosphacan &egrave; un proteoglicano Condroitin solfato che rappresenta la parte extracellulare secreta di una recettore transmembrana della proteina tirosina fosfatasi (RPTP-beta). Queste isoforme sono stati implicati nella cella extracellulare segnalazione eventi associati con mielinizzazione, axon crescita e migrazione delle cellule in via di sviluppo del sistema nervoso centrale e pu&ograve; giocare ruoli critici nel contesto delle patologie cerebrali. Recentemente, abbiamo segnalato l&#39;identificazione di una nuova isoforma di phosphacan, l&#39;isoforma corta phosphacan (PSI), l&#39;espressione di cui cime della settimana secondo postnatale. PSI interagisce con i recettori neuronali L1 e F3/anche e pu&ograve; promuovere la crescita dei neuriti dei neuroni corticali. In questo studio, noi abbiamo valutato, l&#39;ibridazione in situ, il profilo di espressione del PSI nel cervello del ratto a 7 giorno postnatale. PSI &egrave; espressa in gran parte nella sostanza grigia della corteccia cerebrale in via di sviluppo in cui colocalizza con phosphacan, considerando che l&#39;espressione di forme del recettore RPTPbeta &egrave; limitato alla zona ventricolare in cui non &egrave; stato osservato PSI. I neuroni da tutti gli strati della corteccia express PSI. Nel cervelletto, d&#39;altra parte, nessuna espressione del PSI &egrave; stata rilevata, anche se le altre phosphacan/RPTP-beta isoforme mostrano forte espressione PSI qui. Nel complesso, il nostro studio suggerisce che il PSI &egrave; espresso durante il periodo postnatale in differenziata dei neuroni della corteccia ma &egrave; assente dalle strutture in cui si verificano proliferazione e migrazione. Il significato di queste osservazioni &egrave; discusso nel contesto dei modelli precedenti delle funzioni phosphacan/RPTP-beta.

Phosphacan es un proteoglicano de sulfato de condroitina que representa la parte extracelular secretada de un receptor transmembrana tirosina fosfatasa de prote&#xED;nas (RPTP-beta). Estas isoformas han sido implicados en los eventos de c&#xE9;lula-matriz extracelular de se&#xF1;alizaci&#xF3;n asociados con la mielinizaci&#xF3;n, el crecimiento axonal, y la migraci&#xF3;n celular en el desarrollo del sistema nervioso central y puede desempe&#xF1;ar un papel fundamental en el contexto de las patolog&#xED;as cerebrales. The chondroitin sulfate proteoglycan DSD-1-PG/phosphacan represents one of four splice variants of receptor-protein-tyrosine-phosphatase-beta/zeta (RPTPbeta/zeta). This receptor is expressed by glial cells and occurs in two isoforms, RPTPbeta(long) and RPTPbeta(short). The secreted forms phosphacan and phosphacan short isoform (PSI) bind to extracellular matrix and adhesion molecules and might mediate astroglial effects on neuronal differentiation. Phosphacan and RPTPbeta(long) both carry the DSD-1 epitope, a glycosaminoglycan modification that is involved in stimulating neurite outgrowth of embryonic rat mesencephalic and hippocampal neurons in a polycationic environment. Additionally, phosphacan inhibits neurite outgrowth of embryonic DRG neurons in the presence of laminin. In the light of these functional properties we examined the expression patterns of the DSD-1 epitope and phosphacan isoforms in the developing mouse visual system. During retinal development the DSD-1 epitope appears around embryonic day (E)13, peaks around postnatal day (P)6, and is downregulated from P9 to adolescence. By comparison, the phosphacan core protein is first detectable at E12, reaches maximal levels around P14, and persists, although at lower levels, to adulthood. The DSD-1 epitope is restricted to the nerve fiber and the inner plexiform layers. In contrast, the phosphacan core protein immunoreactivity extends from the nerve fiber layer to the outer plexiform layer. The level of expression of the phosphacan/RPTPbeta gene was investigated by reverse-transcriptase polymerase chain reaction. These experiments suggest that there is a shift in the expression patterns of the different phosphacan/RPTPbeta isoforms during late embryonic and postnatal development. In situ hybridization experiments support the conclusion that at least one of the phosphacan/RPTPbeta isoforms in the retina is expressed by neurons.

Die sezernierte Formen Phosphacan und Phosphacan kurze Isoform (PSI) an der extrazellul&auml;ren Matrix und Adh&auml;sion Molek&uuml;le binden und Astroglial Effekte auf neuronale Differenzierung vermitteln k&ouml;nnte. Phosphacan und RPTPbeta(long) beide tragen die DSD-1-Epitop, eine Glykosaminoglykane-Modifikation, die in anregenden Neurit Auswuchs der embryonalen Ratte Aquaeductus und hippocampal Neuronen in einer Polycationic Umgebung beteiligt ist. Il proteoglicano Condroitin solfato DSD-1-PG/phosphacan rappresenta una delle quattro varianti di splicing del recettore-proteina-tirosin-fosfatasi-beta/zeta (RPTPbeta/zeta). Questo recettore &egrave; espresso dalle cellule gliali e si verifica in due isoforme, RPTPbeta(long) e RPTPbeta(short). Le forme secrete phosphacan phosphacan breve isoform (PSI) legano alle molecole di adesione e matrice extracellulare e potrebbe mediare astroglial effetti sulla differenziazione neuronale. Phosphacan e RPTPbeta(long) entrambi portano l&#39;epitopo DSD-1, una modifica di glicosaminoglicano che &egrave; coinvolta nella stimolante escrescenza dei neuriti dei neuroni mesencefaliche e dell&#39;ippocampo di ratto embrionale in un ambiente polycationic. Inoltre, phosphacan inibisce escrescenza dei neuriti dei neuroni DRG embrionali in presenza di laminina. Alla luce di queste propriet&agrave; funzionali abbiamo esaminato il pattern di espressione delle isoforme di epitopo e phosphacan DSD-1 di sviluppo del sistema visivo del mouse. Durante lo sviluppo della retina l&#39;epitopo DSD-1 appare intorno giorno embrionale (E) 13, vette intorno giorno postnatale (P) 6 ed &egrave; downregolata da P9 all&#39;adolescenza. In confronto, la proteina core phosphacan prima &egrave; rilevabile a E12, raggiunge livelli massimali intorno P14 e persiste, anche se a livelli inferiori, fino all&#39;et&agrave; adulta. L&#39;epitopo DSD-1 &egrave; limitata per la fibra del nervo e gli strati interni plessiforme. Al contrario, l&#39;immunoreattivit&agrave; phosphacan core proteico si estende dallo strato della fibra del nervo lo strato plessiforme esterno. Il livello di espressione del gene della phosphacan/RPTPbeta &egrave; stato studiato da reazione a catena della polimerasi trascrittasi inversa. Questi esperimenti suggeriscono che c&#39;&egrave; un cambiamento nei modelli di espressione delle diverse isoforme di phosphacan/RPTPbeta durante lo sviluppo embrionale e postnatale tardo. Esperimenti di ibridazione in situ supportano la conclusione che almeno una delle isoforme di phosphacan/RPTPbeta nella retina &egrave; espresso dai neuroni.

El sulfato de condroitina proteoglicanos DSD-1-PG/phosphacan representa una de las cuatro variantes de empalme de receptor-protein-tyrosine-phosphatase-beta/zeta (RPTPbeta / zeta). Este receptor se expresa en las c&#xE9;lulas gliales y se presenta en dos isoformas, RPTPbeta (largo) y RPTPbeta (corto). El secretada formas phosphacan y phosphacan isoforma corta (PSI) se unen a la matriz extracelular y mol&#xE9;culas de adhesi&#xF3;n y podr&#xED;a mediar los efectos astroglial en la diferenciaci&#xF3;n neuronal. Tissue dissection, cell isolation and a p75(NTR)-antibody-based panning technique, which highly enriches motoneurons within <8 h are described. This protocol is aimed to provide an alternative to the established FACS-based protocols describing p75(NTR)-based enrichments of neurons. This protocol will help in facilitating the research on molecular mechanisms underlying motoneuron development, survival and disease mechanisms.

Mechanisms controlling neuronal survival and regeneration play an important role during development, after birth, and under lesion conditions. Isolated embryonic mouse motoneurons have been a useful tool for studying such basic mechanisms. These cultured motoneurons depend on extracellular matrix (ECM) molecules, which are potent mediators of survival and axonal growth and guidance in the CNS and in vitro, exhibiting either attractive or repellent guidance cues. Additionally, ECM proteoglycans and glycoproteins are components of the glial scar acting as a growth barrier for regenerating axons. Compared with CNS axon outgrowth, less is known about the cues that guide motoneurons toward their peripheral targets. Because we are interested in the effects of glial-derived chondroitin sulfate proteoglycans (CSPGs), we have worked out a model system for investigating the influences of glial-derived matrix molecules on motoneuron outgrowth and survival. We used cultured embryonic mouse motoneurons to investigate axon growth effects of matrix molecules produced by the glial-derived cell lines A7, Neu7, and Oli-neu primary astrocytes as wellas the immortalized Schwann cell line IMS32. The results indicate that molecules of the ECM, especially chondroitin sulfates, play an important role as axon growth-promoting cues. Mecanismos de control de la supervivencia neuronal y la regeneraci&#xF3;n de desempe&#xF1;ar un papel importante durante el desarrollo, despu&#xE9;s del nacimiento, y en condiciones de lesi&#xF3;n. Motoneuronas aislados de embriones de rat&#xF3;n han sido una herramienta &#xFA;til para el estudio de estos mecanismos b&#xE1;sicos. Mechanisms controlling neuronal survival and regeneration play an important role during development, after birth, and under lesion conditions. Isolated embryonic mouse motoneurons have been a useful tool for studying such basic mechanisms. These cultured motoneurons depend on extracellular matrix (ECM) molecules, which are potent mediators of survival and axonal growth and guidance in the CNS and in vitro, exhibiting either attractive or repellent guidance cues. Additionally, ECM proteoglycans and glycoproteins are components of the glial scar acting as a growth barrier for regenerating axons. Compared with CNS axon outgrowth, less is known about the cues that guide motoneurons toward their peripheral targets. Because we are interested in the effects of glial-derived chondroitin sulfate proteoglycans (CSPGs), we have worked out a model system for investigating the influences of glial-derived matrix molecules on motoneuron outgrowth and survival. We used cultured embryonic mouse motoneurons to investigate axon growth effects of matrix molecules produced by the glial-derived cell lines A7, Neu7, and Oli-neu primary astrocytes as well as the immortalized Schwann cell line IMS32. The results indicate that molecules of the ECM, especially chondroitin sulfates, play an important role as axon growth-promoting cues. We could demonstrate a modifying effect of the matrix components on motoneuron survival and caspase3-induced apoptosis.

Mecanismos de control de la supervivencia neuronal y la regeneraci&#xF3;n de desempe&#xF1;ar un papel importante durante el desarrollo, despu&#xE9;s del nacimiento, y en condiciones de lesi&#xF3;n. Motoneuronas aislados de embriones de rat&#xF3;n han sido una herramienta &#xFA;til para el estudio de estos mecanismos b&#xE1;sicos. Estas neuronas motoras cultivadas dependen de la matriz extracelular (ECM) mol&#xE9;culas, que son potentes mediadores de la supervivencia, crecimiento y axonal y orientaci&#xF3;n en el sistema nervioso central e in vitro, que muestran tanto las se&#xF1;ales de orientaci&#xF3;n atractivas o repelentes. Adem&#xE1;s, proteoglicanos y glicoprote&#xED;nas ECM son componentes de la cicatriz glial que act&#xFA;a como una barrera para el crecimiento regeneraci&#xF3;n de los axones. En comparaci&#xF3;n con el ax&#xF3;n nervioso central, se sabe menos sobre las se&#xF1;ales que las neuronas motoras de gu&#xED;a hacia sus objetivos perif&#xE9;ricos. Debido a que estamos interesados &#x200B;&#x200B;en los efectos de la gliales derivadas de los proteoglicanos sulfato de condroitina (CSPGs), hemos desarrollado un sistema modelo para investigar la influencia de mol&#xE9;culas de la matriz gliales derivadas en consecuencia las motoneuronas y la supervivencia. Se utiliz&#xF3; cultivos de motoneuronas embrionarias de rat&#xF3;n para investigar los efectos del crecimiento de axones de las mol&#xE9;culas de la matriz producida por el glial-derivado l&#xED;neas de c&#xE9;lulas A7, Neu7, y Oli-neu astrocitos primarios, as&#xED; como la l&#xED;nea de la c&#xE9;lula de Schwann inmortalizado IMS32. Los resultados indican que las mol&#xE9;culas de la ECM, especialmente sulfatos de condroitina, juegan un papel importante como promotora del crecimiento axonal se&#xF1;ales. Se pudo demostrar un efecto modificador de los componentes de la matriz sobre la supervivencia de las motoneuronas y la apoptosis inducida por caspase3.
