The overall goal of this procedure is to develop a protocol for fabricating an AMLO and chitosan hydrogel for superficial enamel reconstruction. This is accomplished by first preparing etched tooth slices. In the second step, fresh AMLO and kitan hydrogel is prepared.
Next, the AMLO and kitan hydrogel is applied to the tooth slice surface. Finally, the tooth enamel is regrown with the AMLO chizen hydrogel in artificial saliva. Ultimately, ultrasonic treatment and a nano indenture can be used to assess the mechanical strength of the newly formed layer and its binding to natural enamel.
Implications of this technique extends to our developing a technology than when applied on affected tooth enamel, both in adults and children, halts early caries lesion rebuilds loss enamel due to caries preserves tooth structure, and prevents progression of tooth decay. Demonstrating the procedure will be Dr.Chittar one, a postdoctoral fellow from my laboratory. Begin by selecting a human third molar without any restored caries.
Use a water cooled low speed diamond saw to cut the crown longitudinally into two millimeter thick slices, and then remove the root portion of the molar ultrasonically. Clean the slices for two minutes and then rinse them with the ionized water three times to simulate erosive lesions. Etch the two slices with 30%phosphoric acid for 30 seconds.
Then immediately rinse them with the ionized water. Then ultrasonically. Clean the etched slices for two minutes again and rinse them with the ionized water.
Three more times. To prepare the Kazan stock solution. First dissolve 1%weight per volume, Kazan in a 1%volume per volume acetic acid solution, followed by stirring at 80 degrees Celsius overnight.
After cooling the solution to room temperature, strain the esan through a 0.45 micrometer filter, and then add one molar sodium hydroxide to adjust the pH to 5.0. To prepare the calcium and phosphate stock solution, weigh out the calcium chloride and mix a 0.1 molar solution. Next, vortex the solution until it clears and adjust the pH value to 11 with one molar sodium hydroxide.
Then weigh out the diba sodium phosphate. Mix a 0.1 molar solution and vortex until the solution is clear. To prepare the AMLO Genin kaizan hydrogel, add 960 microliters of the freshly prepared 1%esan solution to a tube containing 200 micrograms of recombinant porcine AMLO genin.
Then vortex the solution until clear. Now, add 25 microliters of 0.1 molar calcium chloride solution, followed by 15 microliters of 0.1 molar sodium phosphate diba solution to the AMLO containing ozan solution. Vortex the solution for five minutes.
Then carefully add one molar sodium hydroxide to adjust the pH of the AMLO and ozan solution to 6.5 AMLO and ozan Hydrogel will form when the pH value reaches 6.5 to initiate enamel regrowth in the AMLO andaz, an hydrogel. First, adjust the pH of the artificial saliva solution to 7.0 with one molar sodium hydroxide, and store the solution at four degrees Celsius. Then use a syringe to apply about 20 microliters of AMLO and chizen hydrogel to the edge tooth slice, and place it in a desiccate at room temperature.
After two hours, add 300 PPM of sodium fluoride to the saliva solution, and then transfer the tooth slice to a beaker containing 30 milliliters of the solution. Cover the beaker with aluminum foil and then place the beaker in an oven at 37 degrees Celsius. Finally, remove the tooth slice after three to seven days and place it in a desiccate at room temperature until dry after repair by AMLO and chizen hydrogel for seven days, an enamel like layer with a thickness of 15 micrometers forms on the etched enamel surface.
The newly grown layer is made of highly ordered arrays of crystals with a diameter of about 50 nanometers growing preferentially along the sea axis perpendicular to the surface. Note that these needle-like crystals are organized into bundles similar to the fundamental units of natural enamel. The selected area electron diffraction image of the repaired layer exhibits an arc shaped pattern revealing a hierarchical alignment of the newly grown crystals.
Along the 0 0 2 direction x-ray diffraction analysis confirms that the newly grown layer is composed of appetite, which are aligned along the crystallographic C axis in accordance with the scanning electron microscopy and selected area electron diffraction observations in this image, the microstructure of the interface between the newly grown layer and the natural enamel is shown. Note that the newly grown crystals did not grow in the same direction as the original enamel crystalite, and that the protein mediated appetite growth results in a perpendicular orientation of the newly grown crystals relative to the natural enamel crystals. There is no apparent gap at the interface between the newly grown layer and the enamel substrate.
At the nanoscale, the enamel and the regrown crystals fused together to form a seamless interface. After 10 minutes of ultrasonic treatment, the newly grown layer formed in the AMLO Genin chito hydrogel is still tightly bound to the enamel surface and the organized structure is preserved for the sample treated with tizen hydrogel without am mologen. And however, a large gap between the enamel and the repaired layer is observed to clearly distinguish between the newly grown layer and the natural enamel.
In this experiment, the AMLO genin in the newly grown layer was immuno fluorescently labeled the presence of AMLO genin was then confirmed by the green fluorescence within the newly grown layer. As shown in the graph after acid etching, the hardness and modulus of the enamel surface declines nearly 98 and 88%respectively. The control group treated by ozan hydrogel without AMLO genin only demonstrates limited improvement in both hardness and modulus.
In contrast, after mineralization in the AMLO and ozan hydrogel, an increase of nearly four times in the modulus and nine times in the hardness is observed After its development.
