本論文の動物実験手順はすべて、上海交通医科大学上海交通医院動物ケア委員会によって承認された。
1. 準備
<ol><li>実験には5週齢のオスのスプレイグ・ドーリーラットを2匹使用してください。</li>
	<li>針ホルダー、ピンセット、ハサミを含むすべての器具を高温で殺菌するか、75%エタノールに10分間浸漬します。 注:エタノール浸漬は、細胞の損傷を避けるために長すぎないようにしてください。</li>
	<li>培養培地を事前に準備し、その構成を 表1に示す。以下に説明する各培地を補足する。<ol>
<li>α-MEM培地の調製(10%FBS):10%の胎児ウシ血清および1%ペニシリンおよびストレプトマイシンを用いた最小必須培地アルファ(α-MEM)を補う。</li>
		<li>骨形成分化培地の調製:骨形成分化基底培地を10%胎児ウシ血清、1%グルタミン、1%ペニシリンストレプトマイシン、0.20%アスコルビン酸、1%βグリセロリン酸および0.01%デキサメタゾンを添加する。</li>
		<li>骨形成誘導培地の調製:70%α-MEM培地(10%FBS)と30%の骨形成分化培地を混合する。</li>
		<li>アディポ原性分化培地Aの調製:10%の胎児ウシ血清、1%グルタミン、1%ペニシリンストレプトマイシン、0.20%インスリン、0.10%IBMX、0.10%ロシグリタゾンおよび0.01%デタゾンを有する付加的な分化基底液培地。</li>
		<li>アディポ原性分化培地Bの調製:10%の牛血清、1%のウシ胎児血清、1%のペニシリン連鎖球菌および0.20%インスリンを有する付加的分化基底培地を補足する。</li>
		<li>軟骨発生分化培地の調製:0.01%デキサメタゾン、0.30%アスコルビン酸、1%ITS、0.10%ピルビン酸ナトリウム、0.10%プロリンおよび1%TGF-β3を有するコンドロゲン分化基底培地を補足する。 注: 骨形成分化培地は、構成後1ヶ月以内に使い切る必要があります。構成された軟骨発生分化培地の有効期間は12時間である。</li>
	</ol>
</li>
</ol>2. ラットmMCの分離と栽培
注: 細胞の生存率を維持するために、すべての実験操作を氷上で可能な限り実行する必要があります。
<ol><li>ラット下顎の収穫<ol>
<li>CO2 窒息によって2匹の5週齢の雄のスプレイグ・ドーリーラットを安楽死させる。実験を進める前に、動物の呼吸が止まっていることを確認してください。</li>
		<li>これらのラットを3分間75%エタノールを含むビーカーでリンスする。</li>
		<li>ネズミを清潔な煙のフードの中に入れて下顎を収穫します。 注:使用前に30分間紫外線照射でヒュームフードを殺菌してください。</li>
		<li>ラットをサピーヌの位置に置きます。両側性アングルスオリスから下切歯を有する唯一の可動骨である下顎骨の後部領域に皮膚およびブハネーター筋肉を切開する。 注:明確な操作フィールドを得るためにアングルスの両側に2cmの切開を行うことをお勧めします。</li>
		<li>両親指で上顎および下顎切歯を反対方向に押し出してラットの口を開き、下顎に付着した下顎歯を露出させる。</li>
		<li>頬筋と角筋と下顎下の下の境界に取り付けられた腱を完全に切断します。 注:筋肉の緊張の欠如のために、このステップ中に下顎の移動性の増加が観察されます。</li>
		<li>下顎後歯を押し、両側の顆がはっきりと露出するまで前後に回転します。 注:このステップは、人工的にラットの口を開いて、コンディレを外す可能性のある最大限の範囲で行います。下顎骨は両側の顆を通して頭蓋骨に接続されているので、顆の暴露は頭蓋骨と下顎骨の間の切断につながる。</li>
		<li>下顎骨の体を頭蓋骨から分離します。</li>
		<li>濡れたガーゼを使用して、骨表面の付着性の軟組織を洗浄します。</li>
		<li>生存率を維持するために、氷の上に予冷された最低必須培地α(α-MEM)またはリン酸緩衝液生理食塩水(PBS)で満たされた10cmの無菌ガラス皿に骨を入れます。</li>
	</ol></li>
	<li>mBMSCsの分離と栽培<ol>
<li>下顎体から最初の臼歯の間縁に沿って前骨を切り落とす。骨髄腔を露出させるために、3番目の大臼歯の遠位縁に沿って、コラコイドと顆を含む下顎のラムスを取り除きます。</li>
		<li>10 cmの無菌ガラス皿に10 mLのα-MEM(10%FBS)を充填します。</li>
		<li>1 mL シリンジを使用して、α-MEM培地を吸引します。注射針を骨髄腔に挿入すると、骨髄を皿に繰り返し洗い流す。骨が白くなるまで、骨の前後側から骨腔を少なくとも3回洗い流す。 注:これは、ラット下顎骨の骨髄腔が長い骨ほど明白ではなく、針を挿入するための適切な点を経験的に決定する必要があるため、最も重要なステップです。上記の実験サンプルはすべて、細胞の生存率を維持するために氷の上に保存する必要がありますので、操作時間は2時間以下でなければなりません。</li>
		<li>洗い流された細胞を含む培地を15mL遠心管に移し、4°Cで 800xg の遠心分離機を5分間移動させます。上清を捨てます。</li>
		<li>セルを3 mLのα-MEMで再中断します(FBSが10%)。新しい10cm培養皿に細胞をプレートし、5%CO2インキュベーターで37°Cでインキュベートします。</li>
		<li>培養3日目にこれらの細胞の形態変化と成長を確認してください。非接着性細胞および組織断片を含む培地を除去する。10 mLの新鮮なα-MEM(FBS 10%)をそっと加えます。</li>
		<li>培養7日後、P0 mBMSCは70~80%の合流に達した。</li>
	</ol></li>
	<li>フローセルの並べ替え 注: P0 mBMSC は、フローセルのソートによって、典型的に同定され、主に精製されました。<ol>
<li>吸引し、培養液を捨て、その後PBSで皿を洗います。PBS を破棄します。</li>
		<li>0.25%のトリプシンを1 mL加え、0.02%EDTAを皿に入れます。37°Cで5分間細胞を消化し、2mLのα-MEM(10%FBS)を加えて反応を止めます。</li>
		<li>細胞懸濁液を15mL遠心チューブに移し、800 x g で遠心分離機を5分間移動します。</li>
		<li>遠心分離後、PBSの120 μL(10%FBS)で細胞を再懸濁します。</li>
		<li>これらの細胞懸濁液を、CD16/CD32に対して1μLの抗体を4°Cで15分間ブロックします。</li>
		<li>細胞懸濁液100μLを新しいマイクロ遠心分離管に移し、CD45に対するフィコエリスリン(PE)共役抗体で細胞を染色し、フルオレセイン・イソチオシアネート(FITC)-CD90およびAllophycocyanin(APC)抗体に対して13°Cで13°13°の間にCD29抗体を結合させた抗体この実験で使用した抗体の濃度を表2に示す。他の20μLの細胞懸濁液を無染色陰性対照として使用してください。</li>
		<li>その後、チューブを800 x g で5分間遠心し、懸濁液を捨て、細胞をPBSの0.5 mL(10%FBS)で再懸濁します。</li>
		<li>分析前に10分間0.01mg/mL DAPIの10 μLを加えます。</li>
		<li>遠心分離管に配置された40nmフィルターを使用して細胞をフィルター処理します。</li>
		<li>蛍光活性化細胞ソーター上の細胞を分析する。パネルを次のように設定します。まず、DAPI-細胞、次にCD29+CD90+CD45-を標的mBMSCsとして選択した細胞にゲートして、細胞総数から死んだ細胞を除去する。</li>
		<li>ソートされたCD29+CD90+CD45-細胞を3mLのα-MEM(10%FBS)をあらかじめ用意した15 mL遠心管に集める。</li>
		<li>チューブを800 x g で5分間遠心します。収集バッファーを取り外し、1 mL の新鮮な α-MEM (10% FBS) を追加してセルを再中断します。その後、6 cmの文化皿に入れてお皿に入れます。</li>
	</ol></li>
</ol>3. コロニー形成能力
注: このステップは mBMSCs の区分能力をチェックするために実行されました。15
<ol><li>この実験では、2番目のパッセージmBMSCs(P2)を選択します。細胞合流が80%から90%に達したら、細胞を6ウェルプレートの連続勾配で再播種して、そのクローン増殖能力を評価する。 注:1ウェルあたり1 x 102 から1 x 103 細胞の勾配を設定することをお勧めしますが、人間または動物起源の異なる細胞株の最終的な希釈は経験的に決定されました。</li>
	<li>α-MEM(10%FBS)の細胞を、かなりの数のコロニー形成ユニットが光顕微鏡下で見られるまで約2週間培養した。3日ごとに培地をリフレッシュしてください。</li>
	<li>培地を吸引して廃棄し、一度に5分間PBSでウェルを2回洗浄する。次に、各ウェルに4%パラホルムアルデヒド溶液を加え、少なくとも10分間固定します。</li>
	<li>パラホルムアルデヒドを取り出し、PBSでウェルを2回リンスします。</li>
	<li>結晶紫色染色液で細胞を染色し、37°Cで約10分間インキュベートします。 注:染色時間は、所望のシェードが達成されるまで適切に調整されました。</li>
	<li>染色液を取り出し、蒸留水でサンプルを洗浄して反応を止めます。</li>
	<li>実体顕微鏡下での画像と、少なくとも50個の細胞から成る散乱細胞コロニーを数える。</li>
</ol>4. mMC の多系統分化
注: P2 mBMSCs は、特に説明がない限り、その後の実験に使用されました。
<ol><li>mBMSCsの骨形成誘導<ol>
<li>上記の手順で mBMSCs をダイジェストにします。1 2 mL α-MEM(10%FBS)を補った12ウェルプレートの2.5x 10 5 /cm2 の密度で細胞をシードします。</li>
		<li>細胞合流が60〜70%に達すると、培地を30%骨形成誘導培地に変更する。α-MEM(10%FBS)を陰性対照として細胞を培養し、2日ごとに培地を変化させる。 注:骨形成中に多数のカルシウム結節が出現した後、骨芽細胞が浮遊するのを防ぐために、2日ごとに中量媒体交換に媒体交換パターンを変更することをお勧めします。</li>
		<li>7日間培養した後、アルカリホスファターゼ染色16,17によるこれらの細胞の石灰化を評価する。</li>
		<li>培養培地を取り出し、4%パラホルムアルデヒドで細胞を10分間固定します。1ウェルあたり1 mLのPBSを3分間2回使用して細胞をリンスします。</li>
		<li>次にアルカリホスファターゼ染色液で細胞を染色し、37°Cで10〜30分間インキュベートします。 注:インキュベーションは、必要な色を得るために室温で一晩行うことができます。</li>
		<li>反応を止めるために蒸留水で井戸を洗います。軽顕微鏡で写真を撮る。赤色素色素顆粒はアルカリホスファターゼ(ALP)活性を表す。</li>
		<li>残りの細胞をさらに7日間培養した後、アリザリン赤染色を行い、細胞のミネラル化能力を評価する。</li>
		<li>10分間16、18の細胞固定後に0.5%アリザリン赤染色液で細胞を染色する。3~5分後に蒸留水で発泡反応を止めます。 注:反応時間は、開発した色に応じて延長することができます。</li>
		<li>最後に、培養プレートを光学顕微鏡の下に置き、骨形成染色の効果を観察します。赤い結節は、これらの細胞のカルシウム沈着を示す。</li>
	</ol></li>
	<li>mBMSCsのアディポジェニック誘導<ol>
<li>P2 mBMSCsを上記の12ウェルプレートにシードします。</li>
		<li>ウェルが90%コンフルエントになった後、a.α-MEM培地で培養した細胞(10%FBS)を負の対照として使用して、細胞を培養する。</li>
		<li>2日間の誘導の後、培地Aをウェルから吸引し、各ウェルに1mLのアディポジェニック分化培地Bを加える。</li>
		<li>1日後、培地Bを取り除き、ミディアムAを井戸に戻してサイクルを再開します。培地AとBを用いて細胞を交互に培養する。</li>
		<li>分化培地AとBを3サイクルに交互に適用し、油赤O染色16、18によってこれらの細胞の脂質生成を検出する。 注:丸い、大きな脂質の液滴の多くは、さらに2〜3日間分化培地Bでこれらの細胞を培養することによって観察することができます。</li>
		<li>培地を取り出し、4%パラホルムアルデヒドで細胞を10分間固定します。PBSで細胞を洗い流します。</li>
		<li>油の赤色O染色液で細胞を染色し、37°Cで約3分間インキュベートする。</li>
		<li>染色液を取り除き、蒸留水で井戸を洗います。</li>
		<li>軽顕微鏡での脂質生成を観察します。</li>
	</ol></li>
	<li>mBMSCsの軟骨形成誘導<ol>
<li>P2 mBMSCsを15 mL遠心分離管に5 x 105/cm 2の密度で入れました。</li>
		<li>チューブを300 x g で5分間遠心します。上清を捨てます。</li>
		<li>0.5 mLの軟骨発生分化培地で細胞を再懸濁する。5分間 300xg で細胞を遠心分離する。</li>
		<li>遠心管のキャップを緩め、37°Cで5%CO2インキュベーターでインキュベートします。2日ごとに培地を更新します。 注:細胞は24時間後にペレットを開始します。遠心分離管を48時間動かさないのが推奨されます。培地を変える時は細胞ペレットを吸引しないように注意してください。</li>
		<li>21日間の誘導後、4%パラホルムアルデヒドで細胞ペレットを固定し、続いて脱水、パラフィン埋め込み、切除を行います。</li>
		<li>脱パラフィンと水和後、アルシアブルー溶液中のスライドを少なくとも18、19、20、21分間染色し、蒸留水で洗浄して反応を停止します。写真を光顕微鏡で撮影します。</li>
		<li>コラーゲンII型の免疫蛍光染色については、以下の手順を行う。<ol>
<li>脱パラフィンと水和のステップの後、37 °Cで15分間5μg/mLプロテイナーゼKでスライドをインキュベートします。</li>
			<li>37°Cで1時間ブロッキングバッファーにスライドをインキュベートします。</li>
			<li>200μLのブロッキングバッファーに1μLのコラーゲンII抗体をスライドに塗布し、4°Cで一晩インキュベートします。</li>
			<li>翌日、PBSでスライドを2回洗浄し、37°Cで2次抗体を1時間インキュベートします。</li>
			<li>スライスを40,6-ジミディノ-2-フェニリンドール(DAPI)で10分間インキュベートします。</li>
		</ol>
</li>
		<li>蛍光顕微鏡下で各スライドの写真を撮ります。</li>
	</ol></li>
</ol>5. リアルタイム PCR
<ol><li>市販の試薬に基づいてグアニジウムイソチオシアネートを使用してmBMSCから総RNAを抽出する。</li>
	<li>市販のキットを使用して、RNAを相補的なDNAに逆転写します。逆転写の反応条件は、5分間65°C、15分間37°C、15sで85°Cである。</li>
	<li>表3に記載されているプライマーを用いて骨形成および脂質生成特異的遺伝子を検出するためにリアルタイムPCRを行 う。 PCR増幅手順は、5sの場合は5分間95°C、40サイクルの場合は30sの場合は60°Cの95°Cである。</li>

Isolation, Characterization and Comparative Differentiation of Human Dental Pulp Stem Cells Derived from Permanent Teeth by Using Two Different Methods

Developing wisdom teeth are easy-accessible source of stem cells during the adulthood which could be obtained by routine orthodontic treatments. Human pulp-derived stem cells (hDPSCs) possess high proliferation potential with multi-lineage differentiation capacity compare to the ordinary source of adult stem cells1-8; therefore, hDPSCs could be the good candidates for autologous transplantation in tissue engineering and regenerative medicine. Along with these benefits, possessing the mesenchymal stem cells (MSC) features, such as immunolodulatory effect, make hDPSCs more valuable, even in the case of allograft transplantation6,9,10. Therefore, the primary step for using this source of stem cells is to select the best protocol for isolating hDPSCs from pulp tissue. In order to achieve this goal, it is crucial to investigate the effect of various isolation conditions on different cellular behaviors, such as their common surface markers &amp; also their differentiation capacity.
Thus, here we separate human pulp tissue from impacted third molar teeth, and then used both existing protocols based on literature, for isolating hDPSCs,11-13 i.e. enzymatic dissociation of pulp tissue (DPSC-ED) or outgrowth from tissue explants (DPSC-OG). In this regards, we tried to facilitate the isolation methods by using dental diamond disk. Then, these cells characterized in terms of stromal-associated Markers (CD73, CD90, CD105 &amp; CD44), hematopoietic/endothelial Markers (CD34, CD45 &amp; CD11b), perivascular marker, like CD146 and also STRO-1. Afterwards, these two protocols were compared based on the differentiation potency into odontoblasts by both quantitative polymerase chain reaction (QPCR) &amp; Alizarin Red Staining. QPCR were used for the assessment of the expression of the mineralization-related genes (alkaline phosphatase; ALP, matrix extracellular phosphoglycoprotein; MEPE &amp; dentin sialophosphoprotein; DSPP).14

The method described isolation and characterization of human Dental Pulp Stem Cells (hDPSCs) by using either enzymatic dissociation of pulp (DPSC-ED) or direct outgrowth of stem cells from pulp tissue explants (DPSC-OG). Then followed by in vitro comparative differentiation of both types of hDPSCs into odontoblasts.

Pre-clinical Evaluation of Tyrosine Kinase Inhibitors for Treatment of Acute Leukemia

Receptor tyrosine kinases have been implicated in the development and progression of many cancers, including both leukemia and solid tumors, and are attractive druggable therapeutic targets. Here we describe an efficient four-step strategy for pre-clinical evaluation of tyrosine kinase inhibitors (TKIs) in the treatment of acute leukemia. Initially, western blot analysis is used to confirm target inhibition in cultured leukemia cells. Functional activity is then evaluated using clonogenic assays in methylcellulose or soft agar cultures. Experimental compounds that demonstrate activity in cell culture assays are evaluated in vivo using NOD-SCID-gamma (NSG) mice transplanted orthotopically with human leukemia cell lines. Initial in vivo pharmacodynamic studies evaluate target inhibition in leukemic blasts isolated from the bone marrow. This approach is used to determine the dose and schedule of administration required for effective target inhibition. Subsequent studies evaluate the efficacy of the TKIs in vivo using luciferase expressing leukemia cells, thereby allowing for non-invasive bioluminescent monitoring of leukemia burden and assessment of therapeutic response using an in vivo bioluminescence imaging system. This strategy has been effective for evaluation of TKIs in vitro and in vivo and can be applied for identification of molecularly-targeted agents with therapeutic potential or for direct comparison and prioritization of multiple compounds.

Femoral Bone Marrow Aspiration in Live Mice

Serial sampling of the cellular composition of bone marrow (BM) is a routine procedure critical to clinical hematology. This protocol describes a detailed step-by-step technical procedure for an analogous procedure in live mice which allows for serial characterization of cells present in the BM. This procedure facilitates studies aimed to detect the presence of exogenously administered cells within the BM of mice as would be done in xenograft studies for instance. Moreover, this procedure allows for the retrieval and characterization of cells enriched in the BM such as hematopoietic stem and progenitor cells (HSPCs) without sacrifice of mice. Given that the cellular composition of peripheral blood is not necessarily reflective of proportions and types of stem and progenitor cells present in the marrow, procedures which provide access to this compartment without requiring termination of the mice are very helpful. The use of femoral bone marrow aspiration is illustrated here for cytological analysis of marrow cells, flow cytometric characterization of the hematopoietic stem/progenitor compartment, and culture of sorted HSPCs obtained by femoral BM aspiration compared with conventional marrow harvest.

This protocol describes a procedure for serial sampling of femoral bone marrow (BM) without requiring the sacrifice of mice. This procedure facilitates longitudinal studies of the BM composition of mice over time and provides serial access to cells within the BM for ex vivo and transplantation studies.

Isolation and Enrichment of Rat Mesenchymal Stem Cells (MSCs) and Separation of Single-colony Derived MSCs

MSCs are a population of adult stem cells that is a promising source for therapeutic applications. These cells can be isolated from the bone marrow and can be easily separated from the hematopoietic stem cells (HSCs) due to their plastic adherence. This protocol describes how to isolate MSCs from rat femurs and tibias. The isolated cells were further enriched against two MSCs surface markers CD54 and CD90 by magnetic cell sorting. Expression of surface markers CD54 and CD90 were then confirmed by flow cytometry analysis. HSC marker CD45 was also included to check if the sorted MSCs were depleted of HSCs. MSCs are naturally quite heterogeneous. There are subpopulations of cells that have different shapes, proliferation and differentiation abilities. These subpopulations all express the known MSCs markers and no unique marker has yet been identified for the different subpopulations. Therefore, an alternative approach to separate out the different subpopulations is using cloning cylinders to separate out single-colony derived cells. The cells derived from the single-colonies can then be cultured and evaluated separately.

In Vivo Imaging and Tracking of Technetium-99m Labeled Bone Marrow Mesenchymal Stem Cells in Equine Tendinopathy

Recent advances in the application of bone marrow mesenchymal stem cells (BMMSC) for the treatment of tendon and ligament injuries in the horse suggest improved outcome measures in both experimental and clinical studies. Although the BMMSC are implanted into the tendon lesion in large numbers (usually 10 - 20 million cells), only a relatively small number survive (&#60;10%) although these can persist for up to 5 months after implantation. This appears to be a common observation in other species where BMMSC have been implanted into other tissues and it is important to understand when this loss occurs, how many survive the initial implantation process and whether the cells are cleared into other organs. Tracking the fate of the cells can be achieved by radiolabeling the BMMSC prior to implantation which allows non-invasive in vivo imaging of cell location and quantification of cell numbers.
This protocol describes a cell labeling procedure that uses Technetium-99m (Tc-99m), and tracking of these cells following implantation into injured flexor tendons in horses. Tc-99m is a short-lived (t1/2 of 6.01 hr) isotope that emits gamma rays and can be internalized by cells in the presence of the lipophilic compound hexamethylpropyleneamine oxime (HMPAO). These properties make it ideal for use in nuclear medicine clinics for the diagnosis of many different diseases. The fate of the labeled cells can be followed in the short term (up to 36 hr) by gamma scintigraphy to quantify both the number of cells retained in the lesion and distribution of the cells into lungs, thyroid and other organs. This technique is adapted from the labeling of blood leukocytes and could be utilized to image implanted BMMSC in other organs.

Isolation and Culture of Bone Marrow Mesenchymal Stem Cells from the Human Mandible

Human mesenchymal stem cells (hMSCs) have shown great potential in bone regeneration, immune modulation, and treating refractory chronic diseases. Various origins have been found to obtain hMSCs recently, while bone marrow was still considered the main source. Bone marrow-derived MSCs (BMSCs) from different donor bone sites have distinct characteristics due to microenvironmental factors. Studies have shown that BMSCs from maxillofacial bone may have greater proliferative and osteogenic capacities than BMSCs from long bones or the iliac crest. And maxillofacial BMSCs were considered more suitable for stem cell therapy in the maxillofacial tissues. The mandible, especially the ascending ramal area with sufficient marrow, was a feasible donor site for harvesting BMSCs. This study described a protocol for harvesting, isolating, and culturing human mandibular bone marrow-derived MSCs (hmBMSCs). Furthermore, immunophenotyping of hmBMSCs, proliferation assays, and in vitro induction of osteogenic, adipogenic, and chondrogenic differentiation was performed to identify the cultured cells. Applying this protocol can help the researchers successfully obtain enough high-quality hmBMSCs, which is necessary for further studies of the biological function, microenvironmental effects, and clinical applications.

The present protocol describes an efficient procedure for isolating and culturing of human mandibular bone marrow-derived mesenchymal stem cells using the whole bone marrow adherence method. Isolation of Monocyte-Macrophage Lineage Cells from Rat Bones by Secondary Adherence Method

With a decrease of bone mineral density, bones are more likely to fracture, thus negatively affecting a patient's quality of life. The growth and development of bones are mainly regulated by osteoblasts and osteoclasts. It has been widely accepted that osteoclasts are derived from bone marrow monocyte-macrophage cells (BMMs). BMMs and other hematopoietic stem cells are located in the bone marrow cavity. Therefore, isolating single stable BMMs from different and heterogeneous cell populations is a huge challenge. Here we present a protocol for the isolation of BMMs from SD rats, called the secondary adherence method. Adherent cells cultured for 24-48 h in primary culture were collected. Flow cytometric analysis showed that approximately 37.94% of the cells were CD11b/c+ (monocyte-macrophage surface antigen). Tartrate resistant acid phosphatase (TRAP) staining and western blot analysis demonstrated that BMMs could differentiate into osteoclasts in vitro. The above findings suggested that the secondary adherence cells could be considered as a suitable cellular model for osteoclast differentiation research.

Isolation, Purification, and Differentiation of Osteoclast Precursors from Rat Bone Marrow

Osteoclasts are large, multinucleated, and bone-resorbing cells of the monocyte-macrophage lineage that are formed by the fusion of monocytes or macrophage precursors. Excessive bone resorption is one the most significant cellular mechanisms leading to osteolytic diseases, including osteoporosis, periodontitis, and periprosthetic osteolysis. The main physiological function of osteoclasts is to absorb both the hydroxyapatite mineral component and the organic matrix of bone, generating the characteristic resorption appearance on the surface of bones. There are relatively few osteoclasts compared to other cells in the body, especially in adult bones. Recent studies have focused on how to obtain more mature osteoclasts in less time, which has always been a problem. Several improvements in the isolation and culture techniques have developed in laboratories in order to obtain more mature osteoclasts. Here, we introduce a method that isolates bone marrow in less time and with less effort compared to the traditional procedure, using a special and simple device. With the use of density gradient centrifugation, we obtain large amounts of fully differentiated osteoclasts from rat bone marrow, which are identified by classical methods.

Technique for Isolation and Culture of Rat Jaw Bone Marrow Mesenchymal Stem Cells

Bone marrow mesenchymal stem cells (BMMSCs) are a type of stem cell with multi-directional differentiation potential. Compared with BMMSCs derived from appendicular bones, BMMSCs derived from the jaw have greater proliferative and osteogenic differentiation ability, gradually becoming important seed cells for jaw defect repair. However, the mandible has a complex bony structure and less cancellous content than appendicular bones. It is difficult to acquire a large number of high-quality jaw-derived marrow mesenchymal stem cells using traditional methods. This study presents a &#39;niche-based approach on stemness&#39; for isolating and culturing rat jaw bone marrow mesenchymal stem cells (JBMMSCs). Primary rat JBMMSCs were isolated and cultured using the whole bone marrow adherent method combined with the bone slice digestion method. The isolated cells were identified as JBMMSCs through cell morphology observation, detection of cell surface markers, and multi-directional differentiation induction. The cells extracted by this method exhibit a &#39;fibroblast-like&#39; spindle shape. The cells are long, spindle-shaped and fibroblast-like. The flow cytometry analysis shows these cells are positive for CD29, CD44, and CD90 but negative for CD11b/c, CD34, and CD45, which is congruent with BMMSCs characteristics. The cells show strong proliferation capacity and can undergo osteogenic, adipogenic, and chondrogenic differentiation. This study provides an effective and stable method for obtaining enough high-quality JBMMSCs with strong differentiation ability in a short time, which could facilitate further studies of the exploration of biological function, regenerative medicine, and related clinical applications.
