쥐 장 오가노이드 모델의 개발은 생체 내 장기에서 발견되는 중요한 기능적 특성을 보존하고 전임상 테스트, 약물 스크리닝 및 기능 분석을 위한 유망한 도구입니다. 이 in vitro 모델은 in vivo 전임상 위장병학 연구와 병행하여 사용될 수 있으며, 쥐는 더 큰 장 크기, 인간과 공유되는 생리학적 측면, 그리고 어떤 경우에는 더 나은 질병 모델이기 때문에 종종 선호되는 모델이다38. 여기에서는 쥐 장 크립트의 분리, 쥐 장 오가노이드의 생성 및 장기 배양, 기능적 포스콜린 팽창 분석, 전체 마운트 면역형광, 2D 단층 배양 및 렌티바이러스 유전자 조작을 포함한 다운스트림 응용 분야를 위한 강력한 단계별 프로토콜이 간략하게 설명되어 있습니다. 쥐 장 오가노이드는 마우스 모델의 병태생리학이 부적절하고 마우스 장 오가노이드에 비해 인간 장 생리학에 더 나은 모델을 제공할 수 있는 많은 질병 맥락과 관련이 있을 수 있습니다.
통과 및 확장이 가능한 수명이 긴 오가노이드 배양을 확립하려면 장 상피 증식을 유지하는 데 필요한 주요 성장 인자를 식별하는 것이 필수적입니다. 마우스 오가노이드는 EGF, R-spondin 및 Noggin의 간단한 칵테일에서 가장 자주 성장하지만 Noggin은 장내 오가노이드 배양에 필요하지 않은 것으로 보고되었습니다39. 조절된 배지는 재조합 성장 인자를 대체할 수 있으며, 가장 일반적으로 사용되는 세포주는 Wnt3a, Rspondin-3 및 Noggin39, L-Wnt3a 및 HA-Rspondin1-Fc 293T 세포(40)를 분비하는 L-WRN입니다. L-WRN 조절 배지는 마우스 장39 오가노이드 성장뿐만 아니라 개, 고양이, 닭, 말, 소, 양, 돼지를 포함한 여러 농장 동물 및 반려동물의 장 오가노이드 성장을 지원하기에 충분하다12. 그러나 인간의 장내 오가노이드는 확장 성장 단계(즉, 작은 스페로이드에서 큰 스페로이드로의 진행)와 분화 단계(즉, 분화된 세포 유형의 생성 및 성숙)에 대해 뚜렷한 배지 제형이 필요하기 때문에 성장 인자 요구 사항이 매우 다릅니다10. 쥐 장 오가노이드의 배지 요구 사항은 인간 장 오가노이드에 대한 확장 성장 배지의 요구 사항과 매우 유사하지만, 특히 쥐 오가노이드는 이 배지 환경에서 성장과 분화가 모두 가능하여 배양 요구 사항을 상당히 단순화합니다. 우리의 초기 시도는 L-WRN 조건 배지에서 쥐의 장 오가노이드를 확립하고 성장시키는 데 중점을 두었지만 장기적인 배양은 미약했고 쥐의 장 오가노이드 라인은 견고성 부족으로 어려움을 겪었습니다(데이터 표시 안 됨). 이는 L-WRN 세포주가 R-spondin 3을 분비하도록 조작된 반면, 여기서 권장하는 293T-Rspo1 세포주는 R-spondin 1을 분비하도록 설계되었기 때문일 수 있습니다. 쥐와 인간 오가노이드는 R-spondin 1을 선호할 수 있으며, 이는 잠재적으로 L-WRN 조건 배지에서 쥐 오가노이드 라인의 실패를 설명할 수 있습니다.
in vivo 설정을 가장 밀접하게 요약하려면 줄기 세포의 생존, 유지 및 증식을 허용하고 세포 회전율과 개별 세포 유형으로의 동시 분화 이벤트를 유지할 수 있는 오가노이드 배양 조건을 개발하는 것이 중요합니다. 따라서 재조합 단백질 및/또는 조건화된 배지 내 단백질의 농도는 이러한 완벽한 균형을 이루기 위해 엄격하게 적정되고 제어되어야 합니다. 특히, 최적의 Wnt 수치는 장내 오가노이드 배양물의 손실을 방지하는 데 필수적입니다. 조건화된 배지에 Wnt가 너무 적으면 성장을 지원할 수 없어 줄기 세포가 손실되고 오가노이드가 사멸됩니다. Wnt의 과활성화는 오가노이드를 낭포성 및 미분화 상태로 만든다10. 여기에 상세히 설명되지는 않았지만, Topflash 세포주(41)와 같은 Wnt 리포터 루시페라아제 분석을 사용하여 L-Wnt3a 및 293T-Rspo1 컨디셔닝된 배지의 각 배치를 테스트하는 것이 강력히 권장된다. 이전 연구에서는 L-Wnt3a 매체의 최적 배치가 1% L-Wnt3a 10에 비해 12.5%에서15배 신호 증가, 50%에서 300배 신호 증가를 초래해야 한다고 설명했습니다. 쥐 오가노이드는 배양 요구 사항, 특히 Wnt 활성화 수준에 마우스 오가노이드보다 더 민감하기 때문에 이러한 추가 품질 관리 단계는 쥐 오가노이드 배양의 견고성과 신뢰성을 촉진하는 데 큰 도움이 됩니다. Noggin 컨디셔닝 미디어에서 Bmp 활동 및 상대 Noggin 농도를 테스트하는 데 유사한 리포터 라인을 사용할 수 없기 때문에 Noggin 수준을 정확하게 제어하기 위해 가능한 경우 재조합 Noggin을 사용하는 것이 좋습니다. 마우스 장 오가노이드는 Noggin39가 없는 경우에도 성장 및 유지될 수 있지만 쥐 장 오가노이드 배양에는 시도되지 않았습니다.
세포 배양 요구 사항 외에도 쥐 오가노이드 라인의 성공적인 초기 확립은 크립트 분리 중 분화된 융모의 효율적인 고갈에 크게 좌우됩니다. 높은 수준의 빌라르 오염은 죽어가는 세포의 신호 또는 필수 요소의 격리로 인해 크립트 사멸을 유발합니다. 이러한 분화된 융모를 상피 제제에서 정확하고 일관되게 고갈시키려면 입체경을 사용하여 상피 분리를 수행하는 것이 좋습니다. 방출되는 상피를 육안으로 검사하면 PBS를 폐기하고 교체해야 하는 명확한 단서를 얻을 수 있습니다(그림 1). 융모가 충분히 고갈될 때까지 크립트를 수집해서는 안 됩니다. Villar 세포는 말단 분화되어 배양에서 오가노이드를 생성할 수 없습니다. 또한 쥐의 장 오가노이드를 통과시키고 다운스트림 응용 분야에 사용하려면 섬세한 관리가 필요합니다. 해리 시약에서 더 오랜 시간(10분) 동안 배양하면 상당한 세포 사멸과 오가노이드 라인의 손실이 발생합니다.
여기에서는 쥐 오가노이드에서 장 단층을 생성하기 위한 간단하고 빠른 프로토콜에 대해 설명합니다. EME와 콜라겐 I 기질은 상피에 서로 다른 영향을 미치며, 이는 연구 목적에 따라 활용될 수 있습니다. EME는 단일 셀의 빠르고 효율적인 접착과 셀 돌출부의 형성을 가능하게 합니다. 대조적으로, 표면을 콜라겐 I로 코팅하면 이러한 과정이 지연됩니다. 단층이 약 80%의 밀도에 도달하면 EME에서 성장한 세포가 다시 3D 오가노이드 구조를 생성하기 시작합니다. 그러나 지속적인 성장을 위한 충분한 물리적, 화학적 지원이 부족합니다. 이러한 오가노이드 상태로의 회귀는 EME의 단층을 50%-80%의 밀도로 유지함으로써 방지할 수 있습니다. 단층의 정점 표면에 희석된 EME를 추가하면 de novo 오가노이드의 빠른 회복 및 형성이 촉진되어 수렴 영역을 보다 빠르고 쉽게 생성할 수 있습니다. 콜라겐 I 표면에서 세포는 균일한 단층을 형성하고 작은 클러스터를 생성할 수 있습니다. 그러나 단층 위에 콜라겐 I을 추가하는 것만으로는 오가노이드 형성을 유도하기에 충분하지 않습니다. EME는 초기 오가노이드가 극복하기 위해 더 강한 기계적 저항이 있기 때문에 단층 표면에 추가할 때 희석해야 합니다. 그러나 이 희석된 EME는 대형 오가노이드의 강력한 형성을 허용하지 않습니다. 표면에서 자연적으로 분리되는 de novo 생성 쥐 오가노이드는 즉시 제거하고 희석되지 않은 EME로 옮겨 구조적 지지와 성장을 복원해야 합니다. 이 단계에서 오가노이드의 크기가 작기 때문에 강력한 성장이 확립될 때까지 오가노이드의 통과는 권장되지 않습니다. EME가 오가노이드의 재구성을 지원할 수 있는 이유에 대한 근본적인 생물학적 중요성은 있지만 콜라겐 I이 이를 수행할 수 있는지 여부는 명확하지 않습니다. 그러나 3D 콜라겐에서 성장한 세포는 싹이 트인 오가노이드42,43을 형성하거나 장기적인 유지를 지원할 수 없다는 보고가 있습니다. 상업적으로 이용 가능한 EME 제품은 세포외 단백질, 주로 라미닌 및 콜라겐 IV44의 이질적인 혼합물입니다. 그러므로, 단백질의 뚜렷한 구성과 상피 세포가 다른 세포 복합체를 사용하여 세포외 기질과 관여할 수 있는 능력은 EME에서 리모델링을 허용할 수 있지만 콜라겐 I에서는 허용하지 않을 수 있습니다. 콜라겐 I 유래 단층을 EME에 넣어 오가노이드 형성 및 성장을 지원할 수 있는지 여부는 테스트되지 않았습니다.
쥐 장 오가노이드 모델의 유전자 조작에 대해 설명하고, 3D 오가노이드의 렌티바이러스 transduction 및 2D 단층의 transient transfection에 대한 프로토콜을 간략하게 설명합니다. 렌티바이러스 오가노이드 transduction의 낮은 효율을 극복하기 위해 2D 단층의 transfection을 위한 프로토콜이 개발되었습니다. 단층의 평평한 형태와 노출된 정점 도메인은 바이러스 및 DNA 함유 복합체에 더 쉽게 접근할 수 있도록 합니다. pLJM1-EGFP 벡터를 이용한 EGFP 리포터의 발현은 이 기법의 검증을 위해 사용되었다. GFP 리포터 발현은 24시간 후에 관찰되었고, 단층에서 5-6일 동안 유지되었다. 단층의 렌티바이러스 형질도입에 초점을 맞춘 향후 연구는 3D 오가노이드 형질도입보다 효율성이 높을 것으로 보입니다. 위의 프로토콜을 사용하면 감염된 2D 단층에서 3D 오가노이드를 개질하여 안정적인 라인을 쉽게 생성할 수 있습니다. 쥐의 장내 오가노이드 계통은 여러 통로에서 안정적으로 유지되고, 동결 보존되고, 성공적으로 해동되고, 렌티바이러스 형질도입을 사용하여 유전자 변형되어 1년 이상 성공적으로 유지될 수 있으며, 이를 통해 인간과의 생리학적 관련성을 유지하는 접근 가능하고 다루기 쉬운 체외 장 오가노이드 모델의 필요성을 해결할 수 있습니다.

인간의 소장 상피 구조와 세포 구성은 생리적 기능을 반영하여 복잡합니다. 소장의 주요 역할은 내강을 통과하는 음식물로부터 영양분을 흡수하는 것이다1. 이 기능을 극대화하기 위해 장 표면은 흡수 표면적을 증가시키는 융모라고 하는 손가락 모양의 돌출부와 줄기세포를 수용하고 단열하는 크립트(crypt)라고 하는 컵 모양의 침출부로 구성됩니다. 상피 내에서는 다양한 특수 흡수 및 분비 세포 유형이 생성되어 뚜렷한 기능을 수행합니다1. 이러한 복잡성 때문에 고통로 형질전환된 불멸의 세포주에서 장과 같은 조직을 모델링하는 것은 어려웠습니다. 그러나 줄기세포, 특히 성체 줄기세포와 그 분화 메커니즘에 대한 연구는 3D 장 오가노이드 배양의 개발을 가능하게 했습니다. 오가노이드 모델의 사용은 부분적으로 일부 아키텍처 구성 요소의 재현과 온전한 장에서 발견되는 세포 유형 이질성으로 인해 이 분야를 변화시켰습니다. 장내 오가노이드는 활성 줄기세포 집단의 유지로 인해 in vitro 에서 장기간 배양할 수 있다2.
장내 오가노이드는 줄기세포 생물학, 세포 생리학, 유전 질환 및 영양 3,4을 연구하는 데 적합한 모델로 빠르게 자리잡았을 뿐만 아니라 새로운 약물 전달 방법을 개발하는 도구이기도 하다5. 또한 환자 유래 오가노이드는 질병 모델링, 약물 발견 및 맞춤형 치료 스크리닝에 활용되고 있습니다 6,7,8,9. 그러나 인간의 장내 오가노이드는 여전히 도전 과제를 안고 있습니다. 조직 가용성, 기관 검토 위원회 승인 요건 및 윤리적 문제로 인해 인체 샘플의 광범위한 사용이 제한됩니다. 또한, 장낭샘에서 생성된 인간 장 오가노이드는 미분화 줄기세포의 유지를 위해 또는 성숙한 세포 유형의 분화를 유도하기 위해 두 가지 뚜렷한 배양 조건을 필요로 한다10. 이는 줄기세포와 성숙한 분화 세포 유형이 동시에 존재하고 지속적으로 생성/유지되는 in vivo와 대조됩니다1. 반면에, 성장 인자의 덜 복잡한 칵테일에서 성장한 마우스 장 오가노이드는 배지 조성에서 이러한 전환을 필요로 하지 않으며 동일한 배지 컨텍스트에서 줄기 세포와 분화된 세포를 유지할 수 있습니다 2,11. 그러나 인간과 비교할 때 마우스 장의 주요 차이로 인해 많은 경우 마우스 오가노이드가 차선의 모델이 될 수 있습니다. 전반적으로, 말, 돼지, 양, 소, 개, 고양이를 포함한 대형 포유류의 많은 장 오가노이드는 인간 장 오가노이드의 배양 조건보다 마우스 장 오가노이드와 더 밀접하게 일치하는 배양 조건에서 성공적으로 생성되었습니다12. 마우스와 인간 오가노이드 간의 성장 인자 조건의 차이는 줄기 세포 틈새 구성의 차이와 줄기 세포 생존, 증식 및 유지에 대한 요구 사항의 차이를 반영할 수 있습니다. 따라서 1) 인간의 장내 세포 구성과 매우 유사하고, 2) 인간의 장 오가노이드와 같은 성장 인자 요구 사항을 가진 줄기세포를 포함하며, 3) 미분화 및 분화 구획을 지속적으로 유지할 수 있는 쉽게 접근할 수 있는 모델 오가노이드 시스템이 필요합니다. 이상적으로, 시스템은 일반적으로 사용되는 전임상 동물 모델에서 가져온 것으로, 생체 내 및 시험관 내 실험이 상호 연관되어 함께 사용될 수 있습니다.
랫트는 장 생리학 및 생화학이 인간과 매우 유사하기 때문에 장 생리학 및 약리학 연구에서 일반적으로 사용되는 전임상 모델이다13, 특히 장 투과성과 관련하여14. 생쥐에 비해 상대적으로 크기가 커서 수술 절차에 더 적합합니다. 돼지를 포함한 대형 동물 모델이 때때로 사용되지만, 쥐는 더 저렴한 모델이고, 사육을 위한 공간이 덜 필요하며, 쉽게 상업적으로 이용 가능한 표준 균주를 가지고 있다15. 쥐 모델 사용의 단점은 생체 내 연구를 위한 유전자 툴킷이 마우스에 비해 잘 개발되지 않았고 녹아웃, 넉인, 형질전환을 포함한 새로운 쥐 계통의 생성에 비용이 많이 든다는 것입니다. 강력한 쥐 장 오가노이드 모델의 개발 및 최적화는 인간과의 주요 생리학적 관련성을 유지하는 접근 가능한 모델에서 유전자 조작, 약리학적 치료 및 더 높은 처리량의 연구를 가능하게 할 것입니다. 그러나 한 설치류 오가노이드 모델과 다른 설치류 오가노이드 모델의 장점은 연구 중인 특정 과정이나 유전자에 따라 크게 달라집니다. 인간에서 발견되는 특정 유전자는 쥐의 유사 유전자일 수 있지만 쥐의 유사 유전자는 아닐 수 있다16,17. 또한 종 특이적 세포 아형은 단일 세포 RNAseq18,19,20에 의해 점점 더 밝혀지고 있습니다. 마지막으로, 쥐와 생쥐의 장 질환 모델은 종종 표현형에서 상당한 변화를 보이기 때문에21,22 인간에서 볼 수 있는 증상과 질병 과정을 보다 밀접하게 요약하는 모델을 다운스트림 작업을 위해 선택해야 합니다. 쥐 장 오가노이드 모델의 생성은 연구자가 자신의 상황에 가장 적합한 모델 시스템을 선택할 수 있는 추가적인 유연성과 선택권을 제공합니다. 여기서, 기존 프로토콜23,24는 쥐의 장 오가노이드 생성을 위해 확장되고, 십이지장 또는 제주눔에서 쥐의 장 오가노이드의 생성 및 유지를 위한 프로토콜이 개략적으로 설명된다. 또한 렌티바이러스 감염, 전체 마운트 염색 및 포스콜린 팽창 분석을 포함한 여러 다운스트림 응용 분야에 대해 설명합니다.

The human small intestinal epithelial architecture and cellular composition are complex, reflecting their physiological functions. The primary role of the small intestine is to absorb nutrients from food passing through its lumen1. To maximize this function, the intestinal surface is organized into finger-like protrusions called villi, which increase the absorptive surface area, and cup-like invaginations called crypts, which house and insulate the stem cells. Within the epithelium, various specialized absorptive and secretory cell types are generated to perform distinct functions1. Because of this complexity, it has been difficult to model tissues like the intestine in high passage-transformed immortalized cell lines. However, the study of stem cells, especially adult stem cells and their differentiation mechanisms, has allowed for the development of 3D intestinal organoid cultures. The use of organoid models has transformed the field, in part due to their recapitulation of some architectural components and cell type heterogeneity found in the intact intestine. Intestinal organoids can be cultured long-term in vitro due to the maintenance of the active stem cell population2.
Intestinal organoids have rapidly become an adaptable model to study stem cell biology, cell physiology, genetic disease, and nutrition3,4, as well as a tool to develop novel drug delivery methods5. In addition, patient-derived organoids are being utilized for disease modeling, drug discovery, and personalized treatment screening, among others6,7,8,9. However, human intestinal organoids still present challenges. Tissue availability, requirements for Institutional Review Board approval, and ethical issues limit the widespread use of human samples. Additionally, human intestinal organoids generated from intestinal crypts require two distinct culture conditions for the maintenance of undifferentiated stem cells or to induce the differentiation of mature cell types10. This contrasts with in vivo, where stem cells and mature differentiated cell types are simultaneously present and continuously generated/maintained1. On the other hand, mouse intestinal organoids, which are grown in a less complex cocktail of growth factors, do not require this switch in media composition and can maintain stem cells and differentiated cells in the same media context2,11. However, key differences in mouse intestine when compared to humans can make mouse organoids a suboptimal model in many instances. Overall, many intestinal organoids from larger mammals, including horses, pigs, sheep, cows, dogs, and cats, have been successfully generated in culture conditions more closely aligned with mouse intestinal organoids than the culture conditions of human intestinal organoids12. The differences in growth factor conditions between mouse and human organoids likely reflect differences in stem cell niche composition and differing requirements for stem cell survival, proliferation, and maintenance. Therefore, there is a need for an easily accessible model organoid system that 1) closely resembles human intestinal cell composition, 2) contains stem cells with growth factor requirements like those of human intestinal organoids, and 3) is capable of continuously maintaining undifferentiated and differentiated compartments. Ideally, the system would be from a commonly used preclinical animal model, such that in vivo and in vitro experiments can be correlated and used in tandem.
Rats are a commonly used preclinical model for intestinal physiology and pharmacology studies due to their very similar intestinal physiology and biochemistry to humans13, particularly with regards to intestinal permeability14. Their relatively larger size compared to mice makes them more amenable to surgical procedures. While large animal models, including pigs, are sometimes used, rats are a more affordable model, require less space for husbandry, and have readily commercially available standard strains15. A drawback of using rat models is that the genetic toolkit for in vivo studies is not well developed compared to mice, and the generation of novel rat lines, including knockouts, knock-ins, and transgenics, is often cost-prohibitive. The development and optimization of a robust rat intestinal organoid model would allow for genetic manipulation, pharmacological treatments, and higher throughput studies in an accessible model that retains key physiological relevance to humans. However, the advantages of one rodent organoid model versus another is highly dependent on the particular process or gene being studied; certain genes found in humans may be pseudogenes in mice but not rats16,17. Additionally, species-specific cell subtypes are increasingly being unveiled by single-cell RNAseq18,19,20. Finally, rat and mouse intestinal disease models often display considerable variations in phenotypes21,22 such that the model that more closely recapitulates the symptoms and disease process seen in humans must be selected for downstream work. The generation of a rat intestinal organoid model provides additional flexibility and choice for researchers in selecting a model system most appropriate for their circumstances. Here, existing protocols23,24 are expanded upon for the generation of rat intestinal organoids and a protocol is outlined for the generation and maintenance of rat intestinal organoids from the duodenum or jejunum. In addition, several downstream applications, including lentiviral infection, whole mount staining, and forskolin swelling assays, are described.

In this study we established a rat intestinal organoid model that is robust in long-term cell culture. We also demonstrate the first successful genetic manipulations using both lentiviral infection and transient transfection. This model is an excellent tool for manipulating intestinal biology in the rat where the genetic tools available in vivo are limited.Rat intestinal organoids have previously been difficult to culture long-term. By generating a robust, genetically-manipulatable rat organoid model, we are providing researchers additional flexibility and choice to select an organoid model best suited for their particular circumstances. Mouse intestinal organoids are used to study stem cell behavior, cell fate and physiology, but they can be a suboptimal model.Since there are key differences between mouse and human biology. On the other hand, human intestinal organoids can be difficult to obtain, and have complex cell culture requirements. The rat organoid model we describe here retains key physiological relevance to human biology, while being considerably easier to access and care for than human intestinal organoids.Developing and optimizing the rat intestinal organoid model allows for genetic manipulation, pharmacological treatments, and higher throughput studies of intestinal biology and accessible model with key physiological relevance to humans. Additionally, these rat organoids will allow for the study of species-specific physiology, cell fates, and phenotypic variations and disease models in the intestine.

3D 오가노이드 배양은 정상적인 장 구조의 일부를 재현하기 때문에 선호되는 경우가 많지만, 실시간 이미징, 형질 주입 및 렌티바이러스 형질 주입을 포함한 다른 접근 방식을 기술적으로 더 어렵게 만듭니다. 3D 오가노이드32 (그림 4)로부터 생성된 2D 단층을 사용하면 플라스미드를 더 효율적으로 도입할 수 있습니다. 3D 장 오가노이드는 전통적으로 일시적인 transfection에 내성이 있는 반면, EGFP를 암호화하는 plasmid는 지질 기반 transfection 방법을 사용하여 성공적으로 도입할 수 있습니다. PEI를 사용한 가장 비용 효율적인 접근법은 6.1단계(그림 5)에 요약되어 있지만, 전기천공법 및 상업적으로 이용 가능한 transfection 시약도 유사한 결과를 산출했습니다(데이터는 표시되지 않음). 향후 연구는 이러한 접근 방식이 CRISPR 구조를 단층에 도입하는 데 사용될 수 있는지 여부에 중점을 둘 것입니다.
형질주입 후 2D 단층에서 3D 오가노이드를 개질하여 크립트의 3D 아키텍처 구성 요소와 함께 통과 가능한 라인으로 유지할 수 있도록 하는 것이 중요했습니다. 흥미롭게도, EME에 도금된 2D 단층은 EME를 세포 상단에 다시 추가할 때 작은 스페로이드로 쉽게 재형성되는 반면, 콜라겐 I 기질은 3D 구조의 재구성에 충분하지 않았습니다(그림 6).
일시적인 형질주입은 많은 연구에서 유용하지만, 안정된 세포주를 형성하는 것이 더 유용한 경우가 많기 때문에 세포에 렌티바이러스를 도입해야 합니다. 쥐의 장내 오가노이드는 이전에 발표된 프로토콜을 수정하여 렌티바이러스에 감염되었습니다(그림 7). 프로토콜의 핵심 단계는 오가노이드를 작은 응집체 또는 세포 클러스터로 분할하는 것입니다. 배양이 효율적으로 파괴되지 않고 오가노이드가 손상되지 않은 상태로 유지되면 렌티바이러스 입자가 세포에 들어가지 않습니다. 감염 후 오가노이드는 회복되고 다시 자라야 합니다. 여기에 설명된 프로토콜은 선택 전에 세포의 10%-48%(평균: 19.4% ± 6.5%)까지 바이러스 입자를 흡수할 수 있도록 합니다.
오가노이드의 전체 마운트 염색은 EME 잔류물의 불완전한 제거 또는 불완전한 항체 침투로 인해 어려울 수 있습니다. 여기에 설명된 프로토콜은 오가노이드의 강력한 염색을 가능하게 합니다. 컨포칼 현미경에서 오가노이드를 시각화하는 것도 커버슬립에서 너무 멀리 떨어져 있는 경우 어려울 수 있습니다. VALAP을 사용하면 오가노이드가 커버슬립에 의해 찌그러지지 않고 커버슬립 가까이에 가라앉을 수 있도록 높이가 어느 정도 있는 웰이 생성되어 이미징이 용이합니다. F-actin을 표지하기 위한 정점 음이온 채널 낭성 섬유증 막횡단 전도도 조절제(CFTR) 및 팔로이딘에 대한 대표적인 염색은 그림 8에 나와 있습니다.
마지막으로, 오가노이드는 기능 분석에서 유용합니다. 낭포성 섬유증 환자의 환자 유래 오가노이드는 cAMP 작용제 포스콜린을 사용한 치료가 오가노이드팽창 29,33-37을 유발하는 강력한 CFTR 매개 유체 분비를 유도하기 때문에 CFTR 기능을 선별하는 데 사용되었습니다. 이 연구의 목표 중 하나는 생체 내 전임상 연구와 병행하여 사용할 수 있는 오가노이드 모델을 식별하고 개발하는 것이었습니다. 따라서 쥐의 장 오가노이드가 포스콜린에 의한 팽창을 겪는지 여부를 확인하는 것을 목표로 했습니다. 실제로, 포스콜린 처리 후 30분 이내에 쥐 오가노이드가 부풀어 올랐고, 120분 동안 최대 효과가 관찰되었습니다(그림 9).

When using organoids to assess physiology and cell fate decisions, it is important to use a model that closely recapitulates in vivo contexts. Accordingly, patient-derived organoids are used for disease modeling, drug discovery, and personalized treatment screening. Mouse intestinal organoids are commonly utilized to understand aspects of both intestinal function/physiology and stem cell dynamics/fate decisions. However, in many disease contexts, rats are often preferred over mice as a model due to their greater physiological similarity to humans in terms of disease pathophysiology. The rat model has been limited by a lack of genetic tools available in vivo, and rat intestinal organoids have proven fragile and difficult to culture long-term. Here, we build upon previously published protocols to robustly generate rat intestinal organoids from the duodenum and jejunum. We provide an overview of several downstream applications utilizing rat intestinal organoids, including functional swelling assays, whole mount staining, the generation of 2D enteroid monolayers, and lentiviral transduction. The rat organoid model provides a practical solution to the need of the field for an in vitro model which retains physiological relevance to humans, can be quickly genetically manipulated, and is easily obtained without the barriers involved in procuring human intestinal organoids.

Here, we present a protocol to generate rat intestinal organoids and use them in several downstream applications. Rats are often a preferred preclinical model, and the robust intestinal organoid system fills the need for an in vitro system to accompany in vivo studies.

Establishment and Passaging of Small Intestine Organoids

To begin, place the euthanized rat onto the dissection surface with the ventral side up. Pinch the skin layer with sterile forceps and make cuts at the surface level, avoiding damage to internal organs. Using large sharp dissection scissors, make a longitudinal surface level cut in the center of the abdomen.Then stemming from that cut, make two shorter horizontal cuts one on each side. Using forceps, peel the skin away to expose the abdominal cavity. Cut through the peritoneal membrane to fully expose the internal organs in the abdominal cavity with ready access to the intestine.Using scissors and forceps, locate the stomach and identify the duodenum, approximately two to three centimeters distal to it which appears as a yellowish segment. The proximal jejunum is located approximately four to five centimeters distal to the ligament of Treitz. A landmark between the duodenum and jejunum.Place the isolated intestinal fragment into a 10 centimeter Petri dish. Flush the desired intestinal segment of mesentery with 10 milliliters of ice cold PBS until cleared of luminal contents. On a paper towel, cut the intestinal segment into two centimeter long pieces.Then open each intestinal piece longitudinally to expose the epithelium. Scrape the exposed luminal surface using a glass microscope slide to remove villi. Place the intestinal pieces into the EDTA solution on ice and rotate it four degrees Celsius for 30 minutes on a tube revolver set to 10 RPM.At the dissecting microscope, pour the contents of the tube into a 10 centimeter Petri dish. Add in an additional five milliliters of ice cold PBS. Using fine forceps, hold an intestinal segment and shake vigorously to observe the epithelium release into the PBS.Initially, the PBS will contain mostly villi. Shake the intestinal fragments continuously, periodically discard the PBS containing villi, and add 10 milliliters of fresh PBS to the intestinal fragments. Continue to shake the fragments and repeat the PBS washing step until villi are no longer released into the PBS.But instead, the PBS primarily contains crypts. Discard the remaining intestinal fragments and enrich the remaining PBS in the Petri dish for intestinal crypts. In a tissue culture hood, collect the PBS containing crypts and filter through a 70 micron cells drainer.Centrifuge the filtrate at 250 x g for five minutes. Then remove their supernatant and resuspend the pellet in five millimeters of advanced DMEM plus. Centrifuge again at 250 x g for five minutes.And remove the supernatant, leaving 50 microliters of media with the pellet. Resuspend the pellet in the remaining media and add it to the aliquot of the extracellular matrix extract or EME on ice. Gently pipet up and down to suspend the crypts evenly throughout the EME avoiding making bubbles.Place 50 microliters of the EME domes into a 35 millimeter tissue culture dish. An incubated 37 degrees Celsius with 5%carbon dioxide for 20 minutes. After incubation, add a two milliliters of rat intestinal organoid media, or RIOM, containing 10 micromolar each of Y27632 and CHIR99021.Next, to passage rat intestinal organoids, aspirate the medium from a plate containing organoids and add one milliliter of dissociation reagent to release the organoids from the EME domes. Transfer the dissociation reagent with fragmented organoids to a 15 milliliter conical tube. Wash the culture plate with two milliliters of advanced DMEM plus and add it to the 15 milliliter conical tube containing the fragmented organoids.Using a glass pasture pipette, gently pipette up and down 15 to 20 times to fragment the organoids. Centrifuge the organoids at 350 x g for two minutes and add 50 microliters of solution from the bottom of the tube into the EME. After mixing the solution, plate 50 microliters of the EME domes into a 35 millimeter tissue culture dish and incubate at 37 degrees Celsius with 5%carbon dioxide for 20 minutes.After incubation, add two milliliters of RIOM containing 10 micromolar each of Y27632 and CHIR99021. Representative images of villi fragments and crypts are shown. Crypts are smaller in size compared to villi.Plated crypts expand over the next few days and begin to bud and differentiate by days four to seven. After two days of thawing, a healthy organoid culture showed the presence of both spheroids and budded organoids. The same organoid line after passaging showed the presence of single crypt-like domains.

Generation of Rat Intestinal 2D Monolayers from 3D Organoids

To coat the plate with EME, dilute EME 1 to 20 in cold Advanced DMEM+For coating with collagen, dilute five milligrams per milliliter of collagen I in Advanced DMEM+to 100 micrograms per milliliter. Coat with the plate with 200 microliters of diluted EME or collagen to cover the well surface entirely and incubate for one to two hours at 37 degrees Celsius. To generate monolayers, aspirate the media from a 35 millimeter plate containing organoids.Add one milliliter of PBS and disrupt the EME in the wells with a P1000 tip, pipetting up and down approximately 20 times to loosen all the EME. Transfer the contents to a 15 milliliter conical tube. Add one milliliter of PBS to the plate to collect additional organoids and transfer them to the same conical tube.Centrifuge the sample at 350 G for two minutes and remove the supernatant, including any EME residue. Then, add one milliliter of trypsin solution to the organoid pellet and incubate at 37 degrees Celsius for two minutes. Pipette up and down 10 times using a P1000 tip and add two milliliters of Advanced DMEM+to neutralize trypsin.Centrifuge at 350 G for five minutes and aspirate the supernant before resuspending the pellet in 4.8 milliliters of rIOM2D. Remove excess EME or collagen in Advanced DMEM+from the wells. Then, add 200 microliters of organoids in rIOM2D and 10 micromolar Y27632 to each pre-coated well.After 4 to 16 hours, collect the media and centrifuge at 1, 000 G for one minute. Transfer the supernatant into a new 15 milliliter conical tube and discard the pellet. Wash each well with 300 microliters of PBS before adding 200 microliters of the centrifuged rIOM2D to each well.Change the rIOM2D every two to three days, suspending the use of Y27632.2D monolayers plated on the EME readily reformed into small spheroids when EME was added back to the top of the cells. In contrast, a collagen I substrate was insufficient to reform 3D structures.
