A demanda por anticorpos que atendam às necessidades de ambas as aplicações básicas e clínicas de pesquisa é alto e vai aumentar drasticamente no futuro. No entanto, é evidente que as tecnologias tradicionais não são monoclonais sozinho até esta tarefa. Isto conduziu ao desenvolvimento de métodos alternativos para satisfazer a procura de alta qualidade e reagentes de afinidade renováveis ​​para todos os elementos acessíveis do proteoma. Para este fim, os métodos de alto rendimento para a realização de seleções a partir de bibliotecas de anticorpos sintéticos exibido em fagos foram concebidos para aplicações que envolvem diversos antígenos e otimizado para o rendimento e sucesso rápido. Nisto, um protocolo é descrito em detalhe que ilustra com demonstração de vídeo a selecção de clones paralelo Fab-fago a partir de bibliotecas de alta diversidade contra centenas de alvos quer utilizando um manipulador de líquidos 96 canal manual ou sistema de robótica automatizado. Usando este protocolo, um único usuário pode gerar centenas de antígenos,selecionar anticorpos para-los em paralelo e validar a ligação do anticorpo dentro de 6-8 semanas. Destacam-se: i) um formato de antígeno viável, ii) caracterização antígeno pré-selecção, iii) passos críticos que influenciam a seleção de clones específicos de afinidade e alta, e iv) formas de eficácia seleção monitoramento e fase inicial de anticorpos clone caracterização. Com esta abordagem, temos obtido os fragmentos de anticorpos sintéticos (FAB) para muitas classes alvo, incluindo os receptores de uma só passagem de membrana, hormônios proteína secretada e proteínas intracelulares multi-domínio. Estes fragmentos são facilmente convertidos em anticorpos de comprimento total e foram validados para apresentar uma elevada afinidade e especificidade. Além disso, eles têm sido demonstrados para ser funcional em uma variedade de imunoensaios convencionais, incluindo Western blot, ELISA, imunofluorescência celular, ensaios de imunoprecipitação e afins. Esta metodologia irá acelerar a descoberta de anticorpos e, finalmente, trazer-nos mais perto de realizar o objetivo of gerar anticorpos qualidade renováveis, elevadas para o proteoma.
