Streptococcus agalactiae, groep B Streptococcus (GBS), een ingekapselde, Gram-positieve bacterie die vaak wordt geïsoleerd uit de darm en urogenitale tractus van gezonde volwassenen. In de jaren 1970, GBS naar voren als de belangrijkste agent van besmettelijke neonatale sterfte, met meer dan 7.000 gevallen van neonatale ziekte jaarlijks 1. Early-onset GBS-infectie (EOD) komt in de eerste uren of dagen van het leven, ontstaat als longontsteking of ademhalingsproblemen, en vaak ontwikkelt tot sepsis, terwijl de late vorm van de ziekte (LOD) ontstaat na enkele maanden en presenteert met bacteriëmie, die vaak voorschotten aan meningitis 2. Tal van biologische en sociaal-economische risicofactoren voor GBS vaginale kolonisatiegeïdentificeerd. Biologische factoren omvatten gastrointestinale GBS kolonisatie en afwezigheid van Lactobacillus in de darm. Echter, etniciteit, obesitas, hygiëne en seksuele activiteit ook in verband gebracht met GBS vaginale wagen 7. Hoewel bekend voor het veroorzaken neonatale infecties GBS veroorzaakt ook verschillende maternale infecties zowel peripartum en postpartum. GBS slede is verhoogd bij vrouwen presenteren met vaginitis 8 en in sommige gevallen zelfs het ziektebeeld 9 zijn. Tot voor kort, de meeste werkzaamheden onderzoeken GBS interacties met de cervicovaginal tract werd alleen in vitro celmodellen 13-15. In 2005 werd op korte termijn GBS vaginale kolonisatie gemodelleerd bij muizen om de werkzaamheid van een faag lytisch enzym te onderzoeken tot vaginale GBS behandelen gedurende een periode van 24 uur 23. Enkele jaren later, werd een lange-termijn GBS vaginale kolonisatie muismodel ontwikkeld om gastheer en bacteriële factoren die GBS doorzettingsvermogen te bestuderen. Dit model heeft talrijke GBS factoren die kolonisatie, waaronder oppervlak appendages 17,18 en GBS tweecomponentensystemen 19,24 geïdentificeerd. Dit model heeft bijgedragen tot de identificatie van gastheer respons mechanismen 19,25 en werd gebruikt om verschillende therapeutische strategieën, met inbegrip van immunomodulerende peptiden 26 en probiotica 27 te testen. Dit protocol geeft de nodige begeleiding om GBS te enten in de muis vaginale-darmkanaal en vervolgens kolonisatie te volgen en het verzamelen van monsters voor verdere analyse.

Streptococcus agalactiae (groep B streptococcus, GBS) is een Gram-positieve, asymptomatische kolonisator van het menselijke maagdarmkanaal en vaginale kanaal van 10 - 30% van de volwassenen. In immuun-gecompromitteerde individuen, met inbegrip van pasgeborenen, zwangere vrouwen en ouderen, kan GBS overschakelen naar een invasieve ziekteverwekker die aan sepsis, artritis, longontsteking en hersenvliesontsteking. Dit protocol beschrijft een techniek voor zij hardnekkige GBS vaginale kolonisatie met één β-estradiol voorbehandeling en dagelijks bemonstering bacteriële loa bepalend.
