Mikrodissezierten Explantation Kulturen lassen sich zuverlässig und reproduzierbar zu isolieren Zellpopulationen mit einem sehr hohen Anteil (~ 85%) der proliferative Myf-5 positive Skelettmuskulatur Stammzellen (SMSC) werden. Kultivierte embryonale und adulte Skelettmuskelzellen haben eine Reihe von verschiedenen Anwendungen. Der Nettoeffekt aller Komponenten des Wachstums kann durch Messung der Explantation Auswuchs Preise festgelegt werden. Die Mikro-Explantat-Methode kann verwendet werden, um primäre Kulturen aus einer breiten Palette verschiedener Muskelgruppen Schularten und Altersstufen zu etablieren und, wie hier beschrieben, wurde von den Autoren angepasst worden, um die Isolierung von embryonalen Skelettmuskelzellen Vorstufen zu ermöglichen. Einzigartig sind Mikro-Explantat-Kulturen verwendet worden, um daraus klonalen (einzelne Zelle Herkunft) Skelettmuskulatur Stammzellen (SMSC) Linien, die ausgebaut werden und für in vivo-Transplantation kann. In vivo transplantiert SMSC verhalten, als funktionelle, Gewebe-spezifische, Satelliten-Zellen die dazu beitragen, Skelettmuskel-Faser Regeneration, aber die auch beibehalten werden (in der Satelliten-Zell-Nische) als eine kleine Gruppe von undifferenzierten Stammzellen, die in Kultur re-isoliert werden können mit Hilfe der Mikro-Explantat-Methode.
