1) Einführung Motor Neurone mit einer Muskelfaser an Synapsen, die gemeinsam als neuromuskulären Synapse (NMJ) bezeichnet kommunizieren. NMJs kann leicht in den meisten Krebse Muskel Vorbereitungen zugegriffen werden. Viele der Krebse NMJs demonstrieren nicht spiking exzitatorischen postsynaptischen Potentiale (EPSP) ähnlich wie die abgestufte elektrische Signale in postsynaptischen Dendriten innerhalb der Säuger-ZNS-oder unterschwelligen Reaktionen in Vertebraten NMJs angemerkt erzeugt (Wiersma &amp; Van Harreveld, 1938; Katz &amp; Kuffler, 1946) . Die Krebse NMJs kann als grundlegende synaptischen Modelle dienen dazu, allgemeine Einsichten in die synaptische Übertragung und synaptischen Differenzierung. Im Allgemeinen regeln motorischen Einheiten Aspekte des Verhaltens von Tieren über die Art der synaptischen Kommunikation NMJs und Eigenschaften der Muskeln. Seit der ersten Beobachtung von &quot;schnell&quot; und &quot;langsame&quot; Muskelkontraktionen in Krabben und Krebse näher Muskel (Lucas, 1907, 1917), hat ähnliche muskuläre Kontraktion Differenzierung in andere Krebse Muskel-Typen wie Bauch-Beuger (Kennedy &amp; Takeda, 1965a beschrieben worden , b) Leib und Strecker (Van Harreveld &amp; Wiersma, 1936). &quot;Fast&quot; Wehen einleiten schnellen Antworten. Myofibrilläre Protein-Isoform-Expression ist auch in kontraktilen Unterschiede wichtig, aber in Vorbereitung wie die des Beines Streckmuskel, in dem eine bestimmte Faser, die durch beide Arten von Motoneuronen innerviert ist, liegt der Fokus auf die synaptische Unterschiede der Terminals, da die Terminals Aktien die gleiche Zielzelle (Mykles et al., 2002). Eine frühere Studie untersuchten die beiden erregenden motorischen Axone des Beines extensor und beschrieb die phasischen und tonischen Phänotypen (Bradacs et al., 1997). In diesem Bericht zeigen wir, wie man die Zerlegung durchführen und erhalten Aufnahmen, so dass andere weiter zu untersuchen, können die Eigenschaften der synaptischen Differenzierung dieser Nervenendigungen. Betrachtet mit Transmissions-Elektronenmikroskopie, verschiedene Reihe von Abschnitten aus der tonische und phasische Terminals auf Krebse Bein Streckmuskel erhalten ergaben, dass Tonic Terminals mehr RRP Vesikel als phasische Terminals enthalten, die Mitochondrien sind häufiger in tonische Neuronen und Synapsen auf phasische Terminals sind komplexer als die der Low-Output-Synapsen, da sie enthalten mehrere aktive Zonen mit unterschiedlichen Abständen (Miller et al, 2002; Johnstone et al, 2008;. King et al, 1996;.. Bradacs et al, 1997). Die Low-Output-Tonic-Terminals sind auch anfälliger für die Verbesserung der synaptischen Übertragung mit dem Neuromodulator Serotonin (5-HT) als die phasischen Terminals (Cooper et al., 2003). Die Tatsache, dass die tonische und phasische NMJ auf dem gleichen Muskelfasern sind erleichtert präsynaptischen Unterschiede auf einer bestimmten Muskelfaser zu bewerten und zu Fragen der Muskelermüdung, synaptische Depression und synaptische Übersprechen Adresse. Verschiedene Fragen müssen noch in diesem Präparat behandelt werden, wie zum Beispiel, ob es Unterschiede in der postsynaptischen Rezeptor-Dichte und Glutamat-Rezeptor-Subtypen in postsynaptischen Ziele für die tonische und phasische Terminals, sondern ein besseres Verständnis der grundlegenden Unterschiede in der Anatomie und Physiologie dieser zwei Motor-Einheiten werden in den Aufbau einer tieferen Wissensbasis zu unterstützen. Die Hoffnung ist, dass die grundlegenden Prinzipien in dieser synaptischen Vorbereitung gelernt werden auf andere Synapsen in verschiedenen Zubereitungen und zukünftige Untersuchungen in diesem synaptischen Modell des Krebses zu erhöhen. 2) Die Methoden <ol><li> Alle Experimente werden auf der ersten oder zweiten Laufbeinen mittelständischer Krebse (Procambarus clarkii) durchgeführt. Die Tiere werden einzeln in Kunststoff-Behälter mit oxygenized Wasser untergebracht. Die Temperatur des Tieres Zimmer liegt im Bereich von 13 ° C-16 ° C. Die Zubereitung ist in Kochsalzlösung gelegt. Die Dissektion Gericht sollte eine Sylgard (Dow Corning)-Beschichtung auf der Unterseite (1 cm dick). Die mediale Fläche des Muskels wird in dieser Studie verwendet. Die Trennung der Nerven auf die Extensoren von den Hauptlauf Nerv kann durch leichtes Ziehen am distalen Stumpf der Hauptlauf Nerven an der Seite des Präparats erhöht werden. Beim Schälen der Hauptlauf Nerven wieder über die Extensoren, können kleine Äste von Axon müssen geschnitten werden. Diese sind Zweige von den hemmenden motorischen Neurons auf die Extensoren. Stimulation bei 70 Hz ist die Tonika Axon angewandt, um eine erleichterte Reaktion für die geringe Leistung NMJs zu fördern, oder ein einzelner Impuls (1 Hz) ist die phasische Axon angewendet, um große EPSPs der hohe Leistung zu erhalten NMJs wie gezeigt in Abbildung 12. Abbildung 14 zeigt einen kontinuierlichen 5 Hz Stimulation Drücken der NMJ über 30 min. Die quantal EPSPs direkt über identifizierbare Regionen der Nervenendigung werden, indem das Lumen eines Makro-patch Aufnahme Elektrode auf die synaptische Krampfadern aufgezeichnet visualisiert mit den vitalen Farbstoff 4-Di-2-Asp (5 uM, 5-min-Behandlung, Cooper et al, 1995;. Magrassi et al, 1987).. Die spontane sowie evozierte quantal Antworten können entlang der Nervenendigungen aufgenommen werden. Die evozierte und spontane synaptische Potentiale sind mit der Makro-Patch-Elektrode (Dudel, 1981; Wojtowicz et al, 1991;. Mallart, 1993) erfasst. Kimax Glas (Außendurchmesser: 1,5 mm) gezogen wurde und Feuer-poliert, um Patch-Tipps mit Innen-Durchmessern von 10 bis 20 um (Abbildung 15) zu produzieren. <img src="/files/ftp_upload/2319/2319fig15.jpg" alt="Abbildung 15" /> Abbildung 15: Das Lumen eines Makro-patch Aufnahme Elektrode .. Das Lumen der Elektrode ist mit dem Bade Medium gefüllt ist. Der Verstärker ist die gleiche wie die für die intrazelluläre Aufnahme oben genannten verwendet. Elektrode und Dichtung Widerstand kann durch das Bestehen der Prüfung Stromimpulse durch die Elektrode bestimmt werden. In unseren Experimenten lagen Abdichtwiderstande 0,3 bis 1,0 MOhm und die Elektrode Widerstand reichte von 0,5 bis 1,0 MOhm. Seal Widerstand kann während der Aufnahme kontrolliert werden. </li><li> Direkte Zählung der quantal Ereignisse mit geringer Stimulation Frequenzen möglich. Für jede evozierte, kann die Anzahl der quantal Ereignisse ohne weiteres für die geringe Ausgangsklemmen bestimmt werden (Figure 16). Diese direkten zählt helfen Schätzung der mittleren quantal Inhalt (Del Castillo &amp; Katz, 1954;. Cooper et al, 1995). Membrane Potentials, Synaptic Responses, Neuronal Circuitry, Neuromodulation and Muscle Histology Using the Crayfish: Student Laboratory Exercises

The purpose of this report is to help develop an understanding of the effects caused by ion gradients across a biological membrane. Two aspects that influence a cell's membrane potential and which we address in these experiments are: (1) Ion concentration of K+ on the outside of the membrane, and (2) the permeability of the membrane to specific ions. The crayfish abdominal extensor muscles are in groupings with some being tonic (slow) and others phasic (fast) in their biochemical and physiological phenotypes, as well as in their structure; the motor neurons that innervate these muscles are correspondingly different in functional characteristics. We use these muscles as well as the superficial, tonic abdominal flexor muscle to demonstrate properties in synaptic transmission. In addition, we introduce a sensory-CNS-motor neuron-muscle circuit to demonstrate the effect of cuticular sensory stimulation as well as the influence of neuromodulators on certain aspects of the circuit. With the techniques obtained in this exercise, one can begin to answer many questions remaining in other experimental preparations as well as in physiological applications related to medicine and health. We have demonstrated the usefulness of model invertebrate preparations to address fundamental questions pertinent to all animals.

The experiments demonstrate an easy approach for students to gain experience in examining muscle structure, synaptic responses, the effects of ion gradients and permeability on membrane potentials. Also, a sensory-CNS-motor-muscle circuit is presented to show a means to test effects of compounds on a neuronal circuit.

Muscle Receptor Organs in the Crayfish Abdomen: A Student Laboratory Exercise in Proprioception

The primary purpose of this experiment is to demonstrate primary sensory neurons conveying information of joint movements and positions as proprioceptive information for an animal. An additional objective of this experiment is to learn anatomy of the preparation by staining, dissection and viewing of neurons and sensory structures under a dissecting microscope. This is performed by using basic neurophysiological equipment to record the electrical activity from a joint receptor organ and staining techniques. The muscle receptor organ (MRO) system in the crayfish is analogous to the intrafusal muscle spindle in mammals, which aids in serving as a comparative model that is more readily accessible for electrophysiological recordings. In addition, these are identifiable sensory neurons among preparations. The preparation is viable in a minimal saline for hours which is amenable for student laboratory exercises. The MRO is also susceptible to neuromodulation which encourages intriguing questions in the sites of modulatory action and integration of dynamic signals of movements and static position along with a gain that can be changed in the system.

Electrophysiological Methods for Recording Synaptic Potentials from the NMJ of Drosophila Larvae

In this video, we describe the electrophysiological methods for recording synaptic transmission at the neuromuscular junction (NMJ) of Drosophila larva. The larval neuromuscular system is a model synapse for the study of synaptic physiology and neurotransmission, and is a valuable research tool that has defined genetics and is accessible to experimental manipulation. Larvae can be dissected to expose the body wall musculature, central nervous system, and peripheral nerves. The muscles of Drosophila and their innervation pattern are well characterized and muscles are easy to access for intracellular recording. Individual muscles can be identified by their location and orientation within the 8 abdominal segments, each with 30 muscles arranged in a pattern that is repeated in segments A2 - A7. Dissected drosophila larvae are thin and individual muscles and bundles of motor neuron axons can be visualized by transillumination1. Transgenic constructs can be used to label target cells for visual identification or for manipulating gene products in specific tissues. In larvae, excitatory junction potentials (EJP&#8217;s) are generated in response to vesicular release of glutamate from the motoneurons at the synapse. In dissected larvae, the EJP can be recorded in the muscle with an intracellular electrode. Action potentials can be artificially evoked in motor neurons that have been cut posterior to the ventral ganglion, drawn into a glass pipette by gentle suction and stimulated with an electrode. These motor neurons have distinct firing thresholds when stimulated, and when they fire simultaneously, they generate a response in the muscle. Signals transmitted across the NMJ synapse can be recorded in the muscles that the motor neurons innervate. The EJP&#8217;s and minature excitatory junction potentials (mEJP&#8217;s) are seen as changes in membrane potential. Electrophysiological responses are recorded at room temperature in modified minimal hemolymph-like solution2 (HL3) that contains 5 mM Mg2+ and 1.5 mM Ca2+. Changes in the amplitude of evoked EJP&#8217;s can indicate differences in synaptic function and structure. Digitized recordings are analyzed for EJP amplitude, mEJP frequency and amplitude, and quantal content.

Here we describe electrophysiological methods for measuring synaptic transmission at the neuromuscular junction of Drosophila larva. Evoked release is initiated artificially by stimulating the motor neuron axons, and transmission through the NMJ can be measured by the postsynaptic response evoked in the muscle.

Dissecting and Recording from The C. Elegans Neuromuscular Junction

Neurotransmission is the process by which neurons transfer information via chemical signals to their post-synaptic targets, on a rapid time scale. This complex process requires the coordinated activity of many pre- and post-synaptic proteins to ensure appropriate synaptic connectivity, conduction of electrical signals, targeting and priming of secretory vesicles, calcium sensing, vesicle fusion, localization and function of postsynaptic receptors and finally, recycling mechanisms. As neuroscientists it is our goal to elucidate which proteins function in each of these steps and understand their mechanisms of action. Electrophysiological recordings from synapses provide a quantifiable read out of the underlying electrical events that occur during synaptic transmission. By combining this technique with the powerful array of molecular and genetic tools available to manipulate synaptic proteins in C. elegans, we can analyze the resulting functional changes in synaptic transmission. 
The C. elegans NMJs formed between motor neurons and body wall muscles control locomotion, therefore, mutants with uncoordinated locomotory phenotypes (known as unc s) often perturb synaptic transmission at these synapses 1. Since unc mutants are maintained on a rich supply of a bacterial food source, they remain viable as long as they retain some pharyngeal pumping ability to ingest food. This, together with the fact that C. elegans exist as hermaphrodites, allows them to pass on mutant progeny without the need for elaborate mating behaviors. These attributes, coupled with our recent ability to record from the worms NMJs 2,3,7 make this an excellent model organism in which to address precisely how unc mutants impact neurotransmission. 
The dissection method involves immobilizing adult worms using a cyanoacrylic glue in order to make an incision in the worm cuticle exposing the NMJs. Since C. elegans adults are only 1 mm in length the dissection is performed with the use of a dissecting microscope and requires excellent hand-eye coordination. NMJ recordings are made by whole-cell voltage clamping individual body wall muscle cells and neurotransmitter release can be evoked using a variety of stimulation protocols including electrical stimulation, light-activated channel-rhodopsin-mediated depolarization 4 and hyperosmotic saline, all of which will be briefly described.

Application of electrophysiology to accessible synapses provides a quantifiable measure of synaptic activity, useful in analyzing synaptic mutants. This article describes a dissection method used to expose the neuromuscular junctions (NMJ) of Caenorhabditis elegans (C. elegans) and briefly discusses some of the uses to which this preparation can be applied.

Simultaneous Intracellular Recording of a Lumbar Motoneuron and the Force Produced by its Motor Unit in the Adult Mouse In vivo

The spinal motoneuron has long been a good model system for studying neural function because it is a neuron of the central nervous system with the unique properties of (1) having readily identifiable targets (the muscle fibers) and therefore having a very well-known function (to control muscle contraction); (2) being the convergent target of many spinal and descending networks, hence the name of "final common pathway"; and (3) having a large soma which makes it possible to penetrate them with sharp intracellular electrodes. Furthermore, when studied in vivo, it is possible to record simultaneously the electrical activity of the motoneurons and the force developed by their muscle targets. Performing intracellular recordings of motoneurons in vivo therefore put the experimentalist in the unique position of being able to study, at the same time, all the compartments of the "motor unit" (the name given to the motoneuron, its axon, and the muscle fibers it innervates1): the inputs impinging on the motoneuron, the electrophysiological properties of the motoneuron, and the impact of these properties on the physiological function of the motoneurons, i.e. the force produced by its motor unit. However, this approach is very challenging because the preparation cannot be paralyzed and thus the mechanical stability for the intracellular recording is reduced. Thus, this kind of experiments has only been achieved in cats and in rats. However, the study of spinal motor systems could make a formidable leap if it was possible to perform similar experiments in normal and genetically modified mice.
For technical reasons, the study of the spinal networks in mice has mostly been limited to neonatal in vitro preparations, where the motoneurons and the spinal networks are immature, the motoneurons are separated from their targets, and when studied in slices, the motoneurons are separated from most of their inputs. Until recently, only a few groups had managed to perform intracellular recordings of motoneurons in vivo2-4 , including our team who published a new preparation which allowed us to obtain very stable recordings of motoneurons in vivo in adult mice5,6. However, these recordings were obtained in paralyzed animals, i.e. without the possibility to record the force output of these motoneurons. Here we present an extension of this original preparation in which we were able to obtain simultaneous recordings of the electrophysiological properties of the motoneurons and of the force developed by their motor unit. This is an important achievement, as it allows us to identify the different types of motoneurons based on their force profile, and thereby revealing their function. Coupled with genetic models disturbing spinal segmental circuitry7-9, or reproducting human disease10,11, we expect this technique to be an essential tool for the study of spinal motor system.

Recordings of Neural Circuit Activation in Freely Behaving Animals

The relationship between patterns of neural activity and corresponding behavioral expression is difficult to establish in unrestrained animals. Traditional non-invasive methods require at least partially restrained research subjects, and they only allow identification of large numbers of simultaneously activated neurons. On the other hand, small ensembles of neurons or individual neurons can only be measured using single-cell recordings obtained from largely reduced preparations. Since the expression of natural behavior is limited in restrained and dissected animals, the underlying neural mechanisms that control such behavior are difficult to identify. 
Here, I present a non-invasive physiological technique that allows measuring neural circuit activation in freely behaving animals. Using a pair of wire electrodes inside a water-filled chamber, the bath electrodes record neural and muscular field potentials generated by juvenile crayfish during natural or experimentally evoked escape responses. The primary escape responses of crayfish are mediated by three different types of tail-flips which move the animals away from the point of stimulation. Each type of tail-flip is controlled by its own neural circuit; the two fastest and most powerful escape responses require activation of different sets of large &#8220;command&#8221; neurons. In combination with behavioral observations, the bath electrode recordings allow unambiguous identification of these neurons and the associated neural circuits. Thus activity of neural circuitry underlying naturally occurring behavior can be measured in unrestrained animals and in different behavioral contexts.

Non-invasive measurements of neural activity patterns in freely behaving animals are obtained by combining neurophysiological recordings with high speed videography.

Historical View and Physiology Demonstration at the NMJ of the Crayfish Opener Muscle

Here we present some of the key important discoveries made with the opener neuromuscular (NMJ) preparation of crustaceans and illustrate that there is still much to learn from this model preparation. In understanding the history one can appreciate why even today this NMJ still offers a rich playground to address questions regarding pre- and post-synaptic function and plasticity. The viability and ease of access to the terminal for intracellular as well as extracellular electrophysiology and imaging are significant advantages. The mechanisms behind the modulation of vesicular kinetics and fusion within the high- and low-output terminals are begging for investigation. The preparation also offers a testable model system for computational assessments and manipulations to examine key variables in theoretical models of synaptic function, for example calcium dynamics during short-term facilitation. The synaptic complexity of active zone and statistical nature of quantal release is also an open area for future investigation both experimentally and computationally.

Proprioception and Tension Receptors in Crab Limbs: Student Laboratory Exercises

The primary purpose of these procedures is to demonstrate for teaching and research purposes how to record the activity of living primary sensory neurons responsible for proprioception as they are detecting joint position and movement, and muscle tension. Electrical activity from crustacean proprioceptors and tension receptors is recorded by basic neurophysiological instrumentation, and a transducer is used to simultaneously measure force that is generated by stimulating a motor nerve. In addition, we demonstrate how to stain the neurons for a quick assessment of their anatomical arrangement or for permanent fixation. Staining reveals anatomical organization that is representative of chordotonal organs in most crustaceans. Comparing the tension nerve responses to the proprioceptive responses is an effective teaching tool in determining how these sensory neurons are defined functionally and how the anatomy is correlated to the function. Three staining techniques are presented allowing researchers and instructors to choose a method that is ideal for their laboratory.

Measuring Neuromuscular Junction Functionality

Neuromuscular junction (NMJ) functionality plays a pivotal role when studying diseases in which the communication between motor neuron and muscle is impaired, such as aging and amyotrophic lateral sclerosis (ALS). Here we describe an experimental protocol that can be used to measure NMJ functionality by combining two types of electrical stimulation: direct muscle membrane stimulation and the stimulation through the nerve. The comparison of the muscle response to these two different stimulations can help to define, at the functional level, potential alterations in the NMJ that lead to functional decline in muscle.
Ex vivo preparations are suited to well-controlled studies. Here we describe an intensive protocol to measure several parameters of muscle and NMJ functionality for the soleus-sciatic nerve preparation and for the diaphragm-phrenic nerve preparation. The protocol lasts approximately 60 min and is conducted uninterruptedly by means of a custom-made software that measures the twitch kinetics properties, the force-frequency relationship for both muscle and nerve stimulations, and two parameters specific to NMJ functionality, i.e. neurotransmission failure and intratetanic fatigue. This methodology was used to detect damages in soleus and diaphragm muscle-nerve preparations by using SOD1G93A transgenic mouse, an experimental model of ALS that ubiquitously overexpresses the mutant antioxidant enzyme superoxide dismutase 1 (SOD1).

Ex Vivo Assessment of Contractility, Fatigability and Alternans in Isolated Skeletal Muscles

Described here is a method to measure contractility of isolated skeletal muscles. Parameters such as muscle force, muscle power, contractile kinetics, fatigability, and recovery after fatigue can be obtained to assess specific aspects of the excitation-contraction coupling (ECC) process such as excitability, contractile machinery and Ca2+ handling ability. This method removes the nerve and blood supply and focuses on the isolated skeletal muscle itself. We routinely use this method to identify genetic components that alter the contractile property of skeletal muscle though modulating Ca2+ signaling pathways. Here, we describe a newly identified skeletal muscle phenotype, i.e., mechanic alternans, as an example of the various and rich information that can be obtained using the in vitro muscle contractility assay. Combination of this assay with single cell assays, genetic approaches and biochemistry assays can provide important insights into the mechanisms of ECC in skeletal muscle.
