PST und ähnliche Verbindungen wurde gezeigt, eine anti-Krebs-Eigenschaften 11-15,21. Brustkrebs ist eine der häufigsten Krebserkrankungen bei Frauen in Nordamerika. Viele aktuelle Anti-Krebs-Behandlungen, einschließlich ionisierender Strahlung, die Apoptose über DNA-Schäden. Leider sind solche Behandlungen nicht-selektiven, um Krebszellen zu erzeugen und ähnliche Toxizität in normalen Zellen. Wir haben selektive Induktion von Apoptose in Krebszellen durch die natürliche Verbindung Pancratistatin (PST) berichtet. Kürzlich wurde eine neue PST Analog, ein C-1 Acetoxymethyl-Derivat von 7-deoxypancratistatin (JCTH-4), von de-novo-Synthese hergestellt und weist vergleichbare selektive Apoptose induzierende Aktivität in verschiedenen Krebszelllinien. Vor kurzem hat die Autophagie in bösartigen Erkrankungen wie sowohl pro-Überleben und die pro-Tod-Mechanismen als Reaktion auf Chemotherapie beteiligt. Tamoxifen (TAM) hat stets die Induktion von pro-Überleben Autophagie in zahlreichen Krebsarten demonstriert. In dieser Studie, um die Wirksamkeit von JCTH-4 allein oder in Kombination mit TAM Zelltod in der menschlichen Muttermilch deutung zu induzierenr (MCF7) und Neuroblastom (SH-SY5Y-Zellen) wurde ausgewertet. TAM allein Autophagie, induziert aber unbedeutend Zelltod während JCTH-4 allein signifikante Induktion der Apoptose mit etwas Induktion von Autophagie verursacht. Interessanterweise ergab die kombinatorische Behandlung eine drastische Zunahme der apoptotischen und autophagischen Induktion. Wir überwachten zeitabhängige morphologische Veränderungen in MCF7-Zellen durchlaufen TAM-induzierten Autophagie, JCTH-4-induzierte Apoptose und Autophagie und beschleunigte Zelltod mit kombinatorischen Behandlung mit Zeitraffer-Mikroskopie. Wir haben diese Verbindungen nachgewiesen, dass die Apoptose / Autophagie durch mitochondriale Targeting in diesen Krebszellen zu induzieren. Wichtig ist, dass diese Behandlungen haben keinen Einfluss auf das Überleben der menschlichen Fibroblasten noncancerous. Somit zeigen diese Ergebnisse, dass JCTH-4 in Kombination mit TAM als sicher und sehr potente anti-Krebs-Therapie gegen Brustkrebs und Neuroblastom-Zellen verwendet werden.
