Folglich führt Ablation von Tregs zu tödlichen Autoimmunität und Selbstzerstörung durch aktivierte T-Zellen 1 angetrieben. Umgekehrt kann therapeutische Modulation von Molekülen an Treg Differenzierung blockieren bieten Nutzen in Zukunft Ansätze der kombinierten Behandlung von Tumorpatienten. In vitro Differenzierung Assays sind für die Beschreibung der molekularen Veränderungen, die mit T-Zell-Untergruppe Differenzierung verbunden sind maßgeblich. Im Moment, experimentelle Versuche, Suche oder Bildschirm für die Gen-Produkte, die Kontroll-T-Zell-Differenzierung durch die Tatsache, dass die häufigsten Methoden der ektopische Genexpression in naiven T-Zellen nicht behindert werden. So sind zum Beispiel die Elektroporation und die retrovirale Transduktion nur wirksam in aktivierten T-Zellen. Im Gegensatz zu den ursprünglichen Erwartungen lentiviralen Transduktion, die in der Regel wirksam in ruhenden Zellen ist, erfordert vor Aktivierung naiver T-Zellen durch Zytokine 13. Ferner kann die Übertragung von cDNA oder mRNA bei der Elektroporationbeinhaltet Depolarisation der Zellmembran, die sich verleiht Eigenschaften der T-Zell-Aktivierung und sogar mobilisieren Ca 2 +-Signalisierung und aktivieren NFAT Proteinen (unveröffentlichte Beobachtung und ref. 14). Auch für retrovirale Transduktion haben die naiven T-Zellen für 18 aktiviert werden - 40 Stunden. Während dieser Zeit erfolgt die Auswertung der Kernmembran im Verlauf der Zellteilung ermöglicht und für die nachfolgende genomische Integration des retroviralen Vektors 15. Diese Methoden sind daher nicht in der Lage, um die frühe molekulare Regulation der ersten Begegnung mit T-Zell-Antigen, das die entscheidende Phase der Helfer-T-Zell-Differenzierung ist anzugehen. Adenovirus-Transduktion ist bekannt, ektopische transienten Genexpression in einer Reihe von humanen Zelltypen, die den menschlichen Coxsackie-Adenovirus-Rezeptor (CAR) zum Ausdruck zu verleihen. Es geht ohne Anforderung für Zell-Aktivierung oder Zellzyklus-Progression. Die Oberflächenexpression von CAR ist von wesentlicher Bedeutung für eine effiziente Bindung und Internalisierung Virus, und transgene Expression der verkürzten Version CARΔ1 unter einem T-Zell-spezifischen Promotors wurde festgestellt, dass Mäusethymozyten und T-Zellen anfällig für Adenovirusinfektion 16 machen. Wichtig ist, dass das Transgen nicht ändern Thymozyten Entwicklung oder in vitro Differenzierung von naiven CD4-T-Zellen in verschiedene Untergruppen (Daten nicht gezeigt; ref 17.). Wichtig ist, ermöglicht adenoviralen Transduktion hohe Expression eines Gens von Interesse in naive T-Zellen zu induzieren, ohne offensichtliche Anzeichen der Aktivierung. Die T-Zellen bleiben naive (CD44 niedrig, CD62L hoch) und Ruhe-(CD25 -, CD69 -) nach infectiauf und kann aktiviert und in Treg ähnlich nicht infizierten Zellen unterschieden werden. Herstellung von rekombinanten Adenoviren nach Transfektion von Zellen mit adenoviralen HEK293A Plasmide (Abbildung 1) erreicht werden. Diese Plasmide enthalten in der Regel den menschlichen Adenovirus Typ 5-Genom mit E1 und E3 Gene gelöscht rekombinanten Adenoviren replikationsinkompetent 21 machen. HEK293A Zellen ergänzen Replikationsdefizienz, wie sie durch stabile Integration geschert Adenovirus 22 verewigt haben. Seit Adenovirusvektoren groß (~ 40 kb) und damit nicht gut für traditionelle Restriktionsenzym-vermittelte Klonierung geeignet sind, beschäftigten wir das Gateway-System. Das Gen von Interesse wird zunächst in einem kleineren Eingangsvektor, aus dem sie leicht in das adenovirale Zielvektor über Lambda Rekombination (LR) 23 übertragen werden können kloniert. Wir konstruierten die pCAGAdDu Vektordurch die Kombination der CAG-Promotor (Huhn Actin-Promotor und CMV-Enhancer) mit einer Expressionskassette, enthaltend LR flankieren die prokaryotische ccdB Selektionsmarker 24. Diese Expressionskassette ist mit einer internen Ribosomen-Eintrittsstelle (IRES-Element), die Co-Expression der eukaryontischen Infektionsmarker enhanced green fluorescent protein (eGFP), die zu einer Sequenz, die die Rinder-Wachstumshormon-poly (A)-Signal fusioniert ist ermöglicht fusioniert. Wir haben die CAG cis-regulatorische Sequenzen, da die prototypischen CMV-Promotor wurde als hoch-Aktivierung ab und sind daher ungünstig für die Genexpression in naive T-Zellen. Hier bieten wir ein Protokoll für die effiziente in vitro Treg Differenzierung und eine Methode, um naive CD4 T-Zellen ohne Aktivierung (Abbildung 2) transduzieren. Das Verfahren ermöglicht ektopische Genexpression oder niederzuschlagen vorhergehenden CD4 T-Zell-Differenzierung bei der naiven Zustand. Es ermöglicht die Prüfung der Wirkung einer overexpressed Gen von Interesse in der frühen Signalwege beim erstmaligen TCR Stimulation bis T-Zell-Untergruppe Engagement. Unsere Validierungsexperimente auch die Grundlage, um ähnliche Adenovirus Anwendung bei der Differenzierung von anderen T-Zell-Untergruppen wie Th1, Th2, Th9, Th17, Th22 oder Tfh Zellen zu etablieren.

Regulatorische T-Zellen (Tregs) sind unerlässlich, um Immuntoleranz auf Selbstbestimmung sowie auf bestimmte fremde Antigene zu präsentieren. Tregs kann von naiven CD4 + T-Zellen in vitro mit TCR-und Co-Stimulation in Gegenwart von TGF-und IL-2 erzeugt werden. Dies birgt ein enormes Potenzial für zukünftige Therapien, aber die Moleküle und Signalwege, die Kontrolle Differenzierung sind weitgehend unbekannt. Primären T-Zellen können durch ektopische Expression manipuliert werden, aber üblichen Verfahren nicht die wichtigste naive Zustand der T-Zelle vor dem primären Antigen-Erkennung anzusprechen. Hier bieten wir ein Protokoll zu ektopische Gene in naiven CD4 T-Zellen exprimieren in vitro vor Induktion Treg Differenzierung. Wichtig ist, dass nach der Infektion die T-Zellen bleiben naive (CD44 niedrig, CD62L hoch) und Ruhe-(CD25 -, CD69 -) und kann aktiviert und in Tregs differenziert werden ähnlich nicht-infizierten Zellen. Somit ermöglicht dieses Verfahren Manipulation von CD4 T-Zell-Differenzierung von Anfang an. Es sorgt dafür, dass ektopische Genexpression ist bereits in Kraft, wenn sie früh signalisiert Ereignisse des ersten TCR Stimulation induziert zelluläre Veränderungen, die schließlich in Treg Differenzierung führen.

Retroviral Transduction of T-cell Receptors in Mouse T-cells

T-cell receptors (TCRs) play a central role in the immune system. TCRs on T-cell surfaces can specifically recognize peptide antigens presented by antigen presenting cells (APCs)1. This recognition leads to the activation of T-cells and a series of functional outcomes (e.g. cytokine production, killing of the target cells). Understanding the functional role of TCRs is critical to harness the power of the immune system to treat a variety of immunology related diseases (e.g. cancer or autoimmunity).
It is convenient to study TCRs in mouse models, which can be accomplished in several ways. Making TCR transgenic mouse models is costly and time-consuming and currently there are only a limited number of them available2-4. Alternatively, mice with antigen-specific T-cells can be generated by bone marrow chimera. This method also takes several weeks and requires expertise5. Retroviral transduction of TCRs into in vitro activated mouse T-cells is a quick and relatively easy method to obtain T-cells of desired peptide-MHC specificity. Antigen-specific T-cells can be generated in one week and used in any downstream applications. Studying transduced T-cells also has direct application to human immunotherapy, as adoptive transfer of human T-cells transduced with antigen-specific TCRs is an emerging strategy for cancer treatment6.
Here we present a protocol to retrovirally transduce TCRs into in vitro activated mouse T-cells. Both human and mouse TCR genes can be used. Retroviruses carrying specific TCR genes are generated and used to infect mouse T-cells activated with anti-CD3 and anti-CD28 antibodies. After in vitro expansion, transduced T-cells are analyzed by flow cytometry.

Directed Differentiation of Induced Pluripotent Stem Cells towards T Lymphocytes

Adoptive cell transfer (ACT) of antigen-specific CD8+ cytotoxic T lymphocytes (CTLs) is a promising treatment for a variety of malignancies 1. CTLs can recognize malignant cells by interacting tumor antigens with the T cell receptors (TCR), and release cytotoxins as well as cytokines to kill malignant cells. It is known that less-differentiated and central-memory-like (termed highly reactive) CTLs are the optimal population for ACT-based immunotherapy, because these CTLs have a high proliferative potential, are less prone to apoptosis than more differentiated cells and have a higher ability to respond to homeostatic cytokines 2-7. However, due to difficulties in obtaining a high number of such CTLs from patients, there is an urgent need to find a new approach to generate highly reactive Ag-specific CTLs for successful ACT-based therapies.
TCR transduction of the self-renewable stem cells for immune reconstitution has a therapeutic potential for the treatment of diseases 8-10. However, the approach to obtain embryonic stem cells (ESCs) from patients is not feasible. Although the use of hematopoietic stem cells (HSCs) for therapeutic purposes has been widely applied in clinic 11-13, HSCs have reduced differentiation and proliferative capacities, and HSCs are difficult to expand in in vitro cell culture 14-16. Recent iPS cell technology and the development of an in vitro system for gene delivery are capable of generating iPS cells from patients without any surgical approach. In addition, like ESCs, iPS cells possess indefinite proliferative capacity in vitro, and have been shown to differentiate into hematopoietic cells. Thus, iPS cells have greater potential to be used in ACT-based immunotherapy compared to ESCs or HSCs.
Here, we present methods for the generation of T lymphocytes from iPS cells in vitro, and in vivo programming of antigen-specific CTLs from iPS cells for promoting cancer immune surveillance. Stimulation in vitro with a Notch ligand drives T cell differentiation from iPS cells, and TCR gene transduction results in iPS cells differentiating into antigen-specific T cells in vivo, which prevents tumor growth. Thus, we demonstrate antigen-specific T cell differentiation from iPS cells. Our studies provide a potentially more efficient approach for generating antigen-specific CTLs for ACT-based therapies and facilitate the development of therapeutic strategies for diseases.

Generation of T lymphocytes from induced pluripotent stem (iPS) cells gives an alternative approach of using embryonic stem cells for T cell-based immunotherapy. The method shows that by utilizing either in vitro or in vivo induction system, iPS cells are able to differentiate into both conventional and antigen-specific T lymphocytes.

Accelerated Type 1 Diabetes Induction in Mice by Adoptive Transfer of Diabetogenic CD4+ T Cells

The nonobese diabetic (NOD) mouse spontaneously develops autoimmune diabetes after 12 weeks of age and is the most extensively studied animal model of human Type 1 diabetes (T1D). Cell transfer studies in irradiated recipient mice have established that T cells are pivotal in T1D pathogenesis in this model. We describe herein a simple method to rapidly induce T1D by adoptive transfer of purified, primary CD4+ T cells from pre-diabetic NOD mice transgenic for the islet-specific T cell receptor (TCR) BDC2.5 into NOD.SCID recipient mice. The major advantages of this technique are that isolation and adoptive transfer of diabetogenic T cells can be completed within the same day, irradiation of the recipients is not required, and a high incidence of T1D is elicited within 2 weeks after T cell transfer. Thus, studies of pathogenesis and therapeutic interventions in T1D can proceed at a faster rate than with methods that rely on heterogenous T cell populations or clones derived from diabetic NOD mice.

Isolation and Th17 Differentiation of Na&iuml;ve CD4 T Lymphocytes

Th17 cells are a distinct subset of T cells that have been found to produce interleukin 17 (IL-17), and differ in function from the other T cell subsets including Th1, Th2, and regulatory T cells. Th17 cells have emerged as a central culprit in overzealous inflammatory immune responses associated with many autoimmune disorders. In this method we purify T lymphocytes from the spleen and lymph nodes of C57BL/6 mice, and stimulate purified CD4+ T cells under control and Th17-inducing environments. The Th17-inducing environment includes stimulation in the presence of anti-CD3 and anti-CD28 antibodies, IL-6, and TGF-&#946;. After incubation for at least 72 hours and for up to five days at 37 &#176;C, cells are subsequently analyzed for the capability to produce IL-17 through flow cytometry, qPCR, and ELISAs. Th17 differentiated CD4+CD25- T cells can be utilized to further elucidate the role that Th17 cells play in the onset and progression of autoimmunity and host defense. Moreover, Th17 differentiation of CD4+CD25- lymphocytes from distinct murine knockout/disease models can contribute to our understanding of cell fate plasticity.

Transplantation of Tail Skin to Study Allogeneic CD4 T Cell Responses in Mice

The study of T cell responses and their consequences during allo-antigen recognition requires a model that enables one to distinguish between donor and host T cells, to easily monitor the graft, and to adapt the system in order to answer different immunological questions. Medawar and colleagues established allogeneic tail-skin transplantation in mice in 1955. Since then, the skin transplantation model has been continuously modified and adapted to answer specific questions. The use of tail-skin renders this model easy to score for graft rejection, requires neither extensive preparation nor deep anesthesia, is applicable to animals of all genetic background, discourages ischemic necrosis, and permits chemical and biological intervention. 
In general, both CD4+ and CD8+ allogeneic T cells are responsible for the rejection of allografts since they recognize mismatched major histocompatibility antigens from different mouse strains. Several models have been described for activating allogeneic T cells in skin-transplanted mice. The identification of major histocompatibility complex (MHC) class I and II molecules in different mouse strains including C57BL/6 mice was an important step toward understanding and studying T cell-mediated alloresponses. In the tail-skin transplantation model described here, a three-point mutation (I-Abm12) in the antigen-presenting groove of the MHC-class II (I-Ab) molecule is sufficient to induce strong allogeneic CD4+ T cell activation in C57BL/6 mice. Skin grafts from I-Abm12 mice on C57BL/6 mice are rejected within 12-15 days, while syngeneic grafts are accepted for up to 100 days. The absence of T cells (CD3-/- and Rag2-/- mice) allows skin graft acceptance up to 100 days, which can be overcome by transferring 2 x 104 wild type or transgenic T cells. Adoptively transferred T cells proliferate and produce IFN-&#947; in I-Abm12-transplanted Rag2-/- mice.

Tail-skin transplantation is a powerful model for studying T cell-dependent rejection and tolerance induction during allogeneic immune responses in mice. The advantages of this protocol are minor invasive surgery, and ease of monitoring with no need to sacrifice the recipient mouse.

Induction of Murine Intestinal Inflammation by Adoptive Transfer of Effector CD4+CD45RBhigh T Cells into Immunodeficient Mice

There are many different animal models available for studying the pathogenesis of human inflammatory bowel diseases (IBD), each with its own advantages and disadvantages. We describe here an experimental colitis model that is initiated by adoptive transfer of syngeneic splenic CD4+CD45RBhigh T cells into T and B cell deficient recipient mice. The CD4+CD45RBhigh T cell population that largely consists of na&#239;ve effector cells is capable of inducing chronic intestinal inflammation, closely resembling key aspects of human IBD. This method can be manipulated to study aspects of disease onset and progression. Additionally it can be used to study the function of innate, adaptive, and regulatory immune cell populations, and the role of environmental exposures, i.e., the microbiota, in intestinal inflammation. In this article we illustrate the methodology for inducing colitis with a step-by-step protocol. This includes a video demonstration of key technical aspects required to successfully develop this murine model of experimental colitis for research purposes.

Antigen inexperienced (na&#239;ve) CD4+ T cells undergo expansion and differentiation to effector subsets at the time of T cell receptor (TCR) recognition of cognate antigen presented on MHC class II. The cytokine signals present in the environment at the time of TCR activation are a major factor in determining the effector fate of a na&#239;ve CD4+ T cell. Although the cytokine environment during na&#239;ve T cell activation may be complex and involve both redundant and opposing signals in vivo, the addition of various cytokine combinations during naive CD4+ T cell activation in vitro can readily promote the establishment of effector T helper lineages with hallmark cytokine and transcription factor expression. Such differentiation experiments are commonly used as a first step for the evaluation of targets believed to promote or inhibit the development of certain CD4+ T helper subsets. The addition of mediators, such as signaling agonists, antagonists, or other cytokines, during the differentiation process can also be used to study the influence of a particular target on T cell differentiation. Here, we describe a basic protocol for the isolation of na&#239;ve T cells from mouse and the subsequent steps necessary for polarizing na&#239;ve cells to various T helper effector lineages in vitro.

Retroviral Transduction of Bone Marrow Progenitor Cells to Generate T-cell Receptor Retrogenic Mice

T cell receptor (TCR) signaling is essential in the development and differentiation of T cells in the thymus and periphery, respectively. The vast array of TCRs proves studying a specific antigenic response difficult. Therefore, TCR transgenic mice were made to study positive and negative selection in the thymus as well as peripheral T cell activation, proliferation and tolerance. However, relatively few TCR transgenic mice have been generated specific to any given antigen. Thus, studies involving TCRs of varying affinities for the same antigenic peptide have been lacking. The generation of a new TCR transgenic line can take six or more months. Additionally, any specific backcrosses can take an additional six months. In order to allow faster generation and screening of multiple TCRs, a protocol for retroviral transduction of bone marrow was established with stoichiometric expression of the TCR&#945; and TCR&#946; chains and the generation of retrogenic mice. Each retrogenic mouse is essentially a founder, virtually negating a founder effect, while the length of time to generate a TCR retrogenic is cut from six months to approximately six weeks. Here we present a rapid and flexible alternative to TCR transgenic mice that can be expressed on any chosen background with any particular TCR.

Preparation of CD4+ T Cells for Analysis of GD3 and GD2 Ganglioside Membrane Expression by Microscopy

The methods described herein for activation of na&#239;ve CD4+ T cells in suspension and their adherence in coverslips for confocal microscopy analysis allow the spatial localization and visualization of gangliosides involved in CD4+ T cell activation, that complement expression profiling experiments such as flow cytometry, western blotting or real-time PCR. The quantification of ganglioside expression through flow cytometry and their cellular localization through microscopy can be obtained by the use of anti-ganglioside antibodies with high affinity and specificity. Nonetheless, an adequate handling of cells in suspension involves the treatment of culture plates to promote the necessary adherence required for fluorescence or confocal microscopy acquisition. In this work, we describe a protocol for determining GD3 and GD2 ganglioside expression and colocalization with the TCR during na&#239;ve CD4+ T cell activation. Also, real-time PCR experiments using &#60;40,000 cells are described for the determination of the GD3 and GM2/GD2 synthase genes, demonstrating that gene analysis experiments can be performed with a low number of cells and without the need of additional low input RNA kits.

Retroviral Transduction of Helper T Cells as a Genetic Approach to Study Mechanisms Controlling their Differentiation and Function

Helper T cell development and function must be tightly regulated to induce an appropriate immune response that eliminates specific pathogens yet prevents autoimmunity. Many approaches involving different model organisms have been utilized to understand the mechanisms controlling helper T cell development and function. However, studies using mouse models have proven to be highly informative due to the availability of genetic, cellular, and biochemical systems. One genetic approach in mice used by many labs involves retroviral transduction of primary helper T cells. This is a powerful approach due to its relative ease, making it accessible to almost any laboratory with basic skills in molecular biology and immunology. Therefore, multiple genes in wild type or mutant forms can readily be tested for function in helper T cells to understand their importance and mechanisms of action. We have optimized this approach and describe here the protocols for production of high titer retroviruses, isolation of primary murine helper T cells, and their transduction by retroviruses and differentiation toward the different helper subsets. Finally, the use of this approach is described in uncovering mechanisms utilized by microRNAs (miRNAs) to regulate pathways controlling helper T cell development and function.

Many experimental systems have been utilized to understand the mechanisms regulating T cell development and function in an immune response. Here a genetic approach using retroviral transduction is described, which is economic, time efficient, and most importantly, highly informative in identifying regulatory pathways.
