Im Jahr 2001 wurde eine mutmaßliche Bevölkerung von multipotenten Stammzellen aus Fettgewebe in der Zeitschrift Tissue Engineering 1 beschrieben. Diese Zellen wurden die Namen Verarbeitete Lipoaspirate-oder PLA-Zellen aufgrund ihrer Ableitung aus verarbeitet lipoaspirate Gewebe durch kosmetische Chirurgie erhalten gegeben. Kultivierung dieser SVF über die Zeit wird vorgeschlagen, beseitigen viele dieser kontaminierenden Zellpopulationen und führen zu einer haftenden, Fibroblastenpopulation. Diese Fibroblasten wurden in der Literatur für die letzten 40 Jahre nicht als zuvor identifiziert wordenAdipozyten. Allerdings zeigten unsere Forschungsgruppe, dass diese Zellen besaß mesodermalen Multipotenz und als PLA Zellen umbenannt haft SVF Bevölkerung. Um irgendeine Art von Konsens zu schaffen, wurde der Begriff Fettgewebe gewonnene Stammzellen oder ASC in der 2. Jahres IFATS Konferenz angenommen. Als solche wird der Begriff ASC in diesem Artikel verwendet werden. Die in diesem Artikel beschriebenen Protokoll ist ein relativ einfaches Verfahren, das Standard-Laborausrüstung erfordert und verwendet einfache Reagenzien wie Phosphor-Kochsalzlösung, Standard-Gewebekulturmedien und Reagenzien Kollagenase. Es kann abhängig von der Menge des Ausgangs Fettgewebe Volumen und anschließende c eine große Anzahl von ASCs erzeugenulture Zeit. Jedoch kann die Verarbeitung solcher großen Mengen von Fettgewebe einige physikalische Probleme, die bis zu einem Grad unter Verwendung dieses Protokolls begrenzt werden könnten präsentieren. Darüber hinaus hat dieses Protokoll erfordern sterile Gewebekultureinrichtungen und genehmigt Biosicherheit Hauben, wodurch die Verwendung eines zugelassenen Gewebekultur-Anlage erforderlich. Diese Anforderung kann auch den Nutzen der Bevölkerung zu verringern ASC in der klinischen Anwendung, sofern sie nicht in der guten Herstellungspraxis (GMP) zugelassenen Anlagen zur Isolierung und Ausdehnung von Materialien für den klinischen Einsatz konzipiert isoliert werden. Als Alternative würde automatisierte Systeme, die ASC in einem geschlossenen System im Operationssaal zu isolieren, können diese Schlüsselfrage zu vermeiden und ermöglichen den sofortigen Einsatz von ASC, ohne dass für die anschließende In-vitro-Expansion. Bisher gibt es sechs automatisierten Systemen, die für die Isolation von Zellen aus menschlichem Gewebe sind im Handel erhältlich. Diese Systeme können machen es möglich, ein isolierenerhebliche Anzahl von ASC von großen Mengen an Fettgewebe unmittelbar nach ihrer Ernte. Diese ASC konnte dann in den Patienten für eine Vielzahl von regenerativen Zwecken ohne dass der Patient jemals den Operationssaal verlassen wieder eingeführt werden. Zusätzlich zu diesem Protokoll die manuelle Trennung der ASC beschreibt, ist ein Protokoll für die automatisierte Isolierung von ASC mit dem Celution-System auch in einem Begleitartikel gegeben.

Im Jahr 2001, Forscher an der University of California, Los Angeles, beschrieben die Isolierung einer neuen Population von adulten Stammzellen aus Fettgewebe liposuctioned, die sie anfangs genannt Verarbeitete Lipoaspirate Cells oder PLA-Zellen. Seitdem haben diese Stammzellen aus Fettgewebe gewonnenen Stammzellen oder ASC umbenannt und gegangen auf einer der beliebtesten adulten Stammzellen-Populationen in den Bereichen Stammzellforschung und der regenerativen Medizin zu werden. Tausende von Artikeln werden nun die Verwendung von ASC in einer Vielzahl von Tiermodellen regenerative, einschließlich der Knochenregeneration, periphere Nervenreparatur und Herz-Kreislauf-Engineering. Aktuelle Artikel haben begonnen, die Vielzahl von Anwendungen für ASC in der Klinik zu beschreiben. Die in diesem Artikel gezeigten Protokoll beschreibt das grundlegende Verfahren für manuell und enzymatisch ASC Isolierung von großen Mengen von lipoaspirates von kosmetischen Verfahren erhalten. Dieses Protokoll kann leicht nach oben oder unten skaliert werden, um Unterkunftsaß das Volumen der lipoaspirate und kann auf ASC aus dem Fettgewebe durch abdominoplasties ähnlichen Verfahren erhalten zu isolieren.

Das Protokoll Umriss oben beschrieben, eine manuelle, enzymatische Verfahren zur Isolierung eines SVF aus einem großen Volumen lipoaspirate Probe. Innerhalb dieses SVF gibt zahlreiche Zellpopulationen, einschließlich der ASC. Zahlreiche Studien schlagen vor, dass die Kultivierung dieses SVF unter Standard-Zellkulturbedingungen für eine haft Fibroblasten-Bevölkerung, die hauptsächlich aus der ASC-Typ zusammengesetzt werden soll.
