Für die positive Panning Phase wird das Zielantigen von Interesse auf einer Glimmer-Oberfläche fixiert und gebundenen Phagen werden eluiert und gesiebt, um Phagen, die selektiv das Zielantigen binden identifizieren. Die Zielproteinvariante muss nicht gereinigt werden, sofern die entsprechenden negativen Panning Kontrollen verwendet wurden. Auch Zielproteinvarianten, die nur in sehr niedrigen Konzentrationen in komplexen biologischen Materials in der positiven Panning Schritt verwendet werden vorliegen. Durch Anwendung dieser Technologie übernahmen wir Antikörper gegen Proteinvarianten der TDP-43, die selektiv in menschlichen ALS Hirngewebe gefunden werden. Wir erwarten, dass dieses Protokoll anwendbar sein sollte, um die Erzeugung Reagentien, die selektiv binden Proteinvarianten in einer Vielzahl von verschiedenen biologischen Prozessen und Krankheiten vorkommen.
