Para los modelos in vitro de cultivos celulares para simular con precisión el tejido de interés, el entorno de crecimiento de la cultura es un aspecto crítico a considerar. Comúnmente se utilizan sistemas convencionales de cultivo de células se propagan en las células epiteliales planas superficies impermeables de dos dimensiones (2-D). Aunque mucho se ha aprendido de los sistemas convencionales de cultivo de células, los resultados de muchos que no son reproducibles en los ensayos clínicos humanos o explantes de tejido, potencialmente como resultado de la falta de un microambiente fisiológicamente relevantes. A continuación, describimos un sistema de cultivo que supera muchas de las fronteras culturales de la condición 2-D de cultivos celulares, utilizando el innovador giro de la pared vascular (RWV) la tecnología de biorreactor. Los biorreactores vienen en dos formatos diferentes, un buque de alta aspecto giratorio (HARV) o un buque lento giro lateral (STLV), en el que se diferencian por su fuente de aireación. En este trabajo se describen los pasos detallados de cómo la cultura en 3-D agregados de células epiteliales en el sistema de biorreactor RWV y una variedad de ensayos y análisis posibles que se pueden ejecutar con los agregados de 3-D. Estos análisis incluyen, pero no se limitan a, estructural / manálisis de orphological (confocal, análisis y microscopía electrónica de transmisión), citocinas / quimiocinas y la secreción de señalización celular (matriz de citometría de cuentas y análisis de Western blot), análisis de expresión génica (PCR en tiempo real), análisis toxicológico / drogas y de las interacciones huésped-patógeno. La utilización de estos ensayos de sentar las bases para estudios más en profundidad y expansiva como la metabolómica, la transcriptómica, la proteómica y otras aplicaciones basadas en matrices. Nuestro objetivo es dar a conocer un medio no-convencionales de cultivo de células epiteliales humanas para producir organotípicos modelos 3-D que recapitula el ser humano en el tejido vivo, en un sistema fácil y robusto para ser utilizado por los investigadores con diversos intereses científicos.
