Zellen und Gewebe im Körper Erfahrung Umweltbedingungen, die ihre Architektur zu beeinflussen, interzelluläre Kommunikation, und die allgemeine Funktionen. Für In-vitro-Zellkultur-Modellen simulieren exakt um das Gewebe von Interesse ist das Wachstum der Kultur-Umgebung ein kritischer Aspekt zu betrachten. Üblicherweise verwendete konventionelle Zellkultursystemen ausbreiten Epithelzellen auf flachen, zweidimensionalen (2D) undurchlässige Oberflächen. Obwohl viel von herkömmlichen Zellkultursystemen erfahren haben, sind viele Ergebnisse nicht reproduzierbar in klinischen Studien am Menschen oder Gewebeexplantate, möglicherweise als Folge des Fehlens eines physiologisch relevanten Mikroumgebung. Hier beschreiben wir eine Kultur-System, das viele der Kultur Bedingung Grenzen der 2-D-Zellkulturen überwindet, indem Sie die innovative drehbare Wand Gefäß (RWV) Bioreaktor-Technologie. Wir und andere haben gezeigt, dass organotypischen RWV-abgeleiteten Modelle können structur rekapitulierene, Funktion und authentischen menschlichen Reaktionen auf äußere Reize ähnlich wie bei menschlichen Geweben Explantat 1-6. Die RWV Bioreaktor ist eine Suspensionskultur, das für das Wachstum der Epithelzellen unter niedrigen Scherbedingungen physiologischen Flüssigkeit ermöglicht. Die Bioreaktoren in zwei verschiedenen Formaten, eine mit hohem Aspektverhältnis rotierenden Behälter (HARV) oder eine langsam drehende seitlichen Behälter (STLV), in denen sie sich durch ihre Belüftung Quelle stammen. Epithelzellen sind in den Bioreaktor der Wahl in Kombination mit porösen, Kollagen-beschichteten Mikroträgerkügelchen (1A) zugegeben. Die Zellen nutzen die Kügelchen als Wachstumsförderer Gerüst während der konstanten freien Fall in dem Bioreaktor (1B). Die Mikroumgebung des Bioreaktors vorgesehen ermöglicht es den Zellen dreidimensionalen (3-D) Anzeigen Aggregate in vivo-ähnlichen Eigenschaften zu bilden oft nicht unter Standard-2D-Kulturbedingungen (1D) beobachtet. Zu diesen Merkmalen gehören Tight Junctions, mucuns die Produktion, apikale / basale Orientierung, in vivo Protein-Lokalisierung und zusätzliche epithelialen Zelltyp-spezifische Eigenschaften. Die Progression von einer Monoschicht von Epithelzellen zu einer ausdifferenzierten 3-D-Aggregat hängt vom Zelltyp 1, 7-13 basiert. Regelmäßige Stichproben aus dem Bioreaktor ermöglicht die Überwachung von epithelialen Aggregatbildung, die zelluläre Differenzierung Marker und Lebensfähigkeit (1D). Wenn die zelluläre Differenzierung und Aggregatbildung hergestellt wird, werden die Zellen aus dem Bioreaktor entnommen, und ähnliche Assays 2-D-Zellen durchgeführt werden, um den 3-D-Aggregate mit einigen Überlegungen (1E-G) aufgebracht werden. In dieser Arbeit beschreiben wir eine detaillierte Schritte, wie die Kultur 3-D-Epithelzellen Aggregate in der RWV Bioreaktor und eine Vielzahl von möglichen Tests und Analysen, die mit den 3-D-Aggregate ausgeführt werden kann. Diese Analysen umfassen, sind aber nicht beschränkt auf, strukturelle / morphological Analyse (konfokale, Raster-und Transmissions-Elektronenmikroskopie), Zytokin / Chemokin Sekretion und Zell-Signalisierung (Cytometric Bead Array-und Western-Blot-Analyse), Genexpressionsanalysen (real-time PCR), toxikologischen / Drogen Analyse-und Wirt-Pathogen-Interaktionen. Die Verwendung dieser Assays legte den Grundstein für tiefer gehende Studien und expansiv wie Metabolomik, Transcriptomics, Proteomics und andere Array-basierten Anwendungen. Unser Ziel ist es eine nicht-konventionelle Mittel zur Kultivierung humaner Epithelzellen zu organotypischen 3-D-Modelle, die das menschliche Gewebe in vivo zu rekapitulieren, in einer einfachen und robusten System, um die von Forschern mit vielfältigen wissenschaftlichen Interessen verwendet werden produzieren zu präsentieren.
