In this study, we present a reliable method for the isolation, culture, and adipogenic induction of NCADSCs extracted from the PVAT of Wnt-1 Cre+/-;Rosa26RFP/+ transgenic mice designed to produce RFP+ ADSCs. The prevalence of obesity is increasing worldwide, which increases the risk of related chronic diseases, including cardiovascular disease and diabetes1. PVAT surrounds blood vessels and is a major source of endocrine and paracrine factors involved in vasculature function. Clinical studies show that high PVAT content is an independent risk factor of cardiovascular disease2,3, and its pathological function depends on the phenotype of the constituent adipose-derived stem cells (ADSCs)4.
Although ADSC cell lines like the murine 3T3-L1, 3T3-F442A, and OP9 are useful cellular models to study adipogenesis or lipogenesis5, regulatory mechanisms for adipogenesis differ between cell lines and primary cells. The ADSCs in the stromal vascular cell fraction (SVF) isolated directly from adipose tissues and induced to differentiate into adipocytes most likely recapitulate in vivo adipogenesis and lipogenesis6. However, the fragility, buoyancy, and the variations in size and immunophenotypes of the ADSCs make their direct isolation challenging. In addition, the different isolation procedures can also significantly affect the phenotype and adipogenic potential ability of these cells7, thus emphasizing the need for a protocol that maintains ADSC integrity.
Adipose tissue is typically classified as either the morphologically and functionally distinct white adipose tissue (WAT), or the brown adipose tissue (BAT)8, which harbors distinct ADSCs9. While ADSCs isolated from perigonadal and inguinal subcutaneous WATs have been characterized in previous studies9,10,11,12, less is known regarding the ADSCs from PVAT that is mainly composed of BAT13.
In a recent study, we found that a portion of the resident ADSCs in the periaortic arch adipose tissue (PAAT) are derived from neural crest cells (NCCs), a transient population of migratory progenitor cells that originate from the ectoderm14,15. Wnt1-Cre transgenic mice were used for tracing neural crest cell development16,17. We crossed Wnt1-Cre+ mice with Rosa26RFP/+ mice to generate Wnt-1 Cre+/-;Rosa26RFP/+ mice, in which NCCs and their descendants are labeled with red fluorescent protein (RFP) and are easily tracked in vivo and in vitro15. Here, we describe a method for isolating neural crest derived ADSCs (NC-derived ADSCs, or NCADSCs) from mouse PAAT and induce the NCADSCs to differentiate into white adipocytes or brown adipocytes.

An excessive amount of adipose tissue surrounding the blood vessels (perivascular adipose tissue, also known as PVAT) is associated with a high risk of cardiovascular disease. ADSCs derived from different adipose tissues show distinct features, and those from the PVAT have not been well characterized. In a recent study, we reported that some ADSCs in the periaortic arch adipose tissue (PAAT) descend from the neural crest cells (NCCs), a transient population of migratory cells originating from the ectoderm.
In this paper, we describe a protocol for isolating red fluorescent protein (RFP)-labeled NCCs from the PAAT of Wnt-1 Cre+/-;Rosa26RFP/+ mice and inducing their adipogenic differentiation in vitro. Briefly, the stromal vascular fraction (SVF) is enzymatically dissociated from the PAAT, and the RFP+ neural crest derived ADSCs (NCADSCs) are isolated by fluorescence activated cell sorting (FACS). The NCADSCs differentiate into both brown and white adipocytes, can be cryopreserved, and retain their adipogenic potential for ~3&#8211;5 passages. Our protocol can generate abundant ADSCs from the PVAT for modeling PVAT adipogenesis or lipogenesis in vitro. Thus, these NCADSCs can provide a valuable system for studying the molecular switches involved in PVAT differentiation.

Using the protocol described above, we obtained ~0.5&#8211;1.0 x 106 ADSCs from 5&#8211;6 Wnt-1 Cre+/-;Rosa26RFP/+ mice (48 weeks old, male or female).

The advantage of this method is the inherent fluorescence of the NCADSCs of biotin you need to label the cells with flou-chrome conjugated antibodies for FACS, or magnetic activated cell sorting. This method can be used to acquire abundant ADSC derived from neural crest cells from the ectoderm to study PVAT adipogenesis, or lipogenesis in vitro.Using young mice and having experience with cautery and induced adipogenesis means 3T3 cells is critical to the success of this protocol. Demonstrating the procedure will be Yiding Qi, a grad student, and Lian Xu, a technician from my laboratory. For PAAT dissection, after sterilizing the mouse carcass in 75%ethanol for five minutes, use scissors to snip and separate the skin on the abdomen.Cut along the ventral midline from pelvis to the neck, and open the abdomen. Move the liver to expose the diaphragm, and cut the diaphragm and the ribs on both sides of the midline. Peel back the ribs to expose the heart and lungs.And remove the lungs and the thymus. Extract the PAAT along with the aorta and heart into a petri dish. And cut off the aorta at the aortic root to remove the heart.Then, make a cut between the aortic arch and descending aorta. Carefully separate the adipose tissue surrounding both structures, and the left and right common carotid arteries from the posterior chest wall. It is important to separate the PAAT from the root and as a larger vascular vessels carefully, is the presence of vascular wall cells can affect FACS efficiency.Place the tissue into a petri dish containing ice cold HBSS buffer. And use sterile forceps to remove as much of the vasculature and fascia as possible. Then, transfer the adipose tissue into a two milliliter Eppendorf tube, containing point five milliliters of ice cold HBSS buffer on ice.When all of the PAAT has been collected, transfer the harvested adipose tissue into a new two milliliter micro-centrifuge tube containing one milliliter of freshly prepared digestion medium. And use surgical scissors to mince the tissues. Transfer the tissue slurry into a 50 millimeter tube containing nine milliliters of fresh digestion medium.And use a one milliliter pipette to triturate the tissues 10 times. When a homogenous solution has been obtained, incubate the tube for 30 to 45 minutes at 37 degrees Celsius and 100 revolutions per minute, checking every five to 10 minutes to prevent overdigestion. When a light yellow homogenous adipose tissue solution can be observed upon gentle swirling of the tube, stop the digestion with five milliliters of HDMEM supplemented with 10%fetal bovine serum.After a thorough mixing, centrifuge the adipose tissue sample. The stromal vascular cell fraction will be visible as a brownish pellet. Re-suspend the pellet in 10 milliliters of culture medium, and filter the cell suspension through a 70 micrometer cell strainer.Collect the cells by another centrifugation. And gently re-suspend the pellet in five milliliters of erythrocytes lysis buffer. Transfer the suspension to a 15 milliliter tube.After 10 minutes, stop the reaction with two centrifugations in 10 milliliters of PBS, supplemented with 1%fetal bovine serum. After the second centrifugation, re-suspend the pellet in five milliliters of culture medium on ice. And collect the cells with a final centrifugation.Then, re-suspend the cells in five milliliters of FACS buffer for counting. To set up an NCADSC culture, after their isolation by FACS according to standard protocols, place the cells at a five times 10 to the third cells per centimeters squared concentration in each well, a twelve well culture plate. Incomplete culture medium at 37 degrees Celsius and 5%carbon dioxide for 20 to 24 hours.The next day, wash the cells with 37 degrees Celsius PBS, and feed the culture with fresh culture medium. When the culture reaches 80 to 90%confluency, treat the cells with two milliliters of 0.25%trypsin EDTA per well for three to five minutes at 37 degrees Celsius. When the cells have been detached from the bottom of the plate, neutralize the enzymes with two milliliters of cultured medium, and collect the harvested cells by centrifugation.Re-suspend the pellet in one milliliter of fresh culture medium for counting. And seed the cells in a new 12 well culture plate in a five times 10 to the third cells per centimeters squared concentration. Then, re-suspend the remaining cells in culture medium supplemented with 10%dimethyl sulfoxide for frozen storage in liquid nitrogen.To induce adipogenic differentiation, when the cells reach an 80 to 90%confluency, treat the cells with brown or white adipogenic induction medium for two days as appropriate. At the end of the incubation, gently wash the cells two times with PBS before adding the appropriate fresh adipogenic induction medium to each well, and returning the cells to the cell culture incubator for an additional three to five days of incubation. Cultured NCADSCs reach an 80 to 90%confluency after seven to eight days of culture.And demonstrate an expanded fiberglass like morphology. To further confirm their adipogenic potential, oil red staining of the differentiated NCADSCs can be performed to detect mature adipocytes. Typically, mature adipocytes are observed after eight days of white or brown adipogenic induction with over 60%of the NCADSCs exhibiting adipogenic differentiation.Note that NCADSCs demonstrate a greatly reduced adipogenic potential after passaging. Immunoblotting and quantitative real-time PCR reveal that the expression of adipocyte specific relative proteins and genes significantly increases in adipogenically differentiated NCADSCs after eight days of white adipogenic induction. Similarly, the expression of adipocyte specific genes, and brown adipocyte specific genes, also significantly increases after eight days of brown adipogenic induction.Since most NCADSC isolation protocol is simple, economical, and doesn't require the use of antibodies, for that, the strong fluid reasons intensity of IFP can improve the FACS nourishment efficiency.

Isolation and Enrichment of Human Adipose-derived Stromal Cells for Enhanced Osteogenesis

Bone marrow-derived mesenchymal stromal cells (BM-MSCs) are considered the gold standard for stem cell-based tissue engineering applications. However, the process by which they must be harvested can be associated with significant donor site morbidity. In contrast, adipose-derived stromal cells (ASCs) are more readily abundant and more easily harvested, making them an appealing alternative to BM-MSCs. Like BM-MSCs, ASCs can differentiate into osteogenic lineage cells and can be used in tissue engineering applications, such as seeding onto scaffolds for use in craniofacial skeletal defects. ASCs are obtained from the stromal vascular fraction (SVF) of digested adipose tissue, which is a heterogeneous mixture of ASCs, vascular endothelial and mural cells, smooth muscle cells, pericytes, fibroblasts, and circulating cells. Flow cytometric analysis has shown that the surface marker profile for ASCs is similar to that for BM-MSCs. Despite several published reports establishing markers for the ASC phenotype, there is still a lack of consensus over profiles identifying osteoprogenitor cells in this heterogeneous population. This protocol describes how to isolate and use a subpopulation of ASCs with enhanced osteogenic capacity to repair critical-sized calvarial defects.

Direct Induction of Human Neural Stem Cells from Peripheral Blood Hematopoietic Progenitor Cells

Human disease specific neuronal cultures are essential for generating in vitro models for human neurological diseases. However, the lack of access to primary human adult neural cultures raises unique challenges. Recent developments in induced pluripotent stem cells (iPSC) provides an alternative approach to derive neural cultures from skin fibroblasts through patient specific iPSC, but this process is labor intensive, requires special expertise and large amounts of resources, and can take several months. This prevents the wide application of this technology to the study of neurological diseases. To overcome some of these issues, we have developed a method to derive neural stem cells directly from human adult peripheral blood, bypassing the iPSC derivation process. Hematopoietic progenitor cells enriched from human adult peripheral blood were cultured in vitro and transfected with Sendai virus vectors containing transcriptional factors Sox2, Oct3/4, Klf4, and c-Myc. The transfection results in morphological changes in the cells which are further selected by using human neural progenitor medium containing basic fibroblast growth factor (bFGF) and vascular endothelial growth factor (VEGF). The resulting cells are characterized by the expression for neural stem cell markers, such as nestin and SOX2. These neural stem cells could be further differentiated to neurons, astroglia and oligodendrocytes in specified differentiation media. Using easily accessible human peripheral blood samples, this method could be used to derive neural stem cells for further differentiation to neural cells for in vitro modeling of neurological disorders and may advance studies related to the pathogenesis and treatment of those diseases.

Culturing Human Pluripotent and Neural Stem Cells in an Enclosed Cell Culture System for Basic and Preclinical Research

This paper describes how to use a custom manufactured, commercially available enclosed cell culture system for basic and preclinical research. Biosafety cabinets (BSCs) and incubators have long been the standard for culturing and expanding cell lines for basic and preclinical research. However, as the focus of many stem cell laboratories shifts from basic research to clinical translation, additional requirements are needed of the cell culturing system. All processes must be well documented and have exceptional requirements for sterility and reproducibility. In traditional incubators, gas concentrations and temperatures widely fluctuate anytime the cells are removed for feeding, passaging, or other manipulations. Such interruptions contribute to an environment that is not the standard for cGMP and GLP guidelines. These interruptions must be minimized especially when cells are utilized for therapeutic purposes. The motivation to move from the standard BSC and incubator system to a closed system is that such interruptions can be made negligible. Closed systems provide a work space to feed and manipulate cell cultures and maintain them in a controlled environment where temperature and gas concentrations are consistent. This way, pluripotent and multipotent stem cells can be maintained at optimum health from the moment of their derivation all the way to their eventual use in therapy.

Here is a protocol to grow pluripotent stem cells (PSC) and neural stem cells (NSC) in an enclosed cell culture system that permits maximum sterility and reproducibility, replacing the traditional biosafety cabinet and incubator. Isolation and Differentiation of Adipose-Derived Stem Cells from Porcine Subcutaneous Adipose Tissues

Obesity is an unconstrained worldwide epidemic. Unraveling molecular controls in adipose tissue development holds promise to treat obesity or diabetes. Although numerous immortalized adipogenic cell lines have been established, adipose-derived stem cells from the stromal vascular fraction of subcutaneous white adipose tissues provide a reliable cellular system ex vivo much closer to adipose development in vivo. Pig adipose-derived stem cells (pADSC) are isolated from 7- to 9-day old piglets. The dorsal white fat depot of porcine subcutaneous adipose tissues is sliced, minced and collagenase digested. These pADSC exhibit strong potential to differentiate into adipocytes. Moreover, the pADSC also possess multipotency, assessed by selective stem cell markers, to differentiate into various mesenchymal cell types including adipocytes, osteocytes, and chondrocytes. These pADSC can be used for clarification of molecular switches in regulating classical adipocyte differentiation or in direction to other mesenchymal cell types of mesodermal origin. Furthermore, extended lineages into cells of ectodermal and endodermal origin have recently been achieved. Therefore, pADSC derived in this protocol provide an abundant and assessable source of adult mesenchymal stem cells with full multipotency for studying adipose development and application to tissue engineering of regenerative medicine.

This protocol describes the isolation of pig adipose-derived stem cells (pADSC) from subcutaneous adipose tissues with examination of multipotency. The isolation of adipose-derived stem cells (ASCs) is an important method in the field of adipose tissue biology, adipogenesis, and extracellular matrix (ECM) remodeling. In vivo, ECM-rich environment consisting of fibrillar collagens provides a structural support to adipose tissues during the progression and regression of obesity. Physiological ECM remodeling mediated by matrix metalloproteinases (MMPs) plays a major role in regulating adipose tissue size and function1,2. The loss of physiological collagenolytic ECM remodeling may lead to excessive collagen accumulation (tissue fibrosis), macrophage infiltration, and ultimately, a loss of metabolic homeostasis including insulin resistance3,4. When a phenotypic change of the adipose tissue is observed in gene-targeted mouse models, isolating primary ASCs from fat depots for in vitro studies is an effective approach to define the role of the specific gene in regulating the function of ASCs. Isolation and Expansion of Mesenchymal Stem/Stromal Cells Derived from Human Placenta Tissue

Mesenchymal stem/stromal cells (MSC) are promising candidates for use in cell-based therapies. In most cases, therapeutic response appears to be cell-dose dependent. Human term placenta is rich in MSC and is a physically large tissue that is generally discarded following birth. Placenta is an ideal starting material for the large-scale manufacture of multiple cell doses of allogeneic MSC. The placenta is a fetomaternal organ from which either fetal or maternal tissue can be isolated. This article describes the placental anatomy and procedure to dissect apart the decidua (maternal), chorionic villi (fetal), and chorionic plate (fetal) tissue. The protocol then outlines how to isolate MSC from each dissected tissue region, and provides representative analysis of expanded MSC derived from the respective tissue types. These methods are intended for pre-clinical MSC isolation, but have also been adapted for clinical manufacture of placental MSC for human therapeutic use.

Large-Scale Production of Cardiomyocytes from Human Pluripotent Stem Cells Using a Highly Reproducible Small Molecule-Based Differentiation Protocol

Maximizing the benefit of human pluripotent stem cells (hPSCs) for research, disease modeling, pharmaceutical and clinical applications requires robust methods for the large-scale production of functional cell types, including cardiomyocytes. Here we demonstrate that the temporal manipulation of WNT, TGF-&#946;, and SHH signaling pathways leads to highly efficient cardiomyocyte differentiation of single-cell passaged hPSC lines in both static suspension and stirred suspension bioreactor systems. Employing this strategy resulted in ~ 100% beating spheroids, consistently containing &#62; 80% cardiac troponin T-positive cells after 15 days of culture, validated in multiple hPSC lines. We also report on a variation of this protocol for use with cell lines not currently adapted to single-cell passaging, the success of which has been verified in 42 hPSC lines. Cardiomyocytes generated using these protocols express lineage-specific markers and show expected electrophysiological functionalities. Our protocol presents a simple, efficient and robust platform for the large-scale production of human cardiomyocytes.

Here, we present a robust, fast and scalable cardiomyocyte differentiation protocol for human pluripotent stem cells (hPSCs). Cardiomyocytes derived using this large-scale method can provide sufficient cell numbers for their effective use in human cardiovascular disease modeling, high-throughput drug screening, and potentially clinical applications.

Chondrogenic Differentiation Induction of Adipose-derived Stem Cells by Centrifugal Gravity

Impaired cartilage cannot heal naturally. Currently, the most advanced therapy for defects in cartilage is the transplantation of chondrocytes differentiated from stem cells using cytokines. Unfortunately, cytokine-induced chondrogenic differentiation is costly, time-consuming, and associated with a high risk of contamination during in vitro differentiation. However, biomechanical stimuli also serve as crucial regulatory factors for chondrogenesis. For example, mechanical stress can induce chondrogenic differentiation of stem cells, suggesting a potential therapeutic approach for the repair of impaired cartilage. In this study, we demonstrated that centrifugal gravity (CG, 2,400 &#215; g), a mechanical stress easily applied by centrifugation, induced the upregulation of sex determining region Y (SRY)-box 9 (SOX9) in adipose-derived stem cells (ASCs), causing them to express chondrogenic phenotypes. The centrifuged ASCs expressed higher levels of chondrogenic differentiation markers, such as aggrecan (ACAN), collagen type 2 alpha 1 (COL2A1), and collagen type 1 (COL1), but lower levels of collagen type 10 (COL10), a marker of hypertrophic chondrocytes. In addition, chondrogenic aggregate formation, a prerequisite for chondrogenesis, was observed in centrifuged ASCs.

Rapid Isolation of BMPR-IB+ Adipose-Derived Stromal Cells for Use in a Calvarial Defect Healing Model

Invasive cancers, major injuries, and infection can cause bone defects that are too large to be reconstructed with preexisting bone from the patient's own body. The ability to grow bone de novo using a patient's own cells would allow bony defects to be filled with adequate tissue without the morbidity of harvesting native bone. There is interest in the use of adipose-derived stromal cells (ASCs) as a source for tissue engineering because these are obtained from an abundant source: the patient's own adipose tissue. However, ASCs are a heterogeneous population and some subpopulations may be more effective in this application than others. Isolation of the most osteogenic population of ASCs could improve the efficiency and effectiveness of a bone engineering process. In this protocol, ASCs are obtained from subcutaneous fat tissue from a human donor. The subpopulation of ASCs expressing the marker BMPR-IB is isolated using FACS. These cells are then applied to an in vivo calvarial defect healing assay and are found to have improved osteogenic regenerative potential compared with unsorted cells.

In Vivo Tracking of Human Adipose-derived Mesenchymal Stem Cells in a Rat Knee Osteoarthritis Model with Fluorescent Lipophilic Membrane Dye

In order to support the clinical application of human adipose-derived mesenchymal stem cell (haMSC) therapy for knee osteoarthritis (KOA), we examined the efficacy of cell persistence and biodistribution of haMSCs in animal models. We demonstrated a method to label the cell membrane of haMSCs with lipophilic fluorescent dye. Subsequently, intra-articular injection of the labeled cells in rats with surgically induced KOA was monitored dynamically by an in vivo imaging system. We employed the lipophilic carbocyanines DiD (DilC18 (5)), a far-red fluorescent Dil (dialkylcarbocyanines) analog, which utilized a red laser to avoid excitation of the natural green autofluorescence from surrounding tissues. Furthermore, the red-shifted emission spectra of DiD allowed deep-tissue imaging in live animals and the labeling procedure caused no cytotoxic effects or functional damage to haMSCs. This approach has been shown to be an efficient tracking method for haMSCs in a rat KOA model. The application of this method could also be used to determine the optimal administration route and dosage of MSCs from other sources in pre-clinical studies.
