1.マウスの飼育

</ol>2.マウスの感染
<ol><li>感染の1日前にマウスの背中を体の中央から尾の付け根近くまで剃ります。麻酔薬の使用は必須ではありません。</li>
	<li>前述のように、各マウスに200匹の生存可能な第3段階のN.ブラジリエンシス幼虫(L3)を腰部に100 μLのリン酸緩衝生理食塩水(PBS)10で接種するか、対照として100 μLのPBSを単独で注射します。感染後4日目、7日目、または10日目に動物を犠牲にします。</li>
</ol>3.肺、小腸、縦隔、腸間膜リンパ節の分離
<ol><li>頸部脱臼によってマウスを安楽死させる。マウスを背中に置き、70%エタノールをスプレーします。ハサミを使用して正中切開を行い、皮膚を開いて腹部と胸部を露出させます。 注:イソフルランは代替の安楽死方法として使用できます12。二酸化炭素室は、CO2 が肺組織の損傷や出血を引き起こすため、推奨されません13。</li>
	<li>縦隔リンパ節および肺の分離<ol><li>胸骨を切開し、「V」字型にカットして肋骨と胸筋を取り除きます。胸腔が露出したら、心臓の下の食道の隣に縦隔リンパ節を配置します( 補足図1を参照)。</li>
		<li>縦隔リンパ節を抽出し、12ウェル培養プレート内の1 mLのR-10培地(表1)に回収します。処理するまで光から保護された氷の上に保管してください。 注:これらの実験で使用したレポーターマウスからの蛍光シグナルが減少しないように、サンプルを光から保護する必要があります。</li>
		<li>肺を抽出し、マウス1匹あたり12ウェル培養プレートに1 mLのR-10培地で採取します。処理するまで光から保護された氷の上に保管してください。</li>
	</ol></li>
	<li>腸間膜リンパ節鎖および小腸の分離<ol><li>腹膜腔を露出させ、小腸を慎重に右に動かして、結腸に沿って腸間膜リンパ節(MLN)鎖を露出させます。</li>
		<li>鉗子を使用して、MLNを取り外し、ペーパータオルでそっと転がし、脂肪を引き抜きます。</li>
		<li>MLNを12ウェル培養プレート内の1 mLのR-10培地に移します。処理するまで光から保護された氷の上に保管してください。</li>
		<li>幽門括約筋のすぐ下と盲腸の上の小腸を切断します。鉗子の助けを借りて腸をゆっくりと引き出し、付着した腸間膜と脂肪組織を取り除きます。</li>
		<li>小腸をペーパータオルの上に置き、PBSでたっぷりと湿らせます。ハサミで小腸からパイエル板を取り除きます。 注意: 生存率を維持するために、残りの脂肪組織を取り除き、プロセス全体を通して小腸をPBSで湿らせてください。</li>
		<li>ハサミを使って小腸を縦方向に切り、開いた腸の上に鉗子をそっとスライドさせて糞便と粘液を取り除きます。</li>
		<li>小腸を鉗子で保持し、氷上で5 mLのPBSで数回注意深く沈めて洗浄します。2回繰り返します。</li>
		<li>小腸を短い断片(約5 mm)に切断し、2%FBSを含む10 mLのHBSS14 を含む50 mLの円錐管に収集します(表1)。直ちに上皮内および固有層細胞を単離し続ける。</li>
	</ol></li>
</ol>4.小腸、肺、リンパ節からの単一細胞懸濁液の調製
注:小腸から単一細胞懸濁液を調製する場合、処理期間が長くなると細胞の生存率が大幅に低下するため、1人あたり最大6匹のマウスを処理することが非常に重要です。この方法は、 ヘリグモソモイデス・ポリギラス 感染マウスモデル5から適応した。
<ol><li>振とうインキュベーター、R-20培地、HBSS、およびHBSS-2 mM EDTA(表1)を37°Cで予熱します。</li>
	<li>サンプルあたり10 mLの小腸消化培地(表1)を調製します。</li>
	<li>ステップ3.3.8の腸片を手で激しく振る。</li>

	<li>漏斗からメッシュを取り外し、10 mLの温かいHBSS-2 mM EDTAを含む50 mLのコニカルチューブでメッシュからサンプルを収集します(ステップ4.1)。200rpmで振とうしながら、37°Cで10分間インキュベートします。</li>
	<li>最高速度(3,200 rpm)で10秒間ボルテックスし、ガラス漏斗上でメッシュでサンプルをろ過し、50 mLのコニカルチューブでフロースルーを回収します。</li>
	<li>手順4.5と4.6を2回繰り返し、同じ50 mLコニカルチューブでフロースルーを回収します。上皮内細胞はこの30mL画分に位置する。残りの組織を保存します。</li>
	<li>上皮内細胞からの単一細胞懸濁液調製<ol><li>ステップ4.7で回収した30 mLの細胞懸濁液を遠心分離し、室温(RT)で5分間450 x g で回収します。上澄み液を捨てる。</li>
		<li>5 mLのPBSを加え、450 x g でRTで5分間遠心分離します。 上清を捨てます。</li>
		<li>細胞ペレットを3 mLのRPMI 10%FBS-20 μg/mL DNaseに再懸濁します(表1)。細胞染色まで光から保護された氷上に保管してください。</li>
	</ol></li>
	<li>固有層細胞からの単一細胞懸濁液調製<ol><li>ステップ4.7の残りの小腸組織を、漏斗のメッシュを通して10mL以上の温かいHBSSを注ぐことによって洗浄します。洗浄を繰り返します。10 mLの小腸消化培地を含む50 mLのコニカルチューブのメッシュから組織を収集します。</li>
		<li>200 rpmで振とうしながら、37°Cで30分間インキュベートします。5分ごとに10秒間最高速度で渦を巻きます。</li>
		<li>10 mLのFACS-EDTAバッファー(表1)を加えて消化反応を停止し、氷上に置きます。</li>
		<li>血清学的ピペットを使用して100 μmセルストレーナーを通して各サンプルをろ過し、氷上に置かれた50 mLコニカルチューブで懸濁液を回収します。</li>
		<li>600 x g で4°Cで6分間遠心分離します。</li>
		<li>上澄み液を捨てる。細胞ペレットを5 mLのPBSで洗浄し、600 x g で4°Cで6分間遠心分離します。</li>
		<li>上清を廃棄し、細胞ペレットを1 mLのRPMI 10%FBS-20 μg/mL DNaseに再懸濁します。細胞染色まで光から保護された氷上に保管してください。</li>
	</ol></li>
	<li>肺からの単一細胞懸濁液の調製 注:各肺と小腸は、細胞の生存率が低くなる処理時間の延長を避けるために、2人で並行して処理する必要があります。<ol><li>処理したサンプルごとに4 mLの肺消化培地(表1)を準備します。</li>
		<li>ステップ3.2.3の肺を含むウェルからRPMI培地を取り除きます。細かいハサミで肺を細かく切ります。</li>
		<li>肺片をスパチュラ付きの15mLコニカルチューブに移します。4 mLの肺消化培地を追加します(表1)。</li>
		<li>250 rpmで振とうしながら、37°Cで30分間インキュベートします。終了したら、各サンプルが処理されるまで氷上に保ちます。</li>
		<li>各サンプルを100 μmのセルストレーナーでろ過し、6ウェル培養プレートの単一ウェルに配置します。シリンジプランジャーで組織を解離します。</li>
		<li>各サンプルを15 mLのコニカルチューブに回収し、4 mLのR-2培地を加えます(表1)。他のサンプルが処理されている間、氷上に保管してください。</li>
		<li>600 x g で4°Cで5分間遠心分離します。 上清を捨て、細胞ペレットを1 mLのR-5培地に再懸濁します(表1)。</li>
		<li>遠心分離しながら、造血細胞画分15、16を富化するために、サンプルあたり4mLの27.5%密度勾配溶液(表1)を調製する。単一細胞分離のためのこの戦略は、他の同様の密度勾配媒体17と比較して、より効率的で、費用効果が高く、毒性が低い。</li>
		<li>ステップ4.10.7の細胞懸濁液1 mLに4 mLの27.5%密度勾配溶液を加え、激しく振とうします。</li>
		<li>混合懸濁液の上に1 mLのR-5培地(表1)をゆっくりと加え、2つの相を作ります。</li>
		<li>1,500 x g でRTで20分間遠心分離し、低加速でブレーキをオフにします。2つの相の間に形成されたリングを観察します。</li>
		<li>1 mLマイクロピペットで2つの相の間に形成されたリングを回収し、4 mLのR-2培地に再懸濁します。</li>
		<li>450 x g で4°Cで5分間遠心分離します。 上澄み液を捨てる。細胞ペレットを1mLのACK緩衝液(表1)に再懸濁し、RTで1分間インキュベートします。</li>
		<li>4 mLのR-5培地を加え、450 x g で4°Cで5分間遠心分離します。 上清を捨て、細胞ペレットを1 mLのR-10培地に再懸濁します。フローサイトメトリーで染色するまで光から保護された氷上に保管してください。</li>
	</ol></li>
	<li>リンパ節からの単一細胞懸濁液調製<ol><li>ステップ3.2.2または3.3.3のリンパ節を6ウェル培養プレートのウェル内の2つのメッシュの間に置き、シリンジプランジャーで解離します。</li>
		<li>細胞懸濁液を 1.5 mL コニカルチューブで回収し、450 x g で 4 °C で 5 分間遠心分離します。</li>
		<li>上清を捨て、細胞ペレットを1 mLのR-10培地に再懸濁する。フローサイトメトリーで染色するまで光から保護された氷上に保管してください。 注:塊が見える場合は、100 μmのセルストレーナーでサンプルをろ過します。</li>

</ol>5. フローサイトメトリーのための細胞染色(図2および図3)
注:ステップ4.11.3のリンパ節細胞懸濁液を450 x g で4°Cで5分間遠心分離し、細胞ペレットを500 μLのFACSバッファーに再懸濁します(表1)。
<ol><li>ILC2の同定のための細胞染色(図3 および 補足図2) 注:この染色手順は、IL-9発現ILC2細胞の同定に特異的です。<ol><li>ステップ4.10.14の肺サンプルあたり150 μL(約1.8 x 10 6細胞)、およびステップ4.11.3のリンパ節サンプルあたり50 μL(縦隔および腸間膜リンパ節サンプルの場合はそれぞれ約0.7 x 10 6および2.2 x 106細胞)を96ウェル円錐形底部培養プレートにプレートします。100 μLのFACSバッファーを加え、450 x gで4°Cで5分間遠心分離します。</li>
		<li>小腸サンプルあたり100 μL(上皮内および固有層サンプルの場合はそれぞれ約2.7 x 10 6および0.6 x 106細胞)をステップ4.8.3および4.9.7から96ウェル円錐形底部培養プレートにプレートします。150 μLのFACSバッファーを加え、450 x gで4°Cで5分間遠心分離します。</li>
		<li>上清を捨て、各細胞ペレットをFACS緩衝液で希釈した50 μLのビオチン化抗体カクテル(表2)に再懸濁した。光から保護された4°Cで30分間インキュベートします。 注:上皮内および固有層サンプルには、20 μg/mL DNaseを含むFACSバッファーを使用してください。</li>
		<li>150 μLのFACSバッファーを加えます。450 x g で4°Cで5分間遠心分離します。</li>
		<li>上清を捨て、細胞ペレットを200 μLのFACSバッファーに再懸濁します。再度遠心分離し、上清を廃棄する。</li>
		<li>細胞ペレットを50 μLの抗体/染色カクテル(表2)に再懸濁し、光から保護された4°Cで30分間インキュベートします。</li>
		<li>150 μLのFACSバッファーを加えます。450 x g で4°Cで5分間遠心分離します。</li>
		<li>上清を捨て、細胞ペレットを200 μLのFACSバッファーに再懸濁します。再度遠心分離し、上清を廃棄する。洗浄を繰り返し(ステップ5.1.7)、上清を廃棄します。</li>
		<li>細胞ペレットを300 μLのFACSバッファーに再懸濁し、フローサイトメトリーで分析します。</li>
		<li>小腸および肺のサンプルについては、リンパ球、単一細胞、生細胞、造血細胞、CD90+系統細胞、ST2+細胞、およびIL-9+細胞(図3A、Bおよび補足図2A)のゲーティング戦略を使用します。リンパ節サンプルには、生細胞、単一細胞、CD90+系統細胞、ST2+細胞、およびIL-9+細胞というゲーティング戦略を使用します(補足図2B、C)。</li>

Introducing Shear Stress in the Study of Bacterial Adhesion

During bacterial infections a sequence of interactions occur between the pathogen and its host. Bacterial adhesion to the host cell surface is often the initial and determining step of the pathogenesis. Although experimentally adhesion is mostly studied in static conditions adhesion actually takes place in the presence of flowing liquid. First encounters between bacteria and their host often occur at the mucosal level, mouth, lung, gut, eye, etc. where mucus flows along the surface of epithelial cells. Later in infection, pathogens occasionally access the blood circulation causing life-threatening illnesses such as septicemia, sepsis and meningitis. A defining feature of these infections is the ability of these pathogens to interact with endothelial cells in presence of circulating blood. The presence of flowing liquid, mucus or blood for instance, determines adhesion because it generates a mechanical force on the pathogen. To characterize the effect of flowing liquid one usually refers to the notion of shear stress, which is the tangential force exerted per unit area by a fluid moving near a stationary wall, expressed in dynes/cm2. Intensities of shear stress vary widely according to the different vessels type, size, organ, location etc. (0-100 dynes/cm2). Circulation in capillaries can reach very low shear stress values and even temporarily stop during periods ranging between a few seconds to several minutes 1. On the other end of the spectrum shear stress in arterioles can reach 100 dynes/cm2 2. The impact of shear stress on different biological processes has been clearly demonstrated as for instance during the interaction of leukocytes with the endothelium 3. To take into account this mechanical parameter in the process of bacterial adhesion we took advantage of an experimental procedure based on the use of a disposable flow chamber 4. Host cells are grown in the flow chamber and fluorescent bacteria are introduced in the flow controlled by a syringe pump. We initially focused our investigations on the bacterial pathogen Neisseria meningitidis, a Gram-negative bacterium responsible for septicemia and meningitis. The procedure described here allowed us to study the impact of shear stress on the ability of the bacteria to: adhere to cells 1, to proliferate on the cell surface 5and to detach to colonize new sites 6 (Figure 1). Complementary technical information can be found in reference 7. Shear stress values presented here were chosen based on our previous experience1 and to represent values found in the literature. The protocol should be applicable to a wide range of pathogens with specific adjustments depending on the objectives of the study.

During the infection process, a key step is the adhesion of pathogens with host cells. In most instances this adhesion step occurs in the presence of mechanical stress generated by flowing liquid. We describe a technique that introduces shear stress as an important parameter in the study of bacterial adhesion.

Following Cell-fate in E. coli After Infection by Phage Lambda

The system comprising bacteriophage (phage) lambda and the bacterium E. coli has long served as a paradigm for cell-fate determination1,2. Following the simultaneous infection of the cell by a number of phages, one of two pathways is chosen: lytic (virulent) or lysogenic (dormant)3,4. We recently developed a method for fluorescently labeling individual phages, and were able to examine the post-infection decision in real-time under the microscope, at the level of individual phages and cells5. Here, we describe the full procedure for performing the infection experiments described in our earlier work5. This includes the creation of fluorescent phages, infection of the cells, imaging under the microscope and data analysis. The fluorescent phage is a "hybrid", co-expressing wild- type and YFP-fusion versions of the capsid gpD protein. A crude phage lysate is first obtained by inducing a lysogen of the gpD-EYFP (Enhanced Yellow Fluorescent Protein) phage, harboring a plasmid expressing wild type gpD. A series of purification steps are then performed, followed by DAPI-labeling and imaging under the microscope. This is done in order to verify the uniformity, DNA packaging efficiency, fluorescence signal and structural stability of the phage stock. The initial adsorption of phages to bacteria is performed on ice, then followed by a short incubation at 35&deg;C to trigger viral DNA injection6. The phage/bacteria mixture is then moved to the surface of a thin nutrient agar slab, covered with a coverslip and imaged under an epifluorescence microscope. The post-infection process is followed for 4 hr, at 10 min interval. Multiple stage positions are tracked such that ~100 cell infections can be traced in a single experiment. At each position and time point, images are acquired in the phase-contrast and red and green fluorescent channels. The phase-contrast image is used later for automated cell recognition while the fluorescent channels are used to characterize the infection outcome: production of new fluorescent phages (green) followed by cell lysis, or expression of lysogeny factors (red) followed by resumed cell growth and division. The acquired time-lapse movies are processed using a combination of manual and automated methods. Data analysis results in the identification of infection parameters for each infection event (e.g. number and positions of infecting phages) as well as infection outcome (lysis/lysogeny). Additional parameters can be extracted if desired.

Cutaneous Leishmaniasis in the Dorsal Skin of Hamsters: a Useful Model for the Screening of Antileishmanial Drugs

Traditionally, hamsters are experimentally inoculated in the snout or the footpad. However in these sites an ulcer not always occurs, measurement of lesion size is a hard procedure and animals show difficulty to eat, breathe and move because of the lesion. In order to optimize the hamster model for cutaneous leishmaniasis, young adult male and female golden hamsters (Mesocricetus auratus) were injected intradermally at the dorsal skin with 1 to 1.5 x l07 promastigotes of Leishmania species and progression of subsequent lesions were evaluated for up to 16 weeks post infection. The golden hamster was selected because it is considered the adequate bio-model to evaluate drugs against Leishmania as they are susceptible to infection by different species. Cutaneous infection of hamsters results in chronic but controlled lesions, and a clinical evolution with signs similar to those observed in humans. Therefore, the establishment of the extent of infection by measuring the size of the lesion according to the area of indurations and ulcers is feasible. This approach has proven its versatility and easy management during inoculation, follow up and characterization of typical lesions (ulcers), application of treatments through different ways and obtaining of clinical samples after different treatments. By using this method the quality of animal life regarding locomotion, search for food and water, play and social activities is also preserved.

Use of Galleria mellonella as a Model Organism to Study Legionella pneumophila Infection

Legionella pneumophila, the causative agent of a severe pneumonia named Legionnaires&#39; disease, is an important human pathogen that infects and replicates within alveolar macrophages. Its virulence depends on the Dot/Icm type IV secretion system (T4SS), which is essential to establish a replication permissive vacuole known as the Legionella containing vacuole (LCV). L. pneumophila infection can be modeled in mice however most mouse strains are not permissive, leading to the search for novel infection models. We have recently shown that the larvae of the wax moth Galleria mellonella are suitable for investigation of L. pneumophila infection. G. mellonella is increasingly used as an infection model for human pathogens and a good correlation exists between virulence of several bacterial species in the insect and in mammalian models. A key component of the larvae&#39;s immune defenses are hemocytes, professional phagocytes, which take up and destroy invaders. L. pneumophila is able to infect, form a LCV and replicate within these cells. Here we demonstrate protocols for analyzing L. pneumophila virulence in the G. mellonella model, including how to grow infectious L. pneumophila, pretreat the larvae with inhibitors, infect the larvae and how to extract infected cells for quantification and immunofluorescence microscopy. We also describe how to quantify bacterial replication and fitness in competition assays. These approaches allow for the rapid screening of mutants to determine factors important in L. pneumophila virulence, describing a new tool to aid our understanding of this complex pathogen.

Use of Galleria mellonella as a Model Organism to Study Legionella pneumophila Infection

A Rapid Strategy for the Isolation of New Faustoviruses from Environmental Samples Using Vermamoeba vermiformis

The isolation of giant viruses is of great interest in this new era of virology, especially since these giant viruses are related to protists. Giant viruses may be potentially pathogenic for many species of protists. They belong to the recently described order of Megavirales. The new lineage Faustovirus that has been isolated from sewage samples is distantly related to the mammalian pathogen African swine fever virus. This virus is also specific to its amoebal host, Vermamoeba vermiformis, a protist common in health care water systems. It is crucial to continue isolating new Faustovirus genotypes in order to enlarge its genotype collection and study its pan-genome. We developed new strategies for the isolation of additional strains by improving the use of antibiotic and antifungal combinations in order to avoid bacterial and fungal contaminations of the amoeba co-culture and favoring the virus multiplication. We also implemented a new starvation medium to maintain V. vermiformis in optimal conditions for viruses co-culture. Finally, we used flow cytometry rather than microscopic observation, which is time-consuming, to detect the cytopathogenic effect. We obtained two isolates from sewage samples, proving the efficiency of this method and thus widening the collection of Faustoviruses, to better understand their environment, host specificity and genetic content.

Assessment of the Synaptic Interface of Primary Human T Cells from Peripheral Blood and Lymphoid Tissue

The current understanding of the dynamics and structural features of T-cell synaptic interfaces has been largely determined through the use of glass-supported planar bilayers and in vitro-derived T-cell clones or lines1,2,3,4. How these findings apply to the primary human T cells isolated from blood or lymphoid tissues is not known, partly due to significant difficulties in obtaining a sufficient number of cells for analysis5. Here we address this through the development of a technique exploiting multichannel flow slides to build planar lipid bilayers containing activating and adhesion molecules. The low height of the flow slides promotes rapid cell sedimentation in order to synchronize cell:bilayer attachment, thereby allowing researchers to study the dynamic of the synaptic interface formation and the kinetics of the granules release. We apply this approach to analyze the synaptic interface of as few as 104 to 105 primary cryopreserved T cells isolated from lymph nodes (LN) and peripheral blood (PB). The results reveal that the novel planar lipid bilayer technique enables the study of the biophysical properties of primary human T cells derived from blood and tissues in the context of health and disease.

The Galleria mellonella Waxworm Infection Model for Disseminated Candidiasis

Candida species are common fungal commensals of humans colonizing the skin, mucosal surfaces, and gastrointestinal tract. Under certain conditions, Candida can overgrow their natural niches resulting in debilitating mucosal infections as well as life-threatening systemic infections, which are a major focus of investigation due to their associated high mortality rates. Animal models of disseminated infection exist for studying disease progression and dissecting the characteristics of Candida pathogenicity. Of these, the Galleria mellonella waxworm infection model provides a cost-effective experimental tool for high-throughput investigations of systemic virulence. Many other bacterial and eukaryotic infectious agents have been effectively studied in G. mellonella to understand pathogenicity, making it a widely accepted model system. Yet, variation in the method used to infect G. mellonella can alter phenotypic outcomes and complicate interpretation of the results. Here, we outline the benefits and drawbacks of the waxworm model to study systemic Candida pathogenesis and detail an approach to improve reproducibility. Our results highlight the range of mortality kinetics in G. mellonella and describe the variables which can modulate these kinetics. Ultimately, this method stands as an ethical, rapid, and cost-effective approach to study virulence in a model of disseminated candidiasis.

A Galleria mellonella Oral Administration Model to Study Commensal-Induced Innate Immune Responses

The investigation of the immunogenic potential of commensal bacteria on the host immune system is one essential component when studying intestinal host-microbe interactions. It is well established that different commensals exhibit a different potential to stimulate the host intestinal immune system. Such investigations involve vertebrate animals, especially rodents. Since increasing ethical concerns are linked with experiments involving vertebrates, there is a high demand for invertebrate replacements models. 
Here, we provide a Galleria mellonella oral administration model using commensal non-pathogenic bacteria and the possible assessment of the immunogenic potential of commensals on the G. mellonella immune system. We demonstrate that G. mellonella is a useful alternative invertebrate replacement model that allows the analysis of commensals with different immunogenic potential such as Bacteroides vulgatus and Escherichia coli. Interestingly, the bacteria exhibited no killing effect on the larvae, which is similar to mammals. The immune responses of G. mellonella were comparable with vertebrate innate immune responses and involve recognition of the bacteria and production of antimicrobial molecules. We propose that G. mellonella was able to restore previous microbiota balance, which is well known from healthy mammalian individuals. Although providing comparable innate immune responses in both G. mellonella and vertebrates, G. mellonella does not harbor an adaptive immune system. Since the investigated components of the innate immune system are evolutionary conserved, the model allows a prescreening and first analysis of bacterial immunogenic properties.

Use of the Invertebrate Galleria mellonella as an Infection Model to Study the Mycobacterium tuberculosis Complex

Tuberculosis is the leading global cause of infectious disease mortality and roughly a quarter of the world&#8217;s population is believed to be infected with Mycobacterium tuberculosis. Despite decades of research, many of the mechanisms behind the success of M. tuberculosis as a pathogenic organism remain to be investigated, and the development of safer, more effective antimycobacterial drugs are urgently needed to tackle the rise and spread of drug resistant tuberculosis. However, the progression of tuberculosis research is bottlenecked by traditional mammalian infection models that are expensive, time consuming, and ethically challenging. Previously we established the larvae of the insect Galleria mellonella (greater wax moth) as a novel, reproducible, low cost, high-throughput and ethically acceptable infection model for members of the M. tuberculosis complex. Here we describe the maintenance, preparation, and infection of G. mellonella with bioluminescent Mycobacterium bovis BCG lux. Using this infection model, mycobacterial dose dependent virulence can be observed, and a rapid readout of in vivo mycobacterial burden using bioluminescence measurements is easily achievable and reproducible. Although limitations exist, such as the lack of a fully annotated genome for transcriptomic analysis, ontological analysis against genetically similar insects can be carried out. As a low cost, rapid, and ethically acceptable model for tuberculosis, G. mellonella can be used as a pre-screen to determine drug efficacy and toxicity, and to determine comparative mycobacterial virulence prior to the use of conventional mammalian models. The use of the G. mellonella-mycobacteria model will lead to a reduction in the substantial number of animals currently used in tuberculosis research.

Lentiviral CRISPR/Cas9-Mediated Genome Editing for the Study of Hematopoietic Cells in Disease Models

Manipulating genes in hematopoietic stem cells using conventional transgenesis approaches can be time-consuming, expensive, and challenging. Benefiting from advances in genome editing technology and lentivirus-mediated transgene delivery systems, an efficient and economical method is described here that establishes mice in which genes are manipulated specifically in hematopoietic stem cells. Lentiviruses are used to transduce Cas9-expressing lineage-negative bone marrow cells with a guide RNA (gRNA) targeting specific genes and a red fluorescence reporter gene (RFP), then these cells are transplanted into lethally-irradiated C57BL/6 mice. Mice transplanted with lentivirus expressing non-targeting gRNA are used as controls. Engraftment of transduced hematopoietic stem cells are evaluated by flow cytometric analysis of RFP-positive leukocytes of peripheral blood. Using this method, ~90% transduction of myeloid cells and ~70% of lymphoid cells at 4 weeks after transplantation can be achieved. Genomic DNA is isolated from RFP-positive blood cells, and portions of the targeted site DNA are amplified by PCR to validate the genome editing. This protocol provides a high-throughput evaluation of hematopoiesis-regulatory genes and can be extended to a variety of mouse disease models with hematopoietic cell involvement.
