本研究に用いたMCF-7細胞は、市販の供給源から入手した( 材料表参照)。
1. 細胞培養
<ol><li>MCF-7細胞を加湿した5%CO2 雰囲気、37°C、10%FBSおよび1%ペニシリン(10,000 U/mL)/ストレプトマイシン(10,000 μg/mL)を含むDMEMで培養します( 材料表参照)。 注:ディッシュの底の90%を覆う細胞を実験に用い、対照群、塩酸ドキソルビシン群(DXR、5μM)、Sal群(20μM、40μM、80μM)、または対照群、LY294002(10μM、PI3K阻害剤)群、Sal(80μM)群、LY294002(10μM)+Sal(80μM)群に割り付けた( 材料表参照)。</li>
</ol>2. 細胞生存率アッセイ
注: この手順の詳細については、以前のレポート27 を参照してください。
<ol><li>密度8 x 104 細胞/ウェルのMCF-7細胞を96ウェルプレートに播種し、細胞が接着するまで、Sal(10 μM、20 μM、40 μM、80 μM、160 μM、および320 μM)で24時間またはSal(20、40、80 μM)で12時間/24時間/36時間/48時間インキュベートします。</li>
	<li>処理後、MCF-7細胞を10 μL/ウェルのCCK-8溶液( 材料表を参照)と2時間共インキュベートします。次に、自動マイクロプレートリーダーで450nm(OD450)の光学濃度を測定します。</li>
</ol>3. 細胞の遊走と浸潤
注:この手順の詳細については、以前のレポート35を参照してください。
<ol><li>2 mL の 5 x 10 6 細胞/mL 細胞を6 ウェルプレートに播種し、皿の底の 90% を覆うようにインキュベートします。</li>
	<li>滅菌ピペットチップで細胞単層の中心に沿って線状の引っ掻き傷を巻きます。0時間、24時間、48時間で光学顕微鏡を使用して画像を取得します。</li>
	<li>Image Jソフトウェアを使用して、引っかき傷の幅を測定します。</li>
	<li>トランズウェルアッセイでは、MCF-7細胞を血清培地なしで懸濁し、マトリゲルの有無にかかわらず、プレコートされた上部トランスウェルチャンバーにシードします( 材料表を参照)。</li>
	<li>トランズウェルチャンバーの底部では、化学誘導剤としてDMEM完全培地を使用します。24時間後、上部チャンバー内の細胞を除去し、残りの侵襲性細胞および移動細胞をメタノール35で固定する。</li>
	<li>MCF-7細胞をクリスタルバイオレット溶液で染色します( 材料表を参照)。光学顕微鏡を使用して画像を撮影します。</li>
</ol>4. Na+-K+-ATPaseおよびCa2+-Mg2+-ATPaseの活性評価
<ol><li>ビシンコニン酸(BCA)キットを使用して、製造元の指示に従って、溶解したMCF-7細胞のタンパク質濃度を測定します( 材料表を参照)。<ol><li>溶解した細胞のサンプルを対応するグループの96ウェルプレートに加えます。その後、作業溶液を加えて、さらに37°Cで5分間インキュベートします。最後に、多機能マイクロプレートリーダーでOD636 の値を測定します。</li>
	</ol></li>
</ol>5. アポトーシスと細胞周期のフローサイトメトリー解析
注:この手順の詳細については、以前のレポート31を参照してください。
<ol><li>細胞を消化し、回収してリン酸緩衝生理食塩水(PBS)に再懸濁し、アネキシンV-FITCおよびヨウ化プロピジウム(PI)( 材料表を参照)で20分間染色した後、フローサイトメーターを使用してアポトーシス細胞の数を決定します。</li>
	<li>再懸濁したサンプル溶液とPI溶液を混合して30分間インキュベーションした後、フローサイトメーターで検出します。</li>
</ol>6. DCFH-DAおよびFluo-4 AM蛍光染色
注: この手順の詳細については、以前のレポート29 を参照してください。
<ol><li>MCF-7細胞を10 μM DCFH-DA 蛍光プローブ( 材料表参照)を用いて37°Cで20分間インキュベートします。 PBSで3回洗浄して余剰DCFH-DAを十分に除去した後、励起波長488nm、発光波長525nmの蛍光強度を蛍光顕微鏡で検査します。</li>
	<li>MCF-7細胞を5 μM Fluo-4 AM蛍光プローブ溶液( 材料表参照)と37°Cで45分間インキュベートします。 PBSで3回洗浄した後、蛍光顕微鏡を用いて、励起波長488nm、発光波長516nmの蛍光強度値を求めます。</li>
</ol>7.ウェスタンブロット
<ol><li>異なる薬物を含む5 x 10 6細胞/mL細胞懸濁液2 mLを6 ウェルプレートに加え、37°C、5%CO2 インキュベーターで24時間培養します。 注:ウェスタンブロット解析のグループは、対照群、LY294002(10μM)群、Sal(80μM)群、LY294002(10μM)+Sal(80μM)群として設定しました( 材料表参照)。</li>
	<li>細胞と上清を回収し、560 × g 、4°C、3分間遠心分離します。上澄みを捨て、予冷したPBSで2回洗浄する。各洗浄後、細胞を560 × g 、4°C、3分間遠心分離します。</li>
	<li>ステップ 7.2 の細胞サンプルに 50 μL の溶解バッファーを加え、15 分間のアイスバスを行います。8550 × g 、4°C、10分間遠心分離し、上清タンパク質サンプルを回収します。</li>
	<li>BCA法を用いてタンパク質濃度を検出した後、ステップ7.3のタンパク質サンプルとローディングバッファーを4:1の割合で混合し、金属浴中で100°Cで10分間変性させ、室温まで冷却します。</li>
	<li>ステップ7.4のタンパク質サンプル20 μgを添加し、10% SDS-PAGE(材料表を参照)を使用して分子量35の異なるタンパク質を分離し、0.22 μmのPVDFメンブレンに移します。5% BSAで閉塞した後、メンブレンを対応する一次抗体とともに4°Cで一晩インキュベートします。 注:一次抗体の希薄濃度は1:1,000です:切断カスパーゼ-9/7/3(CC-9/7/3)、Bim、Bax、Bcl-2、p-PI3K/PI3K、p-AKT/AKT、mTOR、HIF-1α、FoxO1、およびβ-アクチン( 材料表を参照)。</li>
	<li>翌日、メンブレンをヤギ抗ウサギIgG二次抗体と37°Cで2時間インキュベートします。 ECL化学発光溶液を使用してメンブレンを現像し、非接触定量ウェスタンブロットイメージングシステムを使用して画像を撮影します( 材料表を参照)。</li>
</ol>8. qRT-PCRの
<ol><li>遠心分離を行い、MCF-7細胞を560 x g 、4°C、3分間回収し、500 μLのバッファーRL1を5 × 106 細胞に加えます。細胞塊が見えなくなるまでピペットで繰り返しブローして混合します。 注:バッファー RL1 は、トータル RNA 単離キットのコンポーネントの 1 つです( 材料表を参照)。</li>
	<li>ステップ8.1の細胞ホモジネートをコレクションチューブに埋め込まれたDNAクリーニングカラムに移し、8,550 × g 、4°C、2分間遠心分離します。DNAクリーニングカラムを取り外し、上清をコレクションチューブに保管します。 注:DNA クリーニングカラムとコレクションチューブは、トータル RNA 分離キットの 2 つのコンポーネントです( 材料表を参照)。</li>
	<li>ステップ8.2の上清500μLに緩衝液RL2 800 μLを加え、穏やかに混合する。 注:バッファー RL2 は、トータル RNA 単離キットのコンポーネントの 1 つです( 材料表を参照)。使用前に、60 mLの緩衝液RL2に120 mLの無水エタノールを加えてください。</li>
	<li>ステップ 8.3 の混合物 700 μL をコレクションチューブに埋め込まれた RNA 専用カラムに移し、8,550 × g で 4 °C で 1 分間遠心分離します。回収管内の廃液を廃棄します。 注:RNA 専用カラムは、トータル RNA 単離キットのコンポーネントの 1 つです( 材料表を参照)。</li>
	<li>ステップ8.4を繰り返して、ステップ8.3の残りの混合物を処理します。</li>
	<li>500 μL のバッファー RW1 を RNA 専用カラムに添加し、8,550 × g で 4 °C、1 分間遠心分離します。回収管内の廃液を廃棄します。 注:バッファー RW1 は、トータル RNA 単離キットのコンポーネントの 1 つです( 材料表を参照)。</li>
	<li>700 μL のバッファー RW2 を RNA 専用カラムに添加し、8,550 × g で 4 ◦C で 1 分間遠心分離します。回収管内の廃液を廃棄します。手順8.7を一度繰り返します。 注:バッファー RW2 は、トータル RNA 単離キットの成分の 1 つです( 材料表を参照)。</li>
	<li>RNA のみのカラムをコレクションチューブに戻し、8550 × g で 4 °C、2 分間遠心分離して、残りのバッファー RW2 を除去します。</li>
	<li>RNA 専用カラムを新しいコレクションチューブに移し、65 °C で予熱した RNase フリー ddH2O 100 μL を RNA 専用カラムのメンブレンの中央に滴下します。25°Cで2分間置き、8,550 × g 、4°C、1分間遠心分離してRNA溶液を回収します。 注:RNaseフリーddH2Oは、total RNA単離キットの成分の1つです( 材料表を参照)。</li>
	<li>5x反応バッファー4 μL、オリゴ(dT)18 プライマー1 μL、ランダム六量体プライマー1 μL、遺伝子特異的プライマー1 μL(表1を参照)、酵素ミックス1 μL、およびステップ8.9のRNA溶液5 μgを100 μLの8チューブストリップに加えます。反応系にRNaseフリーの水を20 μLまで添加します。</li>
	<li>システムの反応条件を次のように設定します:(1):95°C、30秒、1サイクル。(2):95°C、5秒、50サイクル、60°C、34秒;(3):95°C、5秒、1サイクル、65°C、60秒、97°C、1秒;(4):42°C、30秒、1サイクル。qRT-PCR手順を実行します。 注:標的遺伝子の相対発現レベルは、2−ΔΔCT法36,37によって定量的に分析された。qRT-PCR解析に用いた各遺伝子のプライマー情報を表1に示します。</li>
</ol>9. 統計解析
<ol><li>データを 3 つの独立した実験の平均±標準偏差として表します。</li>
	<li>グラフ作成および分析ソフトウェア( 材料表を参照)を使用して、一元配置分散分析(ANOVA)とそれに続くテューキーの検定を使用して、複数のグループ間の比較を分析します。この研究では、 P < 0.05 を統計的に有意なものとして定義しました。</li>

サリドロシドのMCF-7細胞の遊走・増殖抑制機構

乳がんはあらゆる年齢層に罹患し、計り知れない肉体的・精神的負担と大きな経済的圧力を引き起こします1。また、罹患率や死亡率が年々増加している乳がんは、従来の治療法を超えた効果的なハーブベースの複合療法の模索という点でも世界的に注目されています4,5。有望なことに、多くの証拠がSal 24,25,38<sup class="xref"...

最も一般的に診断されるがんの1つであり、最も一般的な悪性腫瘍の1つである乳がんは、最新の統計によると、2020年までに世界中で230万例が発生し、全がん症例の11.7%を占めています1。乳がんの一般的な症状には、乳房の圧痛とうずき、乳房のしこりと痛み、乳頭の分泌物、びらんまたはくぼんだ皮膚、および腋窩リンパ節の肥大が含まれます1,2。さらに憂慮すべきことに、乳がんの新規患者数と全体的な発生率は毎年圧倒的な速度で増加し続けており、がん関連死の6.9%を占めています1。現在、乳がんの介入は、化学療法、手術、放射線療法、および包括的な治療が中心です。治療は患者の再発率と死亡率を効果的に低下させることができますが、長期治療の適用はしばしば多剤耐性、広範囲の脱毛、吐き気と嘔吐を引き起こし、深刻な精神的および心理的負担を引き起こします2,3。特に、乳がんによる多臓器転移の潜在的なリスクは、薬物療法の新しいハーブ源を探すことを人々に強いています4,5。
ホスホイノシチド3キナーゼ(PI3K)を介したシグナル伝達は、複数の遺伝子の発現に影響を与えるスプライシングを介して、乳がんの増殖、増殖、および生存に関与しています6。PI3Kの下流シグナルセンシングタンパク質として、プロテインキナーゼB(AKT)が哺乳類のラパマイシン標的(mTOR)タンパク質と結合して乳がんをさらに増加させる可能性があることを示唆する多くの証拠があります7,8,9。さらに、PI3K / AKT / mTORシグナル伝達の不活性化は、乳がんの悪性増殖を阻害し、アポトーシスを刺激する薬物の重要な要素であるとも主張されています10,11,12。腫瘍微小環境における極端な低酸素状態は、低酸素誘導性第1因子アルファ(HIF-1α)の大規模な急増を余儀なくされ、乳がんの進行をさらに悪化させることはよく知られています13,14,15。並行して、AKT刺激はHIF-1αの過剰蓄積にもつながり、乳がんサンプルのアポトーシスを制限します16,17。興味深いことに、PI3K-AKT-HIF-1αシグナル伝達の活性化は、肺がん18、大腸がん19、卵巣がん20、前立腺がん21など、さまざまながんの病理学的進行と転移に関与していることが確認されています。HIF-1αによって調整されることに加えて、分岐頭部転写因子1(FoxO1)の過剰発現は、AKTシグナル伝達刺激によっても引き起こされ、乳がん細胞の周期停止とアポトーシスの阻害を促進します22,23。まとめると、上記の確かな証拠は、PI3K-AKT-HIF-1α-FoxO1シグナル伝達のカスケードの阻害が、乳がんにおける薬物療法の潜在的な新規標的である可能性があることを示唆しています。
サリドロシド(Sal)は、抗癌24,25、抗低酸素26,27,28,29、および免疫増強薬理活性30を発揮することが広く実証されています。水に溶けやすい薄茶色または褐色の粉末であり、フェニルエタノイド配糖体の一種であり、化学構造式はC14H 20 O7であり、分子量は300.331,32である。現代の薬理学的研究は、SalがPI3K-AKT-mTORシグナル伝達を抑制することにより、胃癌細胞のアポトーシスを促進することができることを実証している24。さらなる証拠はまた、Sal治療によるPI3K-AKT-HIF-1αシグナル伝達の抑制が、化学療法剤に対する感受性を高めることによって癌細胞のアポトーシスに寄与する可能性があることを示唆しています25。証拠はまた、サルが細胞の移動と浸潤を制限し、ヒト乳がんMCF-7細胞のアポトーシスを促進することによって周期停止を引き起こすことを示唆しています33,34。しかし、SalがPI3K-AKT-HIF-1α-FoxO1シグナル伝達を調節し、MCF-7細胞の悪性増殖を阻害できるかどうかは、まだ不明である。従って、このプロトコルはPI3K-AKT-HIF-1α-FoxO1細道によってMCF-7セル移動、浸潤、細胞周期およびapoptosisに対するSalの効果を探検することを向けた。細胞遊走や浸潤の評価、フローサイトメトリーによるアポトーシスや細胞周期の検出、活性酸素種(ROS)、Ca2+蛍光測定など、従来型の低コストで独立した実験からなる統合研究戦略は、従来の漢方薬を用いた抗がん研究の実験の全体的なデザインの参考となります。本研究の実験過程を図1に示す。

As one of the most commonly diagnosed cancers and most common malignancies, the latest statistics indicate that 2.3 million cases of breast cancer emerged around the world by 2020, accounting for 11.7% of all cancer cases1. Common symptoms of breast cancer include breast tenderness and tingling, breast lumps and pain, nipple discharge, erosion or sunken skin, and enlarged axillary lymph nodes1,2. Even more alarming, the number of new cases and the overall incidence of breast cancer continues to increase at an overwhelming rate each year, accounting for 6.9% of cancer-related deaths1. At present, breast cancer intervention still mainly involves chemotherapy, surgery, radiotherapy, and comprehensive treatment. Although treatment can effectively reduce the recurrence rate and mortality rate of patients, long-term treatment application often causes produce multidrug resistance, large-area hair loss, nausea and vomiting, and serious mental and psychological burden2,3. Notably, the potential risk of multiple organ metastases from breast cancer also forces people to seek novel herbal sources of drug therapy4,5.
Phosphoinositide 3 kinase (PI3K)-mediated signaling is implicated in the growth, proliferation, and survival of breast cancer through splicing that affects the expression of multiple genes6. As a downstream signal-sensing protein of PI3K, numerous evidence suggests that protein kinase B (AKT) could couple with the mammalian target of rapamycin (mTOR) protein to further increase breast cancer7,8,9. Moreover, the deactivation of PI3K/AKT/mTOR signaling has also been claimed to be a key component in drugs inhibiting malignant proliferation and stimulating apoptosis in breast cancer10,11,12. It is well known that extreme hypoxia in the tumor microenvironment forces a massive surge in hypoxia-inducible factor 1 alpha (HIF-1&#945;), which further worsens the progression of breast cancer13,14,15. In parallel, AKT stimulation also leads to excessive accumulation of HIF-1&#945;, limiting apoptosis in breast cancer samples16,17. Interestingly, the activation of PI3K-AKT-HIF-1&#945; signaling has been confirmed to be involved in pathologic progression and metastasis in a variety of cancers, including lung cancer18, colorectal cancer19, ovarian cancer20, and prostate cancer21. In addition to being orchestrated by HIF-1&#945;, forked head transcription factor 1 (FoxO1) overexpression is also triggered by AKT signaling stimulation, which promotes cycle arrest and the inhibition of apoptosis in breast cancer cells22,23. Together, the above solid evidence suggests that the inhibition of the cascade of PI3K-AKT-HIF-1&#945;-FoxO1 signaling may be a potential novel target for drug therapy in breast cancer.
Salidroside (Sal) has been widely demonstrated to exert anti-cancer24,25, anti-hypoxia26,27,28,29, and immune-enhancing pharmacological activities30. It is a light brown or brown powder that is easily soluble in water, is a type of phenylethanoid glycoside, and has a chemical structure formula of C14H20O7 and a molecular weight of 300.331,32. Modern pharmacological investigations have demonstrated that Sal can promote the apoptosis of gastric cancer cells by restraining PI3K-AKT-mTOR signaling24. Further evidence also suggests that the suppression of PI3K-AKT-HIF-1&#945; signaling by Sal treatment may contribute to the apoptosis of cancer cells by enhancing their sensitivity to chemotherapeutic agents25. Evidence also suggests that Sal restricts cell migration and invasion and causes cycle arrest by promoting apoptosis in the human breast cancer MCF-7 cells33,34. However, it remains to be seen whether Sal can regulate PI3K-AKT-HIF-1&#945;-FoxO1 signaling and inhibit the malignant proliferation of MCF-7 cells. Therefore, this protocol aimed to explore the effects of Sal on MCF-7 cell migration, invasion, cell cycle, and apoptosis via the PI3K-AKT-HIF-1&#945;-FoxO1 pathway. The integrated research strategies comprising conventional, low-cost, and independent experiments, such as cell migration and invasion assessments, apoptosis and cell cycle detection by flow cytometry, reactive oxygen species (ROS) and Ca2+ fluorescence determination, etc., can provide a reference for the overall design of experiments for anti-cancer research with traditional herbal medicine. The experimental process of this study is shown in Figure 1.

We focus on exploring active compounds derived from natural products and their potential in treating breast cancer. So specifically we aim to elucidate the pharmacologic effects of salidroside and its molecular mechanisms in combining breast cancer. Currently, reliable experimental techniques are lacking to validate salidroside's molecular targets in treating breast cancer.One significant finding in the field is a potential molecular mechanism of salidroside in enhancing the proliferation and migration of MCF7 cells through the regulation of PI3K/AKT/HIF-1 alpha/FoxO1 signaling. The laboratory's future research directly is to explore the pathophysiological mechanisms underlying breast cancer onset, development, prognosis and outcome. Additionally, we screen for active small molecular drugs derived from natural sources that may add in breast cancer prevention and treatment.

Salidroside (Sal) contains anti-carcinogenic, anti-hypoxic, and anti-inflammatory pharmacological activities. However, its underlying anti-breast cancer mechanisms have been only incompletely elucidated. Hence, this protocol intended to decode the potential of Sal in regulating the PI3K-AKT-HIF-1&#945;-FoxO1 pathway in the malignant proliferation of human breast cancer MCF-7 cells. First, the pharmacological activity of Sal against MCF-7 was evaluated by CCK-8 and cell scratch assays. Moreover, the resistance of MCF-7 cells was measured by migration and Matrigel invasion assays. For cell apoptosis and cycle assays, MCF-7 cells were processed in steps with annexin V-FITC/PI and cell cycle-staining detection kits for flow cytometry analyses, respectively. The levels of reactive oxygen species (ROS) and Ca2+ were examined by DCFH-DA and Fluo-4 AM immunofluorescence staining. The activities of Na+-K+-ATPase and Ca2+-ATPase were determined using the corresponding commercial kits. The protein and gene expression levels in apoptosis and the PI3K-AKT-HIF-1&#945;-FoxO1 pathway were further determined using western blot and qRT-PCR analyses, respectively. We found that Sal treatment significantly restricted the proliferation, migration, and invasion of MCF-7 cells with dose-dependent effects. Meanwhile, Sal administration also dramatically forced MCF-7 cells to undergo apoptosis and cell cycle arrest. The immunofluorescence tests showed that Sal observably stimulated ROS and Ca2+ production in MCF-7 cells. Further data confirmed that Sal promoted the expression levels of pro-apoptotic proteins, Bax, Bim, cleaved caspase-9/7/3, and their corresponding genes. Consistently, Sal intervention prominently reduced the expression of the Bcl-2, p-PI3K/PI3K, p-AKT/AKT, mTOR, HIF-1&#945;, and FoxO1 proteins and their corresponding genes. In conclusion, Sal can be used as a potential herb-derived compound for treating breast cancer, as it may reduce the malignant proliferation, migration, and invasion of MCF-7 cells by inhibiting the PI3K-AKT-HIF-1&#945;-FoxO1 pathway.

The present protocol describesa comprehensive strategy for evaluating the pharmacological action and mechanism of salidroside in inhibiting MCF-7 cell proliferation and migration.

Evaluating the Migration and Invasion of Salidroside Treated MCF-7 Cells in a Wound Scratch Assay

To begin, incubate two milliliters of the cultured MCF-7 cells in a six-well plate until 90%confluency. In the grown cell monolayer, create a linear scratch wound down the center using a sterile pipette tip. Then capture the images with an optical microscope at different time intervals.For the transwell assay, suspend MCF-7 cells in a serum-free medium. Then see the cells in the upper transwell chamber, either pre-coded or non-coded with Matrigel. Add DMEM complete medium at the bottom of the transwell chamber to act as a chemical inducer.After 24 hours, remove the cells in the upper chamber and fix the remaining invasive and migrant cells using methanol. Stain the MCF-7 cells using crystal violet solution. Next, image the stained cells with an optical microscope.A dose dependent inhibitory effect of salidroside on cell proliferation was observed, with a 50%decline in cell vitality at 40 micromolar. Further, salidroside could inhibit the vitality of MCF-7 cells over time, with a 50%decrease in MCF-7 cell vitality after 24 hours of co-incubation. The wound-scratch assay showed an inhibitory effect for salidroside treatment on MCF-7 cells.Further, salidroside treatment significantly reduced the undesirable migration and invasion of MCF-7 cells.

Using a bicinchoninic acid kit, measure the protein concentration of lysed MCF-7 cells following the manufacturer's instructions. Add the lysed cell samples into the corresponding groups in a 96-well plate. Then, add the working solution and incubate at 37 degrees Celsius for five minutes.Finally, measure the optical density values at 636 nanometers in a functional microplate reader. Salidroside restricted the enzyme vitalities of sodium potassium ATPases and calcium ATPase, indicating its potential role in promoting mitochondrial dysfunction.

Flow Cytometry Analysis of Apoptosis and the Cell Cycle in Salidroside Treated MCF-7 Cells

To begin, harvest the trypsinized MCF7 cells. Next, stain the PBS suspended cells with an X in five of ITC, and propidium iodide, for 20 minutes. Then determine the number of apoptotic cells using a flow cytometer.For cell cycle analysis, mix the re-suspended cells with propidium iodide solution in a fresh tube, and incubate the mixture for 30 minutes. Then detect the sample using a flow cytometer. Salidroside increased the number of MCF7 cells in the early and late apoptotic stages.Meanwhile, compared with the control group, salidroside treatment also sharply increased the number of cells in the G0 G1 phase, while reducing the proportion of S phase cells.

DCFH-DA and Fluo-4 AM Fluorescence Staining of Salidroside Treated MCF-7 Cells

Add 10 micromolar of Dcfh-DA fluorescence probe to MCF7 cells and incubate at 37 degrees Celsius for 20 minutes. After incubation, remove the excess Dcfh-DA by washing the cells three times with PBS. Next, test the fluorescence intensity using a fluorescence microscope and an excitation wavelength of 488 nanometers, and an emission wavelength of 525 nanometers.Incubate the MC7 cells with a five micromolar Fluo-4 AM fluorescent probe solution for 45 minutes at 37 degrees Celsius. After washing the cells three times with PBS, determine the fluorescence intensity using a fluorescence microscope with excitation at 488 nanometers and emission at 516 nanometers. Salidroside significantly enhanced ROS fluorescence signals shown by Dcfh-DA immunofluorescent staining.Consistently, Salidroside intervention also distinctly elevated calcium production, evidenced by Fluo-4 AM immunofluorescent staining.

Analyzing Protein Expression in Salidroside Treated MCF-7 Cells by Western Blotting

Begin by adding two milliliters of MCF-7 cells containing different drugs into the wells of a six-well plate. Incubate the cell cultures for 24 hours at 37 degrees Celsius and 5%carbon dioxide. Harvest the cells by centrifugation at 560 x g for three minutes at four degrees Celsius.Then wash the cells twice with pre-cool PBS, centrifuging at 560 x g for three minutes between the washes. Add 50 microliters of lysis buffer to the washed cells and place the sample in an ice bath for 15 minutes. Then centrifuge the sample at 8, 550 x g for 10 minutes at four degrees Celsius, and collect the supernatant protein sample.Combine the protein sample with the loading buffer in a four to one ratio. Next, denature the mixture at 100 degrees Celsius for 10 minutes in a metal bath. Then cool the mixture at room temperature.Separate the protein sample using 10%SDS polyacrylamide gel electrophoresis. Transfer the proteins onto a 0.22 micrometer PVDF membrane. After blocking the membrane with 5%BSA, incubate it with the corresponding primary antibodies overnight at four degrees Celsius.The following day, incubate the membrane with goat anti-rabbit IgG secondary antibody at 37 degrees Celsius for two hours. Then develop the membranes using an ECL chemiluminescence solution and capture the images using a contactless quantitative Western blot imaging system. Western blot showed that salidroside treatment promoted the protein expression of the pro-apoptotic factors CC-9, CC-7, CC-3, Bim, and Bax while it inhibited the protein expression of anti-apoptotic BCL-2.Salidroside prominently limited the ratios of p-PI3K to PI3K and p-AKT to AKT. Meanwhile, the protein expression of mTOR, HIF-1 alpha, and Fox01 was also notably suppressed.

Determining Gene Expression in Salidroside Treated MCF-7 Cells Using qRT-PCR

Harvest the MCF-7 cells by centrifugalization at 560-G for three minutes, at four degrees Celsius. Add 500 microliters of buffered RL-1 to five times 10 to the six cells and mix thoroughly until no cell masses are visible. Next, transfer the cell homogenates to a DNA cleaning column embedded in the collection tube.Centrifuge the samples at 85-50-G for two minutes, at four degrees Celsius. After centrifuging, remove the DNA cleaning column, retaining the supernatant in the collection tube. Add 800 microliters of buffer RL-2 and 500 microliters of the supernatant, and mix gently.Next, transfer 700 microliters of the mixture into an RNA-only column embedded in the collection tube. Centrifuge the tube at 8, 550-G for one minute, at four degrees Celsius. Then discard the flow-through in the collection tube.Wash the RNA-only column first with 500 milliliters of buffer RW-1, and then with 700 milliliters of buffer RW-2, by centrifuging for one minute at 8, 550-G, and four degrees Celsius at each wash. After removing the residual RW-2 by centrifuging again, transfer the RNA only column to a new collection tube. Add 100 microliters of RNAs-free deionized water, preheated at 65 degrees Celsius to the center of the membrane in the RNA-only column, and wait for two minutes.Then centrifuge at 8, 550-G for one minute, at four degrees Celsius to collect the RNA solution. Set up the reaction mixture for PCR and then set the PCR reaction conditions of the system. Execute the QRT PCR procedure.QRT PCR showed that cylindricity treatment promoted the gene expression of the pro-apoptotic factors, CC-9, CC-7, CC-3, vim, and vax, while it inhibited the gene expression of anti-apoptotic BCL-2. Further, cylindricide administration reduced the gene expression levels of PI-3K, AKT, M-tor, HIF1-Alpha and Fox-01.
