Drei eng miteinander verkn&uuml;pfte Gen-Cluster (Alpha, Beta und Gamma) ist eine Familie von S&auml;ugetieren Protocadherin (Pcdh) Proteine kodiert. Mehrere Alpha und Gamma-Pcdh-mRNAs werden ausgedr&uuml;ckt in unterschiedlichen Mustern im Nervensystem und entstehen durch alternative Pre-mRNA-Splei&szlig;en zwischen verschiedenen &quot;Variable&quot; Exons und drei &quot;Konstante&quot; Exon innerhalb jedes Clusters. Wir zeigen, dass jede Variable Pcdh-Exon F&ouml;rderer vorangestellt ist und Projekttr&auml;ger Wahl bestimmt, welche Variable Exon in einem Pcdh-mRNA enthalten ist. A family of mammalian protocadherin (Pcdh) proteins is encoded by three closely linked gene clusters (alpha, beta, and gamma). Multiple alpha and gamma Pcdh mRNAs are expressed in distinct patterns in the nervous system and are generated by alternative pre-mRNA splicing between different "variable" exons and three "constant" exons within each cluster. We show that each Pcdh variable exon is preceded by a promoter and that promoter choice determines which variable exon is included in a Pcdh mRNA. In addition, we provide evidence that alternative splicing of variable exons within a gene cluster occurs via a cis-splicing mechanism. However, virtually every variable exon can engage in trans-splicing with constant exons from another cluster, albeit at a far lower level.

Une famille de prot&eacute;ines de mammif&egrave;res PROTOCADHERINE (Pcdh) est cod&eacute;e par trois groupes de g&egrave;nes &eacute;troitement li&eacute;s (alpha, b&ecirc;ta et gamma). Plusieurs ARNm de Pcdh alpha et gamma est exprim&eacute;s dans des profils distincts dans le syst&egrave;me nerveux et est g&eacute;n&eacute;r&eacute;s par l&#39;&eacute;pissage alternatif des pr&eacute;-ARNm entre diff&eacute;rents exons &laquo; variable &raquo; et trois exons &laquo; constants &raquo; au sein de chaque grappe. Nous montrons que chaque exon variable Pcdh est pr&eacute;c&eacute;d&eacute;e d&#39;un promoteur et que le choix du promoteur d&eacute;termine quel exon variable est incluse dans un ARNm Pcdh. Recent landmark experiments have shown that transient overexpression of a small number of transcription factors can reprogram differentiated cells into induced pluripotent stem (iPS) cells that resemble embryonic stem (ES) cells. These iPS cells hold great promise for medicine because they have the potential to generate patient-specific cell types for cell replacement therapy and produce in vitro models of disease, without requiring embryonic tissues or oocytes. Although current iPS cell lines resemble ES cells, they have not passed the most stringent test of pluripotency by generating full-term or adult mice in tetraploid complementation assays, raising questions as to whether they are sufficiently potent to generate all of the cell types in an organism. Whether this difference between iPS and ES cells reflects intrinsic limitations of direct reprogramming is not known. Here we report fertile adult mice derived entirely from iPS cells that we generated by inducible genetic reprogramming of mouse embryonic fibroblasts. Producing adult mice derived entirely from a reprogrammed fibroblast shows that all features of a differentiated cell can be restored to an embryonic level of pluripotency without exposure to unknown ooplasmic factors. Comparing these fully pluripotent iPS cell lines to less developmentally potent lines may reveal molecular markers of different pluripotent states. Furthermore, mice derived entirely from iPS cells will provide a new resource to assess the functional and genomic stability of cells and tissues derived from iPS cells, which is important to validate their utility in cell replacement therapy and research applications.

Produire des souris adultes issues enti&#xE8;rement &#xE0; partir d&#x26;#39;un fibroblaste reprogramm&#xE9; montre que toutes les caract&#xE9;ristiques d&#x26;#39;une cellule diff&#xE9;renci&#xE9;e peut &#xEA;tre restaur&#xE9;e &#xE0; un niveau embryonnaire de la pluripotence, sans exposition &#xE0; des facteurs inconnus ovoplasmique. Recientes experimentos hist&#xF3;ricos han demostrado que la sobreexpresi&#xF3;n transitoria de un peque&#xF1;o n&#xFA;mero de factores de transcripci&#xF3;n pueden reprogramar las c&#xE9;lulas diferenciadas en madre pluripotentes inducidas (iPS) que se asemejan a las c&#xE9;lulas madre embrionarias (ES) las c&#xE9;lulas. Estas c&#xE9;lulas iPS son una gran promesa para la medicina debido a que tienen el potencial de generar en el paciente tipos espec&#xED;ficos de c&#xE9;lulas para la terapia de reemplazo celular y la producci&#xF3;n de los modelos in vitro de la enfermedad, sin necesidad de tejidos de embriones u ovocitos. Aunque las actuales l&#xED;neas de c&#xE9;lulas iPS se parecen a las c&#xE9;lulas madre embrionarias, que no han pasado la prueba m&#xE1;s estricta de pluripotencia por la generaci&#xF3;n de ratones a t&#xE9;rmino o un adulto en ensayos de complementaci&#xF3;n tetraploide, que plantea interrogantes en cuanto a si son lo suficientemente potentes para generar todos los tipos de c&#xE9;lulas en un organismo. Si esta diferencia entre iPS y las c&#xE9;lulas ES refleja las limitaciones intr&#xED;nsecas de reprogramaci&#xF3;n directa no se conoce. Aqu&#xED; mostramos los ratones adultos f&#xE9;rtiles derivados enteramente de c&#xE9;lulas iPS que generaron por la reprogramaci&#xF3;n inducida gen&#xE9;tica de fibroblastos embrionarios de rat&#xF3;n. La producci&#xF3;n de ratones adultos totalmente derivadas de fibroblastos reprogramados una muestra que todas las caracter&#xED;sticas de una c&#xE9;lula diferenciada puede ser restaurado a un nivel embrionario de la pluripotencia sin exposici&#xF3;n a factores desconocidos ooplasmic. Comparando estas l&#xED;neas iPS totalmente de c&#xE9;lulas pluripotentes a las l&#xED;neas de desarrollo menos potentes pueden revelar los marcadores moleculares de los estados pluripotentes diferentes. Adem&#xE1;s, los ratones totalmente derivadas de las c&#xE9;lulas iPS proporcionar&#xE1; un nuevo recurso para evaluar la estabilidad funcional y gen&#xF3;mica de las c&#xE9;lulas y los tejidos derivados de c&#xE9;lulas iPS, lo cual es importante para validar su utilidad en la terapia de reemplazo celular y aplicaciones de investigaci&#xF3;n.

Sensory information may be represented in the brain by stereotyped mapping of axonal inputs or by patterning that varies between individuals. In olfaction, a stereotyped map is evident in the first sensory processing centre, the olfactory bulb (OB), where different odours elicit activity in unique combinatorial patterns of spatially invariant glomeruli. Activation of each glomerulus is relayed to higher cortical processing centres by a set of âˆ¼20-50 'homotypic' mitral and tufted (MT) neurons. In the cortex, target neurons integrate information from multiple glomeruli to detect distinct features of chemically diverse odours. How this is accomplished remains unclear, perhaps because the cortical mapping of glomerular information by individual MT neurons has not been described. Here we use new viral tracing and three-dimensional brain reconstruction methods to compare the cortical projections of defined sets of MT neurons. We show that the gross-scale organization of the OB is preserved in the patterns of axonal projections to one processing centre yet reordered in another, suggesting that distinct coding strategies may operate in different targets. However, at the level of individual neurons neither glomerular order nor stereotypy is preserved in either region. Rather, homotypic MT neurons from the same glomerulus innervate broad regions that differ between individuals. Strikingly, even in the same animal, MT neurons exhibit extensive diversity in wiring; axons of homotypic MT pairs diverge from each other, emit primary branches at distinct locations and 70-90% of branches of homotypic and heterotypic pairs are non-overlapping. This pronounced reorganization of sensory maps in the cortex offers an anatomic substrate for expanded combinatorial integration of information from spatially distinct glomeruli and predicts an unanticipated role for diversification of otherwise similar output neurons.

Jedoch auf der Ebene der einzelnen Neuronen glomerul&auml;re Bestellung weder Klischeehaftigkeit entweder Region beibehalten. Vielmehr anregen homotypischen MT Neuronen aus der gleichen Korbzelle weite Regionen, die zwischen Einzelpersonen unterscheiden. Auff&auml;llig ist, sogar im gleichen Tier zeigen MT Neuronen umfangreiche Vielfalt bei der Verkabelung; Axone homotypischen MT-Paare von einander abweichen ausgeben prim&auml;re Niederlassungen an verschiedenen Standorten und 70-90 % der Zweige der homotypischen und heterotypischen Paaren nicht &uuml;berlappende. Informaci&oacute;n sensorial puede representarse en el cerebro por estereotipados asignaci&oacute;n de entradas axonal o patrones que var&iacute;a entre individuos. En el olfato, mapa estereotipado es evidente en el primer centro de procesamiento sensorial, el bulbo olfatorio (OB), donde diferentes olores provocan actividad en patrones &uacute;nicos combinatorias de glom&eacute;rulos espacial invariantes. Activaci&oacute;n de cada glom&eacute;rulo es retransmitida a los centros de procesamiento cortical superiores por un grupo de neuronas de &sim;20-50 &#39;homot&iacute;picos&#39; mitrales y copetudas (MT). En la corteza, las neuronas objetivo integran informaci&oacute;n de m&uacute;ltiples glom&eacute;rulos para detectar caracter&iacute;sticas distintas de qu&iacute;micamente diversos olores. &iquest;C&oacute;mo esto se logra a&uacute;n no est&aacute; clara, tal vez porque no se ha descrito el mapeo cortical de informaci&oacute;n glomerular por neuronas individuales de MT. Aqu&iacute; utilizamos nuevo trazo viral y m&eacute;todos de reconstrucci&oacute;n tridimensional del cerebro para comparar las proyecciones corticales de conjuntos definidos de neuronas de MT. Demostramos que la organizaci&oacute;n bruto-escala de la OB es preservada en los patrones de proyecciones axonal al centro de un proceso a&uacute;n reordenada en otro, lo que sugiere que distintas estrategias de codificaci&oacute;n pueden operar en diferentes objetivos. Sorprendentemente, incluso en el mismo animal, las neuronas MT exhiben diversidad en cableado; axones de pares homot&iacute;picos MT difieren unos de otros, emiten ramas primarios en distintas localizaciones y 70-90% de las ramas de homot&iacute;picos y heterot&iacute;picos pares son no superpuestos. Esta manifiesta reorganizaci&oacute;n de mapas sensoriales en la corteza ofrece un substrato anat&oacute;mico para integraci&oacute;n ampliada combinatoria de informaci&oacute;n de glom&eacute;rulos espacialmente distintas y predice un papel no previsto para la diversificaci&oacute;n de las neuronas de salida similares.

The biomedical utility of induced pluripotent stem cells (iPSCs) will be diminished if most iPSC lines harbor deleterious genetic mutations. Recent microarray studies have shown that human iPSCs carry elevated levels of DNA copy number variation compared with those in embryonic stem cells, suggesting that these and other classes of genomic structural variation (SV), including inversions, smaller duplications and deletions, complex rearrangements, and retroelement transpositions, may frequently arise as a consequence of reprogramming. Here we employ whole-genome paired-end DNA sequencing and sensitive mapping algorithms to identify all classes of SV in three fully pluripotent mouse iPSC lines. Despite the improved scope and resolution of this study, we find few spontaneous mutations per line (one or two) and no evidence forÂ endogenous retroelement transposition. These results show that genome stability can persist throughout reprogramming, and argue that it is possible to generate iPSCs lacking gene-disrupting mutations using current reprogramming methods.

Hier besch&auml;ftigen wir ganze gepaart-End DNA Genoms und sensible Zuordnung Algorithmen um alle Klassen des SV in drei voll pluripotenten Maus iPSC Zeilen zu identifizieren. Trotz der verbesserten Anwendungsbereich und Aufl&ouml;sung dieser Studie, finden wir einige spontane Mutationen pro Zeile (maximal zwei) und keine Beweise f&uuml;r die endogene Retroelement Umsetzung. Diese Ergebnisse zeigen, dass die Genomstabilit&auml;t kann im gesamten Neuprogrammierung beibehalten und argumentieren, dass iPSCs fehlt Unterbrechung der Gen-Mutationen, die mit aktuelle Neuprogrammierung Methoden generieren kann.
