A inflamação é uma resposta protectora contra a infecção microbiana ou lesão do tecido que requer regulação apertada de entrada e saída de leucócitos a partir do tecido inflamado para permitir a resolução de 1,2. Conversa cruzada entre as células endoteliais (CE) que os navios de linha de sangue, leucócitos circulantes e células do estroma do tecido residente é essencial para coordenar este processo 3. As células estromais regular o recrutamento de leucócitos circulantes durante a inflamação através de cross-talk com células endoteliais vizinhas. Aqui nós descrevemos dois modelos in vitro &quot;vasculares&quot; para estudar o recrutamento de neutrófilos circulantes de fluxo pelas células endoteliais inflamadas. Uma grande vantagem destes modelos é a capacidade de analisar cada passo na cascata de adesão de leucócitos no fim, como poderia ocorrer in vivo. Descrevemos também como ambos os modelos podem ser adaptados para estudar o papel das células estromais, neste caso as células estaminais mesenquimais (MSC), em regular o recrutamento de leucócitos. Células endoteliais primárias foram cultivadas isoladamente ou em conjunto com MSC humana em contacto directo em microslides Ibidi ou em lados opostos de um filtro Transwell durante 24 h. As culturas foram estimuladas com factor de necrose tumoral alfa (TNFa) durante 4 horas e incorporadas num ensaio de adesão com base em fluxo. Um bolus de neutrófilos foi perfundidoao longo do endotélio, durante 4 min. A captura de fluir neutrófilos e suas interações com o endotélio foi visualizado por microscopia de contraste de fase. Em ambos os modelos, a estimulação de citoquinas-endotelial aumento do recrutamento de neutrófilos fluindo de uma maneira dependente da dose. A análise do comportamento dos neutrófilos recrutados mostraram um decréscimo dependente da dose no rolamento e um aumento dependente da dose na transmigração através do endotélio. Em co-cultura, MSC suprimida adesão de neutrófilos ao endotélio estimulada-TNF. Os nossos modelos baseados na aderência de fluxo de imitar as fases iniciais do recrutamento de leucócitos da circulação. Além de leucócitos, que pode ser usado para examinar o recrutamento de outros tipos de células, tais como células tumorais circulantes ou MSC administrados terapeuticamente. Os nossos modelos de multi-camadas de co-cultura MSC demonstraram que comunicam com endotélio para modificar a sua resposta a citocinas pró-inflamatórias, Altering o recrutamento de neutrófilos. Mais pesquisas utilizando esses modelos é necessário para entender completamente como células estromais de diferentes tecidos e condições (doenças inflamatórias ou câncer) influenciam o recrutamento de leucócitos durante a inflamação.

Inicialmente, analisamos o efeito de estimular CE com TNF sobre o recrutamento de neutrófilos de fluxo, utilizando o modelo microlâmina Ibidi (Seção 7-9).
