Os reagentes necessários para preparar MOFs e os ligantes estão listados na Tabela de Materiais.
1. Configuração do processo de troca pós-sintética de ligantes (PSE)
<ol><li>Seque completamente os MOFs UiO-66 pré-sintetizados sob vácuo para remover quaisquer sais metálicos e ligantes não reagidos nos poros e resíduos de solventes remanescentes durante a noite. NOTA: Consulte Arquivo Suplementar 1 para o procedimento de síntese de MOFs UiO-66.</li>
	<li>Preparar ligantes funcionalizados, H 2 BDC-Triazol e H2BDC-Tetrazol(ver Arquivo Suplementar 1 para o processo de preparação; Figura Suplementar 1 e Figura Suplementar 2 para caracterização) em estado isolado e totalmente seco sob vácuo durante a noite.</li>
	<li>Preparar uma solução aquosa de hidróxido de potássio (KOH) a 4% dissolvendo hidróxido de potássio em água deionizada.</li>
	<li>Realizar o processo de PSE em frascos de cintilação de 20 mL com tampas de polipropileno (PP) (ver Tabela de Materiais).</li>
	<li>Medir o H 2 BDC-Triazol (23,3 mg, 0,1 mmol) ou H2BDC-Tetrazol(23,4 mg, 0,1 mmol) e colocar no frasco para injetáveis de cintilação.</li>
	<li>Dissolver o ligante H2BDC em solução aquosa. Use uma pipeta de vidro para transferir 1,0 mL da solução aquosa de KOH a 4% para o frasco de cintilação contendo BDC-Triazol ou BDC-Tetrazólio. CUIDADO: A solução aquosa de 4% KOH é altamente básica. Evite todas as formas de contato e use equipamentos de proteção individual.</li>
	<li>Sonicar a mistura até que todos os sólidos estejam totalmente dissolvidos.</li>
	<li>Neutralizar a solução até pH 7. Utilizar uma pipeta de vidro para transferir uma solução aquosa de ácido clorídrico (HCl) 1 M com agitação para o frasco para injetáveis de cintilação contendo dicarboxilato até atingir o pH 7. Meça o pH por papel de pH (e sua cor) ou por um medidor de pH. NOTA: É crucial manter um pH de 7 durante o processo de troca, uma vez que os MOFs são tipicamente instáveis em condições básicas (>pH 7), e o ligante BDC não é solúvel em condições ácidas (<pH 7). Aproximadamente 1,5 mL de solução aquosa de HCl 1 M é necessária para neutralizar BDC-Triazol ou BDC-Tetrazole-contendo.</li>
	<li>Adicionar MOFs à solução de dicarboxilato e incubar. Adicionar UiO-66 MOF (33 mg, 0,02 mmol) ao frasco para injetáveis de cintilação contendo solução de pH 7. NOTA: Em pH 7, as partículas de UiO-66 MOF não são solúveis em água, resultando em sua suspensão em água.</li>
	<li>Incubar o frasco para injetáveis de cintilação contendo o MOF e o ligante num agitador a 120 rpm e à temperatura ambiente durante 24 horas. NOTA: A agitação pode ser usada como uma alternativa à incubação do agitador para o processo PSE. No entanto, deve-se ter cuidado, uma vez que o contato físico entre a barra de agitação magnética e os MOFs pode resultar em rachaduras ou quebra das partículas de MOF.</li>
</ol>2. Isolamento do MOF trocado e processo de lavagem
<ol><li>Após a incubação, isolar o MOF sólido da mistura por centrifugação (1.166 x g, 5 min, temperatura ambiente).</li>
	<li>Adicionar metanol fresco (10 mL) ao MOF sólido obtido e agitar a mistura para formar uma mistura heterogênea para dissolver os ligantes BDC não trocados restantes.</li>
	<li>Isolar o sólido isolado por centrifugação (1.166 x g, 5 min, temperatura ambiente).</li>
	<li>Repita os passos 2.2-2.3 mais duas vezes para um total de três ciclos de lavagem.</li>
	<li>Secar totalmente o sólido MOF trocado sob vácuo por um período noturno após a última lavagem.</li>
</ol>3. Caracterização do MOF por difração de raios X do pó (PXRD)
<ol><li>Transferir aproximadamente 10 mg do sólido MOF trocado para um suporte de amostra PXRD (ver Tabela de Materiais).</li>
	<li>Coloque o porta-amostras no difratómetro.</li>
	<li>Coletar o padrão PXRD (Figura 2) na faixa de 2θ de 5° a 30°.</li>
	<li>Compare os dados obtidos com o MOF UiO-66 pai e o padrão simulado.</li>
</ol>4. Caracterização da MOF por ressonância magnética nuclear (RMN) após digestão
<ol><li>Transfira aproximadamente 30 mg do sólido MOF trocado para um frasco fresco de 4 ml.</li>
	<li>Use uma micropipeta para transferir 400 μL de DMSO-d 6 para a amostra de MOF.</li>
	<li>Use uma micropipeta para transferir 200 μL da solução 4,14 M NH4F/D2O para uma suspensão DMSO-d 6 de pó de MOF. NOTA: Uma solução aquosa de aproximadamente 40% HF pode ser usada em vez da solução NH4F/D2O. Neste caso, um pico significativo de H2O é observado na RNM. No entanto, uma digestão mais clara é possível no caso da digestão HF. CUIDADO: A HIC é altamente tóxica para o organismo e sistema nervoso central. Todas as formas de contato devem ser evitadas, o trabalho deve ser realizado em uma capela de fumo e equipamentos de proteção individual devem ser usados.</li>
	<li>Sonicar a mistura heterogênea por 30 min até que o MOF seja dissolvido no solvente misturado dimetilsulfóxido (DMSO)-D2O após a digestão.</li>
	<li>Remova quaisquer sólidos insolúveis remanescentes filtrando a solução através de um filtro de seringa de difluoreto de polivinilideno (PVDF) (Φ 13 mm, poro de 0,45 μm; ver Tabela de Materiais) enquanto a transfere do frasco para injetáveis de 4 ml para um tubo de RMN.</li>
	<li>Coloque o tubo de RMN na máquina de RMN. Coletar os dados de RMN (Figura 3).</li>

Estruturas metal-orgânicas (MOFs) são materiais porosos tridimensionais que são formados através de ligações de coordenação entre clusters metálicos e ligantes orgânicos multitópicos. Os MOFs têm recebido atenção significativa devido à sua porosidade permanente, baixa densidade e capacidade de associar componentes orgânicos e inorgânicos, o que possibilita diversas aplicações 1,2. Além disso, a vasta gama de nós metálicos e ligantes orgânicos de escoras oferecem aos MOFs combinações estruturais teoricamente ilimitadas. Mesmo com estruturas de estrutura idênticas, as propriedades físicas e químicas dos MOFs podem ser modificadas através da funcionalização de ligantes com etiquetas químicas. Este processo de modificação oferece uma rota promissora para adequar as propriedades dos MOFs para aplicações específicas 3,4,5,6,7,8,9.
Tanto a pré-funcionalização dos ligantes antes da síntese do MOF quanto a modificação pós-sintética (MSP) dos MOFs têm sido empregadas para introduzir e/ou modificar grupos funcionais nos ligantes doMOF10,11. Em particular, PSMs covalentes têm sido extensivamente estudados para introduzir novos grupos funcionais e gerar uma gama de MOFs com diversas funcionalidades12,13,14. Por exemplo, o UiO-66-NH2 pode ser convertido em UiO-66-AMs funcionalizados com amida com diferentes comprimentos de cadeia (variando da acetamida mais curta à n-hexilamida mais longa) através de reações de acilação com haletos de acila apropriados (como cloreto de acetila ou cloreto de n-hexanoila)15,16. Essa abordagem demonstra a versatilidade de PSMs covalentes para introduzir grupos funcionais específicos em ligantes MOF, abrindo caminho para uma ampla gama de aplicações.
Além das PSMs covalentes, a pós-troca sintética de ligantes (PSE) é uma estratégia promissora para modificar MOFs (Figura 1). Como os MOFs são compostos por ligações de coordenação entre metais e ligantes (como carboxilatos), essas ligações de coordenação podem ser substituídas por ligantes externos de uma solução. A exposição de MOFs a uma solução contendo o ligante desejado com etiquetas químicas pode ser incorporada aos MOFs via PSE 17,18,19,20,21,22. Como o processo de PSE é acelerado pela existência de solventes coordenativos, o fenômeno também é chamado de troca de ligante assistida por solvente (SALE)23,24. Esta abordagem oferece um método flexível e fácil para funcionalizar MOFs com uma ampla gama de ligantes externos, possibilitando um amplo espectro de aplicações 25,26,27,28,29.

O progresso do processo PSE pode ser controlado ajustando a razão do ligante, a temperatura de troca e o tempo. Notavelmente, o PSE à temperatura ambiente pode ser empregado para obter MOFs funcionalizados através da troca de ligantes de uma solução em sólidos MOF20. A estratégia PSE é particularmente útil para introduzir grupos funcionais termicamente instáveis (como grupos azido) e grupos funcionais coordenativos (como grupos fenólicos) em estruturas de MOF18. Além disso, a estratégia PSE tem sido aplicada a vários MOFs com variações metálicas e de ligações de coordenação. Esta troca é um processo universal na química de MOFs30,31,32. Neste estudo, apresentamos um protocolo detalhado para PSE para obter MOFs funcionalizados a partir de MOFs puros e não funcionalizados, e fornecemos uma estratégia de caracterização para confirmar o sucesso da funcionalização dos MOFs. Este método demonstra a versatilidade e conveniência do PSE para modificar MOFs com diversos grupos funcionais.
O ácido benzeno-1,4-dicarboxílico (H 2 BDC-Tetrazólio)33 e o ácido benzeno-1,4-dicarboxílico contendo triazólicos (H2BDC-Triazol)são sintetizados como ligantes-alvo e utilizados no PSE de MOFs UiO-66 para obter novos MOFs contendo triazóis, livres de coordenação. Tanto os triazóis quanto os tetrazóis possuem prótons N-H ácidos em seus anéis heterocíclicos e podem se coordenar com cátions metálicos, possibilitando seu uso na construção de MOFs34,35. No entanto, existem estudos limitados sobre a incorporação de tetrazóis e triazóis livres de coordenação em MOFs e estruturas relacionadas. No caso de Zr-MOFs funcionalizados com triazóis, MOFs do tipo UiO-68 foram investigados quanto às propriedades fotofísicas através da síntese solvotérmica direta com funcionalidades de benzotriazol36. Para Zr-MOFs funcionalizados com tetrazólio, a síntese direta mista foi empregada33. Esses MOFs funcionalizados com heterociclos poderiam fornecer potenciais locais de coordenação em poros MOF para catálise, captação molecular seletiva por afinidade de ligação e aplicações relacionadas à energia, como condução de prótons em células a combustível.

Estruturas metal-orgânicas (MOFs) são uma classe de materiais porosos que são formados através de ligações de coordenação entre clusters metálicos e ligantes orgânicos. Dada a sua natureza coordenativa, os ligantes orgânicos e a estrutura das bielas podem ser facilmente removidos do MOF e/ou trocados com outras moléculas coordenativas. Ao introduzir ligantes-alvo em soluções contendo MOF, MOFs funcionalizados podem ser obtidos com novos tags químicos por meio de um processo chamado troca pós-sintética de ligantes (PSE). O PSE é uma abordagem simples e prática que permite a preparação de uma ampla gama de MOFs com novas etiquetas químicas por meio de um processo de equilíbrio de solução sólida. Além disso, o PSE pode ser realizado à temperatura ambiente, permitindo a incorporação de ligantes termicamente instáveis aos MOFs. Neste trabalho, demonstramos a praticidade do PSE usando ligantes heterocíclicos contendo triazóis e tetrazólicos para funcionalizar um MOF baseado em Zr (UiO-66; UiO = Universidade de Oslo). Após a digestão, os MOFs funcionalizados são caracterizados através de várias técnicas, incluindo difração de raios X em pó e espectroscopia de ressonância magnética nuclear.

Metal-organic frameworks (MOFs) are three-dimensional porous materials that are formed through coordination bonds between metal clusters and multi-topic organic ligands. MOFs have garnered significant attention due to their permanent porosity, low density, and ability to associate organic and inorganic components, which enables diverse applications1,2. Moreover, the vast range of metal nodes and strut organic linkers offer MOFs theoretically unlimited structural combinations. Even with identical framework structures, MOFs&#39; physical and chemical properties can be modified through ligand functionalization with chemical tags. This modification process offers a promising route to tailor the properties of MOFs for specific applications3,4,5,6,7,8,9.
Both the pre-functionalization of ligands prior to MOF synthesis and post-synthetic modification (PSM) of MOFs have been employed to introduce and/or modify functional groups in MOF ligands10,11. In particular, covalent PSMs have been extensively studied to introduce new functional groups and generate a range of MOFs with diverse functionalities12,13,14. For instance, UiO-66-NH2 can be converted to amide-functionalized UiO-66-AMs with different chain lengths (ranging from the shortest acetamide to the longest n-hexyl amide) through acylation reactions with appropriate acyl halides (such as acetyl chloride or n-hexanoyl chloride)15,16. This approach demonstrates the versatility of covalent PSMs to introduce specific functional groups onto MOF ligands, paving the way for a broad range of applications.
In addition to covalent PSMs, post-synthetic ligand exchange (PSE) is a promising strategy for modifying MOFs (Figure 1). Since MOFs are composed of coordination bonds between metals and ligands (such as carboxylates), these coordination bonds can be replaced with external ligands from a solution. Exposing MOFs to a solution containing the desired ligand with chemical tags can be incorporated into the MOFs via PSE17,18,19,20,21,22. Since the PSE process is accelerated by the existence of coordinative solvents, the phenomenon is also called solvent-assisted ligand exchange (SALE)23,24. This approach offers a flexible and facile method for functionalizing MOFs with a wide range of external ligands, enabling a broad spectrum of applications25,26,27,28,29.

The progress of the PSE process can be controlled by adjusting the ligand ratio, exchange temperature, and time. Notably, room temperature PSE can be employed to obtain functionalized MOFs by exchanging ligands from a solution into MOF solids20. The PSE strategy is particularly useful for introducing both thermally unstable functional groups (such as azido groups) and coordinative functional groups (such as phenol groups) into MOF structures18. In addition, the PSE strategy has been applied to various MOFs with metal and coordination bond variations. This exchange is a universal process in the chemistry of MOFs30,31,32. In this study, we present a detailed protocol for PSE to obtain functionalized MOFs from pristine, non-functionalized MOFs, and we provide a characterization strategy to confirm successful functionalization of the MOFs. This method demonstrates the versatility and convenience of PSE for modifying MOFs with diverse functional groups.
Tetrazole-containing benzene-1,4-dicarboxylic acid (H2BDC-Tetrazole)33, and triazole-containing benzene-1,4-dicarboxylic acid (H2BDC-Triazole) are synthesized as target ligands and utilized in the PSE of UiO-66 MOFs to obtain novel, coordination-free, triazole-containing MOFs. Both triazoles and tetrazoles possess acidic N-H protons on their heterocyclic rings and can coordinate with metal cations, thus enabling their use in constructing MOFs34,35. However, there are limited studies on incorporating coordination-free tetrazoles and triazoles into MOFs and related structures. In case of triazole-functionalized Zr-MOFs, UiO-68 type MOFs were investigated to photophysical properties through direct solvothermal synthesis with benzotriazole functionalities36. For tetrazole-functionalized Zr-MOFs, the mixed direct synthesis was employed33. These heterocycle-functionalized MOFs could provide potential coordinating sites in MOF pores for catalysis, selective molecular uptake by binding affinity, and energy-related applications, such as proton conduction in fuel cells.

Our main research topic is functionalizing the porous metal-organic frameworks, MOFs, with interesting functional groups through various techniques. This study is focused on heterocycle installation in MOFs. PSE, post-synthetic ligand exchange is the main technique in this work.The ligand with target functional group could be installed in presynthesized MOFS throughout the solid-solution exchange process. Especially, some of the unstable ligands and ligands with coordinating groups could be efficiently incorporated into MOFs through the PSE techniques. Both triazole and tetrazole functional groups have multiple coordinating nitrogen grooves.Therefore, there is competition for coordination between metal carboxylate and metal triazolate or tetrazolate. The incorporation ratio of target ligands is easily controlled in the PSE process and multiple targets could be installed in a single MOF through this approach.

Metal-organic frameworks (MOFs) are a class of porous materials that are formed through coordination bonds between metal clusters and organic ligands. Given their coordinative nature, the organic ligands and strut framework can be readily removed from the MOF and/or exchanged with other coordinative molecules. By introducing target ligands to MOF-containing solutions, functionalized MOFs can be obtained with new chemical tags via a process called post-synthetic ligand exchange (PSE). PSE is a straightforward and practical approach that enables the preparation of a wide range of MOFs with new chemical tags via a solid-solution equilibrium process. Furthermore, PSE can be performed at room temperature, allowing the incorporation of thermally unstable ligands into MOFs. In this work, we demonstrate the practicality of PSE by using heterocyclic triazole- and tetrazole-containing ligands to functionalize a Zr-based MOF (UiO-66; UiO = University of Oslo). After digestion, the functionalized MOFs are characterized via various techniques, including powder X-ray diffraction and nuclear magnetic resonance spectroscopy.

Setting up the Post-synthetic Ligand Exchange (PSE) Process, Exchanged MOF Isolation and Washing Process

Begin by measuring 23.3 milligrams of H2BDC triazole and place it into the scintillation vial. To dissolve the H2BDC ligand in the aqueous solution, transfer 1.0 milliliters of the 4%potassium hydroxide solution into the vial using a glass pipette. Next, sonicate the mixture until all solids completely dissolve.Add one molar hydrochloric acid solution into the decarboxylate-containing vial and stir until a pH of 7 is reached. Add 33 milligrams of UiO-66 MOF to the scintillation vial. Then incubate the vial with the MOF and ligand at room temperature and 120 RPM in a shaker for 24 hours.Next, isolate the solid MOF from the mixture by centrifugation at 1166g for five minutes. at room temperature. Add 10 milliliters of fresh methanol to the obtained solid MOF and shake the vial, creating a heterogeneous mixture to dissolve any remaining unchanged BDC ligands.Isolate the solid once again by centrifuging the mixture at 1166g for five minutes at room temperature and dry it.

To begin NMR characterization, transfer around 30 milligrams of the exchanged MOF solid to a fresh four milliliter vial.Then move 400 microliters of DMSO-d6 to the MOF sample using a micro pipette. Transfer 200 microliters of 4.14 molar ammonium fluoride deuterium oxide solution to a DMSO-d6 suspension of MOF powder using a micro pipette. Sonicate the heterogeneous mixture for 30 minutes until the MOF is dissolved in the dimethyl sulfoxide deuterium oxide mixed solvent after digestion.Once done, remove any remaining insoluble solids by filtering the solution through a polyvinylidene difluoride syringe filter while transferring it from the four milliliter vial to a nuclear magnetic resonance tube. To determine the success of the exchange, powder X-ray diffraction patterns were measured and compared to pristine UiO-66 MOF for crystallinity. The position of reflection peaks, relative intensity, and broadness in the powder X-ray diffraction patterns of exchanged UiO-66-Triazole and UiO-66-Tetrazole matched with pristine UiO-66 MOFs, indicating that the ligand exchange did not impact the framework structure.The exchange ratios were determined by comparing the integration of nonfunctionalized BDC and BDC-Triazole or BDC-Tetrazole. The exchange ratios for Triazole and Tetrazole were found to be 33%and 30%respectively from post synthetic ligand exchange analysis.
