Bestimmte Mitglieder der Einwärtsgleichrichter Kalium (Kir) Kanal Familie drug targets für eine Vielzahl von Erkrankungen, einschließlich Bluthochdruck, Vorhofflimmern und Schmerzen 1,2 postuliert. Die Entwicklung potenterund selektive niedermolekulare Sonden mit verbesserten pharmakologischen Eigenschaften werden ein Schlüsselfaktor für ein volles Verständnis der Physiologie und therapeutische Potenzial von Kir-Kanälen sein. Das Molecular Libraries Probes Production Center Network (MLPCN) durch die National Institutes of Health (NIH) Common Fund hat Möglichkeiten geschaffen für akademische Wissenschaftler Sonde Entdeckung Kampagnen für molekulare Targets und Signalwege in der Notwendigkeit einer besseren pharmakologischen 6 einzuleiten. Die MLPCN bietet Forschern den Zugang zu Industrie-scale Screening-Zentren und die medizinische Chemie und Informatik zu unterstützen niedermolekularen Sonden, um die Funktion von Genen und Gen-Netzwerke aufzuklären entwickeln. Der entscheidende Schritt in der Zulassung zu dem MLPCN ist die Entwicklung eines robusten Target-oder Pathway-spezifischen Assay, zugänglich für High-Throughput-Screening (HTS) ist. Hier beschreiben wir, wie man eine Fluoreszenz-basierten Thallium (Tl +) flux assa entwickelny von Kir Kanalfunktion für das Hochdurchsatz-Screening Verbindung 7,8,9,10. Der Assay basiert auf der Durchlässigkeit des K +-Kanals Pore der K + Kongener Tl + basiert. Eine handelsübliche Leuchtstofflampen Tl + Reporter-Farbstoff wird verwendet, um transmembrane Fluss der Tl + durch die Pore zu erkennen. Es gibt mindestens drei kommerziell erhältliche Farbstoffe, die sich für Tl + Flux Assays sind: BTC, FluoZin-2 und 7,8 Fluxor. Dieses Protokoll beschreibt Assay-Entwicklung mit FluoZin-2. Wir diskutieren auch die Assays Performance-Benchmarks, die erreicht, um für die Einreise in die MLPCN in Betracht gezogen werden muss. Da Tl + gut durchzieht die meisten K +-Kanäle, sollte der Test sein, anpassbar an die meisten K +-Kanal Ziele.

Die Verwendung eines Tetracyclin-induzierbaren Expressionssystem bietet eine bequeme interne Kontrolle für die Unterscheidung Tl + Fluß durch endogene Wege und der Kir interessierenden Kanal.
