1. Vorbereitung der Tumorzellsuspension
Hinweis: Für die Herstellung und Aufrechterhaltung von primären orthotopischen Gliom-Xenografts sind geeignete institutionelle Genehmigungen für die Verwendung von Patientenmaterial und Tieren erforderlich. Am Vanderbilt University Medical Center wird resektiertes Tumormaterial, das über dem für diagnostische Zwecke erforderlichen Material hinausgeht, mit Zustimmung des Patienten für ein Forschungsgewebe-Repository gesammelt. Die Proben werden mit einer randomisierten 5-stelligen REDcap-Datenbanknummer beschriftet, und alle patientenspezifischen Bezeichner werden entfernt. Die REDcap-Datenbank enthält deidentifizierte klinische Daten für jede Probe im Gewebe-Repository, die Geschlecht, Alter (in Jahren), Rasse, Überlebensstatus, Pathologie, Krebsbehandlungen, Jahr der Resektion und Zeit bis zum Fortschreiten umfasst. Um die Sterilität zu erhalten und den Erfolg der primären Xenograft-Methode zu optimieren, sollten alle Reagenzien, dampfautoklavierten chirurgischen Instrumente und die chirurgische Stelle im Voraus montiert oder vorbereitet werden.
Hinweis: Gliom-Proben enthalten oft große Mengen myelin und Schmutz. Je nach Probengröße kann es notwendig sein, mehr als 4 diskontinuierliche Gradientenrohre für eine angemessene Entfernung von Myelin und Schmutz zu verwenden.
Hinweis: Der Prozess wird abgeschlossen, wenn kein oder wenig, deutlich sichtbares undissoziiertes Gewebe übrig ist. Vermeiden Sie das Einbringen von Blasen während des Triturierungsprozesses.
Hinweis: Die Zelllebensfähigkeit nach Dissoziation beträgt in der Regel >90 %. Geringere Viabilitäten können viele Variablen widerspiegeln, einschließlich suboptimaler Gewebehandhabung oder Transportzeit aus dem Operationssaal, Kryokonservierungsmethode oder Papain-Dissoziationstechnik. Niedrigere zelluläre Viabilitäten können jedoch noch ausreichen, solange eine ausreichende Anzahl lebensfähiger Mittel in dem entsprechenden Volumen transplantiert werden kann. Für jede Maus werden 50.000-200.000 lebensfähige Zellen implantiert. Aufgrund der abgeschrägten Spitze der Spritzennadel sollte im Endvolumen eine zusätzliche Zellsuspension von 5 l enthalten sein, um sicherzustellen, dass für jede Injektion ausreichend Material in die Spritze eingezogen werden kann. Um z. B. für 5 Mäuse 2 l/Maus zu injizieren, sollte das Endvolumen 15 l (15 l = (5 x 2 l) + 5 l) betragen.
<ol><li>Frisch resektiertes, identifiziertes Patientenmaterial in einen sterilen, verschlossenen Behälter auf Eis legen, um es zum Labor zu transportieren. Verarbeiten Sie die Probe direkt zur Transplantation oder kryokonservieren Sie die Probe in flüssigem Stickstoff (siehe unten) zur Lagerung in flüssigem Stickstoff und später zur Verwendung.</li>
	<li>Bereiten Sie die Papain-Dissoziationslösungen (Papainlösung, Wasch-/Protease-Inhibitor-Lösung und diskontinuierliche Gradientenlösung) in einer sterilen Haube mit den vom Hersteller gelieferten Kit-Komponenten und Anweisungen vor. Beschriften Sie sterile 15 ml Polystyrol-Konikonröhrchen und verteilen Sie die Lösungen wie folgt:<ul>
<li>Tube 1: 5 ml Papainlösung</li>
			<li>Tube 2: 3 ml Wasch-/Protease-Inhibitorlösung</li>
			<li>Tuben 3-6: je 5 ml diskontinuierliche Gradientenlösung</li>
		</ul>
</li>
	<li>Die Gliomprobe mit 5 ml Papainlösung in Tube 1 geben und mit einer 5 ml Pipette über einen Zeitraum von 10-20 min triturat.</li>
	<li>Das Material 10 Mal nach oben und unten pfeifen und dann das Material für 2-3 min bei Raumtemperatur inkubieren, bevor der nächste Zyklus der Trituration in Gang gesetzt wird.</li>
	<li>Positionieren Sie die Spitze der 5 ml Pipette näher an der Unterseite des konischen Rohres mit jedem Triturationszyklus, so dass die Spitze den Boden des Rohres für die letzten 2 Zyklen berührt.</li>
	<li>Zentrifuge bei 300 x g für 5 min bei Raumtemperatur. Entfernen Sie den Überstand und Pipette das Pellet nach oben und unten 10x in 3 ml der Wasch-/Protease-Inhibitor-Lösung.</li>
	<li>Schichtung eines gleichen Volumens der Suspension über jede der 5 ml diskontinuierlichen Gradientenlösungen (Rohre 3-6), wobei eine Pipettentur auf die "Gravity"-Dosiergeschwindigkeit eingestellt ist.</li>
	<li>Legen Sie die Rohre in eine Zentrifuge, die mit einem Schwenkschaufelrotor ausgestattet ist, wobei die Beschleunigungsgeschwindigkeit auf die niedrigste Einstellung reduziert und die Bremse ausgeschaltet ist. Zentrifuge bei 76 x g für 12 min bei Raumtemperatur. Wenn die Bremse ausgeschaltet ist, wird die Zentrifugation in der Regel nach 20 min abgeschlossen.</li>
	<li>Den Überstand entfernen, wieder aufhängen und in 5 ml steriler, ausgewogener Salzlösung kombinieren und die Zellen auf Eis legen.</li>
	<li>Entfernen Sie einen Teil (10-100 l) der Zellsuspension und bestimmen Sie die Dichte lebensfähiger Zellen durch Trypan-Blauausschluss mit einem Hämozytometer.</li>
	<li>Zentrifuge bei 300 x g für 5 min. Entfernen Sie den Überstand und setzen Sie das Zellpellet mit einer Dichte von 25.000-125.000 lebensfähigen Zellen pro l wieder aus.</li>
	<li>Übertragen Sie ein ausreichendes Volumen der Zellsuspension auf ein 200-l-Mikrozentrifugenrohr (PCR-Rohr) und legen Sie es auf Eis.</li>
</ol>2. Vorbereitung der chirurgischen Stelle und Instrumente
Hinweis: Während des Eingriffs werden sterile chirurgische Handschuhe getragen. Je nach Bedarf der einzelnen Institutionen können OP-Kleider, Mützen und Gesichtsschutz erforderlich sein.
<ol><li>Bereiten Sie eine desinfizierte Tischplatte für die Nagetierchirurgie mit getrennten angrenzenden Bereichen für die Tieraufbereitung, Das Betriebsfeld und die Tierregeneration vor.</li>
	<li>Sterilisieren Sie chirurgische Instrumente in einem Dampfautoklaven. Anschließend verwenden Sie einen heißen Perlensterilisator, um Instrumente zu blitzsterilisieren, wenn Sie von einem Tier zum anderen wechseln.</li>
</ol>3. Induktion, Anästhesie und Analgesie
Hinweis: Operationen, die mehr als 15 min ab dem Zeitpunkt der Anästhesie der Mäuse sind, erfordern eine Kontaktwärmequelle. Um Unterkühlung zu vermeiden, werden die Mäuse während der prä- und postoperativen Perioden mit einer Wärmequelle (zirkulierende Warmwasserdecke) versorgt. 
<ol><li>Ketamin/Xylazin-Cocktail zur Induktionsanästhesie so zubereiten, dass jede Maus Ketamin 100 mg/kg und Xylazin 10 mg/kg erhält, indem sie 10 l des Cocktails pro Gramm Körpergewicht injiziert.</li>

	<li>Bereiten Sie Ketoprofen-Lösung für Analgesie vor, indem Sie die Stammlösung (10 mg/ml) 1:20 in sterilem, pyogenfreiem Wasser verdünnen, so dass jede Maus eine Dosis von 5 mg/kg erhält, indem sie 10 l der Lösung pro Gramm Körpergewicht injiziert.</li>
	<li>Wiegen und aufzeichnen Sie das prächirurgische Gewicht. Die einzelnen Mausgewichte variieren, reichen aber in der Regel von 18-22 g.</li>
	<li>Mit einer Insulinspritze in den intraperitonealen Raum injizieren Sie 10 l des Ketamin/Xylazin-Cocktails pro Gramm Mauskörpergewicht. Injizieren Sie z. B. 200 l für eine 20 g Maus.</li>
	<li>Bestimmen Sie, ob die Maus vollständig durch Zehenkneifen des Hinterbeins beeinflicht ist. Es dauert in der Regel 3-5 min für die Maus nicht mehr von der Prise zurückziehen.</li>
	<li>Mit einer Insulinspritze in die lockere Haut der Flanke injizieren Sie 10 l der Ketoprofenlösung pro Gramm Mauskörpergewicht subkutan. Injizieren Sie z. B. 200 l für eine 20 g Maus.</li>
	<li>Rasieren Sie Haare mit Clippers über den Schädel, schrubben Sie die freiliegende Kopfhaut mit Povidon-Jod mit einem Baumwoll-Spitzenapplikator und wischen Sie dann ein Alkoholpad ab. Wiederholen Sie diese Schritte, Povidon-Jod-Peeling und Alkohol wischen, zwei weitere Male.</li>
	<li>Tragen Sie die ophthalmologische Salbe auf die Augen der Maus auf.</li>
	<li>Machen Sie einen 1 cm Mittellinienschnitt, der sich von hinter den Ohren bis direkt vor den Augen mit einer sterilen Skalpellklinge erstreckt.</li>
	<li>Platzieren Sie die Maus unter einem sezierenden Bereich und setzen Sie den Schädel aus, um die Nahtlinien zu identifizieren. Mit kreisförmigen Bewegungen mit einem Bohrer, zentrieren Sie ein Gratloch 2,5 mm seitlich zum Bregma und 1 mm hinter der koronalen Naht.</li>
</ol>4. Intrakranielle Transplantation
Hinweis: Injektionsvolumina von mehr als 2,5 l können für Mäuse tödlich sein.
<ol><li>Positionieren Sie die Maus auf einem stereotaktischen Rahmen, indem Sie die oberen Schneidezähne über die Schneidezähne hängen und die Nasenklemme so auftragen, dass der Schädel fest in einer neutralen Position gehalten wird.</li>
	<li>Ziehen Sie 5-10 l der Zellen in einem Mikrozentrifugenrohr mehrmals nach oben und unten in eine 10-l-Hamilton-Spritze, die im Sondenhalter des stereotaktischen Manipulators eingeklemmt ist, um die Suspension zu mischen und Blasen zu beseitigen. Drücken Sie den Kolben, so dass die Spritze mit 2 l (das ausreichende Volumen, um ein Tier zu injizieren) geladen wird.</li>
	<li>Ziehen Sie den seitlichen Rand des Hautschnitts, um das Gratloch freizulegen, und führen Sie mit dem Mikromanipulator die Nadel in das Gratloch ein, so dass sich der abgeschrägte Teil direkt unter der Schädeloberfläche befindet.<ol>
<li>Bewegen Sie die Nadel 3 mm und ziehen Sie dann die Nadel 0,5 mm, um einen Platz für die Injektion zu schaffen.</li>
			<li>Bewegen Sie den Kolben langsam über 30 Sek. und lassen Sie dann die Nadel im Gehirn für weitere 2 min bleiben. Ziehen Sie die Nadel nach und nach ab, indem Sie 0,5 mm aufsteigen und auf weitere 30 Sek. warten, bevor Sie die Nadel aus dem Gehirn entfernen.</li>
		</ol>
</li>
	<li>Entfernen Sie die Maus aus dem stereotaktischen Rahmen und schließen Sie die Wunde mit einem Autoclip.</li>
	<li>Legen Sie die Maus auf ein wärmendes Pad, um die Körpertemperatur zu halten und kontinuierlich das Atmungsmuster und die Farbe von Schleimhäuten und freiliegenden Geweben (Fußsohlen) zu überwachen.</li>
	<li>Legen Sie die Maus in einen sterilen Mikroisolatorkäfig, sobald sie sich vollständig von der Anästhesie (sternal und ambulant) erholt, und bringen Sie sie in die Tierhaltungseinrichtung zurück.</li>
</ol>5. Tierpflege und -beobachtung nach der Implantation
Hinweis: Die zuverlässigste Indikation für Tumorengraftment und infiltratives Wachstum ist allmählich, anhaltende Gewichtsverlust begleitet von der Entwicklung der geknickten Haltung und rauen Haarmantel. In seltenen Fällen, neurologische Defizite festgestellt werden, die Ataxie, Parese, und Krampfanfälle gehören. Orthotopische primäre Xenografts sind hochinvasiv und entwickeln sich über einen langen Zeitraum. Mäuse manifestieren in der Regel Symptome des Tumorwachstums 3-6 Monate nach der Transplantation.
<ol><li>Beobachten Sie Mäuse mit orthotopischen Xenografts täglich für Nahrung und Wasseraufnahme, Verhalten, Pflege und Anzeichen einer Infektion an der Einschnittstelle.</li>
	<li>Ketoprofen (5 mg/kg subkutan, 1-2x pro Tag) verabreichen, wenn Schmerzen oder Leiden postoperativ beobachtet werden.</li>
	<li>Entfernen Sie Autoclips 8-10 Tage nach der Operation.</li>
	<li>Wiegen Sie Mäuse wöchentlich.</li>
	<li>Überwachen Sie Anzeichen und Symptome einer Tumorenpfropte.</li>
</ol>6. Tumorernte
Hinweis: Bei dieser Methode enthält die erste koronale Scheibe (#1) den hinteren Aspekt der Olfaktorzwiebeln und den vorderen Aspekt der Frontallappen. Der transplantierte Tumor ist am häufigsten in der rechten Hemisphäre der folgenden 3 koronalen Scheiben (Scheibe #2, #3 und #4) und der Injektionsblick ist routinemäßig in koronalen Scheiben #3 enthalten. Je nach experimentellem Bedarf kann das Material für Tumorpassagen, gefrorene Gewebeabschnitte, formalin fixierte Paraffin-eingebettete Gewebeabschnitte, Durchflusszytometrie von dissoziiertem Gewebe, Zellkultur oder Lysate für dna-, RNA- oder Proteinanalysen verwendet werden. Teile des Tumors können für zukünftige Bedürfnisse mit Zelllebensfähigkeit von 80-95% kryokonserviert werden.
<ol><li>Euthanisieren Sie Mäuse durch längere Exposition gegenüber Kohlendioxid, gefolgt von zervikalen Dislokationen.</li>
	<li>Enthaupten Sie eingeschläferte Mäuse an der Basis des Gehirns (foramen magnum-Ebene) mit einer größeren chirurgischen Schere, und ziehen Sie die Haut aus dem Schnitt nach vorne, um den Schädel vollständig freizulegen.</li>
	<li>Entfernen Sie das Gehirn.<ol>
<li>Legen Sie eine Spitze der feinen chirurgischen Schere in den seitlichen Aspekt des Foramen magnum, um Ebene der linken externen Auditfleisch, um den okzipitalen Knochen entlang der linken Seite des Schädels zu schneiden. Führen Sie den gleichen Schnitt auf der rechten Seite durch.</li>
			<li>Schneiden Sie den okzipitalen Knochen entlang einer koronalen Ebene von einem externen auditiven Fleisch auf den anderen und entfernen Sie den Knochen, um das Kleinhirn freizulegen.</li>
			<li>Schneiden Sie die Nasenknochenplatten zwischen den Augen.</li>
			<li>Schneiden Sie die sagittale Naht, indem Sie hinter der sagittalen Naht von den Nasenknochenplatten bis zum Bregma schneiden. Vervollständigen Sie den Mittellinienschnitt entlang der sagittalen Naht, indem Sie vordergründ vom Lambda zum Bregma schneiden.</li>
			<li>Legen Sie vorsichtig die Spitze eines feinen Zangenpaares entlang der sagittalen Öffnung auf jeder Seite ein, um jede durale Haftung des Schädels an das Gehirn zu reißen und dann die beiden Schädelhälften seitlich zu schälen, um das Gehirn und die olfaktorischen Glühbirnen dorsal freizulegen.</li>
			<li>Schieben Sie die Zange zwischen dem ventralen Aspekt der olfaktorischen Glühbirnen und dem Schädel, um jegliche Adhäsionen zu befreien.</li>
			<li>Neigen Sie den Schädel zurück, damit das Gehirn zurückgezogen werden kann, so dass die Optik und andere Hirnnerven freigelegt werden. Trennen Sie die Hirnnerven, um das Gehirn in eine Schüssel mit sterilem PBS zu entlassen.</li>
		</ol>
</li>
	<li>Übertragen Sie das Gehirn mit einem Wägespachtel auf einen Matrixschneider und erzeugen Sie 2 mm koronale Scheiben mit Rasierklingen.</li>
	<li>Die koronalen Scheiben in einer Schale mit sterilem PBS für die weitere Sichtprüfung und weitere Gewebeverarbeitung anrichten.</li>
</ol>7. Tumor-Kryokonservierung
<ol><li>Bereiten Sie eine Lagerlösung von Kryokonservierungsmedium vor.<ol>
<li>Fügen Sie 50 ml Proliferationsergänzung, 5 ml 100x Penicillin und Streptomycin-Lösung und 450 l 0,2% Heparin zu 450 ml Basalmedium hinzu.</li>
			<li>Lagerhaltung stämmieren Sie die Lagerlösung bei 4 °C, die vor Licht geschützt ist.</li>
		</ol>
</li>
	<li>Bereiten Sie eine funktionierende Lösung des Kryokonservierungsmediums vor.<ol>
<li>Kombinieren Sie 47,5 ml Kryokonservierungsmedium und 2,5 ml steriles DMSO in einem 50 ml Polypropylen-Konustube und mischen Sie gut.</li>
			<li>Aliquot 1,0 ml Arbeitslösung für jeden Kryovial und lagern bei 4 °C vor Licht geschützt.</li>
		</ol>
</li>
	<li>Legen Sie eine 2 mm koronale Scheibe des xenotransplantierten Gehirns in ein Kryovial, das Kryokonservierungsmedium enthält, auf Eis.</li>
	<li>Die Durchstechflasche fest verschließen und bei -80 °C in einen Gefrierbehälter geben. Übertragen Sie den Kryovial am nächsten Tag in einen flüssigen Stickstofftank für eine längere Lagerung.</li>

Bösartige Gliome sind primäre Gliatumoren des zentralen Nervensystems, die im Gehirn und gelegentlich im Rückenmark auftreten. Gliome werden von der Weltgesundheitsorganisation (WHO) nach histologischer Ähnlichkeit mit Astrozyten, Oligodendrozyten oder ependymalen Zellen klassifiziert und dann numerisch (I bis IV) für pathologische Merkmale der Malignität eingestuft. Die häufigsten histologischen Subtypen sind Astrozytome, Oligodendrogliome und gemischte Oligoastrozytome. Bösartige Gliome, die die KLASSEN II bis IV der WHO umfassen, zeichnen sich durch invasives Wachstum und Widerspenstigkeit gegenüber aktuellen Therapien aus. Jedes Jahr wird in den Vereinigten Staaten etwa 15.750 Personen mit einem bösartigen Gliom diagnostiziert und schätzungsweise 12.740 Patienten erliegen dieser Krankheit. Diese Statistiken unterstreichen die besonders tödliche Natur bösartiger Gliome und den wichtigen Bedarf an verbesserter therapeutischer Wirksamkeit.
Krebsmodelle sind für die Untersuchung der Tumorbiologie und -therapien unerlässlich. Menschliche Krebszelllinien stellen einen wichtigen ersten Schritt für In-vitro-Manipulationen und in vivo Xenografting-Studien (sekundäre Xenografts)1dar. Jedoch, Standard-Krebszellkulturen unterziehen phänotypische und genotypische Transformation2-4, die nicht in sekundären Xenografts wiederhergestellt werden kann5. Darüber hinaus können genetische Veränderungen wie Isocitrate-Dehydrogenase (IDH)Mutationen6, unterschiedliche Stammzellpopulationen7 und Abhängigkeit von wichtigen Signalwegen8 in Krebszellkulturen verloren gehen. Genomprofile können in der Krebssphärenkultur in besserem Maße gepflegt werden, spiegeln aber den Genotyp der Primärtumoren immer noch nicht vollständig wider2,3. Direkte orthotopische Transplantation negiert die Einflüsse der In-vitro-Kultur, bietet eine angemessene Mikroumgebung und bewahrt die Integrität von tumorinitiierenden Zellen9,10. Daher stellen primäre Xenografts ein leistungsfähiges und relevantes präklinisches Modell für die gründliche Prüfung gezielter Wirkstoffe dar, um bei der rationalen Gestaltung zukünftiger klinischer Studien zu helfen5,11,12.

primary orthotopic glioma xenografts recapitulate infiltrative growth and isocitrate dehydrogenase i mutation

Maligne Gliome bilden eine heterogene Gruppe hochinfiltratischer gliaaler Neoplasmen mit ausgeprägten klinischen und molekularen Merkmalen. Primäre orthotopische Xenografts rekapitulieren die histopathologischen und molekularen Merkmale bösartiger Gliom-Subtypen in präklinischen Tiermodellen. Um die WHO-Grade III und IV maligne Gliome in Transplantationstests zu modellieren, werden menschliche Tumorzellen in eine orthotopische Stelle, das Gehirn, von immungeschwächten Mäusen xenografiert. Im Gegensatz zu sekundären Xenografts, die kultivierte Tumorzellen nutzen, werden menschliche Gliomzellen von resezierten Proben getrennt und ohne vorherige Passage in der Gewebekultur transplantiert, um primäre Xenografts zu erzeugen. Das Verfahren in diesem Bericht beschreibt die Tumorprobenvorbereitung, die intrakranielle Transplantation in immungeschwächte Mäuse, die Überwachung der Tumorenpfroptmentierung und die Tumorernte für den späteren Übergang in Empfängertiere oder die Analyse. Tumorzellpräparat erfordert 2 Stunden und chirurgische Eingriffe erfordern 20 min/Tier.

Dissoziierte Gliomzellen werden direkt in das Gehirn immungeschwächter Mäuse transplantiert, um primäre orthotopische Xenograft-Linien zu erhalten. Jeder Tumorprobe wird vor der Transplantation eine randomisierte Zahl zugewiesen, als Teil des Deidentifizierungsprozesses, um geschützte Gesundheitsinformationen zu entfernen. An Orthotopic Model of Murine Bladder Cancer

In this straightforward procedure, bladder tumors are established in female C57 mice through the use of catheterization, local cauterization, and subsequent cell adhesion. After their bladders are transurethrally catheterized and drained, animals are again catheterized to permit insertion of a platinum wire into bladders without damaging the urethra or bladder. The catheters are made of Teflon to serve as an insulator for the wire, which will conduct electrical current into the bladder to create a burn injury. An electrocautery unit is used to deliver 2.5W to the exposed end of the wire, burning away extracellular layers and providing attachment sites for carcinoma cells that are delivered in suspension to the bladder through a subsequent catheterization. Cells remain in the bladder for 90 minutes, after which the catheters are removed and the bladders allowed to drain naturally. The development of tumor is monitored via ultrasound. Specific attention is paid to the catheterization technique in the accompanying video.

Orthotopic Small Bowel Transplantation in Rats

Small bowel transplantation has become an accepted clinical option for patients with short gut syndrome and failure of parenteral nutrition (irreversible intestinal failure). In specialized centers improved operative and managing strategies have led to excellent short- and intermediate term patient and graft survival while providing high quality of life 1,3. Unlike in the more common transplantation of other solid organs (i.e. heart, liver) many underlying mechanisms of graft function and immunologic alterations induced by intestinal transplantation are not entirely known6,7. Episodes of acute rejection, sepsis and chronic graft failure are the main obstacles still contributing to less favorable long term outcome and hindering a more widespread employment of the procedure despite a growing number of patients on home parenteral nutrition who would potentially benefit from such a transplant. The small intestine contains a large number of passenger leucocytes commonly referred to as part of the gut associated lymphoid system (GALT) this being part of the reason for the high immunogenity of the intestinal graft. The presence and close proximity of many commensals and pathogens in the gut explains the severity of sepsis episodes once graft mucosal integrity is compromised (for example by rejection). To advance the field of intestinal- and multiorgan transplantation more data generated from reliable and feasible animal models is needed. The model provided herein combines both reliability and feasibility once established in a standardized manner and can provide valuable insight in the underlying complex molecular, cellular and functional mechanisms that are triggered by intestinal transplantation. We have successfully used and refined the described procedure over more than 5 years in our laboratory 8-11. The JoVE video-based format is especially useful to demonstrate the complex procedure and avoid initial pitfalls for groups planning to establish an orthotopic rodent model investigating intestinal transplantation.

Small bowel transplantation has become an accepted treatment option for patients with irreversible intestinal failure. Orthotopic Liver Transplantation in Rats

Clinical progress in the field of liver transplantation has been largely supported by animal models1,2. Since the publication of the first orthotopic rat liver transplantation in 1979 by Kamada et al.3, this model has remained the gold standard despite various proposed alternative techniques4. Nevertheless, its broader use is limited by its steep learning curve5.
In this video paper, we show a simple and easy-to-establish revision of Kamada's two-cuff technique. The suprahepatic vena cava anastomosis is performed manually with a running suture, and the vena porta and infrahepatic vena cava anastomoses are performed utilizing a quick-linker cuff system6. Manufacturing the quick-linker kit is shown in a separate video paper.

Imaging Glioma Initiation In Vivo Through a Polished and Reinforced Thin-skull Cranial Window

Glioma is the one of the most lethal forms of human cancer. The most effective glioma therapy to date-surgery followed by radiation treatment-offers patients only modest benefits, as most patients do not survive more than five years following diagnosis due to glioma relapse 1,2. The discovery of cancer stem cells in human brain tumors holds promise for having an enormous impact on the development of novel therapeutic strategies for glioma 3. Cancer stem cells are defined by their ability both to self-renew and to differentiate, and are thought to be the only cells in a tumor that have the capacity to initiate new tumors 4. Glioma relapse following radiation therapy is thought to arise from resistance of glioma stem cells (GSCs) to therapy 5-10. In vivo, GSCs are shown to reside in a perivascular niche that is important for maintaining their stem cell-like characteristics 11-14. Central to the organization of the GSC niche are vascular endothelial cells 12. Existing evidence suggests that GSCs and their interaction with the vascular endothelial cells are important for tumor development, and identify GSCs and their interaction with endothelial cells as important therapeutic targets for glioma. The presence of GSCs is determined experimentally by their capability to initiate new tumors upon orthotopic transplantation 15. This is typically achieved by injecting a specific number of GBM cells isolated from human tumors into the brains of severely immuno-deficient mice, or of mouse GBM cells into the brains of congenic host mice. Assays for tumor growth are then performed following sufficient time to allow GSCs among the injected GBM cells to give rise to new tumors-typically several weeks or months. Hence, existing assays do not allow examination of the important pathological process of tumor initiation from single GSCs in vivo. Consequently, essential insights into the specific roles of GSCs and their interaction with the vascular endothelial cells in the early stages of tumor initiation are lacking. Such insights are critical for developing novel therapeutic strategies for glioma, and will have great implications for preventing glioma relapse in patients. Here we have adapted the PoRTS cranial window procedure 16and in vivo two-photon microscopy to allow visualization of tumor initiation from injected GBM cells in the brain of a live mouse. Our technique will pave the way for future efforts to elucidate the key signaling mechanisms between GSCs and vascular endothelial cells during glioma initiation.

An Orthotopic Bladder Tumor Model and the Evaluation of Intravesical saRNA Treatment

We present a novel method for treating bladder cancer with intravesically delivered small activating RNA (saRNA) in an orthotopic xenograft mouse bladder tumor model. The mouse model is established by urethral catheterization under inhaled general anesthetic. Chemical burn is then introduced to the bladder mucosa using intravesical silver nitrate solution to disrupt the bladder glycosaminoglycan layer and allows cells to attach. Following several washes with sterile water, human bladder cancer KU-7-luc2-GFP cells are instilled through the catheter into the bladder to dwell for 2 hours. Subsequent growth of bladder tumors is confirmed and monitored by in vivo bladder ultrasound and bioluminescent imaging. The tumors are then treated intravesically with saRNA formulated in lipid nanoparticles (LNPs). Tumor growth is monitored with ultrasound and bioluminescence. All steps of this procedure are demonstrated in the accompanying video.

Fetal Echocardiography and Pulsed-wave Doppler Ultrasound in a Rabbit Model of Intrauterine Growth Restriction

Fetal intrauterine growth restriction (IUGR) results in abnormal cardiac function that is apparent antenatally due to advances in fetoplacental Doppler ultrasound and fetal echocardiography. Increasingly, these imaging modalities are being employed clinically to examine cardiac function and assess wellbeing in utero, thereby guiding timing of birth decisions. Here, we used a rabbit model of IUGR that allows analysis of cardiac function in a clinically relevant way. Using isoflurane induced anesthesia, IUGR is surgically created at gestational age day 25 by performing a laparotomy, exposing the bicornuate uterus and then ligating 40-50% of uteroplacental vessels supplying each gestational sac in a single uterine horn. The other horn in the rabbit bicornuate uterus serves as internal control fetuses. Then, after recovery at gestational age day 30 (full term), the same rabbit undergoes examination of fetal cardiac function. Anesthesia is induced with ketamine and xylazine intramuscularly, then maintained by a continuous intravenous infusion of ketamine and xylazine to minimize iatrogenic effects on fetal cardiac function. A repeat laparotomy is performed to expose each gestational sac and a microultrasound examination (VisualSonics VEVO 2100) of fetal cardiac function is performed. Placental insufficiency is evident by a raised pulsatility index or an absent or reversed end diastolic flow of the umbilical artery Doppler waveform. The ductus venosus and middle cerebral artery Doppler is then examined. Fetal echocardiography is performed by recording B mode, M mode and flow velocity waveforms in lateral and apical views. Offline calculations determine standard M-mode cardiac variables, tricuspid and mitral annular plane systolic excursion, speckle tracking and strain analysis, modified myocardial performance index and vascular flow velocity waveforms of interest. This small animal model of IUGR therefore affords examination of in utero cardiac function that is consistent with current clinical practice and is therefore useful in a translational research setting.

We describe examination of fetal cardiac function with contemporary functional fetal echocardiography and fetoplacental Doppler ultrasound using the VisualSonics VEVO 2100 microultrasound in a surgically induced model of intrauterine fetal growth restriction in a rabbit.

Bioluminescent Orthotopic Model of Pancreatic Cancer Progression

Pancreatic cancer has an extremely poor five-year survival rate of 4-6%. New therapeutic options are critically needed and depend on improved understanding of pancreatic cancer biology. To better understand the interaction of cancer cells with the pancreatic microenvironment, we demonstrate an orthotopic model of pancreatic cancer that permits non-invasive monitoring of cancer progression. Luciferase-tagged pancreatic cancer cells are resuspended in Matrigel and delivered into the pancreatic tail during laparotomy. Matrigel solidifies at body temperature to prevent leakage of cancer cells during injection. Primary tumor growth and metastasis to distant organs are monitored following injection of the luciferase substrate luciferin, using in vivo imaging of bioluminescence emission from the cancer cells. In vivo imaging also may be used to track primary tumor recurrence after resection. This orthotopic model is suited to both syngeneic and xenograft models and may be used in pre-clinical trials to investigate the impact of novel anti-cancer therapeutics on the growth of the primary pancreatic tumor and metastasis.

Improved understanding of pancreatic cancer biology is critically needed to enable the development of better therapeutic options to treat pancreatic cancer. To address this need, we demonstrate an orthotopic model of pancreatic cancer that permits non-invasive monitoring of cancer progression using in vivo bioluminescence imaging.

Generation of Subcutaneous and Intrahepatic Human Hepatocellular Carcinoma Xenografts in Immunodeficient Mice

In vivo experimental models of hepatocellular carcinoma (HCC) that recapitulate the human disease provide a valuable platform for research into disease pathophysiology and for the preclinical evaluation of novel therapies. We present a variety of methods to generate subcutaneous or orthotopic human HCC xenografts in immunodeficient mice that could be utilized in a variety of research applications. With a focus on the use of primary tumor tissue from patients undergoing surgical resection as a starting point, we describe the preparation of cell suspensions or tumor fragments for xenografting. We describe specific techniques to xenograft these tissues i) subcutaneously; or ii) intrahepatically, either by direct implantation of tumor cells or fragments into the liver, or indirectly by injection of cells into the mouse spleen. We also describe the use of partial resection of the native mouse liver at the time of xenografting as a strategy to induce a state of active liver regeneration in the recipient mouse that may facilitate the intrahepatic engraftment of primary human tumor cells. The expected results of these techniques are illustrated. The protocols described have been validated using primary human HCC samples and xenografts, which typically perform less robustly than the well-established human HCC cell lines that are widely used and frequently cited in the literature. In comparison with cell lines, we discuss factors which may contribute to the relatively low chance of primary HCC engraftment in xenotransplantation models and comment on technical issues that may influence the kinetics of xenograft growth. We also suggest methods that should be applied to ensure that xenografts obtained accurately resemble parent HCC tissues.

Human tumor xenografts in immunodeficient mice are valuable tools to study cancer biology. Specific protocols to generate subcutaneous and intrahepatic xenografts from human hepatocellular carcinoma cells or tumor fragments are described. Liver regeneration induced by partial hepatectomy in recipient mice is presented as a strategy to facilitate intrahepatic engraftment.

Minimally Invasive Establishment of Murine Orthotopic Bladder Xenografts

Orthotopic bladder cancer xenografts are the gold standard to study molecular cellular manipulations and new therapeutic agents&#160;in vivo. Suitable cell lines are inoculated either by intravesical instillation (model of nonmuscle invasive growth) or intramural injection into the bladder wall (model of invasive growth). Both procedures are complex and highly time-consuming. Additionally, the superficial model has its shortcomings due to the lack of cell lines that are tumorigenic following instillation. Intramural injection, on the other hand, is marred by the invasiveness of the procedure and the associated morbidity for the host mouse.
With these shortcomings in mind, we modified previous methods to develop a minimally invasive approach for creating orthotopic bladder cancer xenografts. Using ultrasound guidance we have successfully performed percutaneous inoculation of the bladder cancer cell lines UM-UC1, UM-UC3 and UM-UC13 into 50 athymic nude. We have been able to demonstrate that this approach is time efficient, precise and safe. With this technique, initially a space is created under the bladder mucosa with PBS, and tumor cells are then injected into this space in a second step. Tumor growth is monitored at regular intervals with bioluminescence imaging and ultrasound. The average tumor volumes increased steadily in in all but one of our 50 mice over the study period.
In our institution, this novel approach, which allows bladder cancer xenograft inoculation in a minimally-invasive, rapid and highly precise way, has replaced the traditional model.

The established technique to inoculate primary invasive orthotopic bladder cancer xenografts requires laparotomy and mobilization of the bladder. This procedure inflicts significant morbidity on the mice, is technically challenging and time-consuming. We therefore developed a high-precision, percutaneous approach utilizing ultrasound guidance.

Orthotopic Injection of Breast Cancer Cells into the Mammary Fat Pad of Mice to Study Tumor Growth.

Breast cancer growth can be studied in mice using a plethora of models. Genetic manipulation of breast cancer cells may provide insights into the functions of proteins involved in oncogenic progression or help to discover new tumor suppressors. In addition, injecting cancer cells into mice with different genotypes might provide a better understanding of the importance of the stromal compartment. Many models may be useful to investigate certain aspects of disease progression but do not recapitulate the entire cancerous process. In contrast, breast cancer cells engraftment to the mammary fat pad of mice better recapitulates the location of the disease and presence of the proper stromal compartment and therefore better mimics human cancerous disease. In this article, we describe how to implant breast cancer cells into mice orthotopically and explain how to collect tissues to analyse the tumor milieu and metastasis to distant organs. Using this model, many aspects (growth, angiogenesis, and metastasis) of cancer can be investigated simply by providing a proper environment for tumor cells to grow.

Cancer is a complex disease that is influenced by the tissue surrounding the tumor as well as local pro- and anti-inflammatory mediators. Therefore, orthotropic injection models, rather than subcutaneous models may be useful to study cancer progression in a manner that better mimics human pathology.
