Oxidativer Stress ist mit vielen physiologischen und pathologischen Prozessen, wie auch Fremdstoffmetabolismus assoziiert, was zur Oxidation von Biomakromolekülen, einschließlich DNA. Daher ist eine effiziente Detektion von DNA-Oxidation wichtig für eine Vielzahl von Forschungsbereichen, einschließlich der Medizin und Toxikologie. Eine gemeinsame Biomarker oxidativ geschädigten DNA 8-oxo-7,8-dihydro-2&#39;-Desoxyguanosin (8-oxo-dGuo; oft fälschlicherweise bezeichnet als 8-Hydroxy-2&#39;-Deoxyguanosin (8-OH-dGuo oder 8 oxo-dG)). EINumfassendes Protokoll, wie einen Optimierungsprozess beschreiben, fehlt. Hier wird ein detailliertes Protokoll für die Erfassung von 8-oxo-dGuo durch HPLC-ED aus kultivierten Zellen oder tierische Gewebe beschrieben, in DNA. Sie veranschaulicht, wie die DNA-Aufreinigung kann leicht und schnell optimiert, um unerwünschte DNA-Oxidation, die während der Probenvorbereitung erfolgen kann minimiert werden. Dieses Protokoll zeigt, wie zu erkennen 8-oxo-dGuo in kultivierten menschlichen alveolaren Adenokarzinomzellen (dh, A549-Zellen) mit dem Oxidationsmittel KBrO 3 behandelt und aus der Milz von Mäusen, die polycyclische aromatische Kohlenwasserstoff Dibenzo (DEF, p) ausgesetzt Chrysen (DBC, früher bekannt als Dibenzo (a, l) pyren bekannt, DALP). Insgesamt zeigt diese Arbeit, wie ein HPLC-ED-Methodik kann leicht für die Erfassung von 8-oxo-dGuo in biologischen Proben zu optimieren.

dGuo wurde beobachtet, dass eine Retentionszeit von 4,7 min wohin 8-oxo-dGuo hatte eine Retentionszeit von etwa 6,4 min (2A und B). Es gibt etwa 1000-facher Unterschied in den Peakhöhen der beiden Analyten, wie in 2C zu sehen.
