Protocole

1. Préparer canal microfluidique avec des anti-CD4 ⁺ de surface biotinlayted.
2. Injecter 10 ml de l'échantillon sanguin à 5 ml / min dans le canal microfluidique avec pompe seringue.
3. Rincer le sang à 20 ml / min pendant 20 min avec du PBS.
4. Injecter une solution diluée d'FACS lyse (dilué à 10% de la solution stock avec PBS) à 5ul/min pendant 15 min.
5. Incuber pendant 15 min.
6. Injecter CD3 ⁺ solution d'anticorps primaire (dilué à 10% de solution stock avec du PBS) à 5 ml / min pendant 20 min.
7. Incuber pendant 2 heures.
8. Rincer le canal avec du PBS à 5 ml / min pendant 15 min.
9. Injecter 20% AF488 CD4 ^+, 0,2% d'une solution stock de DAPI, et 10% CD3 ⁺ anticorps secondaires à 5 ml / min pendant 20 min dans le même temps.
10. Incuber pendant 20 min.
11. Rincer le canal avec du PBS à 5 ml / min pendant 15 min.
12. Comptez cellules sous microscope à fluorescence:
    1. Blanc (optique) - Noir et blanc: la cellule et de la poussière
    2. UV (DAPI) - Bleu: seule cellule
    3. Bleu (CD4 ⁺⁾ - Vert: des lymphocytes T et des monocytes
