La reconstitution de les canaux KvAP en différentes membranes a été utilisé dans plusieurs études 8-10. Suite à l&#39;idée d&#39;assurer la distribution des lipides entre détergent / lipides micelles mixtes et la protéine / détergent / lipides micelles mixtes, nous sommes en mesure d&#39;atteindre reconstitution presque complète de la KvAP dans des membranes en très différents lipides. Chaque canal KvAP tétramérique a besoin d&#39;~ 100 molécules lipidiques pour couvrir pleinement son domaine ...

Cellules matériaux d&#39;échange et d&#39;information avec leur environnement par le biais des fonctions des protéines membranaires spécifiques 1. Les protéines membranaires en membranes cellulaires fonctionnent comme des pompes, des canaux, des récepteurs, des enzymes intramembranaires, linkers et les partisans de structure à travers les membranes. Les mutations qui affectent les protéines membranaires ont été liés à de nombreuses maladies humaines. En fait, de nombreuses protéines membranaires ont été les cibles de médicaments primaires parce qu&#39;ils sont importants et facilement accessible dans les membranes cellulaires. Il est donc très important de comprendre la structure et la fonction des différentes protéines membranaires en membranes, et permettre de concevoir de nouvelles méthodes pour atténuer les effets néfastes des protéines mutantes dans les maladies humaines. Les lipides entourent toutes les protéines membranaires intégrées dans des bicouches 2, 3. Dans les membranes eucaryotes, les différents types de lipides sont connus pour être organisés en microdomaines 4, 5.Beaucoup de protéines membranaires ont été présentés à répartir entre ces microdomaines ainsi que la phase fluide volumineux de membranes 3, 6. Le mécanisme sous-jacent à l&#39;organisation des micro-domaines et la fourniture de protéines membranaires en eux et l&#39;importance physiologique de ces distributions sont clairement importants mais restent mal compris. Une difficulté majeure technique à étudier les effets des lipides sur les protéines membranaires est la reconstitution fiable de biochimiquement purifié les protéines membranaires en membranes de composition lipidique bien contrôlée de sorte que presque toutes les protéines reconstituées sont de 7 fonctionnel. Au cours des dernières années, nous avons développé des méthodes pour reconstituer le canal potassique voltage-gated prototype de A. pernix (KvAP) dans différents systèmes membranaires pour les études structurales et fonctionnelles 8-10. Les données des autres et ensemble, nous ont montré que les lipides sont probablement un facteur déterminant dans les changements conformationnels de la détection de tensiondomaines d&#39;un canal ionique et voltage-dépendants peuvent former des structures de certains de ces canaux 11. reconstitution of a kv channel into lipid membranes for structural and functional studies

Pour étudier l&#39;interaction protéine-lipides de manière réductionniste, il est nécessaire d&#39;intégrer les protéines membranaires en membranes de composition lipidique bien défini. Nous étudions les effets de déclenchement de lipides à charge dans un canal potassique voltage-gated prototype (Kv), et nous avons travaillé sur des procédures détaillées pour reconstituer les canaux dans différents systèmes membranaires. Nos procédures de reconstitution prennent considération à la fois la fusion détergent induite par des vésicules et la fusion de protéine / détergent micelles avec le lipide / détergent micelles mixtes ainsi que l&#39;importance de parvenir à une distribution d&#39;équilibre des lipides chez la protéine / détergent / lipides et le détergent / lipides micelles mixtes. Nos données suggèrent que l&#39;insertion des canaux dans les vésicules lipidiques est relativement aléatoire dans les orientations et l&#39;efficacité de la reconstitution est si élevé qu&#39;aucun des agrégats de protéines détectables ont été observés dans des expériences de fractionnement.
