The murine model of DC presented in this protocol reliably reproduces the histopathological features of human DC (e.g., de novo development of lymphoid follicles in the submucosa of inflamed bowel segments, crypt shortening, and reductions in goblet cell numbers). Aside from this advantage, this model is induced by a very similar triggering factor and presents with a clinical course of moderate to mild severity as is the case in most affected humans.

In recent years, a substantial number of non-infectious colitis entities different from classical inflammatory bowel diseases (IBDs; i.e., Crohn&#39;s disease or colitis ulcerosa) have been clinically and histopathologically characterized in humans. The pathophysiological mechanisms leading to the development of these colitis forms are not completely understood partially because appropriate animal models are scarce. Diversion colitis is one of these recently described entities. Although the term was coined in 1980 by Glotzer1, the initial description of a similar phenotype was given in 1972 by Morson2. The disease develops in 50% to 91% of patients with diverting enterostomy, and its clinical intensity varies3,4. Given the annual incidence of around 120,000 colostomy patients in the United States of America, this disease entity constitutes an important health problem.
The overall goal of developing this protocol was to provide a murine DC model that relies on a colitis trigger similar to that seen in human DC and that reproduces the primary histopathological features of the human disease. In contrast to other murine colitis models, colitis induction in our model does not require genetically modified animals (e.g., IL-7 transgenic mice, N-cadherin dominant negative mice, or TGF&#946;-/- mice), the application of chemically irritating substances (e.g., dextran sulfate sodium (DSS)-induced colitis, or trinitrobenzene sulfonic acid (TNBS)-induced colitis), or the transfer of specific cell populations in immune deficient mice (as in the CD45RBhigh transfer model of colitis) (for a review, see5). In contrast to other models, the intact immune system of our DC model allows assessment of the immunological mechanism involved in DC development. The limitation of mucosal inflammation to the excluded bowel segment allows assessment of its repercussion on mucosal immunity in other parts of the gastrointestinal tract, on the immune homeostasis in other immune compartments of the intestinal tract (e.g., Peyer&#39;s patches and mesenteriale lymph nodes), and on the immune homeostasis of the entire organism. Finally, our model constitutes an appropriate tool for investigating the mechanisms controlling local inflammatory stimuli originating from changes in the normal local environment for both the local microbiome as well as alimentary antigens.

Diversion colitis (DC) is a frequent clinical condition occurring in patients with bowel segments excluded from the fecal stream as a result of a diverting enterostomy. The etiology of this disease remains ill-defined but appears to differ from that of classical inflammatory bowel diseases such as Crohn's disease and ulcerative colitis. Research aimed to decipher the pathophysiological mechanisms leading to the development of this disease has been severely hampered by the lack of an appropriate murine model. This protocol generates a murine model of DC that facilitates the study of the immune system's role and its interaction with the microbiome in the development of DC. In this model using C57BL/6 animals, distal parts of the colon are excluded from the fecal stream by creating a distal colostomy, triggering the development of mild to moderate inflammation in the excluded bowel segments and reproducing the hallmark lesions of human DC with a moderate systemic inflammatory response. In contrast to the rat model, a large number of genetically-modified murine models on the C57BL/6 background are available. The combination of these animals with our model allows the potential roles of individual cytokines, chemokines, or receptors of bioactive molecules (e.g., interleukin (IL)-17; IL-10, chemokine CXCL13, chemokine receptors CXCR5 and CCR7, and the sphingosine-1-phosphate receptor 4) to be assessed in the pathogenesis of DC. The availability of congenic mouse strains on the C57BL/6 background largely facilitates transfer experiments to establish the roles of distinct cell types involved in the etiology of DC. Finally, the model offers the opportunity to assess the influences of local interventions (e.g., modification of the local microbiome or local anti-inflammatory therapy) on mucosal immunity in affected and non-affected bowel segments and the on systemic immune homeostasis.

Surgery is well tolerated both in the experimental (colostomy) and sham (colotomy) groups. Perioperative mortality should not exceed 10% when surgery and perioperative management is correctly performed. Murine Distal Colostomy, A Novel Model of Diversion Colitis in C57BL/6 Mice

The overall goal of this protocol is to facilitate the study of the immune system's role and its interaction with the microbiome in the development of Diversion Colitis. This method can help answer key questions regarding Diversion Colitis which occurs in both segments excluded from the fecal stream and affects up to 90%of patients with Diverting Enterostomy. The main advantage of this technique is that it does not require genetically modified animals, the use of chemically irritating substances or the transfer of specific cell populations to immune deficient mice to induce colitis.Upon arrival at the animal facility, divide mice into groups of similar size. Cage each group together and keep the groups constant throughout the experiments. At least one week before surgery, switch to a high energy and high protein feed containing all the essential trace elements and vitamins.To prepare for surgery after anesthetizing the animal according to the text protocol, use tape to stabilize the animal in a supine position on a heating pad to prevent loss of body heat. Then, check that the mouse tolerates mechanical stimulation e.g. toe pinch without a motor response.Before starting surgery, disinfect the operation field three times using alcohol 70%and an iota 4. Drape the operation field to guarantee for aseptic conditions. Perform a 15 mm medium laparotomy by incising the abdominal muscles and the peritoneum along the linea alba thus minimizing blood loss.Then, using two debakey atraumatic forceps, carefully pull the cecum, terminal ileum and descending and transverse colon from the peritoneal cavity. Be careful to limit the mechanical manipulation of the intestine in order to avoid injury to mesenteric structures and to maintain the optimal profusion of the intestine. Next, identify the cecal pole, the ascending colon and the small intestine.If the anatomy of the ileocecal region is ambiguous, the presence of Peyer's patches identifies the small intestine and the presence of forms ature characterizes the colon. Use a ruler to determine the position of the future colostomy. It should be placed 20 mm distal to the ileocecal valve for a distal colostomy.Make a second 3 mm incision in the abdominal wall in the upper right quadrant. Then, pull the previously identified colon segment through this incision to form a loop, taking care not to distort the loop. Carefully pass a 22 gauge, flexible IV canula through the mesocolon, taking care not to damage the mesenteric vascular structures.Then, return the intestine to the peritoneal cavity. Using simple stitches and a resorbable suture, fix both ends of the flexible tube to the skin. Before closing the laparotomy, perform fluid resuscitation using an intra peritoneal injection of 0.5 mL of 0.9%saline.Close the peritoneum and the muscle layer with a continuous resorbable suture. Then, close the skin in a similar manner. Using a fine scissor, open the exteriorized colon loop by performing a subtotal transection.Avoid all injury to the mesentery and do not completely transect the colon. With a monofil absorbable suture, fix each colostomy opening using three single full thickness stitches to the peritoneum and skin. The afferent loop, which is a functional end colostomy and the efferent loop, which is a mucus fistula are clearly separated at this point.To carry out a colotomy, after identifying the cecal pole, ascending colon and small intestine as demonstrated for the distal colostomy, use a ruler to determine the future colotomy position which should be positioned the same distance from the ileocecal valve as the colostomy. Using fine scissors, open the colon at least two-thirds its circumference. Using a monofil absorbable suture, close the colotomy with a full thickness interrupted suture before carrying out fluid resuscitation and closing of the peritoneum and skin as before.Following the procedure, return animals to their cages and place them under an infrared lamp at 37 degrees celsius until they are fully awake before returning them to a temperature and humidity controlled environment. When animals show response to mechanical stimulation, start postoperative analgesia, by injecting 0.1 mg per kg body weight, buprenorphine SC.Carefully check for signs of respiratory depression. Supplement drinking water with 1 mg per mL of tramadol for continued analgesia during the first postoperative week.To compensate for decrease fluid intake due to reduced mobility, supply a solid drink pad in the cage during the first postoperative week. Finally, weigh the animals and score animal behavior daily during the first week, every second day during the remainder of the first month, and every third day during the second month. Surgery is well tolerated both in the experimental and the sham groups.Weight loss is more pronounced in the experimental group reaching 21.7%of initial body weight. In contrast, sham animals lose 10.8%of their initial body weight. Total mortality during the first 60 days postoperatively is around 40%in the colostomy group and about 10%in the sham group.Most deaths occurred during the first postoperative week, with the causes of death shown in this figure. Distal colostomy in mice results in the development of predominantly lymphocytic colitis, reproducing the hallmark histological features of human DC.Crypt length is significantly shortened in excluded bowel segments and reaches statistical significance after 14 days of fecal diversion. The goblet cell-bearing crypt length is reduced after 30 days in excluded bowel segments.Absolute goblet cell numbers are also significantly reduced in crypts in the excluded bowel segment after 14 postoperative days. The hallmark lesion of DC, the development of lymphoid follicles in the mucosa requires a longer duration of stool deviation. Although, an increased number of the lymphoid follicles can be observed as early as two weeks, differences become significant after two months.After watching this video, you should have a good understanding of how to perform a Murine Distal Colostomy. Once mastered, this technique can be done in less than 20 minutes if performed properly. This model can be combined with other genetically modified models or transfer experiments in congenic mouse strains, in order to address the role of inflammatory mediators of specific cell populations, and the pathogenesis of Diversion Colitis.

A Novel Surgical Approach for Intratracheal Administration of Bioactive Agents in a Fetal Mouse Model

Prenatal pulmonary delivery of cells, genes or pharmacologic agents could provide the basis for new therapeutic strategies for a variety of genetic and acquired diseases. Apart from congenital or inherited abnormalities with the requirement for long-term expression of the delivered gene, several non-inherited perinatal conditions, where short-term gene expression or pharmacological intervention is sufficient to achieve therapeutic effects, are considered as potential future indications for this kind of approach. Candidate diseases for the application of short-term prenatal therapy could be the transient neonatal deficiency of surfactant protein B causing neonatal respiratory distress syndrome1,2 or hyperoxic injuries of the neonatal lung3. Candidate diseases for permanent therapeutic correction are Cystic Fibrosis (CF)4, genetic variants of surfactant deficiencies5 and &alpha;1-antitrypsin deficiency6.
Generally, an important advantage of prenatal gene therapy is the ability to start therapeutic intervention early in development, at or even prior to clinical manifestations in the patient, thus preventing irreparable damage to the individual. In addition, fetal organs have an increased cell proliferation rate as compared to adult organs, which could allow a more efficient gene or stem cell transfer into the fetus. Furthermore, in utero gene delivery is performed when the individual's immune system is not completely mature. Therefore, transplantation of heterologous cells or supplementation of a non-functional or absent protein with a correct version should not cause immune sensitization to the cell, vector or transgene product, which has recently been proven to be the case with both cellular and genetic therapies7.
In the present study, we investigated the potential to directly target the fetal trachea in a mouse model. This procedure is in use in larger animal models such as rabbits and sheep8, and even in a clinical setting9, but has to date not been performed before in a mouse model. When studying the potential of fetal gene therapy for genetic diseases such as CF, the mouse model is very useful as a first proof-of-concept because of the wide availability of different transgenic mouse strains, the well documented embryogenesis and fetal development, less stringent ethical regulations, short gestation and the large litter size.
Different access routes have been described to target the fetal rodent lung, including intra-amniotic injection10-12, (ultrasound-guided) intrapulmonary injection13,14 and intravenous administration into the yolk sac vessels15,16 or umbilical vein17. Our novel surgical procedure enables researchers to inject the agent of choice directly into the fetal mouse trachea which allows for a more efficient delivery to the airways than existing techniques18.

We developed a novel surgical approach for intratracheal administration of bioactive agents into the mouse fetus. The delivery route is more efficient in targeting the fetal mouse lungs than the commonly used intra-amniotic injection. This procedure has to date not been described in a mouse model.

Murine Model of Wound Healing

Wound healing and repair are the most complex biological processes that occur in human life. After injury, multiple biological pathways become activated. Impaired wound healing, which occurs in diabetic patients for example, can lead to severe unfavorable outcomes such as amputation. There is, therefore, an increasing impetus to develop novel agents that promote wound repair. The testing of these has been limited to large animal models such as swine, which are often impractical. Mice represent the ideal preclinical model, as they are economical and amenable to genetic manipulation, which allows for mechanistic investigation. However, wound healing in a mouse is fundamentally different to that of humans as it primarily occurs via contraction. Our murine model overcomes this by incorporating a splint around the wound. By splinting the wound, the repair process is then dependent on epithelialization, cellular proliferation and angiogenesis, which closely mirror the biological processes of human wound healing. Whilst requiring consistency and care, this murine model does not involve complicated surgical techniques and allows for the robust testing of promising agents that may, for example, promote angiogenesis or inhibit inflammation. Furthermore, each mouse acts as its own control as two wounds are prepared, enabling the application of both the test compound and the vehicle control on the same animal. In conclusion, we demonstrate a practical, easy-to-learn, and robust model of wound healing, which is comparable to that of humans.

Flexible Colonoscopy in Mice to Evaluate the Severity of Colitis and Colorectal Tumors Using a Validated Endoscopic Scoring System

The use of modern endoscopy for research purposes has greatly facilitated our understanding of gastrointestinal pathologies. In particular, experimental endoscopy has been highly useful for studies that require repeated assessments in a single laboratory animal, such as those evaluating mechanisms of chronic inflammatory bowel disease and the progression of colorectal cancer. However, the methods used across studies are highly variable. At least three endoscopic scoring systems have been published for murine colitis and published protocols for the assessment of colorectal tumors fail to address the presence of concomitant colonic inflammation. This study develops and validates a reproducible endoscopic scoring system that integrates evaluation of both inflammation and tumors simultaneously. This novel scoring system has three major components: 1) assessment of the extent and severity of colorectal inflammation (based on perianal findings, transparency of the wall, mucosal bleeding, and focal lesions), 2) quantitative recording of tumor lesions (grid map and bar graph), and 3) numerical sorting of clinical cases by their pathological and research relevance based on decimal units with assigned categories of observed lesions and endoscopic complications (decimal identifiers). The video and manuscript presented herein were prepared, following IACUC-approved protocols, to allow investigators to score their own experimental mice using a well-validated and highly reproducible endoscopic methodology, with the system option to differentiate distal from proximal endoscopic colitis (D-PECS).

Over the past few years, new generation endoscopes have emerged as important diagnostic research aids for evaluating murine colitis and colorectal tumors. We present herein a detailed protocol for endoscopic assessment of inflammation and colorectal tumors in mice, as well as a novel scoring system that uses decimal identifiers to document the endoscopic severity of colitis and colorectal tumors.

A Murine Model of Subarachnoid Hemorrhage

In this video publication a standardized mouse model of subarachnoid hemorrhage (SAH) is presented. Bleeding is induced by endovascular Circle of Willis perforation (CWp) and proven by intracranial pressure (ICP) monitoring. Thereby a homogenous blood distribution in subarachnoid spaces surrounding the arterial circulation and cerebellar fissures is achieved. Animal physiology is maintained by intubation, mechanical ventilation, and continuous on-line monitoring of various physiological and cardiovascular parameters: body temperature, systemic blood pressure, heart rate, and hemoglobin saturation. Thereby the cerebral perfusion pressure can be tightly monitored resulting in a less variable volume of extravasated blood. This allows a better standardization of endovascular filament perforation in mice and makes the whole model highly reproducible. Thus it is readily available for pharmacological and pathophysiological studies in wild type and genetically altered mice.

A standardized mouse model of subarachnoid hemorrhage by intraluminal Circle of Willis perforation is described. Modeling Stroke in Mice: Permanent Coagulation of the Distal Middle Cerebral Artery

Stroke is the third most common cause of death and a main cause of acquired adult disability in developed countries. Only very limited therapeutical options are available for a small proportion of stroke patients in the acute phase. Current research is intensively searching for novel therapeutic strategies and is increasingly focusing on the sub-acute and chronic phase after stroke because more patients might be eligible for therapeutic interventions in a prolonged time window. These delayed mechanisms include important pathophysiological pathways such as post-stroke inflammation, angiogenesis, neuronal plasticity and regeneration. In order to analyze these mechanisms and to subsequently evaluate novel drug targets, experimental stroke models with clinical relevance, low mortality and high reproducibility are sought after. Moreover, mice are the smallest mammals in which a focal stroke lesion can be induced and for which a broad spectrum of transgenic models are available. Therefore, we describe here the mouse model of transcranial, permanent coagulation of the middle cerebral artery via electrocoagulation distal of the lenticulostriatal arteries, the so-called &#8220;coagulation model&#8221;. The resulting infarct in this model is located mainly in the cortex; the relative infarct volume in relation to brain size corresponds to the majority of human strokes. Moreover, the model fulfills the above-mentioned criteria of reproducibility and low mortality. In this video we demonstrate the surgical methods of stroke induction in the &#8220;coagulation model&#8221; and report histological and functional analysis tools.

A Simple Critical-sized Femoral Defect Model in Mice

While bone has a remarkable capacity for regeneration, serious bone trauma often results in damage that does not properly heal. In fact, one tenth of all limb bone fractures fail to heal completely due to the extent of the trauma, disease, or age of the patient. Our ability to improve bone regenerative strategies is critically dependent on the ability to mimic serious bone trauma in test animals, but the generation and stabilization of large bone lesions is technically challenging. In most cases, serious long bone trauma is mimicked experimentally by establishing a defect that will not naturally heal. This is achieved by complete removal of a bone segment that is larger than 1.5 times the diameter of the bone cross-section. The bone is then stabilized with a metal implant to maintain proper orientation of the fracture edges and allow for mobility. 
Due to their small size and the fragility of their long bones, establishment of such lesions in mice are beyond the capabilities of most research groups. As such, long bone defect models are confined to rats and larger animals. Nevertheless, mice afford significant research advantages in that they can be genetically modified and bred as immune-compromised strains that do not reject human cells and tissue.
Herein, we demonstrate a technique that facilitates the generation of a segmental defect in mouse femora using standard laboratory and veterinary equipment. With practice, fabrication of the fixation device and surgical implantation is feasible for the majority of trained veterinarians and animal research personnel. Using example data, we also provide methodologies for the quantitative analysis of bone healing for the model.

A Novel Murine Model of Arteriovenous Fistula Failure: The Surgical Procedure in Detail

The arteriovenous fistula (AVF) still suffers from a high number of failures caused by insufficient remodeling and intimal hyperplasia from which the exact pathophysiology remains unknown. In order to unravel the pathophysiology a murine model of AVF-failure was developed in which the configuration of the anastomosis resembles the preferred situation in the clinical setting. A model was described in which an AVF is created by connecting the venous end of the branch of the external jugular vein to the side of the common carotid artery using interrupted sutures. At a histological level, we observed progressive stenotic intimal lesions in the venous outflow tract that is also seen in failed human AVFs. Although this procedure can be technically challenging due to the small dimensions of the animal, we were able to achieve a surgical success rate of 97% after sufficient training. The key advantage of a murine model is the availability of transgenic animals. In view of the different proposed mechanisms that are responsible for AVF failure, disabling genes that might play a role in vascular remodeling can help us to unravel the complex pathophysiology of AVF failure.

The bm12 Inducible Model of Systemic Lupus Erythematosus (SLE) in C57BL/6 Mice

Systemic lupus erythematosus (SLE) is an autoimmune disease with diverse clinical and immunological manifestations. Several spontaneous and inducible animal models mirror common components of human disease, including the bm12 transfer model. Upon transfer of bm12 splenocytes or purified CD4 T cells, C57BL/6 mice rapidly develop large frequencies of T follicular helper cells (Tfh), germinal center (GC) B cells, and plasma cells followed by high levels of circulating anti-nuclear antibodies. Since this model utilizes mice on a pure C57BL/6 background, researchers can quickly and easily study disease progression in transgenic or knockout mouse strains in a relatively short period of time. Here we describe protocols for the induction of the model and the quantitation Tfh, GC B cells, and plasma cells by multi-color flow cytometry. Importantly, these protocols can also be used to characterize disease in most mouse models of SLE and identify Tfh, GC B cells, and plasma cells in other disease models.

A Novel Microsurgical Model for Heterotopic, En Bloc Chest Wall, Thymus, and Heart Transplantation in Mice

Exploration of novel strategies in organ transplantation to prolong allograft survival and minimizing the need for long-term maintenance immunosuppression must be pursued. Employing vascularized bone marrow transplantation and co-transplantation of the thymus have shown promise in this regard in various animal models.1-11 Vascularized bone marrow transplantation allows for the uninterrupted transfer of donor bone marrow cells within the preserved donor microenvironment, and the incorporation of thymus tissue with vascularized bone marrow transplantation has shown to increase T-cell chimerism ultimately playing a supportive role in the induction of immune regulation. The combination of solid organ and vascularized composite allotransplantation can uniquely combine these strategies in the form of a novel transplant model. Murine models serve as an excellent paradigm to explore the mechanisms of acute and chronic rejection, chimerism, and tolerance induction, thus providing the foundation to propagate superior allograft survival strategies for larger animal models and future clinical application. Herein, we developed a novel heterotopic en bloc chest wall, thymus, and heart transplant model in mice using a cervical non-suture cuff technique. The experience in syngeneic and allogeneic transplant settings is described for future broader immunological investigations via an instructional manuscript and video supplement.

Cardiopulmonary Bypass in a Mouse Model: A Novel Approach

As prolonged cardiopulmonary bypass becomes more essential during cardiac interventions, an increasing clinical demand arises for procedure optimization and for minimizing organ damage resulting from prolonged extracorporal circulation. The goal of this paper was to demonstrate a fully functional and clinically relevant model of cardiopulmonary bypass in a mouse. We report on the device design, perfusion circuit optimization, and microsurgical techniques. This model is an acute model, which is not compatible with survival due to the need for multiple blood drawings. Because of the range of tools available for mice (e.g., markers, knockouts, etc.), this model will facilitate investigation into the molecular mechanisms of organ damage and the effect of cardiopulmonary bypass in relation to other comorbidities.
