Here we have presented an automated temperature-controlled arena (Figure 1) that produces precise temperature changes in time and space. This method allows exposure of individual Drosophila not only to pre-programmed gradual increases of temperature (Figure 2 and Figure 3), but also to dynamic temperature challenges in which each tile of the fly arena was heated independently to a different temperature (<strong class="xfig"...

Temperature is a constant environmental factor that affects how organisms function and behave1. Differences in latitude and altitude lead to differences in the type of climates organism are exposed to, which results in evolutionary selection for their responses to temperature2,3. Organisms respond to different temperatures through morphological, physiological, and behavioral adaptations that maximize performance under their particular environments4. For instance, in the fruit fly Drosophila melanogaster, populations from different regions have different temperature preferences, body sizes, developmental times, longevity, fecundity, and walking performance at different temperatures2,5,6,7. The diversity observed between flies of different origins is explained in part by genetic variation and plastic gene expression8,9. Similarly, Drosophila species from different areas distribute differently among temperature gradients and show differences in resistance to extreme heat and cold tests10,11,12.
Drosophila has also recently become the model of choice to understand the genetic and neural basis of temperature perception13,14,15,16,17. Broadly, adult flies perceive temperature through cold and hot peripheral temperature sensors in the antennae and through temperature sensors in the brain13,14,15,16,17,18,19,20. The periphery receptors for hot temperatures express Gr28b.d16 or Pyrexia21, while the periphery cold receptors are characterized by Brivido14. In the brain, temperature is processed by neurons expressing TrpA115. Behavioral studies on mutants of these pathways are improving our understanding of how temperature is processed and give insights into mechanisms that vary among populations of Drosophila from different regions.
Here we describe a temperature-controlled arena that produces fast and precise temperature changes. Investigators can pre-program these changes, which allows for standardized and repeatable temperature manipulations without human intervention. Flies are recorded and tracked with specialized software to determine their position and speed at different phases of an experiment. The main measurement presented in this protocol is the walking speed at different temperatures, because it is an ecologically relevant index of physiological performance that can identify individual thermal adaptability5. Together with temperature receptor mutants, this technique can help reveal the mechanisms of thermal adaptation at cellular and biochemical levels.

Drosophila flies also possess a temperature sensing system that has become fundamental to understanding the neural basis of temperature processing in ectotherms. We present here a temperature-controlled arena that permits fast and precise temperature changes with temporal and spatial control to explore the response of individual flies to changing temperatures. Individual flies are placed in the arena and exposed to pre-programmed temperature challenges, such as uniform gradual increases in temperature to determine reaction norms or spatially distributed temperatures at the same time to determine preferences. Individuals are automatically tracked, allowing the quantification of speed or location preference. This method can be used to rapidly quantify the response over a large range of temperatures to determine temperature performance curves in Drosophila or other insects of similar size. In addition, it can be used for genetic studies to quantify temperature preferences and reactions of mutants or wild-type flies. This method can help uncover the basis of thermal speciation and adaptation, as well as the neural mechanisms behind temperature processing.

This method can help us answer key questions about flies'response to temperature changes, such as differences between genotypes, the interaction with other sensory cues, or the function of different temperature receptors. The main advantage of this technique is that it allows for multiple fast and precise temperature changes controlling time and space and in an automated fashion. Begin by placing 20 male and 20 female flies in a rearing bottle containing 45 milliliters of fly food medium, and placing the bottle inside a 25 degree celsius incubator under 12 hour light, 12 hour dark cycles.After 10 days, anesthetize the newly eclosed flies on carbon dioxide pads for a maximum of four minutes. And use a paintbrush to collect virgin flies in 2.5 by 9.5 centimeter fly rearing vials containing 6.5 milliliters of fresh fly food medium, separated by sex into groups of 20 flies per vial. Then return the vials to the incubator for five to seven days.To set up a temperature-controlled arena, turn on the arena and open the temperature phases script in the control computer. Verify that the temperature sequence is properly set and check that the duration of each experimental phase is set to 60 seconds. Under the start experimental block section, confirm these settings:the desired number of phases, iterative on/off setup of the indicative red light emitting diodes, two degrees celsius temperature increase per phase, and 16 degrees celsius as the starting temperature.Next, run the temperature phases script. The software will initialize for five seconds then stop. For temperature behavioral experiments, place a strand of white conductive tape on the top of the copper tiles of the arena, ensuring that all the edges are covered.Place a heated aluminum ring around the copper tiles and use a clean tissue to wipe the glass cover. Place the cover on the top of the aluminum ring, leaving a gap through which a fly can be blown in and tap an acclimated fly vial two times to force the flies to the bottom of the vial. Using a mouth aspirator, trap one fly and close the vial before putting it back into the incubator.Blow the fly into the arena through the gap between the glass cover and the aluminum ring and immediately push the glass clover to close the gap. Place a frame of lights around the arena to ensure symmetric illumination and start recording with the video recording program. Then press the space bar of the control computer to begin running the experimental phases.To track the videos, open the fly steps tracking software and open the configuration file inside the fly tracker folder. Set the location of the videos in video folder, and the names of the videos in video files. Specify the borders of the fly arena in arena settings based on the x/y pixel coordinates of multiple points at the edge of the arena.Specific the location of the inactive red LEDs in LED settings, based on the x/y pixel coordinates of the location of the center of the LEDs. To check the location of the borders of the fly arena, set debug to true in the arena settings, click save, and run the script in the terminal. A screen capture of the video will appear with a blue square formed by the coordinates inputted in arena settings.Then change debug in arena settings to false. Click save, and run the screen in the terminal again to begin the tracking process. In a typical temperature behavioral experiment, single flies exposed to 16, 20, or 24 degrees celsius exhibit a higher locomotion at the beginning of the experiment, than after five minutes.The temperature-controlled arena can also be used to compare fly behavioral responses from different genetic backgrounds to dynamic temperature changes. For example, in this experiment, the speed of all of the tested species increased according to their own response curves as the temperature increased, until reaching a point of maximum performance, after which their speeds decayed and the flies perished. When individual flies were exposed to 40 degrees celsius in the middle and one side tile, with the other side tile at a comfortable 22 degrees celsius the the Wild-type flies quickly stopped moving along the arena and remain in the comfortable location.In contrast, the classic memory mutant Dunce flies keep exploring the arena and spend less time than controls in the comfortable location. Further, testing combinations of temperature and location are also useful in understanding the function of different temperature receptors during dynamic temperature changes. As illustrated in this experiment in which individual d melanogaster mutants were exposed to increasing temperatures while a shifting comfortable location at 22 degrees celsius was also provided.While performing these procedure, remember to perform the steps in order and quickly so you can capture as much of flies behavior as possible. Next to this procedure other methods like negative geotaxis or heat choke assays can be used to answer additional questions such as where do flies move normally or have a normal heat resistance. The implications of this technique extend to us understanding neurological disorders or mutations in which temperature perception or pain play a role.Generally, individuals new to this method will struggle because each step is simple by itself but they're required to be perfectly coordinated so the technique needs to be practiced.

The Crossmodal Congruency Task as a Means to Obtain an Objective Behavioral Measure in the Rubber Hand Illusion Paradigm

The rubber hand illusion (RHI) is a popular experimental paradigm. Participants view touch on an artificial rubber hand while the participants' own hidden hand is touched. If the viewed and felt touches are given at the same time then this is sufficient to induce the compelling experience that the rubber hand is one's own hand. The RHI can be used to investigate exactly how the brain constructs distinct body representations for one's own body. Such representations are crucial for successful interactions with the external world. To obtain a subjective measure of the RHI, researchers typically ask participants to rate statements such as "I felt as if the rubber hand were my hand". Here we demonstrate how the crossmodal congruency task can be used to obtain an objective behavioral measure within this paradigm.
The variant of the crossmodal congruency task we employ involves the presentation of tactile targets and visual distractors. Targets and distractors are spatially congruent (i.e. same finger) on some trials and incongruent (i.e. different finger) on others. The difference in performance between incongruent and congruent trials - the crossmodal congruency effect (CCE) - indexes multisensory interactions. Importantly, the CCE is modulated both by viewing a hand as well as the synchrony of viewed and felt touch which are both crucial factors for the RHI.
The use of the crossmodal congruency task within the RHI paradigm has several advantages. It is a simple behavioral measure which can be repeated many times and which can be obtained during the illusion while participants view the artificial hand. Furthermore, this measure is not susceptible to observer and experimenter biases. The combination of the RHI paradigm with the crossmodal congruency task allows in particular for the investigation of multisensory processes which are critical for modulations of body representations as in the RHI.

We demonstrate how an objective measure can be employed in the widely employed rubber hand illusion paradigm. This measure is obtained by modifying the well-established crossmodal congruency task. Uncovering Beat Deafness: Detecting Rhythm Disorders with Synchronized Finger Tapping and Perceptual Timing Tasks

A set of behavioral tasks for assessing perceptual and sensorimotor timing abilities in the general population (i.e., non-musicians) is presented here with the goal of uncovering rhythm disorders, such as beat deafness. Beat deafness is characterized by poor performance in perceiving durations in auditory rhythmic patterns or poor synchronization of movement with auditory rhythms (e.g., with musical beats). These tasks include the synchronization of finger tapping to the beat of simple and complex auditory stimuli and the detection of rhythmic irregularities (anisochrony detection task) embedded in the same stimuli. These tests, which are easy to administer, include an assessment of both perceptual and sensorimotor timing abilities under different conditions (e.g., beat rates and types of auditory material) and are based on the same auditory stimuli, ranging from a simple metronome to a complex musical excerpt. The analysis of synchronized tapping data is performed with circular statistics, which provide reliable measures of synchronization accuracy (e.g., the difference between the timing of the taps and the timing of the pacing stimuli) and consistency. Circular statistics on tapping data are particularly well-suited for detecting individual differences in the general population. Synchronized tapping and anisochrony detection are sensitive measures for identifying profiles of rhythm disorders and have been used with success to uncover cases of poor synchronization with spared perceptual timing. This systematic assessment of perceptual and sensorimotor timing can be extended to populations of patients with brain damage, neurodegenerative diseases (e.g., Parkinson&#8217;s disease), and developmental disorders (e.g., Attention Deficit Hyperactivity Disorder).

Behavioral tasks that allow for the assessment of perceptual and sensorimotor timing abilities in the general population (i.e., non-musicians) are presented. The Rodent Psychomotor Vigilance Test (rPVT): A Method for Assessing Neurobehavioral Performance in Rats and Mice

The human Psychomotor Vigilance Test (PVT) is a widely used procedure for measuring changes in fatigue and sustained attention. The present article describes a rodent version of the PVT&#8212;termed the &#34;rPVT&#34;&#8212;that measures similar aspects of attention (i.e., performance accuracy, motor speed, premature responding, and lapses in attention). Data are presented that demonstrate both the short- and long-term usefulness of the rPVT when employed with laboratory rats. Rats easily learn the rPVT, and learning to perform the basic procedure takes less than two weeks of training. Once acquired, rat performances in the rPVT show a high degree of similarity to these same performance measures in the human PVT, including similarities in, lapses in attention, reaction times, vigilance decrements across session time (i.e., the human &#34;time-on-task&#34; effects), and the response-stimulus interval (RSI) effect described for humans. Thus the rPVT can be an extremely valuable tool for assessing the effects of a wide range of variables on sustained attention quite similar to human PVT performances, and thus can be useful for developing novel treatments for neurobehavioral dysfunctions.

Behavioral Disturbances: An Innovative Approach to Monitor the Modulatory Effects of a Nutraceutical Diet

In dogs, diets are often used to modulate behavioral disturbances related to chronic anxiety and stress caused by intense and restless activity. However, the traditional ways to monitor behavioral changes in dogs are complicated and not efficient. In the current clinical evaluation, a new, simple monitoring system was used to assess the effectiveness of a specific diet in positively modulating the intense and restless activity of 24 dogs of different ages and breeds. This protocol describes how to easily and rapidly evaluate improvement in a set of symptoms related to generalized anxiety by using a specific sensor, a mobile phone app, a wireless router, and a computer. The results showed that dogs treated with specific diets showed significant improvement in the times spent active and at rest after 10 days (p &lt; 0.01 and p &lt; 0.05, respectively). These dogs also showed an overall significant improvement in clinical and behavioral symptoms. A specific sensor, along with its related hardware, was demonstrated to successfully monitor behavioral changes relating to movement in dogs.

A Method to Test the Effect of Environmental Cues on Mating Behavior in Drosophila melanogaster

An individual's sexual drive is influenced by genotype, experience and environmental conditions. How these factors interact to modulate sexual behaviors remains poorly understood. In Drosophila melanogaster, environmental cues, such as food availability, affect mating activity offering a tractable system to investigate the mechanisms modulating sexual behavior. In D. melanogaster, environmental cues are often sensed via the chemosensory gustatory and olfactory systems. Here, we present a method to test the effect of environmental chemical cues on mating behavior. The assay consists of a small mating arena containing food medium and a mating couple. The mating frequency for each couple is continuously monitored for 24 h. Here we present the applicability of this assay to test environmental compounds from an external source through a pressurized air system as well as manipulation of the environmental components directly in the mating arena. The use of a pressurized air system is especially useful to test the effect of very volatile compounds, while manipulating components directly in the mating arena can be of value to ascertain a compound's presence. This assay can be adapted to answer questions about the influence of genetic and environmental cues on mating behavior and fecundity as well as other male and female reproductive behaviors.

An Automated Rapid Iterative Negative Geotaxis Assay for Analyzing Adult Climbing Behavior in a Drosophila Model of Neurodegeneration

Neurodegenerative diseases are frequently associated with a progressive loss of movement ability, reduced life span, and age-dependent neurodegeneration. To understand the mechanism of these cellular events, and their causal relationships with each other, Drosophila melanogaster, with its sophisticated genetic tools and diverse behavioral features, are used as disease models for assessing neurodegenerative phenotypes. Here we describe a high-throughput method to analyze Drosophila adult negative geotaxis behavior, as an indication for possible motor defects associated with neurodegeneration. An automated machine is designed and developed to drive fly synchronization using an initial electric impulse, later allowing the recording of negative geotaxis behavior over a course of secs to mins. Images from the digitally recorded video are then processed with the self-designed RflyDetection software for statistical data manipulation. Different from the manually controlled negative geotaxis assay based on single fly, this precise, fast, and high-throughput protocol allows data acquisition from more than hundreds of flies simultaneously, providing an efficient approach to advance our understanding in the underlying mechanism of locomotor deficits associated with neurodegeneration.

Using the FishSim Animation Toolchain to Investigate Fish Behavior: A Case Study on Mate-Choice Copying In Sailfin Mollies

Over the last decade, employing computer animations for animal behavior research has increased due to its ability to non-invasively manipulate the appearance and behavior of visual stimuli, compared to manipulating live animals. Here, we present the FishSim Animation Toolchain, a software framework developed to provide researchers with an easy-to-use method for implementing 3D computer animations in behavioral experiments with fish. The toolchain offers templates to create virtual 3D stimuli of five different fish species. Stimuli are customizable in both appearance and size, based on photographs taken of live fish. Multiple stimuli can be animated by recording swimming paths in a virtual environment using a video game controller. To increase standardization of the simulated behavior, the prerecorded swimming path may be replayed with different stimuli. Multiple animations can later be organized into playlists and presented on monitors during experiments with live fish.
In a case study with sailfin mollies (Poecilia latipinna), we provide a protocol on how to conduct a mate-choice copying experiment with FishSim. We utilized this method to create and animate virtual males and virtual model females, and then presented these to live focal females in a binary choice experiment. Our results demonstrate that computer animation may be used to simulate virtual fish in a mate-choice copying experiment to investigate the role of female gravid spots as an indication of quality for a model female in mate-choice copying.
Applying this method is not limited to mate-choice copying experiments but can be used in various experimental designs. Still, its usability depends on the visual capabilities of the study species and first needs validation. Overall, computer animations offer a high degree of control and standardization in experiments and bear the potential to &#39;reduce&#39; and &#39;replace&#39; live stimulus animals as well as to &#39;refine&#39; experimental procedures.

An Improved Assay and Tools for Measuring Mechanical Nociception in Drosophila Larvae

Published assays for mechanical nociception in Drosophila have led to variable assessments of behavior. Here, we fabricated, for use with Drosophila larvae, customized metal nickel-titanium alloy (nitinol) filaments. These mechanical probes are similar to the von Frey filaments used in vertebrates to measure mechanical nociception. Here, we demonstrate how to make and calibrate these mechanical probes and how to generate a full behavioral dose-response from subthreshold (innocuous or non-noxious range) to suprathreshold (low to high noxious range) stimuli. To demonstrate the utility of the probes, we investigated tissue damage-induced hypersensitivity in Drosophila larvae. Mechanical allodynia (hypersensitivity to a normally innocuous mechanical stimulus) and hyperalgesia (exaggerated responsiveness to a noxious mechanical stimulus) have not yet been established in Drosophila larvae. Using mechanical probes that are normally innocuous or probes that typically elicit an aversive behavior, we found that Drosophila larvae develop mechanical hypersensitization (both allodynia and hyperalgesia) after tissue damage. Thus, the mechanical probes and assay that we illustrate here will likely be important tools to dissect the fundamental molecular/genetic mechanisms of mechanical hypersensitivity.

A Simple Technique to Assay Locomotor Activity in Drosophila

Drosophila melanogaster is an ideal model organism for studying various diseases due to its abundance of advanced genetic manipulation techniques and diverse behavioral features. Identifying behavioral deficiency in animal models is a crucial measure of disease severity, for example, in neurodegenerative diseases where patients often experience impairments in motor function. However, with the availability of various systems to track and assess motor deficits in fly models, such as drug-treated or transgenic individuals, an economical and user-friendly system for precise evaluation from multiple angles is still lacking. A method based on the AnimalTracker application programming interface (API) is developed here, which is compatible with the Fiji image processing program, to systematically evaluate the movement activities of both adult and larval individuals from recorded video, thus allowing for the analysis of their tracking behavior. This method requires only a high-definition camera and a computer peripheral hardware integration to record and analyze behavior, making it an affordable and effective approach for screening fly models with transgenic or environmental behavioral deficiencies. Examples of behavioral tests using pharmacologically treated flies are given to show how the techniques can detect behavioral changes in both adult flies and larvae in a highly repeatable manner.

An Automated Assessment of Visual Acuity in Infants and Toddlers

An Automated Method for Assessing Visual Acuity in Infants and Toddlers Using an Eye-Tracking System

Visual acuity measurement is an important visual function test to perform in infancy and childhood. However, accurate visual acuity measurement in infants is difficult because of deficiencies in their communication ability. This paper presents a novel automated method to assess visual acuity in children (5-36 months old). This method, the automated acuity card procedure (AACP), uses a webcam for eye tracking and recognizes children's watching behaviors automatically. A two-choice preferential-looking test is performed when the tested child watches the visual stimuli shown on a high-resolution digital display screen. When the tested child watches the stimuli, their facial pictures are recorded by the webcam. These pictures are used by the set computer program to analyze their watching behavior. With this procedure, the child's eye movement responses to different stimuli are measured, and their visual acuity is assessed without communication. By comparing the results with grating acuity obtained by Teller Acuity Cards (TACs), AACP performance is deemed comparable to that of TACs.
