application of stopped-flow kinetics methods to investigate the mechanism of action of a dna repair protein

Análise cinética transitória é indispensável para a compreensão do funcionamento de macromoléculas biológicas, uma vez que esta abordagem produz informações mecanicista incluindo concentrações sítio ativo e constantes de velocidade intrínseca que governam a função de macromoléculas. Em caso de enzimas, por exemplo, medições estado transiente ou pré-estável identificar e caracterizar os eventos individuais no caminho da reação, enquanto que as medições estado estacionário só o rendimento global de eficiência catalítica e especificidade. Não surpreendentemente, com defeito função MSH2-MSH6 humano é associado com câncer de cólon hereditário não-polipose e outros cânceres esporádicos 6-8.
