Hier berichten wir über Anwendung der Stopped-Flow-Kinetik der Wirkmechanismus von MSH2-MSH6, einer eukaryotischen DNA-Reparatur-Protein, das Basenpaar-Fehlpaarungen und Einfügen / Löschen Schleifen in DNA und Signale Mismatch-Reparatur (MMR) 3-5 erkennt Sonde. T mismatch und Überwachung der daraus resultierende Anstieg in 2-Amino-Fluoreszenz in Echtzeit: DNA-Bindung wird durch rasches Mischen MSH2-MSH6 mit DNA mit einem 2-Amino (2-Ap) Fluorophor benachbart zu einem G gemessen. T (2-Ap) Mismatch-Komplex mit unmarkierten Falle DNA und Überwachung Abnahme der Fluoreszenz im Laufe der Zeit 9: DNA Dissoziation wird durch Mischen von vorgeformten MSH2-MSH6 G gemessen. Pre-Steady-State-ATPase Kinetik werden durch die Veränderung der Fluoreszenz von 7-Diethylamino-3-((((2-maleimidyl) ethyl) amino) carbonyl) Cumarin)-markiertem Phosphat Binding Protein (MDCC-PBP) auf Bindung von Phosphat (gemessen Pi) von MSH2-MSH6 folgenden ATP-Hydrolyse 9,10 freigegeben.
